The performance of compound cold water soluble gel in petrifilm aerobic count plates was evaluated in terms of particle size, pH, transmittance, gel strength, water absorption, and water retention. Results demonstrated that when the ratio of sodium polyacrylate and guar gum is 2:8 in cold water soluble gel, the compound gel has 1.0% concentration, neutral pH, 446 g/cm 2 gel strength, 79.6% transmittance, 103.6 g/g water adsorption, and 14.4% water retention, which have no significant difference with 1.0% agar. After surfactants, somatomedin, glucose, and bean dregs were added, the sample solution was absorbed completely and colony diameter on petrifilm plates reached 0.95 mm, which is appropriate for counting. The petrifilm plate method based on prepared cold water soluble gel can achieve the same limit of detection (10° cfu/mL) and sensitivity (100%) in counting of five strains compared with the National Standards of Aerobe Plate Count Method. The petrifilm plate method also showed high accuracy and linearly dependent coefficient (R 2 = 0.9933). The accordance rate between the petrifilm plate method and the national standard agar plate count method reached as high as 90% in food detection. The compound cold water soluble gel can be used as a good petrifilm plate for aerobic bacterial count.

For the first time, we developed a novel nanocomposite of RGO decorated with α-Mn 2 O 3 nanorods (NRs) for inhibiting microbially influenced corrosion . RGO sheets with 2 nm thick were effectively synthesized by the modified Hummers technique and then a hydrothermal reduction procedure. The fabricated RGO/α-Mn 2 O 3 nanocomposite exhibited controlled morphology, geometry, surface exposure, and orientation. Single crystal α-Mn 2 O 3 NRs with mean diameter of 50 nm and length of 1 μm were produced via the hydrothermal procedure to yield γ-manganite which was calcined at 700 °C. Stable RGO/α-Mn 2 O 3 nanocomposite was successfully prepared via an efficient two-phase ultrasonication approach. The structures of the developed materials and nanocomposite were detected via different characterization techniques. Cost-effectiveness, long-term antimicrobial activity, and biocorrosion inhibition are the major advantages of the produced RGO/α-Mn 2 O 3 nanocomposite. The nanocomposite's biocorrosion-protection was influenced by its high surface area and controlled size and morphology. Its antimicrobial mechanism against different anaerobic and aerobic microbial strains was deeply highlighted. Field emission SEM images showed that the blade-like RGO/α-Mn 2 O 3 nanocomposite cruelly attached into the microbial cell walls forming morphology disruption, cell wrapping, and microbial killing. This nanocomposite exhibited ultrahigh potency against the anaerobic sulfate-reducing bacteria (SRB) strain Desulfovibrio halophilus SL 890 at 150 mg/L. Its highest potency against most of the tested aerobic strains was achieved at 2 μg/mL for Kocuria rhizophila EMCC 1246, Bacillus subtilis EMCCN 1152, Pseudomonas aeruginosa NRRL B-800, Staphylococcus aureus ATCC 25923, Aspergillus brasiliensis ATCC 9642 , and Candida albicans ATCC 24433. The designed RGO/α-Mn 2 O 3 nanocomposite provide a new platform for microbial growth inhibition.

Introduction: In an effort to minimize the usage of fishmeal in aquaculture, novel protein diets, including Tenebrio molitor, cottonseed protein concentrate, Clostridium autoethanogenum, and Chlorella vulgaris were evaluated for their potential to replace fishmeal. Nevertheless, comprehensive examinations on the gut health of aquatic animals under an alternate feeding strategy when fed novel protein diets are vacant. Methods: Five isonitrogenous and isolipidic diets containing various proteins were manufactured, with a diet consisting of whole fishmeal serving as the control and diets containing novel proteins serving as the experimental diets. Largemouth bass (Micropterus salmoides) with an initial body weight of 4.73 ± 0.04g employed as an experimental animal and given these five diets for the first 29 days followed by a fishmeal diet for the next 29 days. Results: The results of this study demonstrated that the growth performance of novel protein diets in the second stage was better than in the first stage, even though only the C. vulgaris diet increased antioxidant capacity and the cottonseed protein concentrate diet decreased it. Concerning the intestinal barriers, the C. autoethanogenum diet lowered intestinal permeability and plasma IL-1β/TNF-α. In addition, the contents of intestinal immunological factors, namely LYS and sIgA-like, were greater in C. vulgaris than in fishmeal. From the data analysis of microbiome and metabolome, the levels of short chain fatty acids (SCFAs), anaerobic bacteria, Lactococcus, and Firmicutes were significantly higher in the C. autoethanogenum diet than in the whole fishmeal diet, while the abundance of Pseudomonas, aerobic bacteria, Streptococcus, and Proteobacteria was lowest. However, no extremely large differences in microbiota or short chain fatty acids were observed between the other novel protein diets and the whole fishmeal diet. In addition, the microbiota were strongly connected with intestinal SCFAs, lipase activity, and tight junctions, as shown by the Mantel test and Pearson’s correlation.Discussion: Taken together, according to Z-score, the ranking of advantageous functions among these protein diets was C. autoethanogenum diet > C. vulgaris diet > whole fishmeal diet > cottonseed protein concentrate > T. molitor diet. This study provides comprehensive data illustrating a mixed blessing effect of novel protein diets on the gut health of juvenile largemouth bass under an alternate feeding strategy.
