Cadmium absorption and translocation of amaranth (Amaranthus mangostanus L.) affected by iron deficiency

Rong Zou,Li Wang,Yuncong C. Li,Zhaohui Tong,Wenmin Huo,Keyu Chi,Hongli Fan

Amaranth ( Amaranthus mangostanus L.) has superior capability for accumulating cadmium (Cd) and has the potential to be used for phytoremediation of Cd contaminated soils. Iron (Fe) is chemically similar to Cd and may mediate Cd-induced physiological or metabolic impacts in plants. The purpose was to investigate the model of time-dependent and concentration-dependent kinetics of Cd absorption under Fe deficiency, understanding the physiological mechanism of Cd absorption in amaranth roots. The kinetic characteristics of Cd uptake by amaranth grown in Cd enriched nutritional solution with or without Fe addition and with methanol-chloroform, carbonyl cyanide 3-chlorophenylhydrazone (CCCP), and lanthanum chloride (LaCl 3 ) were compared using 109 Cd 2+ isotope labeling technique. The results showed that Cd uptake was time-dependent and about 90–93% of uptake occurred during the first 150 min. The kinetics of Cd uptake showed that two stages were involved. The saturation stage fitted the Michaelis-Menten model when concentrations of Cd were lower than 12.71 μmol/L and then the absorption of Cd by roots was increased linearly during the second stage. Only linear absorption was observed with methanol-chloroform treatment while the metabolic inhibitor CCCP inhibited only the saturation absorption process, and the Ca channel inhibitor LaCl 3 partially inhibited the two stages of absorption. These results indicated that the root absorption of 109 Cd 2+ was enhanced under Fe deficiency which induced more Fe transporters in the root cell membrane, and the Ca channel, apoplastic and symplastic pathways enhanced the Cd absorption in roots.

Synergistic Effects of Water Management and Silicon Foliar Spraying on the Uptake and Transport Efficiency of Cadmium in Rice (Oryza sativa L.)

Xiaoyun Huang,Chengwu Fan,Dongyi Xie,Hongxing Chen,Song Zhang,Hui Chen,Song Qin,Tianling Fu,Tengbing He,Zhenran Gao

To study the synergistic effects of water management and silicon (Si) foliar spraying on the uptake and transport of cadmium (Cd) in rice, we designed four treatments: conventional intermittent flooding + no Si foliar spraying (CK), continuous flooding throughout the growth stage + no Si foliar spraying (W), conventional intermittent flooding + Si foliar spraying (Si) and continuous flooding throughout the growth stage + Si foliar spraying (WSi). The results show that WSi treatment reduced the uptake and translocation of Cd by rice and significantly reduced the brown rice Cd content, with no effect on rice yield. Compared with CK, the Si treatment increased the net photosynthetic rate (Pn), stomatal conductance (Gs) and transpiration rate (Tr) of rice by 6.5–9.4%, 10.0–16.6% and 2.1–16.8%, respectively. The W treatment decreased these parameters by 20.5–27.9%, 8.6–26.8% and 13.3–23.3%, respectively, and the WSi treatment decreased them by 13.1–21.2%, 3.7–22.3% and 2.2–13.7%, respectively. The superoxide dismutase (SOD) and peroxidase (POD) activity decreased by 6.7–20.6% and 6.5–9.5%, respectively, following the W treatment. Following the Si treatment, SOD and POD activity increased by 10.2–41.1% and 9.3–25.1%, respectively, and following the WSi treatment, they increased by 6.5–18.1% and 2.6–22.4%, respectively. Si foliar spraying ameliorated the detrimental effects of continuous flooding throughout the growth stage on photosynthesis and antioxidant enzyme activity. We conclude that synergistic continuous flooding throughout the growth stage, combined with Si foliar spraying, can significantly block Cd uptake and translocation and is therefore an effective means of reducing the accumulation of Cd in brown rice. Keywords:water management;foliar spraying;Si;cadmium;photosynthesis

The role of zinc chelate of hydroxy analogue of methionine in cadmium toxicity: effects on cadmium absorption on intestinal health in piglets

