Herein, reduction-responsive disintegratable nanoclusters (NCs) were prepared as a novel nanovehicle for targeted drug delivery. The NCs, with a diameter of ∼170 nm, were self-assembled from hydrophobically modified and iRGD decorated hydroxyethyl starch (iRGD-HES-SS-C18). DOX was loaded into the NCs as a model drug. DOX@iRGD-HES-SS-C18 NCs can disintegrate into smaller ones and release DOX under reduction stimuli. Due to the ligand–receptor binding interactions between iRGD and integrin αV, DOX@iRGD-HES-SS-C18 NCs can specifically bind to the cell membranes of HepG-2 and 4T1 cells (integrin αV positive), resulting in enhanced cellular uptake as compared to DOX@HES-SS-C18 NCs. After cellular internalization, the NCs were transported to endosomes/lysosomes in which the reductive environment triggered the disintegration and DOX release. As a consequence, DOX@iRGD-HES-SS-C18 NCs exhibited an enhanced antitumor effect as compared to DOX@HES-SS-C18 NCs and free DOX, in an in vitro antitumor activity study. The reduction-responsive disintegratable NCs reported here were proved to be a safe and efficient nanoplatform, holding significant translation potential for tumor-targeted drug delivery.

Superparamagnetic iron oxide nanoparticles are of great interest in magnetic targeted drug delivery due to their unique properties. In this paper, size-controlled superparamagnetic iron oxide nanoparticles were synthesized in an ethylene glycol/diethylene glycol (EG/DEG) binary solvent system via a facile solvothermal method. X-ray diffraction (XRD), a scanning electron microscope (SEM), and a vibrating sample magnetometer (VSM) were used to confirm that the prepared samples were superparamagneticFe3O4nanospheres. When theVEG/VDEGwas varied from 100/0 to 80/20, 60/40, and 40/60, the average diameters of the resultingFe3O4nanospheres were approximately 700, 500, 300, and 100 nm, respectively. In addition, the saturation magnetization (Ms) ofFe3O4nanoparticles with a size of 100, 300, 500, and 700 nm was 72.14, 75.94, 80.28, and 85.41 emu/g, and the corresponding remanent magnetization (Mr) was 3.34, 3.97, 3.26, and 4.28 emu/g, respectively. The relevant formation mechanisms ofFe3O4nanoparticles are proposed at the end. These superparamagneticFe3O4nanoparticles with high saturation magnetization may have use as targeted drug carriers. Keywords:targeted drug delivery;superparamagnetic;size-controlled;solvothermal method

A novel micelle based on heparosan and deoxycholic acid (DOCA) conjugate (HD) as drug carrier was reported here. As the surface was negatively charged, this micelle could resist serum adsorption, showing favorable stability. Moreover, fluorescence observation confirmed that it was able to deliver model hydrophobic drug doxorubicin (DOX) into HeLa cells efficiently. The DOX-loaded micelles showed sustained release behavior at pH 7.4, and accelerated release behavior at pH 5.0 or in the presence of β-glucuronidase, which over-expressed in tumor cells. In vitro cytotoxicity assay demonstrated that the half-maximal inhibitory concentration (IC50) of DOX-loaded micelles against HeLa cells was much lower than that of COS7 cells, showing significant therapeutic distinction between tumor cells and normal cells. Combining with the good biocompatibility and biodegradability of heparosan, this micelle may be promising in clinical application for targeted drug delivery.

A novel quantum dots-based multifunctional nanovehicle (DOX-QD-PEG-FA) was designed for targeted drug delivery, fluorescent imaging, tracking, and cancer therapy, in which the GSH–CdTe quantum dots play a key role in imaging and drug delivery. To exert curative effects, the antineoplastic drug doxorubicin hydrochloride (DOX) was loaded on the GSH–CdTe quantum dots through a condensation reaction. Meanwhile, a polyethylene glycol (PEG) shell was introduced to wrap the DOX-QD, thus stabilizing the structure and preventing clearance and drug release during systemic circulation. To actively target cancer cells and prevent the nanovehicles from being absorbed by normal cells, the nanoparticles were further decorated with folic acid (FA), allowing them to target HeLa cells that express the FA receptor. The multifunctional DOX-QD-PEG-FA conjugates were simply prepared using the 'one pot' method. In vitro study demonstrated that this simple, multifunctional nanovehicle can deliver DOX to the targeted cancer cells and localize the nanoparticles. After reaching the tumor cells, the FA on the DOX-QD-PEG surface allowed folate receptor recognition and increased the drug concentration to realize a higher curative effect. This novel, multifunctional DOX-QD-PEG-FA system shows great potential for tumor imaging, targeting, and therapy.

