Synthesis of manganese porphyrins/electrochemical reduction of graphene oxide nanocomposite to simultaneously detect dopamine and uric acid under the interference of ascorbic acid

Jiazheng Zhang,Yuhan Zhang,Tongtong Cao,Yicheng Zhou,Li Dong,Lin Liu,Zhiwei Tong

This study reported the synthesis of ERGO-MnTMPyP and its electrochemical detection performance for dopamine (DA) and uric acid (UA). The nanocomposite was based on the electrostatic interaction between graphene oxide (GO) nanosheets and manganese porphyrins (MnTMPyP). The material was characterized using various techniques including X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), and Fourier transform infrared spectrometer (FT-IR), demonstrating successful synthesis. Various electrochemical tests, including cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), and differential pulse voltammetry (DPV), were performed to analyze the electrochemical characteristics of modified glass carbon electrodes (GCE). The modified electrode exhibited a significant reduction in electron transfer resistance, decreasing to 100.9 Ω. Compared with previous studies, the modified electrode reveals a unique performance to effectively detect DA (208 mV) and UA (328 mV) simultaneously with the presence of ascorbic acid (AA) interference. The detection peaks of DA and UA were apparently separated with the detection limits of 0.29 μM in the linear range of 2.23–10.76 μM and 0.64 μM in the linear range of 2.23–10.76 μM (S/N = 3), respectively.

Manganese porphyrin immobilized on montmorillonite: a highly efficient and reusable catalyst for the aerobic epoxidation of olefins under ambient conditions

Xiantai Zhou,Hongbing Ji

Highly efficient aerobic epoxidation of olefins catalyzed by cationic meso -tetrakis-(1-methyl-4-pyridyl) (TM4PyP) metalloporphyrins immobilized into montmorillonite (MT) interlayer was achieved. The heterogeneous catalysts were characterized by diffuse reflectance UV spectra, infrared, X-ray diffraction, nitrogen adsorption isotherm and scanning electron microscopy. Manganese porphyrin (MnTM4PyP-MT) showed excellent activity and selectivity for the aerobic epoxidation of olefins under ambient conditions, in which more 90% yields of epoxides were obtained. The clay basal spacing played significant role in the catalytic efficiency and selectivity for different olefins. The catalyst could be reused consecutively five times without significant loss of activity.

Construction of porphyrinic manganese-organic frameworks based on structural regulation for electrochemical determination of nitrobenzene in water and vegetable samples

Wang Li,Zhang Mengjie,Li Yuanyuan,Chen Xiumei,Qin Hao,Yang Jin,Fan Suhua,Wu Hai

Nitrobenzene (NB) is one of the major organic pollutants that has seriously endangered human health and the environment even in trace amounts. Therefore, it is of great significance to detect trace NB efficiently and sensitively. Herein, a porphyrinic metal-organic framework (MOF) of Mn-PCN-222 (PCN, porous coordination network) was first synthesized by the coordination between Zr6 cluster and tetrakis (4-carboxyphenyl)-porphyrin-Mn (Ⅲ) (MnTCPPCl) ligand. To regulate its structure and the electrochemical properties, a phenyl group was inserted in each branched chain of TCPP to form the TCBPP organic ligand. Then, we used Zr6 clusters and manganese metalloporphyrin (MnTCBPPCl) to synthesize a new porphyrin-based MOF (Mn-CPM-99, CPM, crystalline porous material). Due to the extended chains of TCPP, the rod-shaped structure of Mn-PCN-222 was switched to concave quadrangular bipyramid of Mn-CPM-99. Mn-CPM-99 exhibited higher porosity, larger specific surface area, better electrochemical performances than those of Mn-PCN-222. By using modular assembly technique, Mn-CPM-99 film was sequentially assembled on the surface of indium-tin-oxide (ITO) to prepare an electrochemical sensor (Mn-CPM-99/ITO). The proposed sensor showed excellent electrochemical reduction of NB and displayed three linear response ranges in the wide concentration ranges. The obtained low limit of detection (LOD, 1.3 nM), high sensitivity and selectivity, and good reproducibility of the sensor for NB detection fully illustrate that Mn-CPM-99 is an excellent candidate for electrochemical sensor interface material. Moreover, the sensor was successfully applied to the detection of NB in lake water and vegetable samples showing satisfactory recovery of 98.9%–101.8%.

