Wound treatment should meet the challenge both of preventing infection and promoting wound healing. To design a sequential delivery system for wound healing, PLGA-glycol chitosan (GC) core-shell microspheres containing chlorhexidine acetate (CHA) at the GC shell and bFGF in the core of PLGA microspheres were fabricated using emulsion-solvent evaporation method. SEM showed that the microspheres were all spherical in shape with a smooth surface. The average size of PLGA-GC microspheres increased due to the GC coating on the surface. The results of release profiles and fluorescence images indicated that PLGA-GC microspheres had an ability to deliver drugs in sequence. The CHA was rapidly released, whereas the proteins presented a sustained release. The release behavior could be modulated by changing the GC amount. Antibacterial assay and cell proliferation tests suggested that the released CHA and bFGF retained their antimicrobial activity and bioactivity during preparation. The microspheres exhibited non-cytotoxicity against 3T3 cells and had a good biocompatibility. These results demonstrated that PLGA-GC core-shell microspheres could be a promising controlled release system of delivering drugs and proteins in sequence for wound healing.

Due to the unclear pathogenesis of Alzheimer's disease (AD) and the lack of a completely cured medication, AD patients need to take medication in order and on time every day all one's life, which is difficult for severe memory impairment patients to strictly follow on time. Traditional AD drug carriers, such as sugar coating and capsules, rely on dissolution or fragmentation to achieve drug release, which lacks the interaction between drug molecules and carriers, thus they cannot achieve sufficient long-acting and sequential drug release. Herein, Mn-MOF-74, which ligand structure is similar to two antioxidants dihydroquercetin (DHQ) and resveratrol (Res) is chosen as the carrier. Due to the differences in adsorption energy between DHQ/MOF and Res/MOF, the release speed of DHQ is much faster than Res. Therefore, Mn-MOF-74 loaded with DHQ and Res (DR@MOF) showed sequential drug release and a long-term antioxidant effect for ≈72 h, with an efficacy time six times longer than that of vitamin E. In 5×FAD transgenic mice, DR@MOF exhibited excellent capacity in maintaining oxidative balance in the brain, ameliorating spatial learning and memory deficits, and showed the potential of an AD agent for long-acting and sequential treatment.

Adsorption is a classical physicochemical process, widely used in heterogeneous catalysis, chemical engineering, and environmental science. Regenerating the exhausted adsorbents or reversible desorption is vital to achieving sustainable adsorption techniques, while the common chemical/thermal regeneration is energy-intensive and environmentally unfriendly. Given that the adsorbents’ surface physicochemical properties like surface charge and electronic structures, significantly affect adsorption capacity and kinetics, which offers opportunities for green desorption by tailoring surface chemical/electronic features. Since a certain adsorbate possesses specific adsorption energy on adsorbents, an electro-desorption idea is therefore proposed to electrically control the (sequential) release of aqueous substances by regulating the interfacial adsorption interaction above the corresponding threshold interfacial potential. Herein, as a proof-of-concept design, a sub-millimeter-sized carbon nanotube-reinforced conductive resin composite adsorbent with high conductivity and mechanical strength is developed, which demonstrates a high electro-desorption efficiency toward benzoic acid (94.1%; 3 V negative bias) and well-controlled electro-desorption kinetics by regulating the surface polarization. Using benzoic acid and terephthalic acid as a model of organic mixtures, the adsorbent exhibits an efficient bias-dependent sequential electro-desorption capability in separating mixtures. This work provides a promising efficient solution for green and efficient desorption/regeneration and a pioneering electrically-driven strategy toward aqueous mixture separation.

