There has been an exponential increase in the rate of incidence of Parkinson's disease (PD) with aging in the global population. PD, the second most common neurodegenerative disorder, results from damaged dopamine neurons in the substantia nigra pars compacta (SNpc), along with the deposition of abnormal α-synuclein (α-Syn), and the progressive degeneration of neurons in striatal regions. Despite extensive investigations to understand the pathophysiology of PD to develop effective therapies to restrict its progression, there is currently no cure for PD. Puerarin (Pue) is a natural compound with remarkable anti-PD properties. However, its poor pharmacological properties, including poor water solubility, inadequate bioavailability, and incomplete penetration of the blood–brain barrier (BBB) have restricted its use for the treatment of PD. Nevertheless, advancements in nanotechnology have revealed the potential advantages of targeted drug delivery into the brain to treat PD. Here, we used Pue-loaded graphene oxide (GO) nanosheets, which have an excellent drug-loading ability, modifiable surface functional groups, and good biocompatibility. Then, Pue was transported across the BBB into the brain using lactoferrin (Lf) as the targeting ligand, which could bind to the vascular endothelial receptor on the BBB. In vivo and in vitro results indicated that this multifunctional brain targeted drug delivery system (Lf-GO-Pue) was an effective and safe therapy for PD.

Two kinds of core–shell structured multifunctional nanocarriers of gold nanoclusters (Au NCs) as core and folate (FA)-conjugated amphiphilic hyperbranched block copolymer as shell based on poly(l-lactide) (PLA) inner arm and FA-conjugated sulfated polysaccharide (GPPS–FA) outer arm (Au NCs-PLA-GPPS-FA) were synthesized for targeted anticancer drug delivery. The structure and properties of Au NCs-PLA-GPPS-FA copolymers were characterized and determined by 1H NMR spectrum, FT-IR spectra, dynamic light scattering (DLS), fluorescence spectroscopy, and transmission electron microscopic (TEM) analyses. The anticancer drug, camptothecin (CPT) was used as a hydrophobic model anticancer drug. In vitro, two kinds of the nanocarriers presented a relatively rapid release in the first stage (up to 1 h) followed by a sustained release period (up to 15 h), and then reached a plateau at pH 5.3, 7.4, and 9.6. The release results indicated that CPT release from two kinds of the nanocarriers at pH 9.6 was much greater than that at both pH 5.3 and 7.4. The cytotoxicity studies showed that the CPT-loaded nanocarriers provided high anticancer activity against Hela cells. Furthermore, nanocarriers gained specificity to target some cancer cells because of the enhanced cell uptake mediated by FA moiety. The fluorescent images studies showed that the nanocarriers could track at the cellular level for advance therapy. The results indicated that the Au NCs-PLA-GPPS-FA copolymers not only had great potential as tumor-targeted drug delivery carrier, but also had an assistant role in the treatment of cancer.

Xylan-based temperature/pH sensitive hydrogels were prepared by the crosslinking copolymerization of xylan with N -isopropylacrylamide (NIPAm) and acrylic acid (AA) using N,Ń -methylenebis-acrylamide (MBA) as a cross-linker and 2,2-dimethoxy-2-phenylacetophenone as a photoinitiator via ultraviolet irradiation. The influence of the NIPAm, AA and MBA amount on properties of xylan-based hydrogels was discussed. The morphology and interactions of hydrogels were characterized by SEM and FTIR. The lower critical solution temperature (LCST) of hydrogels was investigated by DSC. The results indicated that the LCST of hydrogels emerged at around 34 °C and increased with increasing the AA content. The drug encapsulation efficiency of as-prepared hydrogels reached to 97.60% and the cumulative release rate of acetylsalicylic acid was 90.12% and 26.35% in the intestinal and gastric fluid, respectively. Xylan-based hydrogels were proved to be biocompatible with NIH3T3 cell by MTT assay and showed the promising application as drug carriers for the intestinal-targeted oral drug delivery.

Bacterial enteritis is a prevalent disease in Micropterus salmoides . Conventional drug delivery approaches are typically characterized by low efficiency and induce drug resistance. The present study synthesized mesoporous hollow silica nanoparticles (HMSS) and used physical adsorption to load enrofloxacin (ENR) and astaxanthin (ASX) onto the nanoparticles, achieving adsorption efficiencies of 77.39 % and 32.77 %, respectively. To establish the drug-targeted delivery system, ASX@HMSS was functionalized with cell-penetrating peptide (CPP) and ENR@HMSS was encapsulated with hydroxypropyl methylcellulose phthalate (HP55). Positive sustained-release effects were demonstrated by in vitro release studies, which demonstrated that the cumulative release rate of ASX@HMSS-CPP varied from 29.58 to 38.47 % at pH 6.5, whereas the cumulative release rate of ENR@HMSS-HP55 ranged from 70.19 to 88.25 %. The drug-targeted delivery nanoparticles extended the residence time of the drug in the intestines, according to in vivo pharmacokinetic investigations, and the overall metabolic profile was positive. Additionally, these drug-targeted delivery nanoparticles reduced intestinal inflammation and tissue damage, enhanced antioxidant and anti-inflammatory indicators, and improved the survival rate of Micropterus salmoides infected with enteritis. Following the administration of the drug-targeted delivery nanoparticles, the abundance of Aeromonas was significantly regulated, aiding gut microbial homeostasis. By controlling the PI3K-Akt, NF-κB, MAPK, and intestinal IgA immune network signaling pathways, the drug-targeted delivery nanoparticles also reduced intestinal inflammation. These findings demonstrate that the nanoparticle-based drug-targeted delivery system has potential applications in the prevention and treatment of diseases in aquatic animals.

