Effects of ferulic acid and its esterified derivatives on Botryosphaeria dothidea and postharvest kiwifruit quality

Sisi Wu,Xiaomei Cheng,Xinyi Zhang,Yan Liu,Gaoyang Li,Shuangping Chen,Xiangrong Zhu

This study explores the antifungal properties of ferulic acid (FA), ferulic acid methylester (FAM), and ferulic acid ethylester (FAE), against Botryosphaeria dothidea ( B. dothidea ), and their effects on the postharvest quality (decay rate, weight loss rate, total soluble solids, titratable acids, firmness, respiration rate) of “Hongyang” kiwifruit. The results demonstrated that FAE exhibited a stronger in vitro inhibitory effect on B. dothidea than FA and FAM, and offered superior preservation of kiwifruit quality after harvest. Scanning electron microscopy (SEM) images revealed that B. dothidea mycelium treated with FA, FAM, and FAE showed altered surface morphology, with signs of plasmolysis. Transmission electron microscopy (TEM) further confirmed that FAE caused significant damage to the cell membrane and organelles of B. dothidea . Additional analyses using propidium iodide (PI) and calcofluor fluorescence white (CFW) staining, conductivity tests, and alkaline phosphatase (AKP) assays demonstrated that FA, FAM, and FAE treatments compromised cell membrane integrity. In addition, FAE treatment reduced the decay rate of kiwifruit, delayed the softening process and respiratory peak, maintained firmness, and prolonged antioxidant enzyme activity, preserving the fruit's quality over time. These findings suggest that FAE is a potent antifungal agent against B. dothidea with potential applications as an eco-friendly solution for controlling postharvest soft rot in kiwifruit. Practical Application This study explores the antifungal properties of ferulic acid (FA), ferulic acid methylester (FAM), and ferulic acid ethylester (FAE), against Botryosphaeria dothidea , and their effects on the postharvest quality of “Hongyang” kiwifruit. This work provides an eco-friendly solution for controlling postharvest soft rot in kiwifruit.

Antioxidant activity and mechanism of Rhizoma Cimicifugae

Li Xican,Lin Jing,Gao Yaoxiang,Han Weijuan,Chen Dongfeng

Background As a typical Chinese herbal medicine, rhizoma Cimicifugae (RC, 升麻 in Chinese) possesses various pharmacological effects involved in antioxidant activity. However, its antioxidant activity has not been reported so far. The aim of the present study was to systematically evaluate the antioxidant ability of RC in vitro, then discuss the mechanism. Methods Firstly, five RC extracts (i.e. petroleum ether extract PERC, ethyl acetate extract EARC, absolute ethanol extract AERC, 95% ethanol extract 95ERC, and water extract WRC) were prepared and determined by various antioxidant methods, including anti-lipidperoxidation, protection against DNA damage, ·OH scavenging, ·O 2 - scavenging, DPPH· (1,1-diphenyl-2-picryl-hydrazl radical) scavenging, ABTS + · (2,2’-azino-bis (3-ethylbenzo- thiazoline-6-sulfonic acid radical ion) scavenging, Cu 2+ -chelating, and Fe 3+ reducing assays. Subsequently, we measured the chemical contents of five RC extracts, including total phenolics, total saponins, total sugars, caffeic acid, ferulic acid and isoferulic acid. Finally, we quantitatively analyzed the correlations between antioxidant levels (1/IC 50 values) and chemical contents. Results In the study, the antioxidant levels and chemical contents (including total phenolics, total saponins, total sugars, caffeic acid, ferulic acid and isoferulic acid) of five RC extracts were determined by various methods. In all antioxidant assays, five RC extracts increased the antioxidant levels in a dose-dependent manner. However, their antioxidant levels (IC 50 values) and chemical contents significantly differed from each other. Quantitative analysis of the correlation showed that total phenolic was of significant positive correlations (average R value was 0.56) with antioxidant levels; In contrast, total sugars and total saponins had no positive correlation with antioxidant (the average R values were −0.20 and −0.26, for total sugars and total saponins, respectively); Among total phenolics, three phenolic acids (caffeic acid, ferulic acid and isoferulic acid) also displayed positive correlations (the average R values were 0.51, 0.50, and 0.51, for caffeic acid, ferulic acid and isoferulic acid, respectively). Conclusions As an effective antioxidant, Rhizoma Cimicifugae can protect DNA and lipids against oxidative damage. Its antioxidant ability can be responsible for its various pharmacological effects and may be mainly attributed to the existence of total phenolics, among which caffeic acid, ferulic acid and isoferulic acid are regarded as main bioactive components. Rhizoma Cimicifugae exerts its antioxidant effect through metal-chelating, and radical-scavenging which is via donating hydrogen atom (H·) and donating electron (e).

