An ultrasensitive ELISA method for the detection of procalcitonin based on magnetic beads and enzyme-antibody labeled gold nanoparticles

Tao Liao,Fang Yuan,Haoyang Yu,Zigang Li

Sepsis is a potentially fatal systemic body infection with a significant mortality rate worldwide. Procalcitonin (PCT) is a sensitive and specific biomarker for sepsis diagnosis, however, the traditional immune-gold labeling detection of PCT is semi-quantitative with relatively low sensitivity (>0.1 ng mL−1). Recently proposed techniques (such as electrochemical immunosensor, microarray, and total internal reflection based methods) require either a complex sensor or sophisticated equipment. Herein, we report a magnetic bead and enzyme-antibody labeled gold nanoparticle based ELISA strategy for sensitive PCT detection. This developed assay is sensitive enough to detect PCT at 20 pg mL−1, efficient for the direct human serum test. Notably, this assay specifically distinguishes PCT from other sepsis markers and the whole assay takes only 1.5 hours to finish.

Absorption-improving effects of chitosan oligomers based on their mucoadhesive properties: a comparative study on the oral and pulmonary delivery of calcitonin

Hailong Zhang,Xiaoyan Huang,Ya Sun,Jianfeng Xing,Akira Yamamoto,Yang Gao

Effects of chitosan oligomers with different types and varying concentrations on the intestinal and pulmonary absorptions of calcitonin were investigated in rats by an in situ closed loop method and an in vivo pulmonary absorption experiment, respectively. Various chitosan oligomers demonstrated different efficiencies in improving the intestinal and pulmonary absorptions of calcitonin, and chitosan hexamer with the optimal concentration of 0.5% (w/v) showed the greatest absorption enhancing effect. Moreover, pharmacodynamic parameters of calcitonin after its coadministration intrapulmonarily with various chitosan oligomers were consistently larger than that in the intestinal delivery, indicating the superior potential of pulmonary administration for systemic delivery of calcitonin. Furthermore, various chitosan oligomers neither obviously increased release amounts of protein nor activities of lactate dehydrogenase (LDH) in bronchoalveolar lavage fluid (BALF), revealing the safety of these chitosan oligomers to lung tissue. In addition, bioadhesions of various chitosan oligomers were well consistent with their absorption enhancing effects in the absorption experiment, suggesting the contribution of mucoadhesive properties of chitosan oligomers to their absorption improving effects. Taken together, chitosan oligomers, especially chitosan hexamer, can effectively improve the intestinal and pulmonary absorptions of calcitonin partly due to the mucoadhesion between positive chitosan oligomers and negative mucus in the membrane.

Controllable synthesis of sandwich-type magnetic beads for procalcitonin detection on a magnetic enzyme-linked immunosorbent assay

Liang Huang,Tian Tian,Ying Zhang,Yuting Liao,Jing Wang

Although magnetic beads have been widely utilized as a homogeneous carrier for immunoassays, the effectiveness of the immuno-response is often hindered by the physical and chemical characteristics of these beads, such as the challenge of achieving an optimal equilibrium among magnetic response, suspension performance and colloidal dispersibility. Herein, we present the fabrication of monodisperse polystyrene magnetic beads in a sandwich-like configuration and further explore their immunoassay capabilities as capturing substrates in the detection of procalcitonin (PCT) using the enzyme-linked immunosorbent assay (ELISA). The developed two-step magnetic particles assembly process effectively regulates the magnetic content, while the organic/inorganic double silica layer coating imparts desirable stability and dispersion to the assembly structure. Notably, the prepared magnetic beads exhibit high aqueous dispersity (PDI ∼ 0.2), rapid magnetic separation (25 s) and gradual half-sedimentation time (5 h). Leveraging the magnetic bead-based ELISA, we accomplish not only qualitative screening of PCT at an exceptionally low concentration by naked eye (cut-off value: 0.1 ng/mL) but also highly sensitive quantification of PCT using a microplate reader (limit of detection: 0.031 ng/mL). This investigation offers a solution for striking a harmonious balance among the diverse properties of magnetic beads and demonstrates the perspective of the magnetic capturing substrates in the field of immunoassays.

