Fermentation technology presents promising opportunities for food waste valorization. Pomegranate peel ( PP ), a food by-product, has potential applications in fermented feed. This study examined the effects of a 6% dry PP additive on the ensiling characteristics, antioxidant activity, metabolites, bacterial community, and in vitro ruminal fermentation, methane (CH 4 ) emission of corn silage ensiled for 7 days and 60 days using microbiome and metabolome analyses. PP-treated silage inhibited ( P < 0.05) protein degradation by reducing ammonia nitrogen and non-protein nitrogen concentrations during ensiling. The PP additive increased ( P < 0.05) water-soluble carbohydrate content and reduced ethanol production in corn silage. Lactiplantibacillus dominated PP-treated silage at the initial ensiling stage, while Levilactobacillus prevailed at the final stage . Notably, the PP additive exhibited strong antioxidant activity by modulating antioxidant enzymes and flavonoid biosynthesis mediated by key metabolites (epigallocatechin and catechin). Correlation analysis identified Lactiplantibacillus , Citrobacter , Phytobacter and Burkholderia as key microbes in the production of antioxidant metabolites and enzymes in PP-treated silage. Additionally, PP supplementation reduced ( P < 0.05) in vitro ruminal CH 4 and nitrogen losses, while decreasing dry matter (DM) digestibility in corn silage. In summary, PP-treated corn silage enhanced antioxidant properties and reduced the nitrogen losses and in vitro ruminal CH 4 emissions, but lowered DM digestibility. Thus, PP can be recommended as a silage additive, though the dry PP level should be lower than that used in this study.

Broussonetia papyriferaL. (paper mulberry) is an alternative woody plant, which can used to replace part of the protein feed for ruminants. Ensiling is an effective way to preserve fresh pasture and to solve the problem of stable storage and feed conversion of paper mulberry in the rapid growth period. However, low dry matter (DM), water-soluble carbohydrate, and lactic acid bacteria (LAB) reduce the quality of paper mulberry silage. This study assesses the influence of wilting time (0 h and 3.5 h; lighting: 3.43 × 104Lux) and three additives (Enterococcus durans, CL; cellulase, CE; and formic acid, FA) on the fermentation quality, aerobic stability, and bacterial community of whole plantB. papyriferasilage. The whole plantB. papyriferasample was mowed and wilted for 0 h and 3.5 h, and then had CL, CE, or FA added, followed by 60 days of ensiling. The results show all silage samples had high fermentation quality with pH below 4.2, ammonia-nitrogen below 100 g/kg DM, and no detectable butyric acid. The additives protected the DM and the crude protein from protease activity (p< 0.05), and CL was the most effective among them. Furthermore, wilting time influenced the silage’s bacterial communities, but overall, CL treatment had the greatest impact on bacterial communities. Wilting time and formic acid treatment significantly improved aerobic stability (p< 0.05).Enterococcuswas positively correlated with lactic acid (LA), while negatively correlated with LA andWeissella(p< 0.001). Enterococcuswas identified as the main driver of the whole plant paper mulberry ensiling process in the present study. In conclusion, compared to other additives, LAB is the most effective and economical to improve the fermentation quality and reduce the protein degradation of whole plant paper mulberry silage. Our findings provide a theoretical basis to improve the quality and production of paper mulberry silage. Keywords:paper mulberry;silage;wilting;additives;microbial community;aerobic stability

In this study, the influence of two lactic acid bacteria (LAB) strains [Lactobacillus rhamnosusBDy (LR-BDy) andLactobacillus buchneriTSy (LB-TSy)] selected from Southwest China on the fermentation characteristics and in vitro gas production ofBroussonetia papyrifera(paper mulberry) silage were experimentally explored. The experimental groups were a control group (C), an LB-TSy treatment (LB), an LR-BDy treatment (LR), and an LR-BDy + LB-TSy hybrid group (LR × LB). After the LAB were added, the pH value of paper mulberry silage significantly declined (p< 0.05), and the crude protein content was effectively preserved (p< 0.05). However, no significant changes were found in the levels of neutral detergent fiber, acid detergent fiber, and crude ash (p> 0.05). The lactic acid content in paper mulberry silage was evidently increased (p< 0.05). The in vitro gas production in the LR at 36, 48, and 72 h were markedly higher than that in the other treatments (p< 0.05). Owing to the addition of LAB, the microbial diversity in paper mulberry silage was reduced, while the relative bacterial abundance ofLactobacilluswas enhanced. Hence, the addition of LAB selected from the warm and humid region in Southwest China can improve the quality of paper mulberry silage and elevate its feeding value in this region. Keywords:paper mulberry;silage;in vitro gas production;microbial community;lactic acid bacteria

