Hydrogen sulfide alleviates cadmium stress in germinating carrot seeds by promoting the accumulation of proline

Miao Sun,Huan-Xuan Qiao,Tao Yang,Peng Zhao,Jun-Hao Zhao,Jia-Ming Luo,Hai-Ye Luan,Xiang Li,Sheng-Cai Wu,Ai-Sheng Xiong

Carrot ( Daucus carota L.), a widely cultivated economically vegetable from the Apiaceae family, is grown globally. However, carrots can be adversely impacted by cadmium (Cd) pollution in the soil due to its propensity to accumulate in the fleshy root, thus impeding carrot growth and posing health hazards to consumers. Given the potential of hydrogen sulfide (H 2 S) to improve plant resistance against Cd stress, we treated germinating carrot seeds with varying concentrations of sodium hydrosulfide (NaHS), aiming to alleviate the toxic impacts of Cd stress on carrot seed germination. The results revealed that carrot seeds treated with a concentration of 0.25 mM NaHS displayed better seed germination-associated characteristics compared to seeds treated with NaHS concentrations of 0.1 mM and 0.5 mM. Further investigation revealed a rise in the expression levels of L-cysteine desulfhydrase and D-cysteine desulfhydrase , along with enhanced activity of L-cysteine desulfhydrase and D-cysteine desulfhydrase among the NaHS treatment group, thereby leading to H 2 S accumulation. Moreover, NaHS treatment triggered the expression of pyrroline-5-carboxylate synthase and pyrroline-5-carboxylate reductase and promoted the accumulation of endogenous proline, while the contents of soluble sugar and soluble protein increased correspondingly. Interestingly, since the application of exogenous proline did not influence the accumulation of endogenous H 2 S, suggesting that H 2 S served as the upstream regulator of proline. Histochemical staining and biochemical indices revealed that NaHS treatment led to elevated antioxidant enzyme activity, alongside a suppression of superoxide anion and hydrogen peroxide generation. Furthermore, high performance liquid chromatography analysis revealed that NaHS treatment reduced Cd 2+ uptake, thereby promoting germination rate, seed vitality, and hypocotyl length of carrot seeds under Cd stress. Overall, our findings shed light on the application of NaHS to enhance carrot resistance against Cd stress and lay a foundation for exploring the regulatory role of H 2 S in plants responding to Cd stress.

The regulatory mechanism of Chryseobacterium sp. resistance mediated by montmorillonite upon cadmium stress

Huimin Wang,Pingxiao Wu,Juan Liu,Shanshan Yang,Bo Ruan,Saeed Rehman,Liangting Liu,Nengwu Zhu

Cadmium (Cd) is a toxic heavy metal and its uptake by living organisms causes adverse effect, further resulting in cycle pollution of the biosphere. The specific regulatory mechanism between clays and microbes under Cd stress remains unclear. In this study, interface interactions among clays, microbes and Cd were confirmed. Comparative transcriptome was conducted to investigate how it regulated gene expression patterns of microbes ( Chryseobacterium sp. WAL2), which exposed to a series of gradient concentrations of Cd (16, 32, 64 and 128 μg mL −1 ) for 12 d in the presence and absence of clay montmorillonite (Mt) (16 g L −1 ). Cd was highly enriched by the unique interface interactions between Mt and bacteria (67.6–82.1%), leading to a more hostile environment for bacterial cells. However, Mt ultimately enhanced bacterial resistance to Cd stress by stimulating the mechanism of bacterial resistance; namely: (i) Mt increased genes expression connected with ion transport, enhancing the uptake of Cd; (ii) Mt stimulated genes expression related to efflux pump and positively regulated cellular oxidative stress (e.g., glutathione) and Cd accumulation (e.g., cysteine) processes. Further, genes expression related to intracellular metabolic processes was enforced, which supplied a driving force and accelerated electron transfer ; (iii) Mt improved genes expression involved in DNA replication and other biological processes (e.g., terpenoid backbone biosynthesis) to maintain bacterial vitality. Therefore, the study not only optimized a unique Cd resistance mechanism of Mt on Chryseobacterium sp., but also provided a novel insight for environmental mitigation of heavy metals from the perspective of molecular biology .

