Hyaluronic acid-modified mesoporous silica nanoprobes for target identification of atherosclerosis

Ruifan Ma,Liguo Hao,Jianing Cheng,Jun He,Qiangqiang Yin,Zhongtao Li,Guiqiang Qi,Xiaoyang Zheng,Dongxu Wang,Tianyu Zhang,Houyi Cong,Zheng Li,Haifeng Hu,Yuguang Wang

Rupture of vulnerable plaque and secondary thrombosis caused by atherosclerosis are one of the main causes of acute cardiovascular and cerebrovascular events, and it is urgent to develop an in-situ, noninvasive, sensitive and targeted detection method at molecular level. We chose CD44, a specific receptor highly expressed on the surface of macrophages, as the target of the molecular probe, and modified the CD44 ligand HA onto the surface of Gd 2 O 3 @MSN, constructing the MRI imaging nanoprobe HA-Gd 2 O 3 @MSN for targeted recognition of atherosclerosis. The fundamental properties of HA-Gd 2 O 3 @MSN were initially investigated. The CCK-8, hemolysis, hematoxylin-eosin staining tests and blood biochemical assays confirmed that HA-Gd 2 O 3 @MSN possessed excellent biocompatibility. Laser confocal microscopy, cellular magnetic resonance imaging, flow cytometry and immunohistochemistry were used to verify that the nanoprobes had good targeting properties. The in vivo targeting performance of the nanoprobes was further validated by employing a rabbit atherosclerosis animal model. In summary, the synthesized HA-Gd 2 O 3 @MSN nanoprobes have excellent biocompatibility properties as well as good targeting properties. It could provide a new technical tool for early identification of atherosclerosis.

A folic acid-functionalized dual-emissive nanoprobe for “double-check” luminescence imaging of cancer cells

Chaolong Liu,Jianping Liu,Hua Ma,Wenzhu Zhang,Bo Song,Lianying Guo,Run Zhang,Jingli Yuan

Development of luminescent probes for rapid and effective discrimination and detection of cancer cells has the potential to address the current challenges in early diagnosis and treatment monitoring of cancer diseases. In this work, we report the preparation of a unique folic acid (FA)-functionalized dual-emissive nanoprobe, CTMR@BHHBCB-Eu-FA , for steady-state and time-gated luminescence “double-check” imaging of cancer cells. The nanoprobe was engineered by covalently doping two luminescent dyes, 5-carboxytetramethylrhodamine (CTMR) and BHHBCB-Eu 3+ , in core and shell of silica nanoparticles, followed by surface modification of the nanoparticles with FA, a cancer cell-targeting molecule. As-prepared nanoprobe is monodisperse and highly stable in buffer displaying two strong emissions, short-lived emission from CTMR at 584 nm and long-lived emission from BHHBCB-Eu 3+ at 612 nm. The nanoprobe is biocompatible, and can specifically recognize folate receptor (FR)-overexpressed cancer cells through the FA-FR binding interaction. Using the nanoprobe, the “double-check” imaging of HeLa cells was successfully achieved at steady-state and time-gated luminescence modes, indicating the capability of the nanoprobe for cancer cell imaging.

Molecular Engineering of Aptamer Self-Assemblies Increases in Vivo Stability and Targeted Recognition

Fangfang Xia,Axin He,Haitao Zhao,Yang Sun,Qiao Duan,Sk Jahir Abbas,Jianjun Liu,Zeyu Xiao,Weihong Tan

Functionally modified aptamer conjugates are promising tools for targeted imaging or treatment of various diseases. However, broad applications of aptamer molecules are limited by their in vivo instability. To overcome this challenge, current strategies mostly rely on covalent chemical modification of aptamers, a complicated process that requires case-by-case sequence design, multiple-step synthesis, and purification. Herein, we report a covalent modification-free strategy to enhance the in vivo stability of aptamers. This strategy simply utilizes one-step molecular engineering of aptamers with gold nanoclusters (GNCs) to form GNCs@aptamer self-assemblies. Using Sgc8 as a representative aptamer, the resulting GNCs@Sgc8 assemblies enhance cancer-cell-specific binding and sequential internalization by a receptor-mediated endocytosis pathway. Importantly, the GNCs@aptamer self-assemblies resist nuclease degradation for as long as 48 h, compared to the degradation of aptamer alone at 3 h. In parallel, the tumor-targeted recognition and retention of GNCs@aptamer self-assemblies are dramatically enhanced, indicated by a 9-fold signal increase inside the tumor compared to the aptamer alone. This strategy is to avoid complicated chemical modification of aptamers and can be extended to all aptamers. Our work provides a simple, effective, and universal strategy for enhancing the in vivo stability of any aptamer or its conjugates, thus expanding their imaging and therapeutic applications.

