Immobilizing bacitracin on titanium for prophylaxis of infections and for improving osteoinductivity: An in vivo study

Bin’en Nie,Haiyong Ao,Teng Long,Jianliang Zhou,Tingting Tang,Bing Yue

Bacitracin immobilized on the titanium (Ti) surface significantly improves anti-bacterial activity and biocompatibility in vitro . In the current study, we investigated the biologic performance (bactericidal effect and bone-implant integration) of bacitracin-modified Ti in vivo . A rat osteomyelitis model with femoral medullary cavity placement of Ti rods was employed to analyze the prophylactic effect of bacitracin-modified Ti (Ti-BC). Thirty-six female Sprague Dawley (SD) rats were used to establish the Ti implant-associated infection. The Ti and Ti-BC rods were incubated with and without Staphylococcus aureus to mimic the contaminated Ti rod and were implanted into the medullary cavity of the left femur, and sterile Ti rods were used as the blank control. After 3 weeks, the bone pathology was evaluated using X-ray and micro-computed tomography (micro-CT) analysis. For the investigation of the Ti-BC implant osseointegration in vivo , fifteen SD rats were divided into three groups (N   =   5), namely Ti, Ti-dopamine immobilized (Ti-DOPA), and Ti-BC. Ti rods were implanted into the left femoral cavity and micro-CT and histological evaluation was conducted after 12 weeks. The in vivo study indicated that Ti-immobilized bacitracin owned the prophylaxis potential for the infection associated with the Ti implants and allowed for the osseointegration. Thus, the multiple biofunctionalized Ti implants could be realized via immobilization of bacitracin, making them promising candidates for preventing the Ti implant-associated infections while retaining the osseointegration effects.

Lanthanum-Loaded ZIF Nanomaterials for Caries Prevention

Han Xiao,Yuping Qian,Lijun Zhao,Yiman Tang,Yuan Pan,Yunzi Long,Shihan Li,Yukun Chen,Jiale Sun,Chao Yuan,Ludan Zhang,Wenshu Ge

Dental caries represents one of the most widespread oral health challenges worldwide, impacting people of every age demographic. Traditional anticaries strategies primarily rely on fluoride, yet its chronic overuse may lead to health issues such as dental fluorosis. Lanthanum (La), a rare earth element, has emerged as a promising fluoride alternative due to its low toxicity and strong anticaries activity. However, studies have shown that the direct use of free lanthanum under physiological conditions may cause cellular structural damage, dose-dependent hemolytic reactions, and even liver and kidney fibrosis, highlighting the urgent need to optimize its delivery method. In this study, zeolitic imidazolate framework nanoparticles (La@ZIF) were used to encapsulate lanthanum, enabling its controlled and sustained release, thereby proposing a novel fluoride-free anticaries strategy. Three La@ZIF nanoparticles with varying La incorporation levels (20%, 40%, and 60%) were synthesized via a one-pot method and systematically characterized. Results demonstrated their excellent degradation performance and acid-neutralizing capacity under acidic conditions. In vitro experiments confirmed the favorable biocompatibility of La@ZIF and its ability to effectively promote enamel remineralization and restore surface hardness, achieving comparable efficacy to traditional fluoride treatments. In vivo studies further validated the remineralization potential of La@ZIF in a rat model, with no adverse effects observed on major organs. La@ZIF nanoparticles exhibit remarkable anticaries performance and biosafety, offering a new direction for developing fluoride-free anticaries materials.

Effect of wogonin, an active ingredient of Scutellariae Radix, on fish infected with Aeromonas hydrophila

Cui Jun,Zeng Dan,Guan Peipei,Jiang Ning

Exposure to pathogens induces hyperactivate inflammatory immune responses, oxidative stress and even death in fish. Therefore, suppressing excessive inflammatory responses is a means of treating fish diseases. Scutellariae Radix (SR), an herbal medicine, has anti-inflammatory and antioxidant properties. However, the active ingredients of SR that produce a marked effect and their mechanisms are poorly understood. In this study, 32 active ingredients of SR and their 124 target genes associated with anti-inflammatory effect were screened. These target genes in zebrafish were mainly enriched in biological processes associated with response to stress and stimulus. The KEGG enrichment analysis and pathway–pathway network analysis showed that the MAPK signalling pathway and nine other pathways were the key pathways associated with anti-inflammatory effect. A protein–protein interactions (PPI) network of target genes was constructed, and AKT2 and nine other genes were identified as the hub genes by cytoHubba. Combining the above results, we constructed an active ingredient–hub protein–key pathway network, showing that wogonin-AKT2 was the key regulated module. Molecular docking and SPR-based assays were used to investigate the possible interaction of wogonin with the AKT2 protein. The infected zebrafish and grass carp treated with wogonin displayed a higher survival rate and anti-inflammatory and antioxidant effects than the infected fish without wogonin treatment. Dietary wogonin also enhanced grass carp anti-inflammatory and antioxidant effects. These findings further reveal the molecular biological mechanism by which wogonin from SR exerts anti-inflammatory effects and provide a theoretical basis for the prevention of fish diseases.