The aim of this study was to investigate the influence of Perilla frutescens, alone or in combination with Lactobacillus plantarum a214 or citric acid, on forage oat silage quality, bacterial and fungal microbiological profile during ensiling and aerobic exposure. With the exception of Perilla frutescens, all additives could improve silage quality of forage oat based on lower ammonia-nitrogen content and higher residual of water soluble carbohydrates during anaerobic fermentation compared to control silage, especially in Perilla frutescens combined with citric acid (CAPF). Lactobacillus was the dominant bacteria in all silages, while CAPF group increased the relative abundance of Lactobacillus lindneri and Lactobacillus brevis compared to control silage. The application of Perilla frutescens suppressed the relative abundance of yeasts such as Pichia fermentans and Wickerhamomyces anomalus in response to aerobic exposure, especially in CAPF treatment, leading to high acetic acids and lower dry matter loss, as well as good aerobic stability. Therefore, Perilla frutescens, alone or in combination with citric acid, has potential to improve aerobic stability of forage oat silage by shifting bacterial and fungal community composition, and can be used as new additive to prepare high-quality silage for animal production.

AimsThis study aimed to evaluate the feasibility of sorbic acid (SA) as a silage additive and its effects on fermentation quality and aerobic stability of high dry matter (DM) silage. Methods and ResultsHigh DM rice straw was ensiled with distilled water (C), 1 × 106 CFU per gram fresh weight (FW) Lactobacillus plantarum and 1 × 106 CFU per gram FW Lactobacillus buchneri (LP+LB) or SA for 45 days with a subsequent aerobic stability test. After ensiling, LP+LB silage had the highest lactic acid (LA) content and the lowest pH value, whereas SA silage had the highest DM and water‐soluble carbohydrate (WSC) contents, and the lowest ethanol and ammonia nitrogen (NH3‐N) contents among all silages (P < 0·001). Compared to C silage, SA significantly (P < 0·01) reduced the counts of yeasts but not lactic acid bacteria (LAB). During 6‐day aerobic exposure, the continuous pH increase and LA decrease were observed in C and LP+LB silages, and there was no significant change in pH, DM, NH3‐N and WSC contents of SA silage over the whole aerobic exposure. The SA addition slowed the decline of LA and acetic acid (AA) contents as well as the growth of yeasts and aerobic bacteria under aerobic exposure.ConclusionIn this study, L. buchneri could not function in high DM rice straw silage while SA effectively improved both the fermentation quality and aerobic stability.Significance and Impact of the StudyThe SA was more effective than dual‐purpose inoculants to improve the aerobic stability of high DM rice straw silage. Thus, SA can be served as a potential antifungal additive for silage with high DM.

The microbiota in forage silage plays a pivotal role in determining the fermentation quality. Identifying effective microbial additives is essential to help forage producers refine their search for functional inoculants and to support farmers in adopting them for practical ensiling. This study investigated microbial and metabolomic dynamics in king grass silages treated with six commercial inoculants dominated byEnterococcus faecium-like species,Bacillus velezensis, andLactobacillus paraplantarum. The fermentation characteristics, viable microbial diversity, and metabolite profiles were compared between treated and untreated silages using 16S rDNA sequencing and metabolic profiling via LC-QTOF-MS, integrated with multi-omics correlation analyses. Additive-treated silages showed improved fermentation quality, simplified bacterial correlation networks, and distinct microbial successions and interactions. A total of 1523 metabolites were detected, with 56–84 significantly altered in each treated group compared to the control. Metabolites with antimicrobial, antioxidant, and cholesterol-lowering activities were more abundant in treated silages, especially organic acids, amino acids, and short-chain fatty acids. Inoculants distinctly influenced amino acid, energy, nucleotide, and vitamin metabolism during ensiling. This study advances our understanding of how commercial microbial additives reshape the bacterial community structure and function in silages and highlights promising lactic acid bacteria species contributing to silage quality through the production of bio-functional metabolites.

