Arsenic and PAHs impose environmental stress on soil microorganisms, yet their compound effects remain poorly understood. While soil microorganisms possess the ability to metabolize As and PAHs, the mechanisms of microbial response are not fully elucidated. In our study, we established two simulated soil systems using soil collected from Xixi Wetland Park grassland, Hangzhou, China. The As-600 Group was contaminated with 600 mg/kg sodium arsenite, while the As-600-PAHs-30 Group received both 600 mg/kg sodium arsenite and 30 mg/kg PAHs (phenanthrene:fluoranthene:benzo[ a ]pyrene = 1:1:1). These systems were operated continuously for 270 days, and microbial responses were assessed using high-throughput sequencing and metagenomic analysis. Our findings revealed that compound contamination significantly promoted the abundance of microbial defense-related genes, with general defense genes increasing by 11.07 % ∼ 74.23 % and specific defense genes increasing by 44.13 % ∼ 55.74 %. The dominate species Rhodococcus adopts these general and specific defense mechanisms to resist compound pollution stress and gain ecological niche advantages, making it a candidate strain for soil remediation. Our study contributes to the assessment of ecological damage caused by As and PAHs from a microbial perspective and provides valuable insights for soil remediation.

Rice blast disease (RBD) caused by Magnaporthe oryzae , threaten food security by cutting agricultural output. Nano agrochemicals are now perceived as sustainable, cost-effective alternatives to traditional pesticides. This study investigated bioformulation of moringa chitosan nanoparticles (M-CsNPs) and their mechanisms for suppressing RBD while minimizing toxic effects on the microenvironment. M-CsNPs, sized 46 nm with semi-spherical morphology, significantly suppressed pathogen growth, integrity, and colonization at 200 mg L −1 in vitro . Greenhouse tests with foliar exposure to the same concentration resulted in a substantial 77.7 % reduction in RBD, enhancing antioxidant enzyme activity and plant health. Furthermore, M-CsNPs improved photosynthesis, gas exchange, and the nutritional profile of diseased rice plants. RNA-seq analysis highlighted upregulated defense-related genes in treated rice plants. Metagenomic study showcased reshaping of the rice microbiome, reducing Magnaporthe abundance by 93.5 %. Both healthy and diseased rice plants showed increased microbial diversity, particularly favoring specific beneficial species Thiobacillus , Nitrospira , Nocardioides , and Sphingomicrobium in the rhizosphere and Azonexus , Agarivorans , and Bradyrhizobium in the phyllosphere. This comprehensive study unravels the diverse mechanisms by which M-CsNPs interact with plants and pathogens, curbing M. oryzae damage, promoting plant growth, and modulating the rice microbiome. It underscores the significant potential for effective plant disease management.

Soil collembolans live in close proximity to plant roots and may have a role in the phytoextraction of potentially toxic metals from contaminated soils but the underlying mechanisms remain poorly investigated. We hypothesize that soil collembolans may change the root morphology of hyperaccumulators by regulating plant physiological characteristics. Here, a pot experiment was conducted in which a cadmium (Cd) and zinc (Zn) hyperaccumulator ( Sedum plumbizincicola ) was grown with or without a collembolan ( Folsomia candida ), and plant transcriptome and hormones as well as the root characteristics of S. plumbizincicola were analyzed. F. candida promoted the growth and Cd/Zn uptake of S. plumbizincicola , the root and shoot biomass increasing by 53.3 and 34.4%, and the uptake of Cd and Zn in roots increased by 83.2 and 65.4%, respectively. Plant root morphology, total root length, root tip number and lateral root number increased significantly by 40.7, 37.2 and 33.8%, respectively, with the addition of F. candida . Transcriptome analysis reveals that the expression levels of defense-related genes in S. plumbizincicola were significantly up-regulated. In addition, the defensive plant hormones, i.e. salicylic acid in the roots, increased significantly by 338%. These results suggest that the plant in defense of the action of F. candida regulated the expression of the corresponding genes and increased the defensive plant hormones, thus modifying root morphology and plant performance. Overall, this study highlights the importance of the regulation by collembolans of plant growth and metal uptake by interaction with hyperaccumulator roots.

Root-knot nematodes ( Meloidogyne spp.) are highly destructive pests that cause significant yield losses annually. Biological control of nematodes has emerged as a potential alternative in sustainable agriculture. In this study, we originally isolated Bacillus cereus G5 from the rhizosphere soil of rice ( Oryza sativa ). Treatment with the fermentation supernatant of G5 in vitro demonstrated high toxicity to second-stage juveniles (J2) of Meloidogyne graminicola and remarkably inhibited egg hatching. Moreover, G5 steadily colonized rhizosphere soil and rice seedlings, and exhibited excellent biocontrol efficacy against M. graminicola under greenhouse conditions . Notably, the volatile organic compounds (VOCs) produced by G5 displayed high fumigant activity against M. graminicola. The G5 VOCs efficiently reduced the gall index and nematode population in rice roots, while also promoting rice growth in double-layered pot tests. Additionally, the expression of defense genes involved in the salicylic acid ( OsNPR1 , OsWRKY45, OsPAL1 ), jasmonic acid ( OsJaMYB , OsAOS2 ) and ethylene ( OsACS1 ) signalling pathways was significantly upregulated in rice seedlings treated with G5 VOCs. This suggests that G5 VOCs contribute to eliciting plant defense responses. Furthermore, we identified 14 major VOCs produced by G5 using solid-phase micro-extraction gas chromatography and mass spectrometry (SPEM–GC–MS). Notably, allomatrine, morantel, 1-octen-3-ol and 3-methyl-2-butanol displayed strong contact nematicidal activity. Among these, only 1-octen-3-ol demonstrated fumigant activity against J2s of M. graminicola, with an LC 50 value of 758.95 mg/L at 24 h. Overall, these results indicated that the B. cereus G5 and its synthetic VOCs possess high potential as biocontrol agents for managing root-knot nematodes .

Flavone is widely found in plants and plays an important role in plant defense against pests. Many pests, such asHelicoverpa armigera, use flavone as a cue to upregulate counter-defense genes for detoxification of flavone. Yet the spectrum of the flavone-inducible genes and their linkedcis-regulatory elements remains unclear. In this study, 48 differentially expressed genes (DEGs) were found by RNA-seq. These DEGs were mainly concentrated in the retinol metabolism and drug metabolism-cytochrome P450 pathways. Further in silico analysis of the promoter regions of 24 upregulated genes predicted two motifs through MEME and five previously characterizedcis-elements including CRE, TRE, EcRE, XRE-AhR and ARE. Functional analysis of the two predicted motifs and two different versions of ARE (named ARE1 and ARE2) in the promoter region of the flavone-inducible carboxylesterase geneCCE001jverified that the two motifs and ARE2 are not responsible for flavone induction ofH. armigeracounter-defense genes, whereas ARE1 is a new xenobiotic response element to flavone (XRE-Fla) and plays a decisive role in flavone induction ofCCE001j. This study is of great significance for further understanding the antagonistic interaction between plants and herbivorous insects. Keywords:Helicoverpa armigera;flavone;detoxification metabolism;carboxylesterase;cis-transcriptional regulation;AREKey Contribution:The spectrum of flavone up- and downregulated genes was revealed by transcriptomic analysis of the midgut of cotton bollworm treated with flavone, and an ARE-likecis-acting element in the promoter ofCCE001jwas found to determine whether the gene was induced by flavone.