We investigated a novel drug delivery system comprising nanoparticles based on galactosylated chitosan/graphene oxide/doxorubicin (GC–GO–DOX) for the therapeutic treatment of cancer. The drug delivery system was synthesized by loading a drug sample with galactosylated chitosan (GC) on a graphene oxide (GO) carrier. The results showed that the drug loading capacity was as high as 1.08 mg/mg (drug per polymer). The nanoparticles remained stable under physiological conditions, and the drug was released in a low pH environment (i.e., a tumor environment) and was pH-responsive. Cell uptake experiments and a cell proliferation analysis demonstrated that the nanoparticles had higher cytotoxicity for HepG2 and SMMC-7721 cells than chitosan/graphene oxide/doxorubicin (CS–GO–DOX) nanoparticles. Compared with CS–GO–DOX nanoparticles, the GC–GO–DOX nanoparticles exhibited a higher fluorescence intensity in tumor cells. In vivo anti-tumor experiments demonstrated that the GC–GO–DOX nanoparticles inhibit tumors better than the CS–GO–DOX nanoparticles. Nude mouse weight, tumor weight and tumor volume data indicated that the GC–GO–DOX tumor inhibition effect was better than that of the control group and the blank group. In summary, the nanoparticle investigated in this article is significant for tumor therapy.

Anaplastic thyroid cancer (ATC) is aggressive and has a high mortality rate. Doxorubicin (DOX), the first-line chemotherapy drug for ATC, has an insufficient effective concentration and severe cardiac toxicity, limiting its clinical application. Therefore, it is urgent to enhance efficacy of DOX and reduce its toxicity. Designing stimulus-responsive drug delivery systems (DDSs) based on the characteristics of the tumor microenvironment may be a promising approach for the treatment of ATC. We found that heparanase (HPSE) is highly expressed in the tumor microenvironment of ATC but not in normal thyroid tissue. In this study, we successfully constructed and characterized the HPSE-responsive self-assembled carrier TET-HS-NI, which was modified with 3,3,5,5-Tetraiodothyroacetic acid (TET) that targetsαvβ3. DOX was loaded into TET-HS-NI to construct TET-HS-NI/DOX. The zebrafish cell-derived xenograft (CDX) and ATC orthotopic models showed that TET-HS-NI/DOX significantly improved the antitumor efficacy and safety of DOX. In a word , we have constructed a drug delivery system sensitively released DOX in the ATC environment where HPSE overexpressed, increasing the effective concentration at the tumor site and provide a new method for the treatment of ATC.

Background Mesoporous silica nanoparticles (MSNs) have several attractive properties as a drug delivery system, such as ordered porous structure, large surface area, controllable particle size as well as interior and exterior dual-functional surfaces. The purpose of this study was to develop novel lactosaminated mesoporous silica nanoparticles (Lac-MSNs) for asialoglycoprotein receptor (ASGPR) targeted anticancer drug delivery. Results Lac-MSNs with an average diameter of approximately 100 nm were prepared by conjugation of lactose with 3-aminopropyl triethoxysilane modified MSNs. Characterization of Lac-MSNs indicated a huge Brunauer-Emmett-Teller (BET) surface area (1012 m2/g), highly ordered 2D hexagonal symmetry, an unique mesoporous structure with average pore size of 3.7 nm. The confocal microscopy and flow cytometric analysis illustrated Lac-MSNs were effectively endocytosed by ASGPR-positive hepatoma cell lines, HepG2 and SMMC7721. In contrast, non-selective endocytosis of Lac-MSNs was found in ASGPR-negative NIH 3T3 cells. The cellular uptake study showed the internalization process was energy-consuming and predominated by clathrin-mediated pathway. Model drug docetaxel (DTX) was loaded in the mesopores of Lac-MSNs by wetness impregnation method. In vitro cytotoxicity assay showed that DTX transported by Lac-MSNs effectively inhibited the growth of HepG2 and SMMC7721 cells in a time- and concentration- dependent manner. Conclusion sThese results demonstrated that Lac-MSNs could be a promising inorganic carrier system for targeted intracellular anti-cancer drug delivery.

Bioorthogonal chemistry offers a promising approach for advancing chemoimmunotherapy, yet two critical challenges remain: (1) achieving deep tumor penetration of catalysts while ensuring precise tumor-confined prodrug activation, and (2) reversing metabolic immunosuppression to potentiate immunotherapy efficacy. To tackle these issues, here we fabricate a bioorthogonal probiotic nanosystem (NP@ZIF-8@Apt-Lr) by engineering prodrug catalysts and an indoleamine 2,3-dioxygenase (IDO) inhibitor onto AS1411 aptamer-modified Lactobacillus reuteri (Lr). AS1411-guided recognition and GSH-responsive prodrug decaging provide a double guarantee for tumor-selective drug activation, enabling localized tumor killing and further immunogenic cell death (ICD). Also, the inherent tumor-homing properties of Lr endow the nanosystem with the ability of penetrating deep tissue. More crucially, the released IDO inhibitor blocks the immunosuppressive kynurenine pathway of tryptophan metabolism, while Lr redirects tryptophan metabolism toward the immunostimulatory indole-3-aldehyde pathway through host-microbial crosstalk. Upon treatment, NP@ZIF-8@Apt-Lr overcomes immune tolerance and stimulates T lymphocyte infiltration into tumors. Furthermore, it significantly enhances the efficacy of chemoimmunotherapy by suppressing bilateral tumor development and inducing an immune memory response. This work presents a new strategy for enhancing bioorthogonal chemoimmunotherapy through precise prodrug activation and probiotic-mediated metabolic reprogramming by host-microbial crosstalk.