Liver‑targeted drug delivery improves the efficacy of anti‑liver cancer agents and reduces systemic toxicity by limiting the bioavailability of these drugs to within tumors. Liver targeting reagents with galactose residues, which selectively combine to asialoglyco protein receptors, have previously been used to improve liposome‑encapsulated drug accumulation within liver cells. They lead to a reduction in liver cancer cell growth and have been used to cure certain hepatic diseases. In the present study, curcumol, which is the primary active component of Chinese traditional medicine Rhizoma zedoariae, was encapsulated in galactosylated‑liposomes to enhance its anti‑liver cancer efficacy. Galactosylated‑liposomes and normal liposomes were labeled with propidium iodide. Galactosylated‑liposomes with increasing concentrations of galactosylated‑stearate (Gal‑s) had a notably increased level of uptake in HepG2 cells (hepatoblastoma) compared with SGC‑7901 (gastric cancer) and A549 (non‑small cell lung cancer) cells. When the percentage of Gal‑s reached 20%, liposome uptake plateaued. In the in vitro anti‑liver cancer experiment, the anti‑liver cancer efficacy of galactosylated‑curcumol‑liposomes increased significantly more compared with normal curcumol liposomes and free curcumol as indicated by cell survival rate and lactate dehydrogenase release rate. Collectively, these results demonstrate that galactosylated‑liposomes are able to enhance the in vitro liver‑targeting effect and anti‑liver cancer efficacy of curcumol.

Much attention has been drawn to targeted nanodrug delivery systems due to their high therapeutic efficacy in cancer treatment. In this work, doxorubicin (DOX) was incorporated into a zwitterionic arginyl–glycyl–aspartic acid (RGD)-conjugated polypeptide by an emulsion solvent evaporation technique with high drug loading content (45%) and high drug loading efficiency (95%). This zwitterionic nanoformulation showed excellent colloidal stability at high dilution and in serum. The pH-induced disintegration and enzyme-induced degradation of the nanoformulation were confirmed by dynamic light scattering and gel permeation chromatography. Efficient internalization of DOX in the cells and high antitumor activity in vitro was observed. Compared with the free drug, this nanoformulation showed higher accumulation in tumor and lower systemic toxicity in vivo. The DOX-loaded zwitterionic RGD-conjugated polypeptide vesicles show potential application for targeted drug delivery in the clinic.

A novel nanohybrid of hyaluronic acid (HA)-decorated graphene oxide (GO) was fabricated as a targeted and pH-responsive drug delivery system for controlling the release of anticancer drug doxorubicin (DOX) for tumor therapy. For the preparation, DOX was first loaded onto GO nanocarriers via π–π stacking and hydrogen-bonding interactions, and then it was decorated with HA to produce HA–GO–DOX nanohybrids via H-bonding interactions. In this strategy, HA served as both a targeting moiety and a hydrophilic group, making the as-prepared nanohybrids targeting, stable, and disperse. A high loading efficiency (42.9%) of DOX on the nanohybrids was also obtained. Cumulative DOX release from HA–GO–DOX was faster in pH 5.3 phosphate-buffered saline solution than that in pH 7.4, providing the basis for pH-response DOX release in the slightly acidic environment of tumor cells, while the much-slower DOX release from HA–GO–DOX than DOX showed the sustained drug-release capability of the nanohybrids. Fluorescent images of cellular uptake and cell viability analysis studies illustrated that these HA–GO–DOX nanohybrids significantly enhanced DOX accumulation in HA-targeted HepG2 cancer cells compared to HA-nontargeted RBMEC cells and subsequently induced selective cytotoxicity to HepG2 cells. In vivo antitumor efficiency of HA–GO–DOX nanohybrids showed obviously enhanced tumor inhibition rate for H22 hepatic cancer cell-bearing mice compared with free DOX and the GO–DOX formulation. These studies suggest that the HA–GO–DOX nanohybrids have potential clinical applications for anticancer drug delivery.

For a targeted drug delivery, it was great biomedical potential, significance, importance and application value to construct a colloidal-chemistry stable nanocomposite. Functionalized multi-walled carbon (MWNTs-OX) has attracted considerable attention due to the unique atomic structure, strong cell membrane penetration, specific surface area and low toxicity. It has also been used widely for targeted drug delivery as an ideal nanocomposite. In this paper, a facile one-pot strategy to MWNTs-OX with hyperbranched polyglycerol (PG) by surface-initiated ring-opening polymerization of glycidol was used to form MWNTs-OX-PG. Then, the partial periphery of PG layer was converted to carboxy groups and covalently linked to FA to form MWNTs-OX-PG-COOH-FA. Meanwhile, the properties of MWNTs-OX, MWNTs-OX-PG, MWNTs-OX-PG-COOH and MWNTs-OX-COOH-FA were characterized by Fourier transform infrared (FT-IR), Ultraviolet-visible spectrophotometry (UV), Thermogravimetric analysis (TGA), Scanning transmission electron microscopy (STEM) and elemental analysis, and the results indicated that MWNTs-OX-PG-COOH-FA preformed excellent dispersity, interface stability and long-term storage. This paper opened up a new strategy to rational design folic acid-functionalized MWNTs-OX-PG multifunctional nanohybrids as biomaterials.

