Accurate imaging of soft tissues is one of the ultimate goals in biomedical imaging. Different imaging modalities can improve their disadvantages, and promote the imaging ability. However, once an imaging agent has been prepared, it is usually hard to adjust it according to the actual needs. Herein, we developed a supramolecular brush polymer (SBP) as a versatile imaging agent platform. The SBP platform (SBPP) is constructed by the intermolecular inclusion complexation of bridged tris(β-cyclodextrin) (1) with Mn(III)-porphyrin-bearing poly(ethylene glycol) (PEG) side chains (Mn(III)-TPP), and can further bind other functional groups by host–guest interactions of cyclodextrin and adamantine. The SBPP is characterized by UV/vis absorption spectroscopy, NMR, dynamic light scattering (DLS), atomic force microscopy (AFM) and transmission electron microscopy (TEM). We demonstrated that this SBPP not only has no cellular toxicity against NIH 3T3 cells in in vitro cell experiments, but it also shows an efficient enhanced T1 relaxivity in in vitro MR imaging experiments. When used as multifunctional imaging agents, different imaging probes and/or targeting agents can be introduced to this SBPP as needed through simple host–guest interactions. In in vitro imaging experiments, it shows accurate imaging of different kinds of cancer cells by choosing on-demand targeting agents. These results suggest a promising strategy for engineering multifunctional imaging agents with SBPs.

Efficient delivery of anticancer drugs to increase the intracellular drug concentration in targeted tissues is urgently needed in cancer chemotherapy, since clinical drugs usually have serious side effects on normal tissues. In this paper, we developed a new method to fabricate multi-functional porous magnetic nano-cargos by grafting PEG/folate onto the surface and storing doxorubicin in the pores of magnetic supraparticles for accurate delivery of anticancer drugs. The anticancer drug doxorubicin was fixed into the porous magnetic cores with acid-sensitive linkers, which can be broken in acidic intracellular environments or organelles; the superficial PEG chains on the magnetic cores not only enhanced the dispersion stability of the nano-cargos but also immobilized folate modified α-cyclodextrin by inclusion complexation, and the α-cyclodextrin derivatives could be flexibly replaced as needed. Remarkable proliferation inhibition of cancer cells and minor side effects on normal cells were achieved due to the controlled drug release manner of the nano-drug system, indicating that this kind of nano-cargo has great potential in cancer chemotherapy for personalized and accurate treatment.

A synthetic carrier of small interfering RNA (siRNA) with customizable size, pH-responsive degradability and optimized surface population of cell-targeting agents is constructed precisely through a series of interlocked thermodynamically self-regulated processes. This system consists of a unimolecular polyplex core formed from each individual molecule of networked cationic polymer and a multi-functional shell assembled from a rationally designed triblock copolymer. The core-forming polymer of defined size is synthesized via Zeta potential-regulated condensation of branched and linear multi-amine bearing oligomers through pH-responding imidazole conjugated imine linkages. The shell-forming copolymer consists of a multi-carboxyl saccharide end block to guide to the surface assembly, a hydrophobic central block to form an encapsulating layer, and a poly(ethylene glycol) end block equipped with a highly selective active ester for “hooking” various cell-targeting agents. Animal assays confirm the flexibility and convenience of this system to equip with selected functional components for systemic delivery of siRNA.

Molecular imaging is a powerful tool for non-invasive visualization of tumors that plays an important role in their diagnosis and treatment. The specificity of molecular imaging probes for cancer cells is important for accurate tumor visualization, with antibody and polypeptide nanoprobe conjugates having often been used as targeting agents for tumor detection. However, many traditional chemical conjugation methods employ complex conjugation reactions that result in poor efficiency and poor bioactivity. Herein, we describe the use of photo click methodology for the rapid synthesis of nanoprobes comprised of silica nanoparticles functionalized with RGD targeting units (SiO2@T1-RGDk NPs) (∼80 nm) for in vivo prostate cancer fluorescent imaging applications. These SiO2@T1-RGDk NPs exhibit a maximum absorption wavelength of 380 nm in their UV absorption spectra with a maximum fluorescence emission wavelength of 550 nm. Confocal immunofluorescent imaging reveal that SiO2@T1-RGDk NPs exhibit excellent targeting ability for visualizing cancer cells, with in vivo fluorescence imaging intensity in a subcutaneous tumor model of prostate cancer reaching a maxima after 4 h. Biosafety assessments showed that SiO2@T1-RGDk NPs demonstrate no obvious toxicity in mice, thus demonstrated that these novel NPs may prove to be promising fluorescent imaging agents for the accurate detection and treatment of tumors.

With the gradual deep understanding of the tumorigenesis and development process, nanodrug are thought to have great prospects for individualized treatment of tumors. To deliver adequate concentration of active ingredients to targeted tissues, proteins are usually used as carriers to avoid clearance by the immune system. Herein, a new strategy is developed for preparation of the protein-functionalized targeting nanodrugs; different kinds of proteins (albumin, horseradish, transferrin, and ricin) can be quickly loaded in polyacrylic acid nanohydrogels (PAA-NGs) without discrimination within 1 min under the strong driving force of entropy; and the loading efficiency can reach 99% with about 50% loading content. Meanwhile, the activity of the released protein can be well retained. After oriented binding of the targeting agent on the surface of the nanocarriers by a unique and facile technique, the protein-loaded nanodrug exhibits excellent tumor cell uptake and targeting effect. The excellent targeting ability from the oriented binding is further proved by comparing with the non-oriented targeting system. With quick loading of the anti-tumor protein of ricin and oriented binding of transferrin protein (Tf), the targeting nanodrug (PAA-BB@Ricin/Tf) shows a remarkable anti-tumor effect. This study proves a new universal delivery and targeting strategy for improving the nanodelivery system, which has great potentials for clinical application.

The major challenges of immunotherapy for glioblastoma are that drugs cannot target tumor sites accurately and properly activate complex immune responses. Herein, we design and prepare a kind of chemotactic nanomotor loaded with brain endothelial cell targeting agent angiopep-2 and anti-tumor drug (Lonidamine modified with mitochondrial targeting agent triphenylphosphine, TLND). Reactive oxygen species and inducible nitric oxide synthase (ROS/iNOS), which are specifically highly expressed in glioblastoma microenvironment, are used as chemoattractants to induce the chemotactic behavior of the nanomotors. We propose a precise targeting strategy of brain endothelial cells-tumor cells-mitochondria. Results verified that the released NO and TLND can regulate the immune circulation through multiple steps to enhance the effect of immunotherapy, including triggering the immunogenic cell death of tumor, inducing dendritic cells to mature, promoting cytotoxic T cells infiltration, and regulating tumor microenvironment. Moreover, this treatment strategy can form an effective immune memory effect to prevent tumor metastasis and recurrence.
