Quantitative conversion of free, acid-hydrolyzable, and bound ellagic acid in walnut kernels during baking

Xin Guo,Fengying Gu,Tingting Yang,Zhixiao Shao,Qiaozhen Zhang,Jinjin Zhu,Feng Wang

Ellagitannins, the main components of walnut kernel polyphenols, are easily degraded by heating to produce ellagic acid, precursor in the production of urolithins that show multiple physiological functions. To analyze the conversion of ellagitannins to free ellagic acid in walnut kernels during baking, a quantitative method was developed to investigate the ellagic acid content (EAC) in free phenolic acid (FPA), acid-hydrolyzable phenolic acid (AHPA), and bound phenolic acid (BPA) fractions. The results showed that the EAC in FPA reached its maximum (5.17 ± 0.30 mg/g DW) after baking at 165 °C for 30 min, which increased by 99.52% compared with the control. Meanwhile, the content of ellagitannins (ETC) in AHPA and BPA dropped by 89.14% and 26.08%, respectively. It suggested that baking promoted the conversion of ellagitannins in AHPA and BPA to ellagic acid in FPA. Eight ellagitannins were regarded as the main precursors of ellagic acid in walnut kernels.

An Organic Solvent-Free Method for the Extraction of Ellagic Acid Compounds from Raspberry Wine Pomace with Assistance of Sodium Bicarbonate

Ning Jin,Shouyu Zhang,Shibo Sun,Minghuo Wu,Xiaojing Yang,Jianqiang Xu,Kun Ma,Shui Guan,Weiping Xu

Industrial processing of raspberry juice and wine generates considerable byproducts of raspberry pomace. Ellagic acids/ellagitannins, being characterized by their antioxidant and antiproliferation properties, constitute the majority of polyphenolics in the pomace and are valuable for recovery. In the present study, we developed a novel procedure with sodium bicarbonate assisted extraction (SBAE) to recover ellagic acid from raspberry wine pomace. Key parameters in the procedure, i.e., sodium bicarbonate concentration, temperature, time and solid/liquid (S/L) ratio, were investigated by single factor analysis and optimized subsequently by Response Surface Methodology (RSM). Optimal parameters for the SBAE method here were found to be 1.2% (w/v) NaHCO3, 1:93 (w/v) S/L ratio, 22 min and 100 °C. Under these conditions, the ellagic acid yield was 6.30 ± 0.92 mg/g pomace with an antioxidant activity of 79.0 ± 0.96 μmol Trolox eq/g pomace (DPPH assay), which are 2.37 and 1.32 times the values obtained by extraction with methanol–acetone–water solvent, respectively. The considerable improvement in ellagic acid extraction efficiency could be highly attributed to the reactions of lipid saponification and ellagitannin hydrolysis resulted from sodium bicarbonates. The present study has established an organic solvent-free method for the extraction of ellagic acid from raspberry wine pomace, which is feasible and practical in nutraceutical applications. Keywords:ellagic acid;extraction;raspberry;wine pomace;sodium bicarbonate;response surface methodology

Effects of Phenolics on the Physicochemical and Structural Properties of Collagen Hydrogel

Sadia Munir,Wei Yue,Jinling Li,Xiaoyue Yu,Tianhao Ying,Ru Liu,Juan You,Shanbai Xiong,Yang Hu

In the current era, the treatment of collagen hydrogels with natural phenolics for the improvement in physicochemical properties has been the subject of considerable attention. The present research aimed to fabricate collagen hydrogels cross-linked with gallic acid (GA) and ellagic acid (EA) at different concentrations depending on the collagen dry weight. The structural, enzymatic, thermal, morphological, and physical properties of the native collagen hydrogels were compared with those of the GA/EA cross-linked hydrogels. XRD and FTIR spectroscopic analyses confirmed the structural stability and reliability of the collagen after treatment with either GA or EA. The cross-linking also significantly contributed to the improvement in the storage modulus, of 435 Pa for 100% GA cross-linked hydrogels. The thermal stability was improved, as the highest residual weight of 43.8% was obtained for the hydrogels cross-linked with 50% GA in comparison with all the other hydrogels. The hydrogels immersed in 30%, 50%, and 100% concentrations of GA also showed improved swelling behavior and porosity, and the highest resistance to type 1 collagenase (76.56%), was obtained for 50% GA cross-linked collagen hydrogels. Moreover, GA 100% and EA 100% obtained the highest denaturation temperatures (Td) of 74.96 °C and 75.78 °C, respectively. In addition, SEM analysis was also carried out to check the surface morphology of the pristine collagen hydrogels and the cross-linked collagen hydrogels. The result showed that the hydrogels cross-linked with GA/EA were denser and more compact. However, the improved physicochemical properties were probably due to the formation of hydrogen bonds between the phenolic hydroxyl groups of GA and EA and the nitrogen atoms of the collagen backbone. The presence of inter- and intramolecular cross-links between collagen and GA or EA components and an increased density of intermolecular bonds suggest potential hydrogen bonding or hydrophobic interactions. Overall, the present study paves the way for further investigations in the field by providing valuable insights into the GA/EA interaction with collagen molecules. Keywords:collagen;gallic acid;ellagic acid;hydrogels;storage modules