Preparation and characterization of an electrospun colon-specific delivery system for salmon calcitonin

Chen Li,Yun-shan Wei,Peng Wen,Kun Feng,Min-hua Zong,Hong Wu

A novel electrospun colon-specific delivery system for salmon calcitonin (SCT) was developed to improve its stability and bioavailability. Firstly, the pectin-coated SCT liposomes were prepared by film dispersion method and then a liposomes/sodium alginate/polyvinyl alcohol fiber mat was fabricated by electrospining. Scanning electron microscopy analysis indicated that the obtained nanofibers were uniform and smooth with an average diameter of about 350 nm. The release of SCT in different simulated digestive fluids was studied and corresponding release kinetics models were built. It was found that the fiber mat containing pectin-coated SCT liposomes had better stability and colon-specific properties compared with that containing uncoated SCT liposomes and the release of SCT in the colon followed the case II transport mechanism. In addition, there is no significant change in the bioactivity of released SCT measured by ELISA. This study shows that the electrospun colon-specific fiber mat is a potential delivery system for bioactive peptides.

Rapid Colorimetric Determination of Procalcitonin Using Magnetic Separation and Enzymatic Catalysis

Yun Zhang,Xiaohui Si,Miaomiao Zhang,Xue Yang,Huan Yuan,Xiangpeng Wang,Yang Zhang,Hui Wang

Human procalcitonin is an early diagnostic biomarker for sepsis and bacterial infections and can be used in distinguishing bacterial infections from viral infections. In this study, a colorimetric sensing platform for the rapid determination of procalcitonin was developed. The approach involves the capture of procalcitonin by immunomagnetic beads, and a detection antibody labeled with horseradish peroxidase to perform sandwich format, where it catalyzes the oxidation of 3,3′,5,5′–tetramethylbenzidine to produce the colorimetric signal. Under the optimal conditions, a detection limit of 0.04 ng/mL (3σ) was obtained within the calibration range 0.1–10 ng/mL. The proposed method was performed in less than 90 min and exhibited good specificity without interferences from other biomarkers including C-reactive protein and human serum albumin. Overall, the proposed method provided a new alternative strategy for procalcitonin detection due to its sensitive, rapid, specific, and simple characteristics. This method is suitable for rapid screening of various biomedical targets.

One arrow two eagles: Multifunctional nano-system for macrophage reprogramming and osteoclastogenesis inhibition against inflammatory osteolysis

Tong Sha,Ze Wang,Jinwei Li,Yahong Wu,Jinbiao Qiang,Zhenming Yang,Yue Hu,Kaijuan Zheng,Shuyu Zhang,Haizhu Sun,Andrew K. Whittaker,Bai Yang,Hongchen Sun,Quan Lin,Ce Shi

Inflammatory osteolysis poses a significant worldwide threat to public health. However, current monotherapies, which target either the prevention of the inflammatory response or the attenuation of osteoclast (OC) formation, have limited efficacy due to the complexity of the bone immune system being overlooked. Herein, by means of modifying salmon calcitonin (sCT), a multifunctional nano-system (AuNDs-sCT) was designed to synergistically inhibit OC differentiation and reverse the inflammatory microenvironment against inflammatory osteolysis. On the one hand, AuNDs-sCT effectively restrained OC differentiation by binding to the calcitonin receptors on the surface of OC precursors, resulting in the down-regulation of OC-specific genes and proteins. The targeted capacity of AuNDs-sCT provided a more durable and precise therapeutic effect. On the other hand, AuNDs-sCT exhibited antioxidant and anti-inflammatory effects, which regulated the polarization “switch” from the pro-inflammatory M1 phenotype to the anti-inflammatory M2 phenotype in macrophages by the inhibition of NF-κB p65 phosphorylation, thereby effectively reversed the local inflammatory microenvironment. Additionally, AuNDs-sCT served as a promising fluorescent probe, enabling real-time visualization of the therapeutic process. This capability is expected to optimize drug administration and evaluate therapeutic effects. In summary, by inhibiting OC differentiation and reprogramming macrophages, AuNDs-sCT successfully realized drug repurposing and achieved the “one arrow two eagles” therapeutic strategy, which offers a synergistic and effective treatment option for the clinical management of inflammatory osteolysis.

Rapid detection of procalcitonin using dual-color carbon quantum dots as fluorescent probes in FRET assay