Synthesis of a novel polysaccharide natural functional monomer for imprinted hydrogel microsphere preparation and targeted recognition of anti-tumor drug 7-xylulose-10-deacetylpaclitaxel

Xiaodan Wu,Chengdong Nie,Mengqiong Deng,Linan Sun,Ying Wang,Jingru Zhao,Yujie Fu,Yourong Duan,Ying Sun

In this study, natural polysaccharides were first modified to synthesize natural functional monomers (NFMs). Subsequently, a natural novel NFM-based imprinted hydrogel microsphere was prepared for the specific recognition and enrichment of 7-xylulose-10-deacetylpaclitaxel (7-X-10-DTA). Molecular dynamics computational simulations were employed to screen and compare the functional monomers. Subsequently, the optimal preparation and application conditions of NMIHM were determined, with a maximum adsorption capacity of 71.77 mg mL −1 . The adsorption behavior of NMIHM for 7-X-10-DTA conforms to the pseudo-second-order kinetic model and the Langmuir model for monolayer adsorption, representing a spontaneous exothermic process. When used as a solid-phase extraction filler, NMIHM increased the content of 7-X-10-DTA by 13.36 times after enrichment in Taxus × media Rehde. The 7-X-10-DTA extracted in this study exhibited low half-maximal inhibitory concentration (IC50) values of 0.3205 μM and 0.7636 μM against cervical cancer Hela cells and prostate cancer PC-3 cells, respectively, demonstrating a more potent killing effect on these two types of cancer cells. This research is expected to accelerate the research and development of natural functional monomers and the advancement of molecular imprinting technology, thereby promoting the rapid development of practical separation applications.

An integrated approach for studying exposure, metabolism, and disposition of traditional Chinese medicine using PATBS and MDRB tools: a case study of semen Armeniacae Amarum

Dandan Zhang,Junyu Zhang,Simian Chen,Hairong Zhang,Yuexin Yang,Shan Jiang,Yun Hong,Mingshe Zhu,Qiang Xie,Caisheng Wu

Background Deciphering the in vivo processes of traditional Chinese medicine (TCM) is crucial for identifying new pharmacodynamic substances and new drugs. Due to the complexity and diversity of components, investigating the exposure, metabolism, and disposition remains a major challenge in TCM research. In recent years, a number of non-targeted smart mass-spectrometry (MS) techniques, such as precise-and-thorough background-subtraction (PATBS) and metabolomics, have realized the intelligent identification of in vivo components of TCM. However, the metabolites characterization still largely relies on manual identification in combination with online databases. Results We developed a scoring approach based on the structural similarity and minimal mass defect variations between metabolites and prototypes. The current method integrates three dimensions of mass spectral data including m/z , mass defect of MS1 and MS2, and the similarity of MS2 fragments, which was sequentially analyzed by a R-based mass dataset relevance bridging (MDRB) data post-processing technique. The MDRB technology constructed a component relationship network for TCM, significantly improving metabolite identification efficiency and facilitating the mapping of translational metabolic pathways. By combining MDRB with PATBS through this non-targeted identification technology, we developed a comprehensive strategy for identification, characterization and bridging analysis of TCM metabolite in vivo. As a proof of concept, we adopted the proposed strategy to investigate the process of exposure, metabolism, and disposition of Semen Armeniacae Amarum (CKXR) in mice. Significance The currently proposed analytical approach is universally applicable and demonstrates its effectiveness in analyzing complex components of TCMs in vitro and in vivo. Furthermore, it enables the correlation of in vitro and in vivo data, providing insights into the metabolic transformations among components sharing the same parent nucleus structure. Finally, the developed MDRB platform is publicly available for ( https://github.com/933ZhangDD/MDRB ) for accelerating TCM research for the scientific community. Graphical

