Exploring the practical method to grow high quality metal phthalocyanine (MPc) crystals and understanding their growth mechanism is significant for their applications. In this study, manganese phthalocyanine (MnPc) crystals were successfully synthesized using simple substance Mn and dicyanobenzene in the reaction medium of ethanediol without any other chemical reagents. The well shaped quadrangular MnPc crystals more than 500 μm were in situ grown on Mn slice surfaces via a simple solvothermal reaction. XPS analyses show that Mn has the mixed chemical state of +3 and + 4 in MnPc. Ethanediol acted as not only the solvent but also acidic catalyst to boost the ionization of dicyanobenzene. Different from the reported synthesis of MPc crystals, MnPc crystals could be formed at a wide temperature range of 100–190 °C. The reaction time is the only experimental parameter to regulate the crystal growth. To the best of our knowledge, this is the first report on in situ synthesis of the high quality MnPc crystals on Mn substrate by using a simple solvothermal method.

The combination of photodynamic therapy (PDT) and enzyme therapy is a highly desirable approach in malignant tumor therapies as it takes advantage of the spatial-controlled PDT and the effective enzyme-catalyzed bioreactions. However, it is a challenge to co-encapsulate hydrophilic enzymes and hydrophobic photosensitizers, and these two agents often interfere with each other. In this work, a protocell-like nanoreactor (GOx-MSN@MnPc-LP) has been designed for synergistic starvation therapy and PDT. In this nanoreactor, the hydrophilic glucose oxidase (GOx) is loaded in the pore of mesoporous silica nanoparticles (MSNs), while the hydrophobic manganese phthaleincyanide (MnPc) is loaded in the membrane layer of liposome. This spatial separation of two payloads protects GOx and MnPc from the cellular environment and avoids interference with each other. GOx catalyzes the oxidation of glucose, which generates hydrogen peroxide and gluconic acid, leading to the starvation therapy via glucose consumption in cancer cells, as well as the disruption of cellular redox balance. MnPc produces cytotoxic singlet oxygen under 730 nm laser irradiation, achieving PDT. The antitumor effects of the nanoreactor have been verified on tumor cells and tumor-bearing mice models. GOx-MSN@MnPc-LP efficiently inhibits tumor growth in vivo with a single treatment, indicating the robust synergy of starvation therapy and PDT treatment. This work also offers a versatile strategy for delivering hydrophilic enzymes and hydrophobic photosensitizers using a protocell-like nanoreactor for effective cancer treatment.

The current conventional photo-therapeutic agents often show low therapy efficacy because of their single treatment model, the limited penetration depth of excitation light and hypoxia in the tumor microenvironment (TME). Herein, a new type of phthalocyanine manganese (MnPcE 4 ) photosensitizer with strong NIR absorption was designed and fabricated for the first time, and was used to modify pure Bi nanomaterials to obtain an intelligent multifunctional Bi/MnPcE 4 nanocomposites . The Mn 2+ in the Bi/MnPcE 4 nanocomposite could catalyze H 2 O 2 to generate O 2 , thus helping to overcome TME hypoxia and enhancing the photodynamic therapy (PDT) efficacy. Further, the nanocomposites showed excellent T 1 -weighted MRI performance. Our novel use of a pure metal Bi core, offers lower toxicity, higher CT imaging performance , and a photothermal therapy (PTT) effect triggered by 808 nm near infrared (NIR) laser. Moreover, in vivo fluorescence imaging ( in vivo FL) vividly showed that the nanocomposite rapidly accumulates in tumor sites due to the enhanced permeability and retention (EPR) effect and metabolized in the organs. The presence of Bi enables the use of these nanocomposites as a CT contrast agent, and the Mn content enables them to be used in MRI. This triple imaging ability implies that our nanocomposites have a high potential for use in imaging directed tumor therapy.

In recent years, transition metal-nitrogen-carbon (M–N-C) composites are expected to be an alternative to platinum group metal (PGM) among various non-precious metal catalysts investigated. However, the major challenge comes from insufficient electrocatalytic performance and durability for oxygen reduction reaction (ORR). In addition to the selection of suitable central metal active sites, the electrocatalytic activity and stability of the M–N-C catalysts can be enhanced by adjusting the electronic structure of the catalysts. In this work, M–N-C/F composites were synthesized by loading transition metal phthalocyanine complexes onto pre-fluorinated carbon nanotubes through a simple pyrolysis method. Pyrroline-N (PN) and graphite-N (GN) formed after thermal treatment can act as electron acceptors to modulate their charge distribution on the M-N 4 sites, and the use of pre-fluorinated nanotubes also allows for a more controlled introduction of fluoride ions that are well coordinated to transition metals, both of which can modulate and modify the electronic structure of M–N-C catalysts. The obtained manganese phthalocyanine/fluorinated carbon nanotubes at 800 °C (MnPc/FCNT800) exhibit a competitive electrocatalytic ORR performance with the half-wave potential ( E 1/2 ) of 0.9 V and only 12.1% decay after 20-h long-term chronoamperometry (CA) test in 1.0 M KOH electrolyte, outperforming the commercial Pt/C. Overall, this work paves the way of the electronic structure modification and design of such M–N-C composites for sustainable energy applications. Graphical Abstract