H. J. Ni,F. F. Liu,X. Liang,Y.L. Yin,G. Liu

Cadmium (Cd) is a toxic heavy metal that poses a threat to the health of humans and animals. It can cause serious damage to the small intestine, which is the main absorption site of Cd and the primary target organ after oral administration. Our previous study found that zinc chelate of hydroxy analogue of methionine (Zn-HMTB), a new type of feed additive, decreased Cd accumulation in the liver and kidneys. The aim of this study was to investigate the effect of Zn-HMTB on Cd absorption and Cd-induced toxicity in the small intestine of piglets. Twenty-four piglets (Landrace × Large White, 13.22 ± 0.58 kg BW) were randomly divided into four dietary treatment groups: basal diet, and diets containing 30 mg/kg Cd from CdCl2 and 0, 100 or 200 mg/kg Zn from Zn-HMTB. The experiment lasted 27 days. The feed intake and final BW of each piglet were recorded at the end of the experiment. Gastrointestinal (GI) tract tissue and samples of liver, kidney, spleen, heart, lung and longissimus muscle tissue and faeces were collected. The concentrations of Cd and metal trace elements in the GI tract and organs were analysed, as was the relative messenger RNA (mRNA) expression of inflammatory cytokines and metal element transporters in the small intestine, and epithelial apoptosis in the small intestine. The results showed that, compared with Cd-treated piglets, piglets in the Zn-HMTB and Cd cotreatment groups had less Cd deposition in the stomach, ileum, caecum, colon, liver, kidneys, spleen, lungs, heart and muscles (P < 0.05), and lower Cd concentrations in faeces (P < 0.05), suggesting that Zn-HMTB increased Cd absorption and the excretion of Cd in other forms (possibly urine). Zinc chelate of hydroxy analogue of methionine increased Zn deposition in the jejunum and the relative mRNA expression of divalent metal transporters 1 and zinc transporter 5 in the duodenum (P < 0.05), indicating that Zn-HMTB may promote the absorption and transportation of Cd and Zn together by upregulating metal element transporters. Competition between Zn and Cd may be responsible for accelerating Cd excretion. Furthermore, Zn-HMTB reduced Cd-induced apoptosis of enterocytes and inflammatory stimuli in the small intestine, suggesting that Zn-HMTB reduced Cd-induced toxicity to the small intestine. These results suggest that Zn-HMTB can be helpful in decreasing Cd accumulation in the GI tract and organs of piglets and relieving Cd-induced toxicity to the small intestine but cannot reduce the absorption of Cd.

Regulatory roles of indole-3-acetic acid in the physiological and biochemical responses of Cinnamomum camphora seedlings under cadmium stress

Jing Jiang,Chenyang Zhang,Meng Na,Shangqi Xu,Xiaoping Li,Shoubiao Zhou,Jihai Zhou

Soil cadmium (Cd) pollution has become increasingly severe, negatively affecting plant growth, soil safety, the ecological balance, and public health. Phytoremediation technology is an effective method for remediating soil Cd contamination because of its low cost, environmentally friendly nature, and minimal soil disturbance. However, phytoremediation efficiency is affected by factors such as the plant species, Cd concentration, and soil environment. In this study, a 180-day indoor potted experiment was conducted to explore the influences of various indole-3-acetic acid (IAA) levels on the growth, physiological and biochemical responses, Cd uptake, and soil enzyme activity of Cinnamomum (C.) camphora seedlings using 20 mg kg −1 Cd-contaminated soil with or without IAA treatments. The results showed that Cd stress significantly suppressed C. camphora seedling growth, whereas IAA application improved the basal diameter of seedlings by 22.22–133.33 %. This effect of IAA was observed primarily at low concentrations, while higher concentrations showed diminished effects. Compared to the Cd pollution treatment, adding 20 mg kg −1 IAA significantly enhanced the activities of antioxidant enzymes (SOD, POD, CAT), reduced the malondialdehyde (MDA) content, and decreased soil basal respiration (SBR). Moreover, IAA treatment promoted Cd uptake in the roots, stems, and leaves, and reduced the soil Cd content by 22.90–41.17 %. In conclusion, exogenous IAA addition effectively alleviates Cd stress by regulating the plant’s physiological and biochemical characteristics and soil enzyme activities. These findings suggest that the combined use of IAA and C. camphora represents a promising remediation method for soil heavy metal pollution.

