GhPLP2 Positively Regulates Cotton Resistance to Verticillium Wilt by Modulating Fatty Acid Accumulation and Jasmonic Acid Signaling Pathway

Zhu Yutao,Hu Xiaoqian,Wang Ping,Gao Linying,Pei Yakun,Ge Zhaoyue,Ge Xiaoyang,Li Fuguang,Hou Yuxia

Patatin-like proteins (PLPs) have non-specific lipid acyl hydrolysis (LAH) activity, which can hydrolyze membrane lipids into fatty acids and lysophospholipids. The vital role of PLPs in plant growth and abiotic stress has been well documented. However, the function of PLPs in plant defense responses against pathogens is still poorly understood. Here, we isolated and identified a novel cotton (Gossypium hirsutum) PLP gene GhPLP2. The expression of GhPLP2 was induced upon treatment with Verticillium dahliae, the signaling molecules jasmonic acid (JA) and ethylene (ETH) in cotton plants. Subcellular localization revealed that GhPLP2 was localized to the plasma membrane. GhPLP2-silenced cotton plants were more susceptible to infection by V. dahliae, while the overexpression of GhPLP2 in Arabidopsis enhanced its resistance to V. dahliae, which was apparent as mild symptoms, and a decrease in the disease index and fungal biomass. The hypersensitive response, deposition of callose, and H2O2 accumulation triggered by V. dahliae elicitor were reduced in GhPLP2-silenced cotton plants. The overexpression of GhPLP2 in Arabidopsis resulted in the accumulation of linoleic acid (LA, 18:2) and α-linolenic acid (ALA, 18:3) and facilitated the biosynthesis of JA and JA-mediated defensive responses. GhPLP2 silencing in cotton plants consistently reduced the accumulation of linoleic acid (LA, 18:2) and α-linolenic acid (ALA, 18:3) and suppressed the biosynthesis of JA and the defensive responses mediated by JA. These results indicate that GhPLP2 is involved in the resistance of cotton to V. dahliae by maintaining fatty acid metabolism pools for JA biosynthesis and activating the JA signaling pathway.

Steam explosion distinctively enhances biomass enzymatic saccharification of cotton stalks by largely reducing cellulose polymerization degree in G. barbadense and G. hirsutum

Yu Huang,Xiaoyang Wei,Shiguang Zhou,Mingyong Liu,Yuanyuan Tu,Ao Li,Peng Chen,Yanting Wang,Xuewen Zhang,Hongzhong Tai,Liangcai Peng,Tao Xia

In this study, steam explosion pretreatment was performed in cotton stalks, leading to 5–6 folds enhancements on biomass enzymatic saccharification distinctive in Gossypium barbadense and Gossypium hirsutum species. Sequential 1% H 2 SO 4 pretreatment could further increase biomass digestibility of the steam-exploded stalks, and also cause the highest sugar–ethanol conversion rates probably by releasing less inhibitor to yeast fermentation. By comparison, extremely high concentration alkali (16% NaOH) pretreatment with raw stalks resulted in the highest hexoses yields, but it had the lowest sugar–ethanol conversion rates. Characterization of wall polymer features indicated that biomass saccharification was enhanced with steam explosion by largely reducing cellulose DP and extracting hemicelluloses. It also showed that cellulose crystallinity and arabinose substitution degree of xylans were the major factors on biomass digestibility in cotton stalks. Hence, this study has provided the insights into cell wall modification and biomass process technology in cotton stalks and beyond.

Comprehensive Evaluation of Nutritional Quality Diversity in Cottonseeds from 259 Upland Cotton Germplasms

Yiwen Huang,Chengyu Li,Shouyang Fu,Yuzhen Wu,Dayun Zhou,Longyu Huang,Jun Peng,Meng Kuang

Cottonseeds, rich in high-quality protein and fatty acids, represent a vital plant-derived feedstuff and edible oil resource. To systematically investigate genetic variation patterns in nutritional quality and screen superior germplasm, this study analyzed 26 nutritional quality traits and 8 fiber traits across 259 upland cotton (Gossypium hirsutumL.) accessions using multivariate statistical approaches. Results revealed significant genetic diversity in cottonseed nutritional profiles, with coefficients of variation ranging from 3.42% to 26.37%. Moreover, with advancements in breeding periods, the contents of protein, amino acids, and the proportion of unsaturated fatty acids (UFAs) increased, while oil content and C16:0 levels decreased. Correlation analyses identified significant positive associations (p< 0.05) between proteins, amino acids, UFAs, and most fiber traits, except for seed index (SI), fiber micronaire (FM), and fiber elongation (FE). Through a principal component analysis–fuzzy membership function (PCA-FMF) model, 13 elite accessions (F > 0.75) with high protein content, high UFA proportion, and excellent fiber quality were identified. These findings provide both data-driven foundations and practical germplasm resources for value-added utilization of cottonseed and coordinated breeding for dual-quality traits of nutrition and fiber.
