CaF2/SiO2 core–shell nanoparticles as novel fillers with reinforced mechanical properties and sustained fluoride ion release for dental resin composites

Cao Jing,Yang Dan-Lei,Pu Yuan,Wang Dan,Wang Jie-Xin

The good mechanical performances and caries inhibition of resin composites (RCs) are of great importance in dental restoration. Generally, the addition of SiO 2 can improve the mechanical properties of RCs, while the introduction of fluoride (F) ions is capable of preventing caries. In this work, we first report the use of CaF 2 /SiO 2 core–shell nanoparticles (CaF 2 /SiO 2 NPs) as novel fillers for developing RCs with excellent mechanical properties and sustained F release. The results show that the superior performances of RCs can be obtained by filling 50 wt.% CaF 2 /50%SiO 2 NPs, where the flexural strength, flexural modulus, compressive strength, and hardness were, respectively, increased by 111.5%, 41.4%, 61.7% and 66.8%, compared with the RCs filled with CaF 2 nanoparticles. In addition, the initial F-release rate of RCs containing 50 wt.% CaF 2 /50%SiO 2 NPs was 2.5 ± 0.4 μg/(h cm 2 ), while the sustained release rate and the F release ratio were 0.96 ± 0.15 μg/h·cm 2 and 5.22‰ after 1632 h (68 days), respectively. It was much more stable and sustained compared with the counterpart filled with CaF 2 NPs. Therefore, it could be envisioned that CaF 2 /SiO 2 NPs have a great potential as inorganic fillers in dental RCs. Graphical abstract

Synthesis and characterization of an anti-caries and remineralizing fluorine-containing cationic polymer PHMB-F

Y. D. Chen,C. Shu,Z. H. Duan,J. J. Xu,X. J. Li,F. Chen,Q. J. Luo,X. D. Li

Dental caries have become a major global public health problem. Plaque control and remineralization of initial enamel lesions are paramount for the prevention and control of caries. Polyhexamethylene biguanide (PHMB) is a type of cationic amphipathic antibacterial agent with broad-spectrum antibacterial properties and good biological safety. Fluoride delays demineralization and promotes the remineralization of hard dental tissues. However, a high concentration is needed for it to function as an antibacterial agent. In order to create a PHMB with the benefits associated with fluoride, we synthesized a fluorine-containing cationic polymer, PHMB-F. Fourier transform-infrared spectroscopy and solid state nuclear magnetic resonance characterization confirmed the successful synthesis of PHMB-F. Antibacterial tests showed that PHMB-F had better antiseptic efficacy for Streptococcus mutans compared with just PHMB. Moreover, positively-charged PHMB-F allows fluoride ions to exist closer to the enamel surface with negative potential, which markedly lowers the ion concentrations in the microenvironment adjacent to hard dental tissues needed to maintain equilibrium. Thus, only low concentrations of PHMB-F are required for enamel remineralization.

Membrane-Anchored Aggregation-Induced Emission Luminogens: Accurate Labeling and Efficient Photodynamic Inactivation of Streptococcus mutans in Anticaries Therapy

Ting Pan,Yixue Li,Feng-Shou Liu,Huancai Lin,Yan Zhou

Dental caries is a widespread bacterial infectious disease that imposes a significant public health burden globally. The primary culprits in caries development are cariogenic bacteria, notably Streptococcus mutans (S. mutans), due to their robust biofilm-forming capabilities. To address this issue, a series of cationic pyridinium-substituted photosensitizers with aggregation-induced emission have been designed. All of these aggregation-induced emission luminogens (AIEgens) exhibit outstanding microbial visualization and photodynamic killing of S. mutans, thanks to their luminous fluorescence and efficient singlet oxygen generation ability. Notably, one of the membrane-anchored AIEgens (TDTPY) can inactivate planktic S. mutans and its biofilm without causing significant cytotoxicity. Importantly, application of TDTPY-mediated photodynamic treatment on in vivo rodent models has yielded commendable imaging results and effectively slowed down caries progression with assured biosafety. Unlike traditional single-mode anticaries materials, AIEgens integrate the dual functions of detecting and removing S. mutans and are expected to build a new caries management diagnosis and treatment platform. To the best of our knowledge, this is also the first report on the use of AIEgens for anticaries studies both in vitro and in vivo.

Sandwich-structured electrospun pH-responsive dental pastes for anti-caries

Yuna Lang,Baolin Wang,Ming-Wei Chang,Renyuan Sun,Longfei Zhang

Dental caries is the most common chronic infectious disease in the human oral cavity and current anti-caries drugs may cause drug resistance and microecological imbalance. In this study, a sandwich-structured electrospun pH-responsive dental paste was engineered via the layer-by-layer electrospinning technique. The top and bottom layers of the dental paste were pH-sensitive layers prepared with Methylmethacrylate polymers IV (E100), and the middle layer was the polylactic acid (PLA) membrane loading with sodium fluorides (NaF). Herein, the surface morphology , chemical as well as physical properties, drug release behaviors, biocompatibility and antibacterial properties of the dental paste were investigated. It was found that the sandwich liked structure enhanced the mechanical properties of the dental paste. The drug release curves of NaF-loaded dental pastes showed that NaF released rapidly and massively in the simulated oral fluid with a risk of dental caries. While in the simulated healthy oral fluid, NaF released slowly and in small amounts. And the dental paste maintained excellent hydrophobic , biocompatibility and antibacterial properties against Streptococcus mutans. Therefore, this sandwich-structured dental paste could respond to pH environment and achieve anti-caries smartly, indicating clinical potential in caries prevention.

Anti-caries effect of resin infiltrant modified by quaternary ammonium monomers

Jinzhao Yu,Xiaoyu Huang,Xuedong Zhou,Qi Han,Wen Zhou,Jingou Liang,Hockin H.K. Xu,Biao Ren,Xian Peng,Michael D. Weir,Mingyun Li,Lei Cheng

Objectives Resin infiltrant is used in early enamel caries. However, commercial resin infiltrant lacks persistent antibacterial activity. Dimethylaminododecyl methacrylate (DMADDM) was added to resin infiltrant to give it sustainable antibacterial properties and inhibit demineralization. Methods After the application of resin infiltrant to bovine enamel, cytotoxicity, surface roughness, and aesthetics were assessed. A multi-species biofilm was incubated on the enamel disk before and one month after microbial-aging. After a 48-h anaerobic incubation, biomass accumulation, metabolic activity, and lactic acid were analyzed using a crystal violet assay, an MTT (3-(4,5-dimethyl-thiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay, and a lactic acid assay. Biofilm structure and composition were determined by live/dead staining, exopolysaccharide (EPS) staining, scanning electron microscopy (SEM), and quantitative polymerase chain reaction (qPCR). The depth and content of demineralization were tested by transverse microradiography (TMR). Results Incorporating DMADDM did not increase the cytotoxicity or change the physical properties when the mass fraction of the DMADDM was 2.5–10 %. The modification decreased the amount of bacterial biofilm, metabolic activity, lactic acid production, EPS, and the proportion of Streptococcus mutans in the biofilms. It also provided anti-demineralization effects. The surface roughness and antibacterial ability were not changed after one month of microbial-aging. Conclusion The incorporation of DMADDM improved the antibacterial and anti-demineralization effects of the material. It demonstrated a sustained antibacterial effect. Clinical significance The antibacterial modification might be a potential choice for future clinical applications to inhibit early enamel caries.