Regulatory module WRKY33-ATL31-IRT1 mediates cadmium tolerance in Arabidopsis

Cheng Zhang,Chenchen Tong,Lei Cao,Pengpeng Zheng,Xiaofeng Tang,Lihuan Wang,Min Miao,Yongsheng Liu,Shuqing Cao

Cadmium (Cd) is one of the most dangerous environmental pollutants among heavy metals, and threatens food safety and human health by accumulating in plant sink tissues. Here, we report a novel regulatory cascade that profoundly influences Cd tolerance in Arabidopsis. Phenotypic analysis showed that an insertional knockdown mutation at the Arabidopsis Tóxicos en Levadura 31 ( ATL31 ) locus resulted in hypersensitivity to Cd stress, most likely due to a significant increase in Cd accumulation. Consistently, ATL31 -overexpressing lines exhibited enhanced Cd stress tolerance and reduced Cd accumulation. Further, IRON-REGULATED TRANSPORTER 1 (IRT1) was identified, and yeast two-hybrid, co-immunoprecipitation and bimolecular fluorescence complementation assays demonstrated its interaction with ATL31. Biochemical, molecular, and genetic analyses showed that IRT1 is targeted by ATL31 for ubiquitin-conjugated degradation in response to Cd stress. Intriguingly, transcription of ATL31 was strongly induced by Cd stress. In addition, transgenic and molecular analyses showed that WRKY33 directly activated the transcription of ATL31 in response to Cd stress and positively regulated Cd tolerance. Genetic analysis indicated that ATL31 acts upstream of IRT1 and downstream of WRKY33 to regulate Cd tolerance. Our study revealed that the WRKY33-ATL31-IRT1 module plays a crucial role in timely blocking Cd absorption to prevent metal toxicity in Arabidopsis.

Rhizosphere Ecological Characteristics of Trifolium repens L. under Cadmium Stress: with Focus on the Spatial Distribution Pattern of Enzyme Activity

Zhang Chenxi,Li Haibo,Mo Fan,Wang Mingshuai,Bai Yaran,Ma Haotian,Gao Bingqian,Wang Rui

This study aims to illustrate the temporal and spatial patterns of rhizosphere microecological characteristics of plants highly adapted to heavy metals under different levels of heavy metal stress, to reveal the rhizosphere nutrient cycling and the mechanism of enhanced stress tolerance. The dynamic changes of rhizosphere microecology of Trifolium repens L. (white clover) were studied under different concentrations of Cd treatment. The spatial variation of soil enzyme activities was investigated in situ and microscopically by in situ zymography and the spatial distribution models of enzyme activities were developed. The results showed that the rhizosphere ecosystem remained relatively stable under 40 days of stress, with pH ranging from 7.13 to 7.26 and organic matter contents ranging from 14.83 to 18.09 g kg − 1 . In addition, pH and soil organic matter (SOM) were important ecological factors affecting Cd activation in the rhizosphere. Based on soil zymography analysis, both phosphatase and N-acetyl-glucosaminidase hotspots had root effects, and a maximum hotspot area of 21.51 and 10.19% at 10 mg kg − 1 Cd treatment, respectively. Besides, the maximum activities of both enzymes were observed at 5 mg kg − 1 Cd treatment and the rhizosphere extension distance up to 1.82 and 1.59 cm. This study reveals the potential rhizosphere regulatory mechanism of white clover. It was emphasized that the Cd bioavailability was increased in the rhizosphere, the activities of enzymes related to N and P cycling were stimulated under 5–10 mg kg − 1 Cd stress, and thus soil N loss due to Cd could be compensated in the rhizosphere.

