Simply converting color signal readout into thermal signal readout for breaking the color resolution limitation of colorimetric sensor

Mingyue Luo,Xin Xue,Honghong Rao,Hongqiang Wang,Xiuhui Liu,Xibin Zhou,Zhonghua Xue,Xiaoquan Lu

Traditional colorimetric sensors often suffer from the low color resolution and the not well-defined boundary between the yes and no answers, especially for distinguishing the similar color readouts with the naked eye. Herein, a simple strategy is communicated for greenly converting colorimetric sensor into a photothermal sensor, which is based on the simple color-to-thermal (temperature) signal readout conversion. The colorimetric system based on simply mixing commercially available chemicals of 3,3,5,5-tetramethylbenzidine (TMB) and silver ions (Ag + ), provides a straightforward blue color readout owing to the generation of oxidized TMB (TMBox). In the presence of the target alkaline phosphatase (ALP), ascorbic acid phosphate (AAP) in the system could be hydrolyzed into ascorbic acid (AA), subsequently reducing the blue colored TMBox into colorless TMB. The blue-to-colorless change-based colorimetric system also exhibits the highly sensitive near-infrared (NIR) laser-driven photothermal response, allowing the successful thermometer-detectable signal-based assay for point-of-care testing (POCT) of ALP levels just using a common thermometer. This study not only demonstrated a regulatory strategy to convert color-based signal readout into thermal (temperature)-based signal readout, but also established a facile method to design affordable thermometric sensors for POCT and instrument-free bioanalysis , especially in low-resource settings.

Real-time monitoring of mercury(II) in water and food samples using a quinoline-based ionic probe

Siying Che,Linlin Yin,Ming Chen,Yao Fan,Anni Xu,Chunsong Zhou,Haiyan Fu,Yuanbin She

Herein, a novel ionic fluorescent probe for mercury(II) detection is presented consisting of a functional quinoline-based IL. Interestingly, the probe displayed high sensitivity (0.8 nM) and selectivity through the regulation function of electrostatic attraction, where its performance was significantly superior to that of quinoline probes without negative charge. Furthermore, the probe was found to exhibit two different fluorescent signals and colorimetric signals in the presence of different concentrations of mercury(II), which was consistent with the reaction mechanisms of the generation of large conjugated systems and the formation of anion-mercury(II) complexes. Moreover, this probe could be further loaded on a simple filter paper to serve as a visual paper sensor due to its adequate response time of less than 5 s. This regulation function strategy of electrostatic attraction has excellent potential for use in the precise detection of targeted analytes in real complex samples with improved accuracy and selectivity.

Optical Visualization of Cadmium Ion Using a Label-Free Nucleic Acid Aptamer for Recognition

Qing Zhang,Chunrong Zhang,Nan Li,Zhao Dai

Aptamer-based biosensors have been demonstrated to provide real-time physical and chemical signals for biomedicine, food safety, drug identification, and metal ion determination. In this study, cadmium ion (Cd2+) was determined in drinking water by color signaling using the specific binding of the aptamer and target by its regulation of the catalytic capacity of gold nanoparticles (AuNPs). When sodium borohydride (NaBH4) and a probe with a catalytic function were present, the solution containing 4-nitrophenol changed from yellow to colorless. Under optimal conditions, the naked eye was sufficient to determine Cd2+ with a limit of detection (LOD) of 5 μM. The discoloration time showed a positive correlation with Cd2+ concentration up to 30 μM. This method was label-free, efficient, rapid, and selective for Cd2+ in water without expensive instruments, suggesting practical applications.

A dual-readout paper-based sensor for on-site detection of penicillinase with a smartphone

Jia Xu,Xujiang Chen,Habib Khan,Li Yang

A dual-readout paper-based sensor with smartphone detection is developed for quantitative determination of penicillinase. The sensor is accomplished with the aid of penicillinase-mediated alginate hydrogelation, which changes both the viscosity and pH of the reaction solution. The solution is dropped on a mixed cellulose ester filter paper for sensing, which is coated with a color indicator, bromothymol blue-cetyltrimethylammonium bromide-alginate complex. Dual signals of the targets are easily achieved from the sensing spot on the paper since the modulations of viscosity and pH of the reaction solution would result in changes of diffusion diameter and color of the spot, respectively. A smartphone-based readout device is developed to record and analyze both the diffusion diameter and color signal for quantitative detection of penicillinase. The wide linear range (8.00 × 10 −3 - 2.67 mU/μL for distance-readout and 8.00 × 10 -2 to 2.67 mU/μL for color-readout) with low limit-of-detection (2.67 × 10 -3 mU/μL for distance-readout and 2.67 × 10 -2 mU/μL for color-readout) have been achieved using the dual-readout paper-based sensors. The proposed method is successfully applied for the detection of penicillinase in milk samples with excellent sensitivity and reproducibility, showing its potential value for analysis of complex real samples.