Antityrosinase mechanism of ellagic acid in vitro and its effect on mouse melanoma cells

Qian Huang,Wei-Ming Chai,Zuo-Yuan Ma,Wei-Liang Deng,Qi-Ming Wei,Shuang Song,Zheng-Rong Zou,Yi-Yuan Peng

The activities of ellagic acid in inhibiting mushroom tyrosinase and cell proliferation were evaluated in this research. The results of enzyme kinetics indicated that ellagic acid could effectively inhibit tyrosinase activity. The value of the semi-inhibitory rate (IC 50 ) was 0.2 ± 0.05 mM. Ellagic acid inhibited tyrosinase activity in a reversible manner and was a mixed tyrosinase inhibitor. Furthermore, ellagic acid had a good inhibitory effect on the proliferation of mouse melanoma B 16 cells and could induce apoptosis. The results acquired from fluorescence spectroscopy revealed that the interaction of ellagic acid with tyrosinase depended on hydrogen bond and electrostatic force. In addition, computational docking showed that ellagic acid interacted with amino acid residues of tyrosinase (Asn19 and Lys372) by hydrogen bond and produced electrostatic interaction with amino residue Lys18. Practical applications In the present research, the antityrosinase mechanism of ellagic acid and its effect on mouse melanoma cells were investigated. This study suggested that ellagic acid had a strong inhibitory activity against tyrosinase and cell proliferation, which laid an experimental foundation for the development of new drugs and whitening products. The combined multispectral methods used in this research can be applied to the screening of other antityrosinase inhibitors, further promoting the development and utilization of tyrosinase inhibitors.

Oral olsalazine/ellagic acid/ZnCl2‐modified in situ-forming hydrogels alleviates anxiety/depression-like symptoms and brain neural activity in colitis mice

Tingting Li,Nianlu Li,Haoyu Sheng,Luyao Zhang,Qian Luo,Sashuang Liu,Yijing Zhao,Wenqiang Chen,Dexiang Liu,Zhen Wang

Inflammatory bowel disease (IBD) is a kind of chronic non-specific intestinal inflammatory disease, which accompanied by various abnormal psychological states, such as anxiety and depression. Nevertheless, the pathogenesis of IBD with depression is still unclear. Herein, we synthesized an enable a drinkable, liquid in situ-forming hydrogel, which form directly in the stomach through sequential ingestion of a crosslinker solution of CaCl 2 and ε-poly-l-lysine (ε-PL), followed by a drug-containing polymer solution of sodium alginate (SA). Using olsalazine (Olsa) and antioxidant (ellagic acid (EA) and ZnCl 2 ) as small molecule drug (referred as Olsa/EA/Zn), the resulting hydrogel (referred as Olsa/EA/Zn@SA/ε-PL hydrogel) could form rapidly in gastric fluid and exhibit remarkable stability under acidic conditions, unique low swelling properties and a rational release of drug in entering the intestine. We demonstrated that the oral administration of drug-loaded in situ-forming hydrogel with excellent antioxidant, anti-inflammatory activities, cytocompatibility, intestinal retention capacity of the drug, significantly enhanced the IBD therapeutic outcomes, and relieve accompanying depression-like symptoms in colitis mice. Overall, this designed in situ-forming hydrogel oral delivery system will open up new pathway in combined gastrointestinal therapy and psychological comorbidities for IBD patients, especially those with dysphagia.