Interactions of ferulic acid and ferulic acid methyl ester with endogenous proteins: Determination using the multi-methods

Ying Yang,Shuqin Wang,Xingyan Liu,Wenbin Zhang,Wenhua Tong,Huibo Luo,Liming Zhao

Ferulic acid (FA) and ferulic acid methyl ester (FAM) are important phenolic compounds in Baijiu. In this study, the interaction of FA and FAM with human serum albumin (HSA) and lysozyme (LZM) was investigated using multispectral methods and molecular dynamics simulation. FA and FAM could interact with HSA and LZM, changing the conformation and hydrophilicity of the protein. The quenching mechanisms of FA-HSA, FA-LZM, FAM-HSA, and FAM-LZM were all static-quenching. In the FA-HSA, FAM-HSA, and FA-LZM systems, the interaction forces were mainly hydrophobic interactions and hydrogen bonding. In the FAM-LZM system, the interaction forces were mainly hydrophobic interactions, hydrogen bonding, and van der Waals force. Common metal ions such as K + , Ca 2+ , Cu 2+ , Mg 2+ , and Mn 2+ could affect the binding ability of FA and FAM to HSA and LZM. Moreover, FA and FAM could increase the stability of HSA and LZM, and the protein bound to FA/FAM was more stable than the free protein. FA and FAM had varying degrees of impact on the physiological activities of HSA and LZM. This study provides relevant information on the interactions and metabolic mechanisms of FA and its derivatives with endogenous proteins.

Immobilization of feruloyl esterases on magnetic nanoparticles and its potential in production of ferulic acid

Fuming He,Song Zhang,Xinli Liu

Feruloyl esterase plays an indispensable role in hydrolyzing plant cell walls, and ferulic acid will be released as by-product, which has potential applications in food and medicine industry. This study presents immobilization of partially purified feruloyl esterases from the fermentation liquor of recombinant Pichia on magnetic Fe 3 O 4 nanoparticles. Furthermore, the optimal conditions for the immobilization and some characteristics of immobilized enzyme were studied. The optimal immobilization conditions observed were enzyme 0.2 mg (1 mg/mL, 0.2 mL), magnetic Fe 3 O 4 nanoparticles 4 mg, pH 6.0, immobilization time 3 h. The results showed that the optimal reaction temperature of immobilized enzyme was increased from 45°C to 55°C. Thermal stability of the immobilized enzyme also had an improvement, the residual activity retained 80% and 69% after 120 h at 40°C and 50°C, respectively, while free enzymes only showed 45% and 40% remnant activity at the same condition. The immobilized enzyme also exhibited good operational stability and 52.4% of its initial activity was observed during the fifth cycle. In terms of the thermal and operational stability, the immobilized enzyme could be better used in many more applications than the free enzymes. At last the destarched wheat bran was taken as substrate for immobilized and free feruloyl esterases to produce ferulic acid, the maximum ferulic acid yield was 11.2% and 12.3%, respectively, indicating a great potential in industrial application.

Inhibitory Effect of Apigenin on Losartan Metabolism and CYP2C9 Activity in vitro

Wang Zhe,Gong Yun,Zeng Da-li,Chen Lian-guo,Lin Gao-tong,Huang Cheng-ke,Sun Wei,Chen Meng-Chun,Hu Guo-xin,Chen Rui-jie

Background: CYP2C9 is one of the most important phase I drug-metabolizing enzymes in liver. The objective of this work was to investigate the effects of apigenin on the metabolism of losartan and human CYP2C9 and rat CYP2C11 activity in vitro. Methods: Different concentrations of apigenin were added to a 100 mmol/l Tris-HCl reaction mixture containing 2 pmol/ml recombinant human CYP2C9.1, 0.25 mg/ml human liver microsomes or 0.5 mg/ml rat liver microsomes to determine the half maximal inhibition or a half-maximal inhibitory concentration (IC50) on the metabolism of losartan. In addition, diclofenac used as CYP2C9 substrate was performed to determine the effects of apigenin on CYP2C9. Results: The results showed that apigenin has the inhibitory effect on the metabolism of losartan in vitro, the IC50 was 7.61, 4.10 and 11.07 μmol/l on recombinant CYP2C9 microsomes, human liver microsomes and rat liver microsomes, respectively. Meanwhile, apigenin's mode of action on human CYP2C9 activity was competitive for the substrate diclofenac. In contrast to its potent inhibition of CYP2C9 in humans (9.51 μmol/l), apigenin had lesser effects on CYP2C11 in rat (IC50 = 15.51 μmol/l). Conclusion: The observations imply that apigenin has the inhibitory effect on the metabolism of losartan and CYP2C9 activity in vitro. More attention should be paid as to when losartan should be administrated combined with apigenin

Directed evolution of feruloyl esterase from Lactobacillus acidophilus and its application for ferulic acid production