Hydrogen sulfide enhances Cd tolerance through persulfidation of SlGSTU24 in Solanum lycopersicum L

Yi Yuying,Li Wenhui,Wang Jinzhao,Sun Guozhi,Ma Ying,Li Fali,Shi Wei,Hu Yanfeng,Jia Honglei,Li Jisheng

Aims Hydrogen sulfide (H₂S), a gaseous signaling molecule, plays a pivotal role in facilitating plant resistance to cadmium (Cd) stress. Glutathione S-transferases (GSTs) are critical enzymes to tolerate Cd stress in plants. However, the regulatory interplay between H₂S and GSTs activity remains elusive. Here, we identified the persulfidation of SlGSTU24 as a previously uncharacterized mechanism regulating Cd detoxification. Methods To investigate the effect of H₂S on the growth of tomato seedlings under Cd stress, 1, 3, and 5 mg/L CdCl 2 and 200 μM NaHS (an H₂S donor) were used to treat three-week-old seedlings for 7 days in this study. Morphological characteristics, malondialdehyde (MDA) content, non-protein thiol (NPT) levels, metallothioneins (MTs) and protein content of treated seedlings were analyzed. GSTs activity were assessed, and persulfidation proteomics was conducted on GSTs. Mass spectrometry and the modified biotin switch method (MBSM) identified persulfidated residues in SlGSTU24. Cys132 was mutated to alanine for functional analysis. Results Cd exposure inhibited the growth of tomato seedlings, while treatment with NaHS alleviated growth inhibition, decreased malondialdehyde (MDA) content and electrolyte leakage (EL) under Cd stress. The application of NaHS increased non-protein thiols glutathione (GSH) and phytochelatins (PCs) contents, and protein thiols metallothionein (MTs) content. The endogenous activity of GSTs was enhanced by NaHS. Cys132 in SlGSTU24 was identified as a persulfidated residue. Persulfidation of Cys132 increased the SlGSTU24 wild type (WT) activity. When Cys132 was mutated to alanine, H₂S-induced SlGSTU24 activity were compromised in SlGSTU24 Cys(C)132 Ala(A) . Conclusions H₂S-mediated persulfidation of Cys132 in SlGSTU24 enhances its activity and improves tomato resistance to Cd stress.

Comparative transcriptomic analysis provides key genetic resources in clove basil (Ocimum gratissimum) under cadmium stress

Bin Wang,Yukun Wang,Xiao Yuan,Yuanyuan Jiang,Yunna Zhu,Xinmiao Kang,Jinming He,Yanhui Xiao

Planting aromatic plant might be a promising strategy for safely utilizing heavy metal (HM)-contaminated soils, as HMs in essential oil could be completely excluded using some special technologies with ease. Clove basil ( Ocimum gratissimum L.) is an important aromatic plant used in essential oil production. Improving cadmium (Cd) tolerance in clove basil can increase its production and improve the utilization efficiency of Cd-contaminated soils. However, the lack of genomic information on clove basil greatly restricts molecular studies and applications in phytoremediation. In this study, we demonstrated that high levels of Cd treatments (0.8, 1.6 and 6.5 mg/L) significantly impacted the growth and physiological attributes of clove basil. Cd contents in clove basil tissues increased with treatment concentrations. To identify Cd stress-responsive genes, we conducted a comparative transcriptomic analysis using seedlings cultured in the Hoagland’s solution without Cd ion (control) or containing 1.6 mg/L CdCl 2 (a moderate concentration of Cd stress for clove basil seedlings). A total of 104.38 Gb clean data with high-quality were generated in clove basil under Cd stress through Illumina sequencing. More than 1,800 differential expressed genes (DEGs) were identified after Cd treatment. The reliability and reproducibility of the transcriptomic data were validated through qRT-PCR analysis and Sanger sequencing. KEGG classification analysis identified the “MAPK signaling pathway,” “plant hormone signal transduction” and “plant-pathogen interaction” as the top three pathways. DEGs were divided into five clusters based on their expression patterns during Cd stress. The functional annotation of DEGs indicated that downregulated DEGs were mainly involved in the “photosynthesis system,” whereas upregulated DEGs were significantly assigned to the “MAPK signaling pathway” and “plant-pathogen interaction pathway.” Furthermore, we identified a total of 78 transcription factors (TFs), including members of bHLH, WRKY, AP2/ERF, and MYB family. The expression of six bHLH genes, one WRKY and one ERF genes were significantly induced by Cd stress, suggesting that these TFs might play essential roles in regulating Cd stress responses. Overall, our study provides key genetic resources and new insights into Cd adaption mechanisms in clove basil.