In plant cell wall, ferulic acid (FA) andp-coumaric acid (pCA) are commonly linked with arabinoxylans and lignin through ester and ether bonds. These linkages were deemed to hinder the access of rumen microbes to cell wall polysaccharides. The attachment of rumen microbes to plant cell wall was believed to have profound effects on the rate and the extent of forage digestion in rumen. The objective of this study was to evaluate the effect of bound phenolic acid content and their composition in corn silages on the nutrient degradability, and the composition of the attached bacteria. Following an in situ rumen degradation method, eight representative corn silages with different FA andpCA contents were placed into nylon bags and incubated in the rumens of three matured lactating Holstein cows for 0, 6, 12, 24, 36, 48, and 72 h, respectively. Corn silage digestibility was assessed by in situ degradation methods. As a result, the effective degradability of dry matter, neutral detergent fibre, and acid detergent fibre were negatively related to the ether-linked FA andpCA, and their ratio in corn silages, suggesting that not only the content and but also the composition of phenolic acids significantly affected the degradation characteristics of corn silages. After 24 h rumen fermentation, Firmicutes, Actinobacteria, and Bacteroidota were observed as the dominant phyla in the bacterial communities attached to the corn silages. After 72 h rumen fermentation, the rumen degradation of ester-linked FA was much greater than that of ester-linkedpCA. The correlation analysis noted thatErysipelotrichaceae_UCG-002,Olsenella,Ruminococcus_gauvreauii_group,Acetitomaculum, andBifidobacteriumwere negatively related to the initial ether-linked FA content whilePrevotellawas positively related to the ether-linked FA content and the ratio ofpCA to FA. In summary, the present results suggested that the content of ether-linked phenolic acids in plant cell walls exhibited a more profound effect on the pattern of microbial colonization than the fibre content. Keywords:corn silage;rumen-attached bacteria;phenolic acids;in situ degradation

Simple SummaryThe leguminous forageChamaecrista rotundifoliacontains a high crude protein content and can be an important feed source for livestock. Forage is preserved mainly as silage, and microbial fermentation is key to high-quality silage. The fermentation quality of silage depends on the microbial community structure and the metabolites produced during silage fermentation. The current study provides evidence that the addition of malic or citric acid and/orLactobacillus acidophilus(L) improves the quality of silage and inhibits the growth of harmful microorganisms.AbstractThis study aimed to investigate the effects of citric acid, malic acid, andLactobacillus acidophilus(L) on fermentation parameters and the microbial community of leguminousChamaecrista rotundifoliasilage. FreshC. rotundifoliawas treated without any additive (CK), or with L (106CFU/g fresh weight), different levels (0.1, 0.3, 0.5, and 1% fresh weight) of organic acid (malic or citric acid), and the combinations of L and the different levels of organic acids for 30, 45, and 60 days of ensiling. The effects of malic acid and citric acid were similar during the ensiling process. Treatment with either citric or malic acid and also when combined with L inhibited crude protein degradation, lowered pH and ammonia nitrogen, and increased lactic acid concentration and dry matter content (p< 0.05). The neutral detergent fiber and acid detergent fiber increased initially and then decreased with fermentation time in all treatments (p< 0.05). Increasing the level of organic acid positively affected the chemical composition ofC. rotundifoliasilage. In addition, the addition of 1% organic acid increased the relative abundance ofLactobacillus, while the relative abundances ofClostridiumandEnterobacterdecreased at 60 days (p< 0.05). Moreover, both organic acids and combined additives increased (p< 0.05) the relative abundance ofCyanobacteriaat 60 days of fermentation. We concluded that adding malic acid, citric acid, and L combined with an organic acid could improve the quality ofC. rotundifoliasilage and increase the relative abundance of beneficial bacteria. The addition of organic acid at a level of 1% was the most effective. Keywords:citric acid;Chamaecrista rotundifoliasilage;Lactobacillus acidophilus;malic acid;microbial diversity

The aim of the present study was to characterize the fermentation properties of Lactobacillus rhamnosus GG (LGG; ATCC 53103) in vitro, and investigate the effects of inoculation with LGG on fermentation quality of whole plant corn silage (WPCS) and fermented total mixed ration (FTMR). The antifungal activity of LGG against 4 prevalent fungi isolated from corn stalk was determined using the Oxford cup method. The binding capacity of LGG to aflatoxin B 1 , zearalenone and deoxynivalenol was examined by in vitro adsorption experiments using a batch mode in phosphate buffered saline. The antioxidant activity of LGG in vitro was evaluated by determining the resistance to hydrogen peroxide, free radical scavenging capacity and reducing activity. Three batches of whole plant corn and total mixed ration were collected separately. Each batch of raw material was divided into equal portions and applied to 2 treatments: (1) control with distiller water (CON), and (2) treatment inoculated with LGG (LGG). After 48 d of ensiling, the chemical composition, ruminal degradability, microbial community and mycotoxin concentrations of WPCS and FTMR were determined. The results showed that lactic acid was the predominant component among organic acids in LGG culture process, with a rapid generated occurring at 4 h after inoculation and reaching a stable state at approximately 48 h. Lactobacillus rhamnosus GG could effectively inhibit the growth of 4 prevalent fungi isolated from corn stalk, the diameter of inhibition zone was higher for Mucor circinelloides than that of the other moulds ( P = 0.016). With increasing of LGG cell amount, the binding capacity to 3 mycotoxins increased ( P < 0.01); the heat and acid treated LGG also enhanced the binding capacity to mycotoxins ( P < 0.01). Lactobacillus rhamnosus GG displayed tolerance to hydrogen peroxide (0.8 mmol/L) for 8 h at 37 °C (96%, viability), but the viability decreased sharply (52.39%, viability) under the concentration of H 2 O 2 increased to 1.2 mmol/L ( P < 0.001). The free radical scavenging activity and reducing activity of viable LGG were greatest, followed by cell-free extracts and inactivated cells ( P < 0.001). Ensiling of WPCS and FTMR inoculated with LGG decreased dry matter loss ( P < 0.05), increased lactic acid and acetic acid production ( P < 0.05), enhanced some nutrients ruminal degradability ( P < 0.05). Meanwhile, the inoculation of LGG reduced the counts of yeast, mold and coliform bacteria ( P < 0.05) and, more importantly, alleviated mycotoxin contamination ( P < 0.05). These results indicated that LGG could be a novel inoculant to improve the quality and safety of WPCS and FTMR.