Deciphering Cd Stabilization Pathways by Organosilicon in Soil-Pakchoi Systems Through Isotope-Tracing

Jing Sun,Shuxin Tu,Zhenli Zhu

Cadmium (Cd) pollution in farmland soils threatens food safety, making it essential to study soil-crop Cd transport mechanisms and develop remediation strategies. While organosilicon-modified materials demonstrate potential in blocking metal uptake by crops, their underlying mechanisms remain underexplored. This study employed stable isotope 114 Cd tracing to investigate Cd absorption, translocation, allocation in soil-pakchoi system mediated by organosilicon (DMDCS and D6). Results revealed that organosilicon induced Cd redistribution in soils, enhancing its sequestration into non-reactive fractions. Specifically, DTPA-Cd decreased by 13.14-24.19%, while RES-Cd increased by 70.3-106.8%. Organosilicon also inhibited Cd uptake and translocation in pakchoi, reducing root and shoot Cd concentrations by 30.0-33.2% and 54.6-57.1%, respectively, with a 35% decline in translocation factor (TF). This reduction stemmed from diminished Cd accumulation (12.09-17.73% decrease) in soluble subcellular fractions. Among the plant parts, Cd isotopes were markedly fractionated: shoots were isotopically heavier than roots. Silicone application attenuated this fractionation intensity, with Δ 114/110 Cd shoot-root decreased from 0.33 to 0.08. We suggest that organosilicon suppressed the enrichment of exogenous isotopes in plant edible parts due to a 28.1% decrease in Cd isotope exchangeability (E- value) and vacuolar compartmentalization. This study provides new insights into organosilica-mediated Cd dynamics in pakchoi, laying the foundation for future Cd regulation strategies.

Synergistic effects of arbuscular mycorrhizal fungi and biochar are highly beneficial to Ligustrum lucidum seedlings in Cd-contaminated soil

Li Tiantian,Yang Huan,Zhang Naili,Dong Lijia,Wu Aiping,Wu Qiqian,Zhao Mingshui,Liu Hua,Li Yan,Wang Yanhong

Cadmium (Cd) contamination is a widespread environmental issue. There is a lack of knowledge about the impacts of applying arbuscular mycorrhizal fungi (AMF) and biochar, either alone or in their combination, on alleviating Cd phytotoxicity in Ligustrum lucidum . Therefore, a pot experiment was conducted in a greenhouse, where L. lucidum seedlings were randomly subjected to four regimes of AMF treatments (inoculation with sterilized AMF, with Rhizophagus irregularis , Diversispora versiformis , alone or a mixture of these two fungi), and two regimes of biochar treatments (with or without rice-husk biochar), as well as three regimes of Cd treatments (0, 15, and 150 mg kg −1 ), to examine the responses of growth, photosynthetic capabilities, soil enzymatic activities, nutritional concentrations, and Cd absorption of L. lucidum plants to the interactive effects of AMF, biochar, and Cd. The results demonstrated that under Cd contaminations, AMF alone significantly increased plant total dry weight, soil pH, and plant nitrogen (N) concentration by 84%, 3.2%, and 13.2%, respectively, and inhibited soil Cd transferring to plant shoot by 42.2%; biochar alone significantly enhanced net photosynthetic rate, soil pH, and soil catalase of non-mycorrhizal plants by 16.4%, 9%, and 11.9%, respectively, and reduced the soil Cd transferring to plant shoot by 44.7%; the additive effect between AMF and biochar greatly enhanced plant total dry weight by 101.9%, and reduced the soil Cd transferring to plant shoot by 51.6%. Furthermore, dual inoculation with D. versiformis and R. irregularis conferred more benefits on plants than the single fungal species did. Accordingly, amending Cd-contaminated soil with the combination of mixed-fungi inoculation and biochar application performed the best than either AMF or biochar alone. These responses may have been attributed to higher mycorrhizal colonization, soil pH, biomass accumulation, and biomass allocation to the roots, as well as photosynthetic capabilities. In conclusion, the combined use of mixed-fungi involving D. versiformis and R. irregularis and biochar addition had significant synergistic effects on enhancing plant performance and reducing Cd uptake of L. lucidum plants in Cd-contaminated soil. The plant-growth promoting bacteria promote cadmium uptake by inducing a hormonal crosstalk and lateral root formation in a hyperaccumulator plant Sedum alfredii

