An ion chromatography (IC) system with on-line sample pretreatment using valve-switching technique was developed for the determination of hexavalent chromium (Cr(VI)) in Colla corii asini . Colla corii asini is a complicated sample with organics as main matrix. In this work, a polymer-based reversed-phase column was used as a pretreatment column. Via valve-switching technique, sample solution with target ions were eluted from a collection loop to analytical columns, with matrix eliminated on-line. Under the optimized separation conditions, the method showed good linearity ( r = 0.9998) in the range of 0.004–1.0 mg/L and satisfactory repeatability (RSD < 3%, n = 6). The limit of detection (LOD) was 1.4 μg/L (S/N = 3). The average spiked recoveries of Cr(VI) were 93.4–102.0%. The result showed that the on-line sample pretreatment IC system was convenient and practical for the determination of trace Cr(VI) in Colla corii asini samples

Addition of nanomaterials into polymer matrix is a promising avenue to reinforce the mechanical properties of composites. In this work, chitosan-based hydrogels reinforced by cellulose nanofibers (CNFs) have been prepared by chitosan as matrix. The effect of CNFs (1vt%, 1.5vt% and 2vt%) on the structure-property relationship in mechanical, swelling capability, pH sensitivity and antibacterial have been investigated, respectively. Different from normal reinforced particle, CNFs with special fiber shape and abundant hydrogen bonding can form interconnected porous structure with chitosan hydrogel, which can spread stress and defer permanent damage under compressing. And the compressive strength increases nearly 20 % and swelling capacity achieve 140 %, when 1.5vt% CNFs added. Thanks to CNFs which obtained from apocynum venetum generated positive effect on the antibacterial rate against E. coli and S. aureus of composite hydrogel. It proves that CNFs display an excellent mechanical and antibacterial enhancement in composite, and it provided a new prospect for the rational selection of the different shapes and aspect ratio of reinforced materials. The obtained CNFs reinforced composite hydrogels could be potentially applied in antibacterial biological and food packaging area.

Colla Corii Asini (Ejiao) peptide-iron (EPI) chelates formed during digestion have been revealed as potential hematopoietic components. However, it remains unclear how EPI regulates endogenous metabolism in vivo. Iron deficiency anemia (IDA) mice were gavaged with EPI chelates in a gradient dosage, while Ejiao hydrolysate-iron (EJI) chelates and FeSO 4 were used as positive controls. The effect of EPI on hematopoietic recovery was evaluated. For the plasma untargeted metabolomic analysis, liquid chromatography-tandem mass spectrometry (LC-MS/MS) were applied to identify metabolites in the samples. EPI significantly increased the levels of hemogram in IDA mice in a dose-dependent manner. The relative iron bioavailability in EPI was 123.45% of that in FeSO 4 in vivo . After high dose of EPI (EPI-H) supplementation, the levels of 22 potential metabolic markers in the IDA group returned to normal, including sphingomyelin and arachidonic acid . EPI-H affected endogenous metabolites by regulating the metabolism of energy, lipids , and amino acids , promoted hematopoiesis , regulated metabolic pathways, and improved iron homeostasis . It’s a comprehensive nutritional intervention of IDA by EPI supplementation.

The objective of this study was to fabricate and characterize antimicrobial films based on Agar/Konjac glucomannan (KA) incorporated with carvacrol (CV) (1%, 1.5% and 2%). The physical, structural and antimicrobial properties were studied to evaluate the effects of carvacrol on the composite films. Results indicated that the addition of 2% CV markedly enhanced the tensile strength (36.72 MPa), elongation at break (100.19%), hydrophobic properties and UV barrier properties compared with KA film. X-ray diffraction, Fourier transform infrared spectroscopy , and scanning electron microscopy indicated good compatibility and the formation of intermolecular hydrogen bonds. Additionally, the addition of 2% CV showed strong antimicrobial activities against the most common food spoiling bacteria, Staphylococcus aureus and Escherichia coli . Finally, the potential application of the films was evaluated by the packing test of chicken, where KA-2% CV was able to prolong the shelf life of refrigerated chicken breast from 5 days to 9 days. Overall, the composite films incorporating 2% carvacrol was a promising antimicrobial packing material to displace petrochemical-based plastics to extend the shelf life of foods .