Phenolic hydroxyl derived copper alginate microspheres as superior adsorbent for effective adsorption of tetracycline

Xiaonuo Zhang,Xiaoyan Lin,Yu He,Xuegang Luo

Significant concerns had been given raise to the pollution of tetracycline in aquatic environment in recent years. In this study, a novel phenolic hydroxyl (bayberry tannin) functionalized copper alginate microspheres adsorbent for the adsorption of tetracycline from contaminated water environment had been synthesized by a facile gelation and solidification process. The structure and chemical properties were characterized by the various analytical methods. The as-prepared adsorbent displayed the excellent adsorption performance with the maximum adsorption capacity of 153.89 mg·g −1 , and the equilibrium data were well fitted the Langmuir isotherm model and pseudo-second-order kinetic model. In addition, the thermodynamic values (Δ H 0  > 0, Δ S 0  > 0, Δ G 0  < 0) demonstrated that the tetracycline adsorption process was feasible, endothermic and spontaneous in nature. The possible preparation and adsorption mechanisms of as-prepared adsorbent were the result of hydrogen bond, cation bonding bridge, n-π EDA interaction, hydrophobic interaction and π-π EDA interaction. Compared with other adsorbents, the as-prepared adsorbent could be considered a high performance and promising candidate for the effective removal of tetracycline from aquatic environment.

Ellagic acid-based non-isocyanate polyurethane water-soluble sizing agents for enhancing the interfacial properties of carbon fiber/epoxy composites

Hao Liu,Lu Wang,Xiaoxue Zhang,Junbo Zang,Song Xu,Chunhong Zhang,Yuhui Ao,Weifeng Du,Lei Shang

The preparation of environmentally friendly and efficient waterborne polyurethane sizing agents for carbon fiber (CF) is difficult, especially to avoid the use of isocyanates. Herein, ellagic acid and CO 2 were used to prepare a five-membered cyclic carbonate compound (EARBO). The polymerization between EARBO and diamines enabled the preparation of highly efficient and green aqueous non-isocyanate polyurethane (WNIPU) sizing agents. WNIPU sizing agent exhibits excellent heat resistance (T 5% :244.5 °C). The hydroxyl group and tertiary amine effectively enhance the polarity and wettability of CF (CF surface energy is as high as 55.42 mJ/m 2 ) and show excellent compatibility with epoxy resin. Carbon fiber/epoxy composites treated with this sizing agent showed exceptional interfacial properties, achieving interfacial shear strength, ply shear strength, flexural strength, and tensile strength of 116.84 MPa, 71.58 MPa, 1352.79 MPa, and 1719.27 MPa, respectively—representing 112.05 %, 42.05 %, 86.68 %, and 87.11 % improvements over traditional composites. The WNIPU sizing agent’s efficiency and environmental advantages indicate it as a promising alternative to commercial polyurethanes.

Novel Metal Polyphenol Framework for MR Imaging-Guided Photothermal Therapy

Gaozheng Zhao,Huihui Wu,Ruilu Feng,Dongdong Wang,Pengping Xu,Peng Jiang,Kang Yang,Haibao Wang,Zhen Guo,Qianwang Chen

Phothermal therapy has received increasing attention in recent years as a potentially effective way to treat cancer. In pursuit of a more biocompatible photothermal agent, we utilize biosafe materials including ellagic acid (EA), polyvinylpyrrolidone (PVP), and iron element as building blocks, and we successfully fabricate a homogeneous nanosized Fe-EA framework for the first time by a facile method. As expected, the novel nanoagent exhibits no obvious cytotoxicity and good hemocompatibility in vitro and in vivo. The microenvironment responsiveness to both pH and hydrogen peroxide makes the NPs biodegradable in tumor tissues, and the framework should be easily cleared by the body. Photothermal potentials of the nanoparticles are demonstrated with relevant features of strong NIR light absorption, moderately effective photothermal conversion efficiency, and good photothermal stability. The in vivo photothermal therapy also achieved effective tumor ablation with no apparent toxicity. On the other hand, it also exhibits T2 MR imaging ability originated from ferric ions. Our work highlights the promise of the Fe-EA framework for imaging-guided photothermal therapy.

Maillard Browning Inhibition by Ellagic Acid via Its Adduct Formation with the Amadori Rearrangement Product

Heping Cui,Ziyan Wang,Mengyu Ma,Khizar Hayat,Qiang Zhang,Yan Xu,Xiaoming Zhang,Chi-Tang Ho

The Maillard reaction performed under a stepwise increase of temperature was applied for researching the inhibition of Maillard browning caused by ellagic acid. Ellagic acid was found effective for the inhibition of melanoidin formation in the xylose–glycine Maillard reaction but depended on its dosage and the point of time it was added in the reaction system. The lightest color of the Maillard reaction products was observed when ellagic acid was added at the 90th min, which was the point of time when the Amadori rearrangement product (ARP) developed the most. LC-ESI-MS/MS analysis results showed a significant tendency of the ellagic acid hydrolysis product to react with the predominant intermediate ARP to yield an adduct. The adduct stabilized the ARP and delayed its decomposition and inhibited the downstream reactions toward browning. After the ARP was depleted, ellagic acid also showed an effect on scavenging some short-chain dicarbonyls which contributed to the inhibition of Maillard browning.