Modified casein-stabilized amorphous calcium phosphate nanoparticles prevent dental caries by inhibiting the growth of Streptococcus mutans and promoting the remineralization of tooth enamel

Pengchao Fang,Xiaoxing Ye,Leijie Tian,Yibiao Chen,Xiaolan Li,Haiyan Hu

Dental caries, a prevalent oral disease, is caused by acid- producing plaque microorganisms-notably Streptococcus mutans ( S. mutans )-which drive enamel demineralization. Current prevention strategies, such as fluoride and chlorhexidine, primarily target planktonic bacteria but exhibit limited efficacy against biofilms and neglect enamel remineralization, thereby reducing their therapeutic efficacy. To address these limitations, we developed dual-functional nanoparticles (AXCP NPs) that can eradicate S. mutans biofilm and promote enamel remineralization. The amphiphilic polymer Arg-XOS-CPP (AXC) was synthesized by covalently conjugating oligosaccharide carbonyl groups to the amino residues of L-arginine and casein. Self-assembly of AXC with calcium and phosphate ions yielded hybrid nanoparticles (AXCP NPs). Experimental results indicated that AXCP NPs had no effect on the viability of HaCaT cells. Under acidic conditions, protonation of arginine triggered a surface charge reversal from negative to positive, enabling targeted binding to negatively charged biofilms. As expected, AXCP NPs exhibited potent anti- S. mutans activity (MIC: 640   μg/mL) and remarkable biofilm eradication efficacy (>80% clearance). In vitro release and remineralization assays demonstrated that AXCP NPs continuously released calcium and phosphorus ions, regulating the mineralization process and effectively restoring enamel functionality. Furthermore, in mice caries model, localized application of AXCP NPs significantly suppressed biofilm accumulation on tooth surface and restored mineral density in demineralized enamel, inhibiting caries progression.

Mussel-Inspired Clickable Antibacterial Peptide Coating on Ureteral Stents for Encrustation Prevention

Qin Yao,Jinyi Zhang,Guoqing Pan,Binghai Chen

Long-term indwelling catheters or stents often cause complications like infection, encrustation, hematuria, pain, and so on. The source of these problems is bacteria, which can form biofilms on the stents to reduce antibiotic sensitivity and produce urease to form encrustation by increasing the urine pH. Urinary tract infection (UTI) can aggravate the body damage and even seriously endanger lives, and the encrustation will block the stents, which can cause hydronephrosis and renal function damage. Therefore, the prevention of UTI and encrustation represents a great challenge in clinical ureteral stent uses. In this work, a clickable mussel-inspired peptide and antimicrobial peptide (AMP) were used to functionalize the commercial stents’ surfaces to inhibit long-term infection and encrustation caused by bacteria. Copper (Cu) ions were used to coordinate the mussel-inspired peptide to improve the stability. The AMP with an azido group was clicked to the mussel-inspired Cu-coordinated peptide coating through click chemistry. The bio-inspired antibacterial coating was constructed with excellent stability, bactericidal properties, and improved biological compatibility. In in vitro and in vivo experiments, it was further found that the coating showed bactericidal and encrustation reduction abilities. This study thus developed an effective, safe, and stable AMP coating on urinary stents/catheters capable of long-term antibacterial and encrustation inhibition.