Comparative effects of silicon and silicon nanoparticles on the antioxidant system and cadmium uptake in tomato under cadmium stress

Guochao Yan,Han Jin,Chang Yin,Yuchen Hua,Qingying Huang,Guanfeng Zhou,Yunmin Xu,Yong He,Yongchao Liang,Zhujun Zhu

Cadmium (Cd) pollution is an important threat to agricultural production globally. Silicon (Si) and silicon nanoparticles (Si NPs) can mitigate Cd stress in plants. However, the mechanisms underlying the impacts of Si and Si NPs on Cd resistance, particularly in low-Si accumulators, remain inadequately understood. Accordingly, we conducted a comparative investigation into the roles of Si and Si NPs in regulating the antioxidant system (enzymes and antioxidants) and Cd uptake (influx rate, symplastic and apoplastic pathways) in tomato (a typical low-Si accumulator). The results revealed that Si and Si NPs improved tomato growth under Cd stress, and principal component analysis (PCA) demonstrated that Si NPs were more effective than Si. For oxidative damage, redundancy analysis (RDA) results showed that Si NPs ameliorated oxidative damage in both shoots and roots, whereas Si predominantly alleviated oxidative damage in roots. Simultaneously, Si and Si NPs regulated antioxidant enzymes and nonenzymatic antioxidants with distinct targets and strengths. Furthermore, Si and Si NPs decreased Cd concentration in tomato shoot, root, and xylem sap, while Si NPs induced a more significant decline in shoot and xylem sap Cd. Noninvasive microtest and quantitative estimation of trisodium-8-hydroxy-1,3,6-pyrenetrisulfonic (PTS, an apoplastic tracer) showed that Si and Si NPs reduced the Cd influx rate and apoplastic Cd uptake, while Si NPs induced a more significant reduction. Moreover, Si regulated the expression of genes responsible for Cd uptake ( NRAMP2 and LCT1 ) and compartmentalization ( HMA3 ), while Si NPs reduced the expression of NRAMP2 . In conjunction with RDA, the results showed that Si and Si NPs decreased Cd uptake mainly by regulating the symplastic and apoplastic pathways, respectively. Overall, our results indicate that Si NPs is more effective in promoting tomato growth and alleviating oxidative damage than Si in tomato under Cd stress by modulating the antioxidant system and reducing apoplastic Cd uptake.

CRISPR/Cas9 Mutant Rice Ospmei12 Involved in Growth, Cell Wall Development, and Response to Phytohormone and Heavy Metal Stress

Zhaoyang Li,Muhammad Junaid Rao,Jiaying Li,Yanting Wang,Peng Chen,Hua Yu,Chongjian Ma,Lingqiang Wang