Yuxin Tian,Yuwei Du,Zhang Chen,Li Li,Ruhong Yan,Xiaoqiang Li,Juan Yue

FRET, known as Förster Resonance Energy Transfer, serves as a versatile technique enabling precise quantitative assessment of protein concentrations with minimal disruption to biological processes. In this study, we have developed two types of fluorescent carbon quantum dots, B-CDs and G-CDs, serving as donor and acceptor probes, respectively, for FRET-based procalcitonin (PCT) detection. The B-CDs exhibit strong blue fluorescence (excitation/emission peaks at 360/442 nm) as donors, while the G-CDs display faint green fluorescence (excitation/emission peaks at 449/520 nm) as acceptors. These CDs are conjugated with PCT antibodies to form immune probes. Upon antigen binding, the antibody quickly binds and reacts with the antigen. Concurrently, the fluorescence energy from the donor is transferred to the acceptor, leading to a decrease in the blue fluorescence intensity emitted by B-CDs. The ratio of the fluorescence intensity between the green probe (G-CDs-Ab2) and the blue probe (B-CDs-Ab1) exhibited a positive correlation with the concentration of PCT antigen in the low concentration range of 0.5 to 5 ng/mL. Consistent results were observed in both phosphate-buffered saline (PBS) and diluted human serum. In the concentration range of 0.5–5 ng/mL in PBS buffer, the linear equation is expressed as F A /F D  = −0.0045 ×  c [PCT] ^2 + 0.0021 ×  c [PCT]  + 0.3232, with a limit of detection (LOD) of 0.1 ng/mL based on 3σ K −1 . For concentrations spanning from 5-80 ng/mL, the equation is F A /F D  = 0.0021 ×  c [PCT]  + 0.4212. These findings suggest that carbon dot-labeled immune probes could offer a novel approach for sensitive detection of disease biomarkers.

Mechanism of calcitonin gene related peptide against acute pancreatitis in rats by modulating amino acid metabolism based on metabonomics

Hu Jian-Xiong,Chen Ying-Kai,Chen Shi-Jun,Lin Yan-Ya,Chen Jun-Nian,Xie Ying,Zhao Cheng-Fei,Chen Cun-Rong

To study the mechanism of calcitonin gene related peptide(CGRP) protecting acute pancreatitis based on metabolomics. 24 adult male rats were randomly divided into control group (Con), acute pancreatitis model group (AP), CGRP treatment group (CGRP + AP, abbreviated as CGRP) and CGRP antagonist(CGRP(8–37)) pretreatment group (preCGRP(8–37) + AP, abbreviated as CGRP37), with 6 rats in each group. After different interventions, pancreases of rats in each group were collected for pathological analysis, and serum was collected for metabolomics analysis. Pathological examination of the pancreas suggested that the inflammation of pancreatitis in AP group was significant, the inflammation of pancreatitis in CGRP group was significantly reduced, and the pancreatitis in CGRP37 group was aggravated. Metabolomics of rat serum suggested that the differences in metabolites in each group were mainly related to amino acid metabolism, coenzyme/vitamin metabolism, carbohydrate metabolism, lipid metabolism, digestive system and other metabolic pathways. According to the trend of metabolite changes, we found 6 differential metabolites that were significantly correlated with CGRP intervention, including L-Valine, 5-Aminopentanoic acid, 4-oxo-L-proline, L-glutamine, L-proline, and Ornithine, all of which were related to amino acid metabolism. CGRP can effectively protect acute pancreatitis, possibly by regulating amino acid metabolism to alleviate acute pancreatitis.

Ratiometric electrochemical immunosensor for the detection of procalcitonin based on the ratios of SiO2-Fc–COOH–Au and UiO-66-TB complexes

Juncong Miao,Kang Du,Xuan Li,Xiaoting Xu,Xue Dong,Jinglong Fang,Wei Cao,Qin Wei

Procalcitonin (PCT) as a disease marker is of great significance in the early diagnosis of septicemia and pyemia. Biosensor have good prospects for analysis and detection of disease markers, but developing highly sensitive detection methods for detecting PCT remains a daunting task. In this paper, we develop a ratiometric electrochemical immunosensor with Au NPs modified SiO 2 -Fc-COOH complex as matrix and UiO-66 loaded with Toluidine blue (TB) as a marker for quantitative detection of procalcitonin (PCT). The SiO 2 modified by APTES not only has a large specific surface area but also contains abundant amino groups, which can be connected to the electrochemical probe of ferrocenecarboxylic acid (Fc-COOH). UiO-66 has the advantages of large specific surface area, high porosity and adjustable structure, which can adsorb toluidine blue electrochemical probe through electrostatic attraction . Measurement and analysis of the prepared immunosensor by Differential Pulse Voltammetry (DPV) show that the oxidation peak currents of Fc-COOH and TB appear at potentials of 0.30 V and −0.30 V respectively. As the PCT concentration increases, the oxidation peak current of TB increases and the oxidation peak current of Fc-COOH decreases. The ratios ( ΔI = ΔI TB /ΔI Fc-COOH ) between the double signals could show a certain linear relationship with the concentration of the PCT within a certain range. Under optimal conditions, the linear range of detection obtained by the immunosensor was 1 pg/mL-100 ng/mL and the detection limit was 0.3 pg/mL. In this work, the developed ratiometric electrochemical immunosensor not only provides a simple, reliable and sensitive strategy for quantitative detection of PCT but also provides a useful method for clinical detection of other disease markers.