Aim Sequential treatment with paclitaxel (PTXL) and gemcitabine (GEM) is considered clinically beneficial for non-small-cell lung cancer. This study aimed to investigate the effectiveness of a nano-system capable of sequential release of PTXL and GEM within cancer cells. Methods PTXL-ss-poly(6-O-methacryloyl-d-galactopyranose)-GEM (PTXL-ss-PMAGP-GEM) was designed by conjugating PMAGP with PTXL via disulfide bonds (-ss-), while GEM via succinic anhydride (PTXL:GEM=1:3). An amphiphilic block copolymer N-acetyl-d-glucosamine(NAG)-poly(styrene-alt-maleic anhydride)58-b-polystyrene130 acted as a targeting moiety and emulsifier in formation of nanostructures (NLCs).Results The PTXL-ss-PMAGP-GEM/NAG NLCs (119.6 nm) provided a sequential in vitro release of, first PTXL (redox-triggered), then GEM (pH-triggered). The redox- and pH-sensitive NLCs readily distributed homogenously in the cytoplasm. NAG augmented the uptake of NLCs by the cancer cells and tumor accumulation. PTXL-ss-PMAGP-GEM/NAG NLCs exhibited synergistic cytotoxicity in vitro and strongest antitumor effects in tumor-bearing mice compared to NLCs lacking pH/redox sensitivities or free drug combination. Conclusion This study demonstrated the abilities of PTXL-ss-PMAGP-GEM/NAG NLCs to achieve synergistic antitumor effect by targeted intracellularly sequential drug release.

Quercetin (Que) and resveratrol (Res) exhibit significant synergistic effects in multiple health benefits. However, their sites of action in the gastrointestinal tract are not consistent. To achieve the targeted release of Que and Res into the small intestine and colon, respectively, core–shell–shell nanoparticles with programmed sequential release properties were constructed by layer-by-layer assembly technique in this work. Compared with hordein/pectin nanoparticles, hordein/pectin/carboxymethyl chitosan nanoparticles (HPCCS NPs) possessed higher physicochemical stability, encapsulation efficiency and loading capacity. Besides, the existence of CCS layers also significantly improved the co-delivery and programmed sequential release properties. To further optimize the performance of NPs, ultrasonic treatment was innovatively applied to Que/Res–HPCCS NPs. It was found that ultrasonic treatment enhanced the hydrogen bonding, hydrophobic interactions and electrostatic adsorption between molecules, thus improving the physicochemical stability, encapsulation efficiency and loading capacity of NPs. Furthermore, in vitro simulated gastrointestinal studies demonstrated that Que/Res–HPCCS NPs (ultrasound) had better programmed sequential release properties, which further improved the release rates of Res and Que in the small intestine and colon, respectively. This study is of great significance for the development of co-delivery systems with programmed sequential release of nutraceuticals, which can provide novel ideas for the development of related functional foods.

Co-administration of natural anti-inflammatory active ingredients is a prospective strategy for intervening in ulcerative colitis (UC). Resveratrol (Res) possesses synergistic efficacy with vitamin D3 (Vd) in combating UC. However, it is difficult for Vd and Res to sequentially reach the small intestine and colon after oral administration to exert their synergistic anti-colitis effects due to their poor solubility, instability, and sensitivity to the gastrointestinal environment. Herein, Res and Vd were co-encapsulated in solid-in-oil-in-water emulsions (S/O/W-E), where the aqueous phase consisted of carboxymethyl starch/propylene glycol alginate complexes, the oil phase was coconut oil, and the solid phase was carboxymethyl konjac glucomannan-coated gliadin nanoparticles. The encapsulation efficiency of Res and Vd was determined to be 95.1 % and 94.7 %, respectively. The in vitro release results confirmed that the prepared S/O/W-E not only effectively protected Res and Vd from degradation under gastric conditions but also ensured that the encapsulated Vd and Res are released sequentially in the small intestine and colon. Furthermore, co-encapsulating Res and Vd in S/O/W-E could dramatically relieve symptoms in dextran sodium sulfate-induced UC mice and ameliorate UC by regulating inflammatory cytokine expression and oxidative stress levels. The present study demonstrates that S/O/W-E with programmed sequential release is a promising delivery vehicle to alleviate colitis by combining different action modes of different anti-inflammatory active ingredients, providing new insights into the synergistic amelioration of UC.