Fu Yang,Shuying Gao,Cuirong Xiong,Haiqing Wang,Jin Chen,Yan Kong

The different amounts of [5,10,15,20-tetrakis-(pentafluorophenyl)porphyrin] manganese chloride (TF 20 PPMnCl) were immobilized on amino-functionalized MCM-41 for catalysis of the hydroxylation of naphthalene . The samples were characterized by X-ray powder diffraction, N 2 adsorption/desorption isotherms, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, scanning electron microscopy, diffuse reflectance ultraviolet-visible spectroscopy, thermogravimetric and differential scanning calorimetry , and inductively coupled plasma mass spectrometry . The results indicated that the manganese porphyrins were axially coordinated on amino-functionalized MCM-41. The prepared samples showed remarkable catalytic activity in the hydroxylation of naphthalene with meta -chloroperbenzoic acid as the oxidant. The catalyst could be reused several times without loss of its activity.

A Portable Electrochemical Sensor Based on Manganese Porphyrin-Functionalized Carbon Cloth for Highly Sensitive Detection of Nitroaromatics and Gaseous Phenol

Andrews Boakye,Kun Yu,Benjamin K. Asinyo,Huining Chai,Tahir Raza,Tailin Xu,Guangyao Zhang,Lijun Qu

Organic pollutants (OPs) have garnered a considerable amount of attention in recent times due to their extreme toxicity toward humans and the ecosystem. The need for an inexpensive yet robust, sensitive, selective, and easy-to-operate method for detecting OPs remains a challenge. Herein, a portable electrochemical sensor is proposed based on manganese porphyrin-functionalized carbon cloth (CC). To explain the electrochemical performance of the sensor, cyclic voltammetry (CV) and differential pulse voltammetry (DPV) were employed. The presence of manganese(III) ion in the center of the porphyrin ligand acted as an agent for the transfer of electrons and enhanced sensitivity toward analyte-specific redox catalysis. Moreover, it allowed for the concurrent detection of multiple analytes in a complex environment. The modified CC electrode can selectively detect nitroaromatic and phenolic compounds with accessible data collected through wireless means onto a smartphone device. The as-synthesized electrode demonstrated a higher sensitivity toward the detection of nitrobenzene (NB) and aqueous phenol with a limit of detection (LOD) found to be 5.9268 × 10–10 M and 4.0178 × 10–10 M, respectively. Additionally, our proposed portable electrochemical sensor demonstrates a high selectivity and reproducibility toward nitroaromatic and phenolic compounds, which can be employed in real complex water samples. With regard to the sensor’s remarkable electrochemical performance, it is envisaged that such a sensor could pave the way for environmental point of care (POC) testing.

Electrocatalytic reduction of trace nitrobenzene using a graphene-oxide@polymerized-manganese-porphyrin composite

Huanhuan Li,Can Huang,Yingying Li,Weijun Yang,Fan Liu

A core–shell structure electrocatalyst for trace nitrobenzene reduction was prepared with Mn(II)[5,10,15,20-tetra(4-aminophenyl)porphyrin] (MnTAPP) and graphene oxide (GO) as raw materials. Firstly, MnTAPP and GO were combined together by covalent bonds, and then the supported MnTAPP was coupled together through p-dibromobenzene, a conjugated bridging agent, to obtain a more stable graphene-oxide@polymerized-manganese-porphyrin composite (GMPP@AMP). The structure and morphology of the GMPP@AMP were characterized by FT-IR, Raman spectroscopy, SEM and TEM. The GMPP@AMP modified glassy carbon electrode (GMPP@AMP/GCE) was prepared and the electrochemical activity of GMPP@AMP towards nitrobenzene reduction was evaluated by cyclic voltammetry (CV). The results showed that GMPP@AMP/GCE had a more positive reduction potential than MnTAPP/GCE and GO/GCE, and the reduction current responded more sensitively. Electrocatalytic reduction currents of nitrobenzene were found to be linearly related to concentration over the range 0.04 to 0.24 mM using a differential pulse voltammogram (DPV) method. Nitrobenzene is easily compatible with polymerized MnTAPP which has rich nitrogen-containing functional groups and porous structure, and the highly conductive GO combined with the polymerized MnTAPP having excellent electron transfer ability. This produced a significant synergistic catalytic effect during the electrocatalytic reaction of trace nitrobenzene. The novel composite has good application prospects in electrochemical detection of trace nitrobenzene compounds in the environment.