Shujun Liu,Lubna Soomro,Xue Wei,Xufeng Yuan,Tianyi Gu,Zhen Li,Yulu Wang,Yuming Bao,Fengzhong Wang,Boting Wen,Fengjiao Xin

Producing ferulic acid (FA) from the natural substrate with feruloyl esterase is promising in industries, screening and engineering new enzymes with high efficiency to increase the FA yield is of great concern. Here, the feruloyl esterase of Lactobacillus acidophilus (FAELac) was heterologous expressed and the FAELac with different oligomerization states was separated. Interestingly, the activity of dimer was 37-fold higher than high-polymer. To further enhance the efficiency of FAELac, eight mutants were generated based on the simulated structure, of which Q198A, Q134T enhanced the catalytic efficiency by 5.4- and 4.3-fold in comparison with the wild type. Moreover, higher yields of FA (2.21, 6.60, and 1.67 mg/g substrate, respectively) were released by the mutants from de-starched wheat bran, insoluble wheat arabinoxylan, and steam-exploded corn stover. These results indicated that improving the purification process, engineering new FAELac and substrates bias studies hold great potential for increasing FA production yield.

Characterization of the sunflower oil-based oleogel developed by the ferulic acid/monoglyceride mixture and its application in chocolate preparation

Sheng Zhang,Sheng Geng,Hanjun Ma,Benguo Liu

The ferulic acid (FA) / monoglyceride (MG) mixture could act as a gelator to structure sunflower oil at the gelator concentration ( c ) ≥ 4 % and the FA/MG ratios ( r ) of 0:100, 25:75, 50:50 and 75:25. The rectangular FA and needle-shaped MG crystals in the oleogel interlock with each other to form a 3D network, restricting the flow of oil. The gel strength and rheological performance of the oleogel were positively correlated with c and negatively correlated with storage temperature ( t ). Its gelation temperature was determined by c and r . Its β-carotene protective ability against ultraviolet irradiation was proportional to the FA concentration. The oleogel with r  = 25:75, c  = 5 % and t  = 4 °C could partially substitute cocoa butter for chocolate production. With the increasing substitution rate (W CBS ), the appearance, texture properties and enthalpy change of chocolate gradually deteriorated. But when W CBS  ≤ 40 %, the oleogel had no significant effect on the sensory characteristics.

Ferulic Acid Exerts Neuroprotective Effects via Autophagy Induction in C. elegans and Cellular Models of Parkinson’s Disease

Tao Long,Qian Wu,Jing Wei,Yong Tang,Yan-Ni He,Chang-Long He,Xue Chen,Lu Yu,Chong-Lin Yu,Betty Yuen-Kwan Law,Jian-Ming Wu,Da-Lian Qin,An-Guo Wu,Xiao-Gang Zhou

Parkinson’s disease (PD) is a complex neurological disorder characterized by motor and nonmotor features. Although some drugs have been developed for the therapy of PD in a clinical setting, they only alleviate the clinical symptoms and have yet to show a cure. In this study, by employing the C. elegans model of PD, we found that ferulic acid (FA) significantly inhibited α-synuclein accumulation and improved dyskinesia in NL5901 worms. Meanwhile, FA remarkably decreased the degeneration of dopaminergic (DA) neurons, improved the food-sensing behavior, and reduced the level of reactive oxygen species (ROS) in 6-OHDA-induced BZ555 worms. The mechanistic study discovered that FA could activate autophagy in C. elegans, while the knockdown of 3 key autophagy-related genes significantly revoked the neuroprotective effects of FA in α-synuclein- and 6-OHDA-induced C. elegans models of PD, demonstrating that FA exerts an anti-PD effect via autophagy induction in C. elegans. Furthermore, we found that FA could reduce 6-OHDA- or H2O2-induced cell death and apoptosis in PC-12 cells. Moreover, FA was able to induce autophagy in stable GFP-RFP-LC3 U87 cells and PC-12 cells, while bafilomycin A1 (Baf, an autophagy inhibitor) partly eliminated the protective effects of FA against 6-OHDA- and H2O2-induced cell death and ROS production in PC-12 cells, further confirming that FA exerts an anti-PD effect via autophagy induction in vitro. Collectively, our study provides novel insights for FA as a potent autophagy enhancer to effectively prevent neurodegenerative diseases such as PD in the future.