Yingjie Wu,Luyao Ma,Qizhen Liu,Mette Vestergård,Olivera Topalovic,Qiong Wang,Qiyao Zhou,Lukuan Huang,Xiaoe Yang,Ying Feng

Plant growth-promoting bacteria (PGPB) that inhabit hyperaccumulating plants assist cadmium (Cd) absorption, but the underlying mechanism has not been comprehensively studied. For this reason, we combined the fluorescence imaging, and transcriptomic and metabolomic methods in a Cd hyperaccumulator, Sedum alfredii , inoculated or not with PGPB Pseudomonas fluorescens . The results showed that the newly emerged lateral roots, that were heavily colonized by P. fluorescens , are the main entry for Cd influx in S. alfredii . Inoculation with P. fluorescens promoted a lateral root formation of its host plant, leading to a higher Cd phytoremediation efficiency. Furthermore, the plant transcriptome revealed that 146 plant hormone related genes were significantly up-regulated by the bacterial inoculation, with 119 of them showing a complex interaction, which suggests that a hormonal crosstalk participated root development. The targeted metabolomics analysis showed that P. fluorescens inoculation significantly increased indole acetic acid concentration and significantly decreased concentrations of abscisic acid, brassinolide, trans-zeatin, ethylene and jasmonic acid in S. alfredii roots, thereby inducing lateral root emergence. Altogether, our results highlight the importance of PGPB-induced lateral root formation for the increased Cd uptake in a hyperaccumulating plant.

Strain-specific endophytes modulate physiological and metabolic responses of Perilla frutescens to cadmium stress

Gen Li,Simiao Wang,Yijin Wang,Yibang Zhang,Qing Dang,Yashuo Tian,Longfei Li,Lijuan Wang,Li Han

Perilla frutescens is a medicinal plant that bioactive compounds such as perillaldehyde and luteolin, but cadmium (Cd) contamination threatens its safe cultivation and medicinal quality. While dark septate endophytes (DSEs) are known to increase plant stress tolerance, their strain-specific roles in regulating Cd uptake and secondary metabolite biosynthesis remain poorly understood. This study aimed to investigate three DSE strains, DSE6 ( Paraphoma radicina ), DSE7 ( P. chlamydocopiosa ), and MD3-1 ( Acrocalymma aquatica ), to determine their effects on growth, physiological traits, Cd accumulation, and key metabolite production in P. frutescens under 0, 1, 5, and 10 mg/kg Cd stress. DSE6 and DSE7 significantly reduced root Cd content and improved photosynthetic pigments, antioxidant activity, and biomass under moderate Cd levels. Notably, MD3-1 did not reduce Cd uptake but increased luteolin content by 22.67 % under high Cd stress, suggesting a decoupling between Cd accumulation and metabolite production. All three strains increased perillaldehyde content, with DSE7 showing the greatest increase (262 %) at 5 mg/kg Cd. These findings demonstrate that DSEs regulate Cd tolerance and medicinal quality via distinct mechanisms, and that internal metabolic tolerance may sustain bioactive compound synthesis under heavy metal stress. This highlights their potential as microbial agents for cultivating medicinal plants in contaminated soils.

Heterogeneity of humic/fulvic acids derived from composts explains the differences in accelerating soil Cd-hyperaccumulation by Sedum alfredii

Mao Gao,Fan Tang,Kaidi Wang,Fanjian Zeng,Yan Wang,Guangming Tian

The hyperaccumulating mechanism concerning heavy metal activation or passivation and plant response triggered by fulvic acid (FA) and humic acid (HA) recruitments are investigated herein. We carefully examine the Cd activation effect by various FA and HA, tracing from pig, goat, and duck manure composts to straw compost and commercial materials (i.e., PC, GC, DC, SC, and CM), as well as their roles in plant growth promotion and Cd uptake. Our results indicate that due to the decrease of soil pH and their multiple functional groups, the contents of available Cd (AE–Cd) increased by 4.3–4.8% and 3.6–6.3% when all FA and HA sources were applied for 30 days. A 13.1–19.9% increase in AE–Cd was observed when CFA , DFA , and PFA were applied for five days, and a 9.5% increment was found when PHA was applied for 10 days. In the pot experiment, the Cd accumulation in plants increased by 2.78 and 2.17 folds with PFA and PHA applications, respectively, compared to the blank control group. This result can be attributed to the stimulative effects of the simultaneous Sedum alfredii growth and Cd phytoavailability. Notably, the Cd accumulation increased by 2.26 times with the SFA amendment due to the predominant stimulation effect to the phytoavailable Cd rather than plant growth. However, slight inhibitory effects were observed upon plant growth or Cd uptake, which led to the reduction of the Cd accumulation with DHA, SHA, and CHA employments. Consistently, the corresponding soil Cd removal efficiencies were 43.5% and 34.6% with PFA and PHA, respectively, which hold abundant O- and N-containing groups. Our research aims to gain insights into the ternary interaction in the presence of heavy metal, humic substances , and S. alfredii to simultaneously accelerate Cd activation and hyperaccumulation .