Electrochemical Detection of Superoxide Anion Released by Living Cells by Manganese(III) Tetraphenyl Porphine as Superoxide Dismutase Mimic

Cui Min,Ren Jujie,Wen Xiaofang,Li Na,Xing Yifei,Zhang Cong,Han Yuanyuan,Ji Xueping

Superoxide anion, one of the most active reactive oxygen species, is associated with the development of many diseases. So monitoring superoxide anion in living cells is of great significance for the pathological research of many diseases. In this work, a new non-enzymatic sensor for the detection of superoxide anion(O •− 2 ) was developed, which was fabricated by the nanocomposites composed of manganese(III) tetraphenyl porphine(MnTPP) as superoxide dismutase mimic and electrochemical reduced graphene oxide(ERGO) as electrode support material to modify the glassy carbon electrode(GCE). The electrochemical behavior of the fabricated electrode(MnTPP/ERGO/GCE) was performed by electrochemical impedance spectroscopy(EIS) and cyclic voltammetry(CV), which revealed that MnTPP/ERGO/GCE possessed good catalytic ability to the electrochemical reduction of O •− 2 . The MnTPP/ERGO/GCE showed excellent electroanalysis performance towards O •− 2 using the technique of differential pulse voltammetry(DPV) with a linear relationship in the range of 0.2–110.0 µmol/L, a sensitivity of 445 µA·L·mmol −1 ·cm −2 and a detection limit of 0.039 µmol/L(S/N=3). The real-time monitoring of O •− 2 from MCF-7 breast cancer cells stimulated by zymosan was realized in this work, which indicates that the MnTPP/ERGO/GCE hold potential application for electrochemical quantification of superoxide anions in biological applications.

Hyperbranched Supramolecular Polymer of Tris(permethyl-β-cyclodextrin)s with Porphyrins: Characterization and Magnetic Resonance Imaging

Mo Sun,Hengyi Zhang,Xinyue Hu,Bowen Liu,Yu Liu

A hyperbranched supramolecular polymer (HSP) is constructed based on the intermolecular inclusion complexation of bridged tris(permethyl- β -cyclodextrin) with Mn III -porphyrin bearing poly(ethylene glycol) (PEG) side chains (Mn III -TPP), and characterized by UV/vis absorption spectroscopy, isothermal titration calorimetry (ITC), NMR, dynamic light scattering (DLS), atomic force microscopy (AFM) and transmission electron microscopy (TEM), respectively. The presence of permethyl- β -cyclodextrin can stabilize the low-valent Mn II -TPP. In vitro cell experiment indicates that this HSP has no remarkable cellular toxicity against NIH 3T3 cells. In vitro MR imaging experiment indicates a slightly increased T 1 relaxivity for this HSP compared to that for the linear supramolecular polymer.

Selective regulation of oxygen transfer over Mn porphyrin-based organic polymers enables the mild aerobic epoxidation of isobutylene

Xiaodao Liang,Yaorong He,Chao Xiong,Hongbing Ji,Yan Yang

Background Oxygen activation is a critical step in oxidation reactions for energy conversion and fine chemical synthesis, with its efficiency directly determining catalytic performance and industrial applicability. The main challenges are the high activation energy barriers and low target product selectivity caused by the inherent inertness of molecular oxygen. To address these issues, this study draws on the advantages of biomimetic catalysis using metal porphyrins and synthesizes a series of porphyrin-based organic polymers (Metal-Por-POPs) to achieve efficient aerobic epoxidation of olefins. Method A series of metalloporphyrin-based organic polymers were synthesized and thoroughly characterized using techniques such as SEM, XRD, and XPS. The catalytic performance was systematically evaluated under various conditions, including catalyst types, oxidant amount, reaction time, and temperature. Significant finding Using Mn-Por-POP as a catalyst, isobutylene was converted to the corresponding epoxide within 1 h at room temperature with a selectivity of up to 96.4 %. The catalyst also demonstrated excellent recyclability and broad substrate adaptability, maintaining over 96 % epoxide selectivity after five consecutive reaction cycles. Mechanistic studies via EPR and HRMS revealed that Mn-Por-POP promoted the generation of acyl radicals from isobutyraldehyde under aerobic conditions. These radicals subsequently reacted with molecular oxygen to form acylperoxy radicals, which facilitate the highly selective epoxidation of alkenes.

Manganese corrole catalyzed selective oxidation of styrene to benzaldehyde: sodium nitrite functions as an oxidant and cocatalyst

Bei Wan,Fan Cheng,Hua-Hua Wang,Atif Ali,Yan-Mei Sun,Hai-Yang Liu,Chi-Kwong Chang

Catalytic oxidation using manganese corrole is a hot topic of contemporary porphyrin chemistry, in which PhIO, TBHP, PhI(OAc)2, KHSO5 and m-CPBA are usually used as oxidants. This article reports the first selective oxidation of styrene to benzaldehyde using a manganese(III) corrole catalyst and sodium nitrite (NaNO2) as oxidant and cocatalyst at room temperature. The yield was 158.1% in air and 96.5% under a nitrogen atmosphere, showing oxygen might be involved in the reaction and that NaNO2 is an oxygen source and cocatalyst in the system. The peripheral electron-withdrawing substituents of the manganese corrole were favorable to the catalytic reaction. Radical inhibition and H218O experiments proved that the catalytic reaction was a free radical and hydrolysis-involved reaction.