Ferulic Acid Induces Autophagy and Apoptosis in Colon Cancer CT26 Cells via the MAPK Pathway

Shanbin Chen,Dong Zhao,Chunguang Luan,Jia Zheng,Wei Liu,Zheng Feng,Ruiqi Luo,Xinglin Han,Deliang Wang

Ferulic acid (FA) is a bioactive compound found in traditional Chinese herbal medicine; for example, it is present in Xinjiang Ferula, but also in strong-flavor Chinese baijiu. FA has been shown to play a crucial role in treating oxidative stress, skin whitening, and eye diseases. In this study, the potential role of FA as a means of inducing apoptosis and inhibiting colon cancer induced by the transplantation of CT26 cells was investigated. The results show that FA adjuvant treatment caused an upregulation in the expression of genes related to autophagy while simultaneously suppressing the expression of inflammatory response elements and improving the bodyweight, glutamic pyruvic transaminase (ALT), and glutamic oxaloacetic transaminase (AST) in vivo. Furthermore, FA inhibited the proliferation of CT26 cells and induced apoptosis, specifically by activating the phosphorylation of ERK and JNK to enhance the essential proteins BCL-2 and BAX in the apoptosis pathway. These results suggest that FA could be a promising auxiliary therapeutic agent for the treatment of colon cancer. Further research is needed to better understand the mechanisms underlying the beneficial effects of FA and its synergistic effects with other compounds. Keywords:ferulic acid;apoptosis;autophagy;MAPK pathway

Rapid determination of ferulic acid in three kinds of Chinese herbs by direct analysis in real-time mass spectrometry

Zhongping Huang,Xiaoli Huang,Huijun Liu,Lili Wang,Yilei Fan,Charles Liu,Yiping Ren

A new method of direct analysis in real-time integrated with mass spectrometry was established and applied to the rapid determination of ferulic acid in Chinese herbs. Instrument parameter settings were optimized to obtain the sensitive and accurate determination of ferulic acid. At the sample introduction speed of 0.5 mm/s, high signal intensity of [M-H] − ( m/z 193) for ferulic acid was achieved in the negative ion mode by utilization of high purity helium gas at 350°C. The calibration curve was linear over the range of 5 – 100 mg/L with the correlation coefficient r = 0.9846 and relative standard deviation (n = 6) in the range of 2.0 to 6.1%. The limit of detection ( S/N = 3) and limit of quantitation ( S/N = 10) were 0.4 and 1.2 mg/L respectively. The recoveries were in the range of 90.6–114.2%. The results of direct analysis in real time-mass spectrometry were in agreement with those obtained by high performance liquid chromatography. The proposed direct analysis in real time mass spectrometry method is a simple, rapid and high-throughput approach for the determination of ferulic acid in Chinese herbs.

Pharmacokinetic Study of Ferulic Acid Following Transdermal or Intragastric Administration in Rats

Yan Na,Tang Zhan,Xu Yuqi,Li Xiumin,Wang Qiao

Ferulic acid is contained in some Chinese herbal medicines such as Ligusticum chuanxiong or Angelica sinensis. Studies have focused on the treatment of inflammatory diseases and pain using ferulic acid. However, little is known about its pharmacokinetics after transdermal administration. The present research investigated the pharmacokinetic behavior of ferulic acid in rat plasma and skin microdialysate after ferulic acid transdermal or intragastric administration. Samples collected at predetermined time points were determined by a simple and sensitive HPLC-UV method. The pharmacokinetic parameters were estimated using non-compartmental analysis with DAS 2.0 software. The values of AUC 0- t and C max after intragastric administration (20 mg/kg) in plasma were 281.47 ± 46.76 min mg/L and 12.20 ± 2.46 mg/L, respectively. After emulsion transdermal administration (117 mg/kg, 35 mg/4 cm 2 ), the values of AUC 0- t and C max in plasma and skin microdialysate were 953.90 ± 175.30 min mg/L, 7630.47 ± 1410.33 min mg/L, 3.00 ± 0.61 mg/L, and 19.08 ± 4.39 mg/L, respectively. Here, we show a promising delivery system for ferulic acid that could replace traditional administration, and a better understanding of the transdermal pharmacokinetics of ferulic acid, which may be helpful for further clinical and laboratory studies.

Enhancing oxidative stability of encapsulated fish oil by incorporation of ferulic acid into electrospun zein mat

Huan Yang,Kun Feng,Peng Wen,Min-Hua Zong,Wen-Yong Lou,Hong Wu

A composite zein nanofibrous mat containing fish oil and ferulic acid was successfully fabricated by electrospinning. The process became more fluent and continuous by adding 30 g/L glycerol into the polymer solutions and using modified coaxial electrospinning. The average diameter of nanofibers was 440 nm. The loading capacity and encapsulation efficiency of fish oil were 20% and 94%, respectively. FTIR data demonstrated that fish oil and ferulic acid were successfully embedded into the nanofibers and there were interactions among the molecules of zein, fish oil and ferulic acid. Addition of ferulic acid into the nanofibers significantly improved the oxidative stability of encapsulated fish oil; moreover, it did not change the release behavior of fish oil. The release of encapsulated fish oil was controlled by a combination of diffusion and macromolecular chain relaxation. This composite nanofibrous mat with favorable oxidation stability and release property is potential in application as nutrition additive.