Resources recycle of traditional Chinese medicine (TCM) wastewater 2: The UF-FO-MD hybrid system and wetting prevention

Guanhua Li,Sheng Li

Wastewater discharge of traditional Chinese medicine production is a serious threat to the environment in China. This study investigated the effectiveness of the UF-FO-MD hybrid system for resources recycle from the TCM wastewater. Real TCM wastewater was treated with the self-assemble UF-FO-MD hybrid system. Moreover, the natural surfactant rejection and the FO fouling impact were evaluated by separate FO operation with synthetic TCM wastewater, and MD wetting was also assessed. Results show that the UF-FO-MD hybrid system successfully isolated the macromolecular substances, bioactive compounds and water in the TCM wastewater for recovering useful resources. Natural surfactants in the TCM wastewater could be effectively rejected by FO, avoiding the subsequent MD wetting problem which has occurred in our previous study on the resource recovering with the UF-MD hybrid system. However, FO membrane fouling might be an issue affecting the recovery efficiency of bioactive compounds from TCM wastewater. The bioactive compounds could not be completely concentrated due to potential adsorption on the fouling layer .

Evaluation on prevention and treatment of cuminaldehyde in culture of shrimp against white spot syndrome virus

Xu Zhang,Li-Hua Hu,Da-Wei Song,Yang Hu,Jiong Chen

With the continuous expansion of cultural scale and the deterioration of cultural environment recently, various viral diseases occur frequently in cultured shrimp industry. White spot disease (WSD) caused by white spot syndrome virus (WSSV) results in a high mortality as well as a huge economic loss of shrimp culture. As a kind of Chinese herbal medicine extract, the application of cuminaldehyde in prevention and treatment of WSSV was investigated in this study. The data showed that cuminaldehyde at 40 μmol/L could significantly inhibit WSSV replication in shrimp within the highest inhibition rate of 89% at 72 hpi. The survival rate of WSSV-infected shrimp was increased by >50% after continuous treatment of cuminaldehyde versus the virus group at 144 hpi. Obviously, the horizontal transmission of WSSV was inhibited by cuminaldehyde in a static cohabitation challenge model, further weakening the spread of the virus to reduce the mortality of the virus-infected shrimp. Meanwhile, cuminaldehyde increased resistance of shrimp to WSSV by activating the innate immune response, which was essential for prevention of WSSV. Thus, cuminaldehyde could potentially be used as a therapeutic agent with a great potential in culture of shrimp seedling and may also be suitable for applications in pond aquaculture settings against WSSV transmission.

Study of Huperzine A derivatives with extended protection against soman intoxication

Yalan Cui,Xuejun Chen,Jingjing Shi,Qian Jin,Ruihua Zhang,Tong Shi,Chen Wang,Liqin Li

Pre-administration of huperzine A (Hup A) was validated to prevent poisoning from exposure to nerve agents (NAs) by reversibly inhibiting acetylcholinesterase (AChE). However, like the currently commonly used reversible inhibitors, Hup A has a short half-life and is unable to produce a long-term preventative effect. To extend the protective time of Hup A against NAs, 42 derivatives with a C N bond were designed based on the structure of Hup A in this study. All designed derivatives showed good binding capability with AChE via molecular docking. Six compounds (H3, H4, H11, H14, H16, and H25) with representative structures were selected for synthesis by Schiff base reaction, and their structures were stable. The modified Ellman's method showed the six compounds concentration-dependently inhibited AChE, and the half maximal inhibitory concentration (IC 50 ) were higher than that of Hup A. Pretreatment of AChE with the derivatives significantly increased the IC 50 of soman . In vivo experiments demonstrated H3, H4, H14, H16, and H25 had longer protective capacities against 1 × LD 95 soman-induced death in mice than Hup A. The 12 h protective index showed that the protective ratios of H3, H4, H14 and H16 were 2.31, 1.85, 2.23 and 1.99 respectively, better than that of Hup A. The extended protection of the derivatives against soman may be explained by their transformation to Hup A in vivo. Furthermore, all six compounds showed lower acute oral toxicity than Hup A. Overall, our study provided an optional strategy to acquire pretreatment agents for NAs with extended action and low toxicity.

Multi-Omics Approaches for Liver Reveal the Thromboprophylaxis Mechanism of Aspirin Eugenol Ester in Rat Thrombosis Model

Qi Tao,Ning Ma,Liping Fan,Wenbo Ge,Zhendong Zhang,Xiwang Liu,Jianyong Li,Yajun Yang