Transdermal Delivery of Salmon Calcitonin Using a Dissolving Microneedle Array: Characterization, Stability, and In vivo Pharmacodynamics

Zhang Lu,Li Yingying,Wei Fang,Liu Hang,Wang Yushuai,Zhao Weiman,Dong Zhiyong,Ma Tao,Wang Qingqing

Salmon calcitonin (sCT) is a polypeptide drug, possessing the ability to inhibit osteoclast-mediated bone resorption. Just like other bioactive macromolecules, sCT is generally administered to the patients by either injection for poor compliance or through nasal spray for low bioavailability, which limits its use as therapeutic drugs. In the present study, to overcome the limitations of the conventional routes, two new dissolving microneedle arrays (DMNAs) based on transdermal sCT delivery systems were developed, namely sCT-DMNA-1 (sCT/Dex/K90E) and sCT-DMNA-2 (sCT/Dex-Tre/K90E) with the same dimension, meeting the requirements of suitable mechanical properties. An accurate and reliable method was established to determine the needle drug loading proportion in sCT-DMNAs. The stability study exhibited that the addition of trehalose could improve the stability of sCT in DMNA under high temperature and humidity. Further, in vivo pharmacodynamic study revealed that DMNA patch could significantly enhanced relative bioavailability to approximately 70%, and the addition of trehalose was found to be beneficial for sCT transdermal delivery. Therefore, sCT-DMNA is expected to replace traditional dosage form, providing a secure, efficient, and low-pain therapeutic strategy for bone disorders.

Design and evaluation of solid lipid nanoparticles modified with peptide ligand for oral delivery of protein drugs

Tingting Fan,Chunhui Chen,Han Guo,Juan Xu,Jian Zhang,Xi Zhu,Yang Yang,Zhou Zhou,Lian Li,Yuan Huang

Designing feasible and effective peptide ligand modified solid lipid nanoparticles (SLNs) to improve oral bioavailability of protein drugs and evaluating the influence of mucus remains important. In the present work, two kinds of peptide ligand modified SLNs loaded with salmon calcitonin (sCT), namely, sCT CSK-SLNs and sCT IRQ-SLNs, were prepared by coupling the peptide ligand CSKSSDYQC (CSK) which was reported to show affinity with goblet cells, or IRQRRRR (IRQ), a cell penetrating peptide, to polyoxyethylene (40) stearate (SA-PEG 2000 ). Compared with unmodified SLNs, CSK or IRQ modified SLNs with better drug protection ability could facilitate the internalization of drug on Caco-2/HT29-MTX co-cultured cells and permeation in excised rat duodenum mucosa. The internalization mechanism of two kinds of peptide ligand modified SLNs was mainly active transport via both clathrin- and caveolae-dependent endocytosis. Although mucus was an impediment to the transport of SLNs, the peptide ligand modified SLNs still showed improved drug absorption. The absolute bioavailability of sCT CSK-SLNs (12.41   ±   3.65%) and sCT IRQ-SLNs (10.05   ±   5.10%) raised to 2.45-fold and 1.98-fold compared with unmodified SLNs (5.07   ±   0.54%), implying the feasibility and effectiveness of CSK and IRQ peptide modification for the enhancement of the oral bioavailability of protein drugs. In summary, the nanoparticles modified with CSK or IRQ peptide ligand could be the potential carriers for the transport of protein drugs across intestinal barriers.

Composite Separable Microneedles for Transdermal Delivery and Controlled Release of Salmon Calcitonin for Osteoporosis Therapy

Yao Li,Xiao-Jie Ju,Han Fu,Chang-Hai Zhou,Yi Gao,Jun Wang,Rui Xie,Wei Wang,Zhuang Liu,Liang-Yin Chu

A composite separable microneedles (MNs) system consisting of silk fibroin (SF) needle tips and hyaluronic acid (HA) base is developed for transdermal delivery of salmon calcitonin (sCT) for therapy of osteoporosis. Poly(ethylene glycol) (PEG) is used to modulate the conformation structure of SF to achieve controllable sustained release of sCT. The prepared MNs can effectively penetrate the skin stratum corneum. After application to the skin, the HA base is dissolved within 2 min, allowing these SF drug depots to be implanted into the skin for controllable sustained release of sCT. The release kinetics of sCT can be controlled by regulating the conformation of SF with PEG and the interaction between sCT peptide and SF proteins. Compared with traditional needle injection, delivery of sCT using optimized HA-PEG/SF MNs shows better trabecular bone repair for ovariectomized-induced osteoporosis in mice. The proposed MNs system provides a new noninjection strategy for therapy of osteoporosis.
