Effects of Compound Elicitors on the Biosynthesis of Triterpenoids and Activity of Defense Enzymes from Inonotus hispidus (Basidiomycetes)

Jiao Zhou,Xinyue Lin,Shuangshuang Liu,Zhanbin Wang,Dongchao Liu,Yonghong Huo,Dehai Li

Inonotus hispidushas various health-promoting activities, such as anticancer effects and immune-stimulating activity. The commercialization of valuable plant triterpenoids faces major challenges, including low abundance in natural hosts and costly downstream purification procedures. In this work, orthogonal design was used to compound methyl jasmonate (MeJA), salicylic acid (SA), oleic acid, and Cu2+, and the effects of combinations on the total triterpenes biosynthesized were studied. The optimal combination was screened out and its effect on the activity of PAL, CAT, and SOD was studied. The optimal concentration of oleic acid was 2% when MeJA was 100 mol/L, and the total triterpenoid content and mycelia production were 3.918 g and 85.17 mg/g, respectively. MeJA treatment induced oxidative stress, and at the same time increased the activity of related defense enzymes. Oleic acid is thought to regulate cell permeability by recombining cell membranes. It promotes the material exchange process between cells and the environment without affecting cell growth. When oleic acid was used in combination with MeJA, a synergistic effect on triterpene production was observed. In conclusion, our findings provide a strategy for triterpenoid enrichment ofI. hispidus. Keywords:Inonotus hispidus;combination of elicitors;defense enzyme;triterpenoid

cAMP-induced Sporidiobolus pararoseus Y16 alleviates postharvest sour rot of grapes and the potential action mechanisms

Xiaoyun Zhang,Yu Zhao,Esa Abiso Godana,Jun Li,Solairaj Dhanasekarana,Yuanyuan Xie,Lina Zhao,Hongyin Zhang

Sour rot caused by Aspergillus carbonarius is a common postharvest disease of grapes, which leads to the declined fruit quality and serious economic losses of grape industry. In this study, we investigated the control effectiveness of Sporidiobolus pararoseus Y16 induced by cyclic adenosine monophosphate (cAMP) against postharvest sour rot of grapes, and explored the potential action mechanisms. The outcomes presented that S. pararoseus Y16 induced by 0.05 mM cAMP for 24 h further mitigated sour rot compared with S. pararoseus Y16 cultured without cAMP. cAMP could enhance the colonization ability of S. pararoseus Y16. More importantly, cAMP-induced yeast further boosted the activities of antioxidant enzymes and the expression level of the corresponding genes, thus improving the ability of scavenging reactive oxygen species of grapes. Moreover, S. pararoseus Y16 induced by cAMP further mounted the activities of defensive enzymes in grapes, as well as the expression level of the corresponding genes, thereby strengthening the disease resistance of grapes. Additionally, the accumulation of resistant secondary metabolites was further augmented, which also contributed to improving the disease resistance of grapes. This research revealed that cAMP-induced culture is helpful to improve the effectiveness of S. pararoseus Y16 in controlling postharvest sour rot of grapes. This research provides a novel and effective biocontrol strategy for the grape industry to mitigate postharvest sour rot. By using cAMP-induced antagonistic microorganisms, the reliance on chemical fungicides could be potentially reduced, thereby promoting sustainable agricultural practices and enhancing the overall quality and safety of grape products.

Design, synthesis and Anti-PVY activity of planar chiral thiourea derivatives incorporated with [2.2]Paracyclophane

Liangzhen Shu,Ya Lv,Zhongyin Chen,Yixian Huang,Meng Zhang,Zhichao Jin,Tingting Li,Yonggui Robin Chi

BACKGROUND Potato virus Y (PVY) is a prominent representative of plant viruses. It can inflict severe damage upon Solanaceae plants, leading to global dissemination and substantial economic losses. To discover new antiviral agents, a class of planar chiral thiourea molecules through the key step of N -heterocyclic carbene-catalyzed nitrile formation reaction was synthesized with excellent optical purities for antiviral evaluations against plant virus PVY. RESULTS The absolute configurations of the planar chiral compounds exhibited obvious distinctions in the anti-PVY activities. Notability, compound ( S )- 4u exhibited remarkable curative activities against PVY, with a half maximal effective concentration (EC 50 ) of 349.3 μg mL −1 , which was lower than that of the ningnanmycin (NNM) (EC 50 = 400.8 μg mL −1 ). Additionally, The EC 50 value for the protective effects of ( S )- 4u was 146.2 μg mL −1 , which was superior to that of NNM (276.4 μg mL −1 ). Furthermore, the mechanism-of-action of enantiomers of planar chiral compound 4u was investigated through molecular docking, defensive enzyme activity tests and chlorophyll content tests. CONCLUSION Biological mechanism studies have demonstrated that the configuration of planar chiral target compounds plays a crucial role in the molecular interaction with PVY-CP, enhancing the activity of defense enzymes and affecting chlorophyll content. The current study has provided significant insights into the roles played by planar chiralities in plant protection against viruses. This paves the way for the development of novel green pesticides bearing planar chiralities with excellent optical purities. © 2024 Society of Chemical Industry.

Acyl hydrazone derivatives with trifluoromethylpyridine as potential agrochemical for controlling plant diseases

Ya Wang,Renfeng Zhang,Xiaoqiu Guo,Ying Xu,Wei Sun,Shengxin Guo,Jian Wu

BACKGROUND Crops are consistently under siege by a multitude of pathogens. These pathogenic microorganisms, including viruses and bacteria, result in substantial reductions in quality and yield globally by inducing detrimental crop diseases, thus posing a significant challenge to global food security. However, the biological activity sepectrum of commercially available pesticides is limited and the pesticide efficacy is poor, necessitating the urgent development of broad-spectrum and efficient strategies for crop disease prevention and control. RESULTS The bioassay results revealed that certain target compounds demonstrated outstanding in vivo antiviral efficacy against cucumber mosaic virus and tobacco mosaic virus. In particular, compound D6 showed remarkable antiviral activity against CMV, significantly higher than that of the control agent ningnanmycin. Mechanism of action studies have shown that compound D6 could enhance the activity of defense enzymes and upregulate the expression of genes related to disease resistance, thereby enhancing the antiviral effects in plants. In addition, these compounds displayed superior inhibitory activity against plant bacterial diseases. For Xoo , compound D10 showed an excellent inhibitory effect that was better than that of the control agent bismerthiazol. Scanning electron microscopy and fluorescence double-staining experiments revealed that compound D10 effectively inhibited bacterial growth by disrupting the cell membrane. CONCLUSION A series of trifluoromethyl hydrazone derivatives were designed and synthesized, and it was found that they have control effects on plant viruses and bacterial diseases. In addition, this study revealed the mechanism of action of the active compounds and demonstrated their potential as multifunctional crop protectants. © 2024 Society of Chemical Industry.