Novel Photo-STING Agonists Delivered by Erythrocyte Efferocytosis-Mimicking Pattern to Repolarize Tumor-Associated Macrophages for Boosting Anticancer Immunotherapy

Zhiyan Li,Xianghui Li,Yanjun Lu,Xudong Zhu,Wenxuan Zheng,Kai Chen,Xingzhou Wang,Tao Wang,Wenxian Guan,Zhi Su,Song Liu,Jinhui Wu

Immunotherapy has emerged as a highly effective therapeutic strategy for cancer treatment. Cyclic guanosine monophosphate-adenosine monophosphate synthase (cGAS)-stimulator of interferon gene (STING) pathway activation facilitates tumor-associated macrophage (TAM) polarization toward M1 phenotype, and Mn 2+ are effective agents for this pathway activation. However, the high in vivo degradation rate and toxicity of Mn 2+ hamper clinical application of immunotherapy. Here, this work has newly synthesized and screened manganese porphyrins for Mn 2+ transport, referred to as photo-STING agonists (PSAs), and further encapsulate them into core-shell nanoparticles named Rm@PP-GA with dual specificity for tumor tissue and TAMs. Not only do PSAs achieve higher Mn 2+ delivery efficiency compared to Mn 2+ , but they also generate reactive oxygen species under light exposure, promoting mitochondrial DNA release for cGAS-STING pathway activation. In Rm@PP-GA, globin and red blood cell membranes (Rm) are used for erythrocyte efferocytosis-mimicking delivery. Rm can effectively prolong the in vivo circulation period while globin enables PSAs to be taken up by TAMs via CD163 receptors. After Rm rupture mediated by perfluorohexane in nanoparticles under ultrasonication, drugs are specifically released for TAM repolarization. Further, dendritic cells mature, as well as T lymphocyte infiltrate, both of which favor tumor eradication. Therefore, cancer immunotherapy is optimized by novel PSAs delivered by erythrocyte efferocytosis-mimicking delivery pattern.

Sono-promoted drug penetration and extracellular matrix modulation potentiate sonodynamic therapy of pancreatic ductal adenocarcinoma

Hong Xiao,Xiaoxia Li,Bo Li,Yin Zhong,Jingya Qin,Yong Wang,Shisong Han,Jie Ren,Xintao Shuai

Pancreatic ductal adenocarcinoma (PDAC) exhibits difficult penetration of most drugs, leading to a very poor therapeutic outcome with a quite low five-year survival rate. The foremost reason is the highly-dense extracellular matrix (ECM) with abundant collagen and fibronectin secreted by the activated pancreatic stellate cells (PSCs). Here, we constructed a sono-responsive polymeric perfluorohexane (PFH) nanodroplet to elicit a deep drug penetration in PDAC via the combination of exogenous ultrasonic (US) exposure and endogenous ECM modulation for potent sonodynamic therapy (SDT) of PDAC. Under US exposure, the rapid drug release and deep penetration in PDAC tissues were realized. The released and well penetrated all-trans retinoic acid (ATRA) as an inhibitor of activated PSCs successfully reduced the secretion of ECM components to form a non-dense matrix conducive to drug diffusion . Meanwhile, the sonosensitizer, manganese porphyrin (MnPpIX), was triggered to produce robust reactive oxygen species (ROS) to exert the SDT effect under US exposure. Furthermore, oxygen (O 2 ) delivered by PFH nanodroplets alleviated tumor hypoxia and enhanced the eradication of cancer cells. Overall, the sono-responsive polymeric PFH nanodroplets were successfully developed as an efficient strategy for PDAC therapy. Statement of significance Pancreatic ductal adenocarcinoma (PDAC) is a representative refractory cancer with a highly dense extracellular matrix (ECM), making it difficult for most drugs to penetrate the nearly impenetrable desmoplastic stroma. Seeking methods for deep drug penetration is an extremely pressing matter for the treatment of PDAC and many other solid tumors. Herein, we designed a fluoroalkane-modified polymer to prepare a sono-responsive polymeric perfluorohexane (PFH) nanodroplet for loading sonosensitizers, and inhibitors of activated PSCs and O 2 . Under ultrasonic exposure, the nanodroplet elicited deep drug penetration in PDAC via ultrasonic disturbance and stromal remodeling, inducing potent sonodynamic therapy (SDT) of PDAC. By combining exogenous ultrasonic exposure and endogenous ECM modulation, this work successfully alleviated the severe physiological barrier of PDAC and achieved a favourable treatment effect.
