The hazardous effects of plastic and plastic leachates on organisms, even bacteria, have attracted widespread attention, but only a limited effort has been devoted to explore the response of fungi to plastic leachate induced by light irradiation. Here, we performed plastic leaching experiments to obtain leachates from polyethylene (PE), polyethylene terephthalate (PET) and polypropylene (PP), and optical properties of plastic leachates were analysed to determine the influence of light conditions and plastic materials on that. The effects of plastic leachates on the production of fungal enzyme and the biodegradation of heterocyclic dye by fungi were evaluated. Results indicated that the UV light greatly enhanced the release of leachates from the three plastics. Both plastic polymers and light irradiation affected the plastic-derived dissolved organic carbon (DOC) and their aromaticity, but the molecular weight of plastic leachates showed no dependency on light irradiation types, and PE was the easiest to photo age and leached more DOC. Plastic leachates had no dose-effect on the production of extracellular enzymes by fungi. PE leachates showed long-term toxicities to fungi, and no manganese peroxidase activities were detected after a 42-day incubation, while that of controls were up to 73.64 ± 8.81 U/L. However, the PE and PP leachates greatly promoted methylene blue degradation by the fungi, but PET leachates relieved the decolouration of methylene blue, probably because of the benzene ring structure in the PET monomer . Fusarium oxysporum had a stronger degradation ability than Phanerochaete chrysosporium. Our results indicate that plastic leachates can influence the production and secretion of fungi ligninolytic extracellular enzymes, and regulate the fungal degradation of heterocyclic dye.

The non-invasive imaging of the gastrointestinal (GI) tract is highly desired for clinical research due to the various GI tract bacterial infection-induced diseases. To treat GI tract infections, various antibiotics have been used in the clinic. The growing problem of multidrug-resistant bacteria calls for effective antibiotic alternatives. Here, we construct a dual-functional Bi2S3@mSiO2@Ag nanocomposite for simultaneous enhanced X-ray computed tomography (CT) imaging and efficient antibacterial activity in the GI tract. The nanocomposite also has good stability, low cytotoxicity, and negligible hemolysis. Moreover, the investigation of the long-term toxicity and biodistribution of the Bi2S3@mSiO2@Ag nanocomposite after oral administration confirms its safety at the tested dosage. In particular, Ag nanoparticles (NPs) well dispersed on a silica substrate can reduce the antibacterial dosage and enhance the antibacterial activity of the Bi2S3@mSiO2@Ag nanocomposite. Furthermore, we have established bacterially infected enteritis animal models to confirm the antibacterial ability of the nanocomposite. This work opens up a new avenue for the design of a nanotheranostic agent that acts as both a contrast agent for the enhanced visualization of the GI tract and an antibacterial agent as an alternative to antibiotics.

Nanoscale porphyrinic metal–organic frameworks (NMOFs) have emerged as promising therapeutic platforms for the photodynamic therapy (PDT) of cancer in recent years. However, the relatively large sizes of current NMOFs ranging from tens to hundreds of nanometers usually lead to inefficient body clearance and unsatisfactory PDT effect, thus amplifying their long-term toxicity and restricting their further usage. To overcome these shortcomings, herein, ultrasmall porphyrinic metal–organic framework nanodots (MOF QDs) prepared from NMOFs are rationally synthesized via a facile method and used as renal-clearable nanoagents for the enhanced PDT of cancer. Compared with the precursor NMOFs, our well-prepared MOF QDs can generate 2-fold effective toxic reactive oxygen species (ROS) upon the same light irradiation and greatly decrease the inefficacy of PDT caused by the inefficient use of ROS generated from the interior of NMOFs. Meanwhile, the IC50 value of ultrasmall MOF QDs is nearly one-third that of NMOFs, and in vivo results demonstrate that our MOF QDs exhibit better PDT efficacy than NMOFs under the same treatment owing to their overcoming the limited ROS diffusion distance. Significantly, these ultrasmall MOF QDs show efficient tumor accumulation and rapid renal clearance in vivo, indicating their potential in biomedical utility. Last but not least, comprehensive investigations of long-term toxicity of these MOF QDs well demonstrate their overall safety. Therefore, this study will offer valuable insight into the development of safe and high-performance PDT nanoplatforms for further clinical translation.