Pectin is one of the constituents of the cell wall, distributed in the primary cell wall and middle lamella, affecting the rheological properties and the cell wall stickiness. Pectin methylesterase (PME) and pectin methylesterase inhibitor (PMEI) are the most important factors for modifying methyl esterification. In this study, 45PMEIgenes from rice (Oryza sativaL.) were screened by bioinformatics tools, and their structure, motifs, cis-acting elements in the promoter region, chromosomal distribution, gene duplication, and phylogenetic relationship were analyzed. Furthermore, CRISPR/Cas9 was used to edit theOsPMEI12(LOC_Os03G01020) and two mutantpmei12lines were obtained to explore the functions ofOsPMEIin plant growth and development, and under cadmium (Cd) stress. Compared to wild type (WT) Nipponbare, the second inverted internodes of the mutant plants shortened significantly, resulting in the reduction in plant height at mature stage. The seed setting rate, and fresh and dry weights of the mutants were also decreased in mutant plants. In addition, the pectin methylation ofpmei12lines is decreased as expected, and the pectin content of the cell wall increased at both seedling and maturity stages; however, the cellulose and hemicellulose increased only at seedling stage. Interestingly, the growth of thepmei12lines was better than the WT in both normal conditions and under two phytohormone (GA3and NAA) treatments at seedling stage. Under Cd stress, the fresh and dry weights were increased inpmei12lines. These results indicated thatOsPMEI12was involved in the regulation of methyl esterification during growth, affected cell wall composition and agronomic traits, and might play an important role in responses to phytohormones and stress. Keywords:Oryza sativaL.;pectin methylesterase inhibitor;cell wall;plant growth regulators;Cd stress

A fish scale-like magnetic nanomaterial as a highly efficient sorbent for monitoring the changes in auxin levels under cadmium stress

Qingqing Ding,Hui Chen,Chuanhui Huang,Qiaomei Lu,Ping Tong,Wenmin Zhang,Lan Zhang

Sorbents with high surface utilization and good dispersibility are of great importance for the extraction performance of magnetic solid-phase extraction (MSPE). In this study, a fish scale-like magnetic nanomaterial (Co@Co3O4/OCN) was synthesized, which can be used as a highly efficient MSPE sorbent due to its strong magnetism, special morphology, doping of N element, numerous micro–mesopore cavities and organic functional groups (hydroxyl and carboxyl). Furthermore, a Co@Co3O4/OCN-based MSPE method for monitoring the changes in the levels of three auxins (indole-3-acetic acid, indole-3-propionic acid and 3-indole butyric acid) was successfully established. Wide linear ranges (1.0–1000.0 pg mL−1) with good correlation coefficients (R > 0.9992), low limits of detection (LODs, 0.2–4.0 pg mL−1) and satisfactory repeatability (RSD ≤5.9%, n = 3) were obtained. Using the developed method, various growth parts and different growth periods of plants under Cd stress were monitored. The results showed that auxins in various parts of plants showed differential response under Cd stress, and there was a threshold for the changes in auxin levels against Cd stress. This indicates that the developed fish scale-like Co@Co3O4/OCN nanomaterial has a good application prospect for enriching small molecular targets containing hydroxyl and carboxyl groups.

Indole-3-butyric acid mediates antioxidative defense systems to promote adventitious rooting in mung bean seedlings under cadmium and drought stresses

Shi-Weng Li,Xiao-Ying Zeng,Yan Leng,Lin Feng,Xiao-Hu Kang

In vitro experiments were performed to determine whether auxin can mediate the formation of adventitious roots in response to heavy metal and drought stresses using a model rooting plant, mung bean [ Vigna radiata (L.) Wilczek]. The treatments with CdCl 2 or mannitol alone significantly inhibited the formation and growth of adventitious roots in mung bean seedlings. In contrast, when CdCl 2 or mannitol was applied together with indole-3-butyric acid (IBA), IBA considerably cancelled the inhibition of adventitious rooting by stresses. Treatment with CdCl 2 or mannitol alone significantly increased the soluble protein and malondialdehyde (MDA) contents. CdCl 2 and mannitol stress each induced differentially significant changes in the activities of antioxidative enzyme and antioxidant levels during adventitious rooting. Notably, both CdCl 2 and mannitol stress strongly reduced the peroxidase (POD) and ascorbate peroxidase (APX) activities and glutathione (GSH) and phenols levels. Catalase and superoxide dismutase (SOD) activity were enhanced by CdCl 2 but reduced by mannitol. CdCl 2 increased the ascorbate acid (ASA) level, which was decreased by mannitol. Furthermore, when CdCl 2 or mannitol was applied together with IBA, IBA counteracted the CdCl 2 - or mannitol-induced increase or decrease in certain antioxidants, MDA, and antioxidative enzymes. These results suggest that Cd and mannitol stress inhibition of adventitious rooting is associated with the regulation of antioxidative enzymes and antioxidants in cells to defense the oxidative stress . Moreover, IBA alleviates the effects of Cd and mannitol stress on the rooting process partially through the regulation of antioxidative defense systems.