Graphical Targeted drug delivery : Nanoscale metal–organic frameworks modified with folic acid (FA-NMOFs) have been prepared as potential drug carriers for targeted delivery to tumor cells (see figure). The anticancer drug 5-fluorouracil (5-FU) was encapsulated in the pores of the FA-NMOFs. The 5-FU-loaded FA-NMOFs showed excellent biocompatibility and were found to effectively kill tumor cells through the targeting effect of the FA-NMOFs. Nanoscale metal–organic frameworks (NMOFs) have proven to be a class of promising drug carriers as a result of their high porosity, crystalline nature with definite structure information, and potential for further functionality. However, MOF-based drug carriers with active tumor-targeting function have not been extensively researched until now. Here we show a strategy for constructing active tumor-targeted NMOF drug carriers by anchoring functional folic acid (FA) molecules onto the metal clusters of NMOFs. Two zirconium-based MOFs, MOF-808 and NH 2 -UiO-66, were chosen as models to reduce to the nanoscale for application as drug carriers, and then the terminal carboxylates of FA molecules were coordinated to Zr 6 clusters on the surfaces of the nanoparticles by substitution of the original formate or terminal -OH ligands. The successful modification with FA was confirmed by solid-state 13 C MAS NMR and UV/Vis spectroscopy and other characterization methods. Drug loading and controlled release behavior at different pH were determined by utilizing the anticancer drug 5-fluorouracil (5-FU) as the model drug. Confocal laser scanning microscopy measurements further demonstrated that 5-FU-loaded FA-NMOFs have excellent targeting ability through the efficient cellular uptake of FA-NMOFs. This work opens up a new avenue to the construction of active tumor-targeted NMOF-based drug carriers with potential for cancer therapies.

Conventional cancer chemotherapy presents relevant limitations associated with the non-selectivity of cytotoxic drugs, their narrow therapeutic indices and limited cellular penetration. Molecularly imprinted polymers (MIP) prepared using molecular imprinted technology has the advantages such as high stability against chemical and enzymatic attack, high selectivity for a specific template, and the capability of resistance to harsh environment. In this research, the MIP with cancer antigen 125 (CA125) tumor markers as template molecule, combined with molecular imprinting technique was prepared on the surface of graphene oxide for drug delivery. As a result the prepared composite had the properties of specific binding to CA125 and biocompatibility. Along with this, the composites could target specific tumor cell by the capture of CA125. CA125-molecularly imprinted polymers as a recognition element for the tumor cell could eliminate the use of antibody, enhance stability and prolong the retention time of drugs surrounding them. Ultimately, this method developed a cost-effective and stable way to target tumor cell for drug delivery.

In this paper, a plasma membrane engineering approach is reported for tumor targeting drug delivery and contact-cell-enhanced photodynamic therapy (“CONCEPT”) by anchoring functionalized conjugates to cell vehicles. The membrane anchoring conjugates are comprised of a positively charged tetra-arginine peptide sequence, a palmitic-acid-based membrane insertion moiety, and a lysine linker whose ε-amine is modified with camptothecin (CPT), protoporphyrin IX (PpIX), or fluorescein (FAM). The amphipathic CPT, PpIX, or FAM conjugates (short as aCPT, aPpIX, or aFAM, respectively) can easily and steadily anchor or coanchor on the cell membrane of RAW264.7 cells (short as RCs), red blood cells, or mesenchymal stem cells. After anchoring aPpIX in RC cells, the tumor targeting ability and therapeutic effect of aPpIX-anchored RC cells (short as aPRCs) is demonstrated in vitro and in vivo. Importantly, aPRCs exhibit the “CONCEPT” effect, which can enhance the therapeutic efficacy and reduce side effects at the single cell level. Due to the good tumor-targeting ability, aPRCs can efficiently inhibit the tumor growth with no systemic toxicity after photoirradiation by photodynamic therapy.

The fabrication of supramolecular host in combination biomolecules is an interesting idea in modern drug delivery for development of new polymer with advanced chemical and biological properties. Herein, hyper-crosslinked copolymer was fabricated from β-cyclodextrin (β-CD) and sialic acid (SA) monomers , which can be undoublty considered as a new class of copolymer. The as-synthesized copolymer has complexation properties, which could cover the drug within the structure and deliver to the site of action. The well-known inclusion capability of β-CD and targeted efficacy of SA made it more appropriate for targeted drug delivery. The copolymer was characterized using a wide range of spectroscopic and microscopic techniques such as synchrotron radiation based FTIR spectroscopy (SR-FTIR), differential scanning calorimetry (DSC), thermogravimetric analysis (TGA), and powder X-ray diffraction (PXRD). The surface area and porosity were calculated by using Nitrogen adsorption method. Doxorubicin (Dox) was selected as a model drug to evaluate the loading efficiency and cellular penetration ability of the copolymer. The copolymer showed high adsorption towards Dox with no significant cytotoxic effects on HeLa cells as proved by cell viability assay. High cellular penetration of Dox loaded copolymer was also recorded by confocal microscopy when compared with free Dox in HeLa cells at 4 h of exposure. Thus, β-CD-SA copolymer could be a useful carrier for targeted drug delivery of cancer and has the potential for further investigation in viral and nervous disease due to the targeting ability of SA.