High energy ionizing radiation was widely used in medical diagnosis and cancer radiation therapy. The high dose of X ray or γ ray can cause the damage of cancerous tissue as well as healthy tissue during therapy. Therefore, it is urgent to develop chemical agents to protect
the healthy tissue from high energy ray invasion. Here, the ultrasmall Pt clusters were employed as the anti-radiation agents for protecting healthy cells and improving survival rate of irradiated mice. It was found that Pt clusters can reduce the DNA damages in irradiated cells. In vivo
experiments show that the Pt clusters treatment can improve the survival rate of irradiated mice up to 30%. As a contrast, only-irradiated mice without Pt clusters treatment completely died after 15 days. The detailed biological experiments showed that Pt clusters can recover the bone marrow
DNA level and superoxide dismutase activities via scavenging free radicals. Importantly, the ultrasmall Pt clusters can be excreted rapidly by kidney and do not cause long-term toxicity.

Background Reactive oxygen species (ROS) overproduction and excessive hypoxia play pivotal roles in the initiation and progression of ulcerative colitis (UC). Synergistic ROS scavenging and generating O2 could be a promising strategy for UC treatment. Methods Ceria nanozymes (PEG-CNPs) are fabricated using a modified reverse micelle method. We investigate hypoxia attenuating and ROS scavenging of PEG-CNPs in intestinal epithelial cells and RAW 264.7 macrophages and their effects on pro-inflammatory macrophages activation. Subsequently, we investigate the biodistribution, pharmacokinetic properties and long-term toxicity of PEG-CNPs in mice. PEG-CNPs are administered intravenously to mice with 2,4,6-trinitrobenzenesulfonic acid-induced colitis to test their colonic tissue targeting and assess their anti-inflammatory activity and mucosal healing properties in UC.Results PEG-CNPs exhibit multi-enzymatic activity that can scavenge ROS and generate O2, promote intestinal epithelial cell healing and inhibit pro-inflammatory macrophage activation, and have good biocompatibility. After intravenous administration of PEG-CNPs to colitis mice, they can enrich at the site of colonic inflammation, and reduce hypoxia-induced factor-1α expression in intestinal epithelial cells by scavenging ROS to generate O2, thus further promoting disrupted intestinal mucosal barrier restoration. Meanwhile, PEG-CNPs can effectively scavenge ROS in impaired colon tissues and relieve colonic macrophage hypoxia to suppress the pro-inflammatory macrophages activation, thereby preventing UC occurrence and development. Conclusion This study has provided a paradigm to utilize metallic nanozymes, and suggests that further materials engineering investigations could yield a facile method based on the pathological characteristics of UC for clinically managing UC.Graphical Abstract

Infiltration of inflammatory cells, especially the M1 macrophages that secrete various types of inflammation cytokines, play crucial roles in the pathogenesis of rheumatoid arthritis (RA). To relief synovial inflammation, M1 macrophages must be eliminated or switched to anti-inflammatory M2 phenotype. We herein developed folic acid modified silver nanoparticles (FA-AgNPs) that can actively deliver into M1 macrophages to synergistically induce M1 macrophages reduction and M2 macrophages polarization for effective RA treatment. The AgNPs was facilely prepared, PEGylated and modified with FA to realize M1 macrophages targeting delivery via folate receptor overexpressed on M1 macrophages surface. After entering cells, FA-AgNPs dissolved and released Ag + in response to intracellular glutathione (GSH), which is the key element to exert a series of anti-inflammatory functions, such as M1 macrophages apoptosis and reactive oxygen species (ROS) scavenging to facilitate M2 macrophages polarization, both of which contributed to RA treatment. This nano-system could passively accumulate into inflamed joints , permit potent anti-inflammatory activity, and impose strong therapeutic efficacy in mice RA models with high biosafety. After treatment, FA-AgNPs could be gradually cleared from the body mainly via feces without tissue accumulation , and did not show any appreciable long-term toxicity. This work declares the first example of using bio-active nanoparticles for RA treatment without loading any drugs, and highlights the potential of FA-AgNPs for targeted RA therapy via simultaneous M1 macrophage apoptosis and M1-to-M2 macrophages re-polarization.