Aspirin eugenol ester (AEE) is a novel medicinal compound synthesized by esterifying aspirin with eugenol using the pro-drug principle. Pharmacological and pharmacodynamic experiments showed that AEE had excellent thromboprophylaxis and inhibition of platelet aggregation. This study aimed to investigate the effect of AEE on the liver of thrombosed rats to reveal its mechanism of thromboprophylaxis. Therefore, a multi-omics approach was used to analyze the liver. Transcriptome results showed 132 differentially expressed genes (DEGs) in the AEE group compared to the model group. Proteome results showed that 159 differentially expressed proteins (DEPs) were identified in the AEE group compared to the model group. Six proteins including fibrinogen alpha chain (Fga), fibrinogen gamma chain (Fgg), fibrinogen beta chain (Fgb), orosomucoid 1 (Orm1), hemopexin (Hpx), and kininogen-2 (Kng2) were selected for parallel reaction monitoring (PRM) analysis. The results showed that the expression of all six proteins was upregulated in the model group compared with the control group. In turn, AEE reversed the upregulation trend of these proteins to some degree. Metabolome results showed that 17 metabolites were upregulated and 38 were downregulated in the model group compared to the control group. AEE could reverse the expression of these metabolites to some degree and make them back to normal levels. The metabolites were mainly involved in metabolic pathways, including linoleic acid metabolism, arachidonic acid metabolism, and the tricarboxylic acid (TCA) cycle. Comprehensive analyses showed that AEE could prevent thrombosis by inhibiting platelet activation, decreasing inflammation, and regulating amino acid and energy metabolism. In conclusion, AEE can have a positive effect on thrombosis-related diseases.

Magnetically-drivable, pH-responsive Fe3O4/ZIF-8 coating on implant for biofilm prevention and treatment

Kecheng Quan,Yupu Lu,Ting Zhang,Shuren Wang,Hao Tang,Chunhao Yu,Yongsheng Zhou,Yunsong Liu,Dandan Xia,Yufeng Zheng

Antimicrobial coatings with the capabilities to prevent and treat biofilm infections on implant are promising surface treatment strategies, but it is difficult to eradicate an already-formed biofilm by the coating. Magnetically-drivable coatings can physically destruct the surface-attached biofilm to enhance antimicrobial killing. However, current magnetic coatings cannot inhibit the initial bacteria growth and the demand of coating stability restricts their magnetic controllability. Herein, a magnetically-drivable, pH-responsive coating is developed by simply mixing Fe 3 O 4 and ZIF-8 nanoparticles on titanium surface. In physiological pH, Fe 3 O 4 nanoparticles are stable and non-magnetically drivable on surface due to charge attractions and electrostatic interactions. While in acidic pH caused by bacterial growth, ZIF-8 degradation is significantly accelerated, releasing Zn 2 + and recovering the magnetic controllability of Fe 3 O 4 nanoparticles. The released Zn 2+ kills the initially-attached bacteria, inhibiting subsequent biofilm formation. Once a biofilm has formed, Fe 3 O 4 nanoparticles can be magnetically pulled-off to destruct the biofilm structure and then enhance antibiotic killing. The Fe 3 O 4 /ZIF-8 coating shows excellent biocompatibility and osteogenesis to compare with bare Ti disc. Animal experiments further proved the in-vivo antimicrobial and osteogenic performances of the coating. Therefore, this work put forward a new surface strategy to solve biofilm-related infections on implants.

A multi-functional drug delivery system based on polyphenols for efficient tumor inhibition and metastasis prevention

Si Chen,Jin-Xuan Fan,Di-Wei Zheng,Fan Liu,Xuan Zeng,Guo-Ping Yan,Xian-Zheng Zhang

Although chemotherapy is the most common method in clinical therapeutics with a straightforward mechanism, conventional anti-tumor drugs are still almost incapable of preventing the occurrence of tumor metastasis. In this study, we developed a multi-functional drug delivery system EINP@DOX consisting of a tea-derived polyphenol EGCG, iron ions and DOX. The system integrated the functions of tumor inhibition, diagnosis and metastasis prevention to achieve a systematic tumor treatment. The nanoscale size of EINP@DOX facilitated its accumulation in tumor tissues by means of the enhanced permeability and retention (EPR) effect, and it was then transferred to endosomes. The weakly acidic microenvironment in the endosomes of the tumor cells could destroy the coordination structure of EINP@DOX to realize the release of DOX for tumor therapy. Furthermore, the dissociative EGCG played the role of an adjuvant to restrain EMT and down-regulate the MMP levels, which could prevent the occurrence of tumor metastasis. Meanwhile, iron ions as superior magnetic resonance imaging (MRI) contrast agents provided visual evidence for the accurate location of EINP@DOX. In vitro and in vivo studies demonstrated that EINP@DOX showed a remarkable performance in tumor diagnosis and excellent therapeutic efficacy, inhibiting the metastasis of tumor cells effectively at the same time.