Jasmonic acid alleviates cadmium toxicity through regulating the antioxidant response and enhancing the chelation of cadmium in rice (Oryza sativa L.)

Yan Li,Shengnan Zhang,Qiongli Bao,Yutan Chu,Hongyu Sun,Yizong Huang

Cadmium (Cd) is a potentially hazardous element with substantial biological toxicity, adversely affecting plant growth and physiological metabolism. Therefore, it is necessary to explore practical and environment-friendly approaches to reduce toxicity. Jasmonic acid (JA) is an endogenous growth regulator which helps plants defend against biological and abiotic stresses. To determine how JA help relieve Cd toxicity in rice, both laboratory and field experiments were implemented. In the seedling stage, the role of JA in mediating rice Cd tolerance was investigated via a fluorescent probe in vivo localization, Fourier Transform Infrared Spectroscopy (FTIR), and colorimetry . At the mature growth stage of rice, field experiments were implemented to research the effects of JA on the Cd uptake and translocation in rice. In the seedling stage of rice, we found that JA application increased the cell wall compartmentalization of Cd by promoting the Cd combination on chelated-soluble pectin of rice roots and inhibited Cd movement into protoplasts , thereby reducing the Cd content in the roots by 30.5% and in the shoots by 53.3%, respectively. Application of JA reduced H 2 O 2 content and helped relieve Cd-induced peroxidation damage of membrane lipid by increasing the level of catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX), and glutathione (GSH), but had no significant effect on the superoxide dismutase (SOD) activity. Additionally, field experiments showed that foliar spraying of JA inhibited rice Cd transport from the stalk and root to the grain and reduced Cd concentration in grain by 29.7% in the high-Cd fields and 28.0% in the low-Cd fields. These results improve our understanding of how JA contributes to resistance against Cd toxicity in rice plants and reduces the accumulation of Cd in rice kernels.

Prevention of Cadmium Contamination by Microbial Inoculant and Its Potential Mechanism

Ximei Xu,Xiaofeng Yue,Du Wang,Mengxue Fang,Li Yu,Fei Ma,Nanri Yin,Xuefang Wang,Baocheng Xu,Liangxiao Zhang,Peiwu Li

Cadmium is the main heavy metal contaminant of food in the world. The extent of cadmium pollution in peanut in China remains unclear. To determine the cadmium pollution level in peanut, samples from the main producing regions in China were assessed. The findings revealed that the cadmium pollution level in Chinese peanuts was relatively low. Moreover, the Aflatoxin Rhizobia CoupleB. amyloliquefaciens,B. laterosporu,B. mucilaginosus,E. ludwiggi(ARC-BBBE) microbial inoculants on cadmium contamination in peanut were evaluated. The fertilization methods were categorized into conventional fertilization and conventional fertilization supplemented with 60 kg/hectare of microbial inoculant ARC-BBBE as the base fertilizer. The cadmium contents in the soil and peanut plant parts were detected and analyzed. The results demonstrated that the microbial inoculant ARC-BBBE significantly reduced the total cadmium content in peanut, as well as the available cadmium and exchangeable cadmium in soil. Furthermore, the pH and urease and alkaline phosphatase activities in soil were significantly enhanced, suggesting that the microbial inoculant ARC-BBBE decreased cadmium content in soil and reduced the cadmium uptake by plants through a combination of the action of the bacteria itself and the secretion of extracellular substances. This ultimately achieves the goal of reducing the cadmium content in peanut seeds.