Local administration of platelet-derived growth factor-BB (PGDF-BB) and bone morphogenetic protein-2 (BMP-2) in a sequential release manner could substantially promote bone healing. To achieve this goal, a delivery system that could sustain the release of PGDF-BB and BMP-2 by way of temporal separation was developed. One type of PGDF-BB-encapsulated alginate microsphere and another type of BMP-2-encapsulated microsphere with a core-shell structure were respectively produced using emulsification methods. These two types of microspheres were then embedded into chitosan/glycerophosphate hydrogel for constructing composite gels. Some of them were found to be injectable at ambient temperature and had thermo-sensitive features near physiological temperature and pH. The optimally formulated composite gels showed the ability to control the release of PGDF-BB and BMP-2 in a sequential fashion in which PDGF-BB was released earlier than BMP-2. In vitro release patterns indicated that the release rates could be significantly regulated by varying the embedded amount of the factor-encapsulated microspheres, which can in turn mediate the temporal separation release interval between PGDF-BB and BMP-2. The released PDGF-BB and BMP-2 were detected to be bioactive based on their respective effects on Balb/c 3T3 and C2C12 cells. These results suggest that the presently developed composite gels have the potential for bone repair by synergistically utilizing the early chemotactic effect of PDGF-BB and the subsequent osteogenic and angiogenic functions of PDGF-BB and BMP-2.Keywords:microspheres;growth factor;composite hydrogel;sequential delivery;platelet-derived growth factor-BB;bone morphogenetic protein-2

Endogenous repair of osteochondral defect is usually limited by the insufficient number of cells in the early stage and incomplete cell differentiation in the later stage. The development of drug delivery systems for sequential release of pro-migratory and pro-chondrogenic molecules to induce endogenous bone marrow-derived mesenchymal stem cells (BMSCs) recruitment and chondrogenic differentiation is highly desirable for in situ osteochondral regeneration. In this study, a novel, all-silk-derived sequential delivery system was fabricated by incorporating the tunable drug-loaded silk fibroin (SF) nanospheres into a SF porous matrix. The loading efficiency and release kinetics of biomolecules depended on the initial SF/polyvinyl alcohol (PVA) concentrations (0.2%, 1% and 5%) of the nanospheres, as well as the hydrophobicity of the loaded molecules, resulting in controllable and programmed delivery profiles. Our findings indicated that the 5% nanosphere-incorporated matrix showed a rapid release of E7 peptide during the first 120 h, whereas the 0.2% nanosphere-incorporated matrix provided a slow and sustained release of Kartogenin (KGN) longer than 30 days. During in vitro culture of BMSCs, this functional SF matrix incorporated with E7/KGN nanospheres showed good biocompatibility, as well as enhanced BMSCs migration and chondrogenic differentiation through the release of E7 and KGN . Furthermore, when implanted into rabbit osteochondral defect, the SF nanosphere matrix with sequential E7/KGN release promoted the regeneration of both cartilage and subchondral bone. This work not only provided a novel all-silk-derived drug delivery system for sequential release of molecules, but also a functional tissue-engineered scaffold for osteochondral regeneration.

Background A sequential release co-delivery system is an effective strategy to improve anti-cancer efficacy. Herein, multicomponent-based liposomes (TET-CTM/L) loaded with tetrandrine (TET) and celastrol (CEL)-loaded coix seed oil microemulsion (CTM) were fabricated, which showed synergistic anti-liver cancer activities. By virtue of Enhanced Permeability and Retention (EPR) effect, TET-CTM/L can achieve efficient accumulation at the tumor site. TET was released initially to repair abnormal vessels and decrease the fibroblasts, and CTM was released subsequently for eradication of tumor tissue. Methods TEM (transmission electron microscopy) and DLS (dynamic light scattering) were adopted to characterize the TET-CTM/L. Flow cytometry was adopted to examine the cellular uptake and cytotoxicity of HepG2 cells. The HepG2 xenograft nude mice were adopted to evaluate the anti-tumor efficacy and systemic safety of TET-CTM/L.Results TEM images of TET-CTM/L showed the structure of small particle size of CTM within large-size liposomes, indicating that CTM can be encapsulated in liposomes by film dispersion method. In in vitro studies, TET-CTM/L induced massive apoptosis toward HepG2 cells, indicating synergistic cytotoxicity against HepG2 cells. In in vivo studies, TET-CTM/L displayed diminished systemic toxicity compared to celastrol or TET used alone. TET-CTM/L showed the excellent potential for tumor-targeting ability in a biodistribution study. Conclusion Our study provides a new strategy for combining anti-cancer therapy that has good potential not only in the treatment of liver cancer but also can be applied to the treatment of other solid tumors.