BACKGROUND The limited water solubility and instability of myricetin (Myr) restrict its application in functional food. Peptides from colla corii asini (CCAP) have recognized potential for the design of hybrid nanoparticles due to their therapeutic bioactivity. In this study, a dual-protein (animal–plant hybrid) delivery system was developed by the integration of CCAP with Zein (Z), with the objective of enhancing the efficiency of Myr for diabetes management. RESULTS Size-exclusion chromatography combined with multi-angle light scattering and zeta potential revealed small- and medium-sized CCAP fragments (83.0–100.5 and 100.5–785.6 Da, 10.0% and 80.0%, respectively; degree of hydrolysis: 78.3%) enabling electrostatic interactions with Z. CCAP-Z-Myr nanoparticles (98.2 nm) exhibited enhanced encapsulation efficiency (91.4%) and thermal stability (endothermic peak shift from 71.8 °C (Z-Myr) to 138 °C (CCAP-Z-Myr)). Fourier transform infrared spectroscopy and X-ray diffraction confirmed Myr amorphization (disappearance of crystalline peaks at 5.8°, 11.4°, 14.1°, 16.6°, 26.3° and 28.4°) and electrostatic interactions between CCAP and Z (CO peak shift from 1657 cm −1 (Z) to 1633 cm −1 (CCAP-Z), amide II from 1542 cm −1 (Z) to 1536 cm −1 (CCAP-Z)). Antioxidant activity of CCAP-Z-Myr surpassed that of Z-Myr (ABTS: 60.4% versus 25.3%, DPPH: 57.6% versus 33.0%, FRAP: 0.65 versus 0.42, hydroxyl radicals: 50.4% versus 30.2%, superoxide anions: 50.9% versus 29.7%). α -Glucosidase inhibitory activity of CCAP-Z-Myr was notably improved compared with Z-Myr (78.5% versus 64.8%). CONCLUSION This study demonstrated a dual animal–plant protein strategy to enhance hydrophobic bioactive Myr delivery for diabetes management, offering novel insights for functional foods and therapeutic formulations. © 2025 Society of Chemical Industry.

Background: Equus asinus L. [Equidae; Asini Corri Colla] (donkey-hide gelatin, E-Jiao) is a traditional Chinese medicine renowned for its exceptional blood-supplementing effect. However, the specific components that contribute to its efficacy remain elusive. This study aimed to demonstrate that peptides are responsible for E-Jiao’s blood-supplementing effect and to explore the specific peptides contributing to its efficacy. Methods: The low molecular weight peptides of E-Jiao (LMEJ) were obtained using an in vitro digestion method. LMEJ and peptides in the rat bloodstream were characterized by peptidomics analysis. The blood-supplementing effect of LMEJ was assessed using blood-deficient zebrafish and mouse models. The effect of the peptides detected in rat blood was evaluated using the same zebrafish model, and network pharmacology analysis was performed to investigate the underlying mechanisms. Results: A total of 660 unique peptides were identified within LMEJ. Both E-Jiao and LMEJ significantly alleviated myelosuppression in mice but only LMEJ attenuated myelosuppression in zebrafish. After the administration of E-Jiao to rats, 67 E-Jiao-derived peptides were detected in the bloodstream, 41 of which were identical to those identified in LMEJ. Out of these 41 peptides, five were synthesized. Subsequent verification of their effects revealed that two of them were able to alleviate myelosuppression in zebrafish. Network pharmacology study suggested that E-Jiao may exert a blood-supplementing effect by regulating signaling pathways such as JAK-STAT, IL-17 and others. These results indicated that peptides are at least partially responsible for E-Jiao’s efficacy. Conclusion: This study provides a crucial foundation for further exploration of the bioactive components of E-Jiao.