Xanthine oxidase inhibitory kinetics and mechanism of ellagic acid: In vitro, in silico and in vivo studies

Jianmin Chen,Zemin He,Sijin Yu,Xiaozhen Cai,Danhong Zhu,Yanhua Lin

Ellagic acid (EA), which is widely distributed in many foods, has been found to possess inhibitory activity against xanthine oxidase (XO). However, there is ongoing debate about the difference in XO inhibitory activity between EA and allopurinol. Additionally, the inhibitory kinetics and mechanism of EA on XO are still unclear. Herein, the authors systematically studied the inhibitory effects of EA on XO. The authors’ findings showed that EA is a reversible inhibitor with mixed-type inhibition, and its inhibitory activity is weaker than allopurinol. Fluorescence quenching experiments suggested that the generation of EA-XO complex was exothermic and spontaneous. In silico analysis further confirmed that EA entered the XO catalytic centre. Furthermore, the authors verified the anti-hyperuricemia effect of EA in vivo. This study elucidates the inhibition kinetics and mechanism of EA on XO, and lays a theoretical foundation for the further development of drugs and functional foods containing EA for the treatment of hyperuricemia.

Transdermal BQ-788/EA@ZnO quantum dots as targeting and smart tyrosinase inhibitors in melanocytes

Xiao Huang,Chun Chen,Xinting Zhu,Xi Zheng,Sanhua Li,Xingquan Gong,Zuli Xiao,Nian Jiang,Cangyan Yu,Caixia Yi

Tyrosinase inhibitors could effectively limit the activity of tyrosinase in melanocytes to reduce the excessive synthesis and deposition of melanin. However, low skin permeability and lacking in targeting greatly restricted their application. Herein, ZnO quantum dots were synthesized by gel-sol method and grafted with BQ-788, which have been employed as transdermal and targeting carrier to delivery ellagic acid to melanocytes. Ellagic acid loaded ZnO quantum dots with the size distribution of around 9 nm could targetedly bind to melanocytes and enter the melanocytes by endocytosis within 1 h. The ellagic acid release behavior was controlled by the decreasing of pH via the rapid dissolution of ZnO. When the concentration of BQ-788/EA@ZnO was 12.5 μg/mL, the inhibition rate on tyrosinase activity and melanin deposition were up to 44.23 ± 4.97% and 37.50 ± 5.23%, respectively. In view of their good biocompatibility, they were of great potential in clinically external application for tyrosinase inhibition.

Sulfonated carbon derived from the residue obtained after recovery of essential oil from the leaves of Cinnamomum longepaniculatum using Brønsted acid ionic liquid, and its use in the preparation of ellagic acid and gallic acid

Zaizhi Liu,Yanlong Qi,Mengling Gui,Chunte Feng,Xun Wang,Yang Lei

A Brønsted acid ionic liquid, 3-methyl-1-(4-sulfonylbutyl) imidazolium hydrogensulfate ([HO3S(CH2)4mim]HSO4), was used for the first time for the preparation of a sulfonated carbon catalyst. The catalyst was prepared from the residue obtained after recovery of the essential oil from the leaves of Cinnamomum longepaniculatum. The sulfonated carbon catalyst with an amorphous structure attained high acidic efficiency at a sulfonation temperature of 200 °C for 2 h of sulfonation time, and was characterised. SEM morphologies revealed that the carbon catalyst consisted of uniform carbon microspores. FTIR analysis, elemental analysis, and X-ray photoelectron spectroscopy revealed that the sulfonic acid group was successfully introduced on the surface of the sulfonated carbon catalyst. The result of TG analysis showed that the obtained sulfonated carbon catalyst has high thermal stability. Good acid and catalytic activity of the obtained sulfonated carbon catalyst were observed for the preparation of ellagic acid and gallic acid, which is comparable to those of diluted sulfuric acid and a sulfonated carbon catalyst that had been prepared with concentrated sulfuric acid. The excellent reusability of the sulfonated carbon catalyst was also confirmed by repeated experimental trials. In summary, the sulfonated catalyst derived from the residue obtained after recovery of essential oil from the leaves of C. longepaniculatum is an economic, eco-benign and promising substitute for traditional mineral acid catalysts for acidic catalysis in industrial applications.