In this study, tetracarboxylic manganese phthalocyanine coated nano-zinc oxide (MnC 4 Pc-ZnO) composite material was prepared by in-situ growth method and modified with indium tin oxide (ITO) glass electrode to construct a photoelectrochemical (PEC) sensor. A PEC sensor for the determination of amlodipine besylate (AB) was developed for the first time based on the principle of precipitation reaction between heavy metal ions and dihydropyridine and the recombination suppression effect of the material. The morphology and optical properties of the MnC 4 Pc-ZnO composites were characterized by scanning electron microscopy (SEM) and ultraviolet-visible diffuse reflectance spectroscopy (UV-vis DRS). Chronoamperometry (i-t) and electrochemical impedance spectroscopy (EIS) were used to study the PEC behavior of ITO electrodes modified by MnC 4 Pc-ZnO composite material. The study found that the MnC 4 Pc-ZnO composite material has a good photocurrent response to AB, and there was a good linear relationship between the concentration range of 75 nM-250 μM, the linear equation was I (μA)=−5.2×10 −8 ×lg C +3.2×10 −8 (r=0.9947), a limit of detection (LOD) of 20 nM. In addition, MnC 4 Pc-ZnO/ITO also has good selectivity and stability. The PEC sensor detects amlodipine besylate tablets, amlodipine besylate dispersible tablets, and biological samples, with standard addition recovery rates of 96.11 % and 103.96 %, respectively. The determination result has good accuracy, and the PEC sensor provides a new method for detecting AB.

Graphical Delivered by nanobody : Site-specific conjugation of nanobody to ferritin generates a targeted drug-delivery platform; the encapsulation of a photosensitizer results in targeted photodynamic therapy. Ferritin is an iron-storage protein nanocage that is assembled from 24 subunits. The hollow cavity of ferritin enables its encapsulation of various therapeutic agents; therefore, ferritin has been intensively investigated for drug delivery. The use of antibody-ferritin conjugates provides an effective approach for targeted drug delivery. However, the complicated preparation and limited protein stability hamper wide applications of this system. Herein, we designed a novel nanobody-ferritin platform (Nb-Ftn) for targeted drug delivery. The site-specific conjugation between nanobody and ferritin is achieved by transglutaminase-catalyzed protein ligation. This ligation strategy allows the Nb conjugation after drug loading in ferritin, which avoids deactivation of the nanobody under the harsh pH environment required for drug encapsulation. To verify the tumor targeting of this Nb-Ftn platform, a photodynamic reagent, manganese phthalocyanine (MnPc), was loaded into the ferritin cavity, and an anti-EGFR nanobody was conjugated to the surface of the ferritin. The ferritin nanocage can encapsulate about 82 MnPc molecules. This MnPc@Nb-Ftn conjugate can be efficiently internalized by EGFR positive A431 cancer cells, but not by EGFR negative MCF-7 cells. Upon 730 nm laser irradiation, MnPc@Nb-Ftn selectively killed EGFR positive A431 cells by generating reactive oxygen species (ROS), whereas no obvious damage was observed on MCF-7 cells. Given that ferritin can be used for encapsulation of various therapeutic agents, this work provides a strategy for facile construction of nanobody-ferritin for targeted drug delivery.

Reactive oxygen species (ROS) play a fundamental role in antibacterial therapeutic strategies. However, excessive ROS generation can disrupt redox homeostasis and induce a severe inflammatory response, causing immense stress on surrounding cells and tissues. Herein, an intelligent nanoheterojunction established with Prussian blue (PB) and manganese polyphthalocyanine (MnPPc), PB@MnPPc (PM), was established to achieve sequential antibacterial and anti-inflammatory functions via controllable ROS regulation. With spatiotemporal control of near-infrared (NIR, 808 nm) light, PM demonstrates the potential for on-demand generation and scavenging of ROS. When exposed to NIR irradiation, PM exhibits prominent bactericidal abilities, which result from its augmented singlet oxygen generation capacity and photothermal effects. These advantages originate from the narrow band gap as well as the enhanced electron-hole separation and transfer efficiency enabled by its unique heterostructure. Upon cessation of NIR irradiation, PM efficiently achieves ROS elimination and inflammation alleviation through its superior superoxide dismutase-like and catalase-like activities. In vivo studies in a mouse Staphylococcus aureus -infected skin wound model further confirmed the superior therapeutic effects of PM, which effectively eliminated bacteria, relieved inflammation, promoted collagen deposition, and angiogenesis. This work provides an attractive therapeutic modality for equipping nanoheterojunctions with antibacterial and anti-inflammatory properties through programmed ROS regulation.