In this study, a novel beta-cypermethrin (beta-cyp)-degrading strain Lactobacillus pentosus 3–27 (LP3–27) was screened from beta-cyp-contaminated silage. The strain could degrade 96% of beta-cyp (50 mg/L) in MSM medium after 4 d of culture, while the strain lost its degradation ability when the beta-cyp concentration reached 250 mg/L. The effects of LP 3–27 on fermentation, bacterial community, and bioremediation of contaminated alfalfa silage at two dry matter (DM) contents were studied. The results showed that inoculation with LP3–27 not only degraded beta-cyp, but also improved the fermentation quality of alfalfa silage after 60 d of ensiling. Meanwhile, L. pentosus dominated the bacterial community during ensiling in LP3–27 inoculated silages, whereas Pediococcus acidilactici was the dominant species in the control silage. LP3–27 inoculation also simplified the bacterial interaction networks of ensiled alfalfa. Beta-cyp degradation was positively correlated with L. pentosus in LP- inoculated silages, which confirmed the function of beta-cyp degradation by L. pentosus . In addition, higher beta-cyp degradation was observed in silage with 35% versus 43% DM. In summary, strain LP3–27 could be used as a candidate inoculum for bioremediation of beta-cyp-contaminated silage and to produce safe silage for animal production.

For the first time, Lactobacillus plantarum strains carrying heterologous genes encoding multifunctional glycoside hydrolases were constructed and used as additives for alfalfa silage. The chemical characteristics, nonstructural carbohydrate composition, and fermentation quality of alfalfa silage were examined. The supernatant of L. plantarum expressing CbXyn10C and Bgxg1 (LP11AG) showed activities on xylan, Avicel, and carboxymethylcellulose (CMC), while the supernatant of the wild-type L. plantarum showed no activity. When LP11AG was used as silage additive, the water-soluble carbohydrate content of alfalfa silage increased by 72%, 55%, and 155% compared with control when the silage was stored at 20 °C, 30 °C, and 40 °C, respectively. With LP11AG being used as an additive for the alfalfa silage stored at 20 °C, the hemicellulose, cellulose, and acid detergent ligninin (ADL) contents decreased by 17%, 6%, and 14% compared with the control ( p < 0.05), respectively. Compared with the corresponding original contents, the contents of glucose, arabinose, galactose, and fructose detected in silage treated with LP11AG after 45 days of ensiling increased by 55%, 1494%, 68%, and 5% , respectively, when stored at 40 °C. Raffinose and stachyose, originally present in alfalfa, disappeared after ensiling. In conclusion, our results suggest that LP11AG provides a substantial benefit as a silage additive.

Oregano essential oil (OEO), primarily composed of carvacrol, serves as a significant bactericide and represents a promising alternative to antibiotics for use as feed additives worldwide. However, the extraction of OEO generates substantial solid residues that require proper treatment. We found that residues obtained after the distillation of high-yield carvacrol-type OEO (RDCO) still contain numerous active compounds, including non-volatile components such as flavonoids and phenolic acids, along with volatile compounds like thymoquinone and carvacrol. This study demonstrates that incorporating RDCO into Medicago sativa L. silage fermentation optimizes the microbial community by suppressing undesirable microorganisms (i.e., 18.17-fold reduction in Pseudomonas with 6 % RDCO vs . control) while promoting beneficial probiotics (i.e., 4.26-fold enrichment of Lactobacillus ). Also, 6 % RDCO addition significantly increased lactic acid production by 31 % vs . control. Furthermore, treated silages exhibited elevated levels of volatile and non-volatile secondary metabolites alongside enhanced fermentation quality. Metaproteomic analysis revealed that RDCO promoted lactic acid production by enhancing bacterial glycolysis/gluconeogenesis, pyruvate metabolism, and TCA cycle. This study establishes a scientific foundation for the sustainable utilization of plant essential oil extraction residues and explores new avenues for high-value applications of these industrial by-products, thus emphasizing the valorization and recycling of the waste residues.
