Viral nervous necrosis (VNN) caused by nervous necrosis virus (NNV) is a major threat to the global aquaculture industry, resulting in huge economic losses. In this study, we evaluated a naturally anthraquinone, rhein, as a potential therapeutic agent against NNV. Rhein effectively inhibited NNV-induced cytopathic effects (CPE) at a concentration of 18.84 μM, with an inhibitory concentration (IC 50 ) against NNV replication of 8.77 μM. Moreover, the study revealed a possible anti-NNV mechanism. Rhein could disrupt NNV-induced S-phase cell cycle arrest by inhibiting ATP synthesis, and cell cycle arrested in S phase is favorable for NNV replication, which in turn inhibits NNV replication. The therapeutic effect of rhein was also evaluated in vivo , where the survival rate of NNV infected grouper was increased by 26.7% with the treatment of 10 mg/kg rhein. Our findings demonstrated the potential of rhein as a pharmacological compound that exerts significant antiviral activity against NNV by modulating the cell cycle and ATP synthesis. Consequently, rhein has shown significant promise as a medication for the prevention and treatment of viral infections in aquaculture.

Background Non-small-cell lung cancer (NSCLC) comprises about 85% of all lung cancers and is usually diagnosed at an advanced stage with poor prognosis. The IL-6/STAT3 signaling pathway plays a pivotal role in NSCLC biology. Rhein is a lipophilic anthraquinone extensively found in medicinal herbs. Emerging evidence suggests that Rhein has significant antitumor effects, supporting the potential uses of Rhein as an antitumor agent. Methods Cell viability and colony formation were performed to examine Rhein’s potent anti-proliferative effect in human NSCLC cell lines PC-9, H460 and A549. Flow cytometry-based assay was employed to study whether Rhein could affect cell apoptosis and cycle. The expression level of P-STAT3, apoptosis and cycle-related proteins Bcl-2, Bax, MDM2, CDC2, P53 and CyclinB1 were detected by Western blotting. The xenograft models were used to evaluate the in vivo effect of Rhein. Results We found that Rhein could significantly reduce the viability and stimulate apoptosis in human NSCLC cells in a dose-dependent manner. Western blot analysis results suggested that the antitumor effect of Rhein might be mediated via STAT3 inhibition. Rhein upregulated the expression of the proapoptotic protein Bax and downregulated the expression of the antiapoptotic protein Bcl-2. In addition, Rhein induced the arrest of NSCLC cells in the G2/M phase of the cell cycle and dose dependently inhibited the expression of cycle-related proteins. The Rhein also inhibited tumor growth in H460 xenograft models. Conclusion Rhein shows potent efficacy against NSCLC through inhibiting the STAT3 pathway. Our results also suggest that Rhein has a promising potential to be used as a novel antitumor agent for the treatment of NSCLC.

Reline, which is composed of choline chloride and urea in a molar ratio of 1:2, is the first and most extensively studied deep eutectic solvent (DES). In certain applications, reline is blended with organic solvents, dimethyl sulfoxide (DMSO) in most cases, to gain improved properties. Therefore, it is crucial to have a profound understanding of the impact of DMSO on the dynamics and structures of the species in the binary mixtures. In this study, neat reline and ten reline/DMSO mixtures, with DMSO molar fraction ranging from 0.1 to 0.95, were investigated primarily through a combined approach utilizing nuclear magnetic resonance (NMR) and Fourier transform infrared (FT-IR) spectroscopic techniques. Based on our investigation, we probed a significant transition of the binary mixtures from large molecule solutions or viscous liquids to nonviscous small-molecule solutions at a DMSO molar fraction of 0.7. Specifically, upon analyzing the self-diffusion coefficient, 1H T1 and 1H T2, we observed a notable increase in the molecular mobility of the species within the reline/DMSO system, particularly when the DMSO molar fraction exceeded 0.7. Drawing upon the FT-IR findings, we suggest that the enhanced molecular mobility, as evidenced by NMR analysis, is correlated with the disruption of molecular hydrogen-bonding interactions involving the −NH2 and −OH groups. Furthermore, based on 1D 1H, 1D 15N, and 2D 1H–1H COSY spectra, it was revealed that the interaction between urea and choline remains relatively stable until the DMSO fraction exceeds 0.7, whereupon it exhibited a notable weakening as the DMSO fraction increases from 0.7 to 0.95. In the meantime, DMSO molecules predominantly engage in hydrogen bond interactions with urea and choline when the DMSO molar fraction exceeds 0.7. Our results align well with previous molecular dynamics (MD) simulation studies and provide profound insights into the significant transition in the reline/DMSO mixture system.