Regulatory roles of indole-3-acetic acid in the physiological and biochemical responses of Cinnamomum camphora seedlings under cadmium stress

Jing Jiang,Chenyang Zhang,Meng Na,Shangqi Xu,Xiaoping Li,Shoubiao Zhou,Jihai Zhou

Soil cadmium (Cd) pollution has become increasingly severe, negatively affecting plant growth, soil safety, the ecological balance, and public health. Phytoremediation technology is an effective method for remediating soil Cd contamination because of its low cost, environmentally friendly nature, and minimal soil disturbance. However, phytoremediation efficiency is affected by factors such as the plant species, Cd concentration, and soil environment. In this study, a 180-day indoor potted experiment was conducted to explore the influences of various indole-3-acetic acid (IAA) levels on the growth, physiological and biochemical responses, Cd uptake, and soil enzyme activity of Cinnamomum (C.) camphora seedlings using 20 mg kg −1 Cd-contaminated soil with or without IAA treatments. The results showed that Cd stress significantly suppressed C. camphora seedling growth, whereas IAA application improved the basal diameter of seedlings by 22.22–133.33 %. This effect of IAA was observed primarily at low concentrations, while higher concentrations showed diminished effects. Compared to the Cd pollution treatment, adding 20 mg kg −1 IAA significantly enhanced the activities of antioxidant enzymes (SOD, POD, CAT), reduced the malondialdehyde (MDA) content, and decreased soil basal respiration (SBR). Moreover, IAA treatment promoted Cd uptake in the roots, stems, and leaves, and reduced the soil Cd content by 22.90–41.17 %. In conclusion, exogenous IAA addition effectively alleviates Cd stress by regulating the plant’s physiological and biochemical characteristics and soil enzyme activities. These findings suggest that the combined use of IAA and C. camphora represents a promising remediation method for soil heavy metal pollution.

Effects of Duckweed Species Diversity on Physiological Responses and Removal Efficiency under Cadmium Stress

Cao Qin,Tan Ai-Juan,Lan Yan,Zou Wen-Bo,Yang Gui-Li

Cadmium (Cd) contamination threatens aquatic ecosystems and human health. Duckweeds are ideal for toxicological studies due to their rapid growth and high metal uptake. While interactions between single duckweed species and Cd has been extensively studied, the impact of duckweed diversity on Cd tolerance remains unknown. This study examines three duckweed species ( Lemna minor , Landoltia punctata , and Spirodela polyrhiza ) under Cd stress, assessing growth, chlorophyll content, Cd accumulation, removal efficiency, and metabolic adaptations in monoculture and mixed-culture systems. The results showed that the growth of duckweeds was inhibited across all cultivation modes under 10 mg L⁻ 1 Cd stress, with growth rates remaining below 0.1 g d⁻ 1 and experiencing more than 80% inhibition. Moreover, chlorophyll content declined with increasing Cd concentration under all cultivation modes, exceeding 64% inhibition under 10 mg L⁻ 1 Cd stress. Cd accumulation peaked in L. minor monoculture at 10 mg L⁻ 1 Cd (1601.33 mg kg⁻ 1 ), with bioenrichment coefficients exceeding 50 in all systems. The combination of S. polyrhiza and L. minor exhibited the highest Cd removal efficiency, achieving 50.60% at a Cd concentration of 1 mg L⁻ 1 . However, the Cd removal rate of all culture modes was below 30% when the Cd concentration increased to 10 mg L⁻ 1 . Elevated antioxidant enzyme activities (POD, CAT) and malondialdehyde (MDA) levels indicated enhanced oxidative stress responses. This study provides support for the application of duckweed polycultures in water contaminated with heavy metals.