Photodynamic Therapy of Up-Conversion Nanomaterial Doped with Gold Nanoparticles

Wei Zhang,Yang Zang,Yanli Lu,Jinghui Han,Qingyun Xiong,Jinping Xiong

Two key concerns exist in contemporary cancer chemotherapy: limited therapeutic efficiency and substantial side effects in patients. In recent years, researchers have been investigating the revolutionary cancer treatment techniques of photodynamic therapy (PDT) and photothermal therapy (PTT) proposed by many scholars. A photothermal treatment of cancer was synthesized using the hydrothermal method which has high photothermal conversion efficiency and can generate reactive oxygen species (ROS) in cells. Photothermal treatment of tumors has a good short-term effect and photodynamic therapy lasts longer. However, both PTT and PDT have their inevitable shortcomings and it is difficult to completely eradicate a tumor using a single mode of treatment. PTT and PDT synergistic treatment not only inherits the advantages of low toxicity and side effects of phototherapy but also enables the two treatment methods to complement each other. It is an effective strategy to improve curative effects and reduce toxic and side effects. Furthermore, gold doped UCNPs have an exceptionally high target recognition for tumor cells. The gold doped UCNPs, in particular, are non-toxic to normal tissues, endowing the as-prepared medications with outstanding therapeutic efficacy and exceptionally low side effects. These findings may encourage the creation of fresh, effective imaging-guided approaches to meet the goal of photothermal cancer therapy.Keywords:up-conversion;nanomaterials;photodynamic;photothermal

Targeted recognition and enhanced biotransformation of phytochemicals by a dual-functional cellulose-based hydrogel bioreactor

Lina Fu,Ye Li,Xinlin Zhang,Jihong Cui,Xuanting Zhao,Jiandong Wang,Quan Zhou,Litao Wang,Yujie Fu

The combined technology of molecular imprinting and immobilization is a promising strategy for improving biocatalytic performance. In this work, a dual-functional cellulose-based hydrogel bioreactor with targeted recognition and transformation functions was innovatively designed by cellulose-based molecularly imprinted polymers (CM-MIPs) hydrogel coupled with layered double hydroxide immobilized enzyme (LDH-E). Firstly, CM-MIPs hydrogel was prepared by using phlorizin, 4-pinylpyridine, and cellulose microspheres as template molecule, functional monomer, and support material, respectively. Meanwhile, layered double hydroxide (LDH) taken as a carrier to immobilize β-glucosidase. The prepared LDH was layered sheet-like structure with ultra-thin thickness of approximately only 1.5 nm, and β-glucosidase was immobilized on both sides of the LDH. Further, the dual-functional bioreactor was constructed by the anion exchange, which possessed maximum targeted adsorption capacity to phlorizin of 15.25 mg/g, exhibited 1.2 folds higher transformation efficiency than LDH-E, and the phloretin content increased 26 folds to that in Lithocarpus litseifolius (Hance) Chu leaves extracts. Moreover, the transformation efficiency remained above 70 % even after five consecutive transformations. Overall, the dual-functional bioreactor has broad prospects for the application in targeted recognition and transformation of phytochemicals, and provides a new insight on multifunctional bio-based reactors in natural production field.

Modulation of Electron Transport via Target Recognition in Asymmetric Nanochannel Arrays for Enantiomer Detection

Xuemei Shang,Mei Yang,Tiantian Su,Junli Guo,Chenxi Zhao,Pei Song,Yan-Yan Song,Yucen Li

Chirality pervades biological architecture, and therapeutics that interface with living systems likewise manifest as stereoisomeric pairs whose divergent metabolic fates engender distinct pharmacological outcomes. Accordingly, the creation of facile, cost-effective platforms for enantiomer discrimination is pivotal both to chiral-drug development and to elucidating biochiral-material interactions. Here we engineer an asymmetric nanoelectrode by partitioning a single TiO2 nanochannel membrane (TiO2M) into spatially discrete “recognition” and “reporting” domains. Gold nanoparticle decoration enables covalent anchoring of l-cysteine (Cys) via Au–S bonds at one face of TiO2M, establishing a stereospecific binding interface; copper-based metal–organic frameworks (Cu-MOFs) are grown on the trans face to serve as redox-active reporters. Using l-/d-dihydroxyphenylalanine (DOPA) as the model enantiomers, selective binding of l- or d-DOPA at the l-Cys interface initiates electrochemical oxidation that injects electrons into the Au/TiO2 conduit; these electrons traverse the nanochannel and reduce Cu(II) to Cu(I) within the distal MOF layer, thus modulating the surface charge and producing a discernible transmembrane current shift. Differential hydrogen-bonding competencies of l- versus d-DOPA with l-Cys translate into quantitatively distinct charge perturbations, thereby affording high-fidelity enantiomer recognition. This longitudinally segmented TiO2 nanochannel architecture thus converts stereospecific molecular interactions into amplified electronic signatures, offering a versatile blueprint for next-generation electrochemical sensors targeting chiral analytes.