Ethnopharmacological relevance Classical prescriptions are not only a primary method of clinical treatment in traditional Chinese medicine (TCM) but also represent breakthroughs in the inheritance and development of this field. Kuntai capsule (KTC), a formulation based on a classical prescription, comprises six TCMs: Rehmanniae Radix Praeparata, Coptidis Rhizoma , Paeoniae Radix Alba , Scutellariae Radix , Asini Corii Colla, and Poria . This formulation possesses various beneficial effects, such as nourishing yin and blood, clearing heat and purging fire, and calming the nerves and relieving annoyance. The investigation of the efficacy and mechanism of KTC in regulating anti-aging factors in the treatment of premature ovarian insufficiency (POI) is not only a prominent topic in classical prescription research but also a crucial issue in the treatment of female reproductive aging using TCM. Aim of the study To evaluate the therapeutic effect of KTC on POI and its underlying mechanism. Materials and methods Healthy and specific pathogen-free (SPF) female Kunming mice aged 6–8 weeks were selected. After acclimatization, the mice were randomly divided into a control, model, and high, middle, and low dose groups of KTC (1.6, 0.8, and 0.4 mg/kg, respectively). Except for the control group, the animals in the other groups were administered a single intraperitoneal injection of 120 mg/kg cyclophosphamide and 30 mg/kg Busulfan to induce the model of POI. After modeling, the mice were treated with the corresponding drugs for 7 days. Serum and ovarian tissues were collected, and the levels of serum follicle-stimulating hormone (FSH), estradiol (E2), and superoxide dismutase 2 (SOD2) were determined using enzyme-linked immunosorbent assay (ELISA). The chemical composition of KTC was characterized and analyzed using ultra-high-pressure liquid chromatography-linear ion trap-Orbitrap tandem mass spectrometry. A “drug-component-target-pathway-disease” network was constructed using network pharmacology research methods to identify the key active components of KTC in treating POI and to elucidate its potential mechanism. The protein expression of the FOXO3/SIRT5 pathway was detected by western blotting . Results Compared to the model group, the high-dose group of KTC showed a significant increase in ovarian index, significant increase in levels of E2 and SOD2, and a significant decrease in FSH levels. Through systematic analysis of the chemical constituents of KTC, 69 compounds were identified, including 7 organic acids, 14 alkaloids, 28 flavonoids , 15 terpenoids , 2 lignans , 2 phenylpropanoids, and 1 sugar. Based on network pharmacology research methods, it was determined that KTC exerts its therapeutic effect on POI through multiple components (paeoniflorin and malic acid), multiple targets (FOXO3 and SIRT5), and multiple pathways (prolactin signaling pathway, longevity regulating pathway, and metabolic pathways). The accuracy of the network pharmacology prediction was further validated by detecting the protein expression of SIRT5 and FOXO3a, which showed a significant increase in the middle and high-dose groups of KTC compared to the model group. Conclusions KTC may effectively treat POI through a multi-component, multi-target, multi-pathway approach, providing an experimental basis for using KTC based on classical prescriptions in the treatment of POI.

Fufang E ’ jiao Jiang (FEJ) as a healthy food consisting of medicine food homology materials approved by China’s Ministry of Health has been extensively applied to replenish qi and nourish blood, and it has a positive impact on women’s health. To find out the material basis and mechanism of FEJ, a systematic “compound-effect-target” analysis including chemical composition resolution, zebrafish, network pharmacology, molecular docking, transcriptome, and bibliometric analysis was adopted. 124 chemical components including ginsenosides, and phenylethanoid glycosides in FEJ were discovered, and effects of FEJ on promoting the generation of immune cells, erythropoiesis and angiogenesis in zebrafish were exhibited. Based on network pharmacology, molecular docking and in vivo activity assay, 6 compounds including jionoside A1, isoacteoside, echinacoside, acteoside, lobetyolin, and rehmannioside D were identified as active components of FEJ. Transcriptome data showed that several pathways such as complement and coagulation cascades, ECM-receptor interaction, and PI3K-Akt signaling pathway were associated with proangiogenic effect of FEJ. 19 common targets were obtained through combined analysis of network pharmacology and transcriptomics, and 5 targets of them were verified by PCR. The bibliometric analysis of these common targets revealed that FEJ was related to energy metabolism, pathway in cancer, etc., which was consistent with the results of network pharmacology and transcriptome. The studies suggested that FEJ could replenish qi and nourish blood through multi-compound and multi-targets.