Bladder tissue engineering holds promise for addressing bladder defects resulting from congenital or acquired bladder diseases. However, inadequate vascularization significantly impacts the survival and function of engineered tissues after transplantation. Herein, a novel bilayer silk fibroin (BSF) scaffold was fabricated with the capability of vascular endothelial growth factor (VEGF) and platelet derived growth factor-BB (PDGF-BB) sequential release. The outer layer of the scaffold was composed of compact SF film with waterproofness to mimic the serosa of the bladder. The inner layer was constructed of porous SF matrix incorporated with SF microspheres (MS) loaded with VEGF and PDGF-BB. We found that the 5% (w/v) MS-incorporated scaffold exhibited a rapid release of VEGF, whereas the 0.2% (w/v) MS-incorporated scaffold demonstrated a slow and sustained release of PDGF-BB. The BSF scaffold exhibited good biocompatibility and promoted endothelial cell migration, tube formation and enhanced endothelial differentiation of adipose derived stem cells (ADSCs) in vitro. The BSF patch was constructed by seeding ADSCs on the BSF scaffold. After in vivo transplantation, not only could the BSF patch facilitate the regeneration of urothelium and smooth muscle, but more importantly, stimulate the regeneration of blood vessels. This study demonstrated that the BSF patch exhibited excellent vascularization capability in bladder reconstruction and offered a viable functional bioengineered patch for future clinical studies.

Inhibiting the function of P-glycoprotein (P-gp) transporter, which causes drug efflux through adenosine triphosphate (ATP)-dependent manner, has become an effective strategy to conquer multidrug resistance (MDR) of cancer cells. However, there remains challenges for effective co-delivery, sequential release of P-gp modulator and chemotherapeutic agent. In this work, a novel type of core–shell nanoparticle is reported. It can independently encapsulate a high amount (about 683 µg mg −1 ) of chemotherapeutic agent doxorubicin (DOX) in the mesoporous polydopamine (MPDA) core and glucose oxidase (GOx) in the zeolite imidazolate frameworks-8 (ZIF-8) shell, namely MPDA@ZIF-8/DOX+GOx. The fast release of GOx triggered by acid-sensitive degradation of the ZIF-8 shell consumes glucose to starve cancer cells for ATP deprivation and effective suppress ATP-dependent drug efflux in advance, and then effectively facilitates the accumulation of DOX in MCF-7/ADR cancer cells. Experiments in vitro and in vivo demonstrate that the fabricated nanosystem can dramatically improve anticancer effects for MDR through sequential release property and exhibit excellent biocompatibility. Overall, this work reveals new insights in the use of GOx for MDR treatment.

Periodontitis is a pathological disorder characterized by inflammation and tissue destruction; it is primarily attributed to bacterial infection and immune responses. Nevertheless, existing strategies for drug delivery in periodontal treatment are insufficient to accommodate the intricate anatomical morphology of the periodontium and the complex oral environment. Furthermore, the disordered release of antibacterial agents and bone-promoting substances are not conducive to the natural healing of periodontal tissues, thus resulting in suboptimal therapeutic outcomes for periodontitis. In this study, we successfully co-loaded methylene blue (MB) and poly(lactic-co-glycolic acid) microspheres encapsulating Sema3A into a dopamine-grafted sodium alginate hydrogel (SA-DA). Experimental evidence indicates that MB/Sema3A@SA-DA exhibits favorable injectability, adhesion, and compression resistance characteristics, thus allowing it to adapt to the intricate therapeutic conditions of the oral cavity. The prompt release of MB (within 14 h) demonstrates its high antibacterial efficacy against periodontal pathogens upon exposure to 660 nm light, thereby establishing a microenvironment conducive to osteogenesis devoid of bacterial interference. Furthermore, the protracted release of Sema3A (within 28 d) shows noteworthy osteogenic capabilities and facilitates the polarization of macrophages toward the M2 phenotype. In a rat model of periodontitis, MB/Sema3A@SA-DA can be perfectly applied in situ to local periodontal lesions, and the sequential release of MB and Sema3A effectively achieved antibacterial, anti-inflammatory, and osteogenic effects. This therapeutic system avoids the abuse of antibiotics, surgical trauma, and repetitive administration. Hence, MB/Sema3A@SA-DA emerges as a safe, effective, minimally invasive, comfortable, and cost-effective treatment strategy with considerable potential for clinical implementation in periodontitis management.
