Biofilm inhibition and eradication activity of citral and borneol against foodborne bacteria

Wen Wang,Tom Coenye,Jianyu Su,Sheng-Xiang Qiu

Biofilms growing on food surfaces, packaging, equipment and processing environments bring huge risks to food safety and result in economic losses. Natural components are receiving increasing interest as “green” antimicrobial agents due to their biocompatibility, broad-spectrum antimicrobial properties, and antibiofilm potential. As a traditional adjuvant in Chinese medicine, borneol has the potential to enhance citral’s antibiofilm efficacy. In this study, the antibiofilm activities of citral, borneol, and the combination citral and borneol (BC) against various foodborne bacteria were evaluated using multiple techniques. Light microscopy, crystal violet (CV) staining, and the 2,3-bis (2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylaminocarbonyl]-2H-tetrazolium hydroxide (XTT) assay were employed to monitor biofilm formation. Additionally, biofilm eradication was assessed against Staphylococcus aureus and Escherichia coli biofilms grown on polycarbonate membranes. Protein leakage was measured, and field emission scanning electron microscopy (FESEM) was used to examine cell membrane integrity. Results showed that borneol and citral acted synergistically to inhibit biofilms. Borneol, citral, alone or together inhibited biofilm formation in a concentration dependent manner, and they have the ability to inactivate or kill at least 90.7 ± 1.9 % bacteria in preformed biofilms. Both borneol and citral disrupted the membrane integrity of the bacteria, increased permeability, and ultimately led to cell death. These findings suggest that the combination citral and borneol has the potential to be developed as a food preservative or bactericidal agent for controlling foodborne bacterial contamination.

Nucleic acid strand displacement for indirect determination of foodborne bacteria by capillary electrophoresis and its application in antagonism and bacteriostasis studies

Shaoyan Zhang,Qingqing Wang,Jinfeng Ning,Wei Wang

Foodborne bacteria threaten human's health. Capillary electrophoresis (CE) is a powerful separation means for the determination of bacteria. Direct separation of bacteria suffers from the shortages of low resolution, channel adsorption, and bacterial aggregation. In this work, a method of nucleic acid strand displacement was developed to indirect separate the bacteria by CE. DNA complexes, consisting of probes and aptamers, were mixed with the three bacteria Staphylococcus aureus , Escherichia coli , and Pseudomonas aeruginosa . The aptamers could specifically bond with bacteria and release the probes. Through the separation of the probes, the bacteria could be indirectly determined by CE. This method avoided the shortages of direct separation of bacteria. Under the optimized conditions, the three probes for the bacteria could be separated and detected within 2.5 min by high-speed CE with laser-induced fluorescence detection. The limits of detection for the bacteria were in the range 4.20 × 10 6 to 1.75 × 10 7 CFU/mL. Finally, the developed method was applied on the study of antagonism of the coexistent bacteria to reveal the relationship between them. Furthermore, the efficiency of bacteriostasis of three traditional Chinese medicines, Coptis chinensis , Schisandra chinensis , and honeysuckle, was also studied by this method.

Ultrasensitive electrochemical biosensor for bacteria detection based on Fe3O4@COF-AuNPs and trigging isothermal circular amplification

Jialin Zhang,Ming Zhou,Linna Yang,Bin Huang,Kangqiang Lu,Herui Wen,Jiali Ren

Foodborne bacterial infections pose serious threats to human health, leading to illnesses and even fatalities. The key to solve this problem is to develop fast and effective sensor for sensitive detection of bacteria. In this study, an electrochemical biosensor based on magnetic nanocomposite material Fe 3 O 4 @COF-AuNPs and trigging isothermal circular amplification (TICA) was constructed for detection of foodborne bacteria. In this design, a hairpin aptamer probe (HAP)-bound DNA monolayer was designed, serving as the substrate for the electrode to increase surface area. The Escherichia coli ( E. coli ) was selected as the target. The presence of E. coli caused the configuration change of the HAP and allowed the occurrence of TICA with the use of the Klenow Fragment polymerase (KF-Poly), thus the HAPs were away from the DNA monolayer. After adding the catalytic magnetic nanocomposite material Fe 3 O 4 @COF-AuNPs, it was immobilized on the DNA monolayer of the electrode through DNA complementary pairing and exhibited high electrocatalytic behavior towards the reduction of 4-nitrophenol. This enabled ultrasensitive detection of E. coli , with the detection limit (LOD) as low as 10 CFU/mL. The detection time was 1 h. This designed electrochemical sensor showed high selectivity, stability, and reproducibility, which offered a new avenue for detection of foodborne bacteria.

Food-borne bacteria analysis using a diatomite bioinspired SERS platform

Yikai Chen,Binggang Ye,Mengling Ning,Meng Li,Yixuan Pu,Zhiming Liu,Huiqing Zhong,Chaofan Hu,Zhouyi Guo

Rapid and sensitive detection of food-borne bacteria has remained challenging over the past few decades. We propose a surface-enhanced Raman scattering sensing strategy based on a novel bioinspired surface-enhanced Raman scattering substrate, which can directly detect dye molecular residues and food-borne pathogen microorganisms in the environment. The surface-enhanced Raman scattering platform consists of a natural diatomite microporous array decorated with a metal-phenolic network that enables the in situ reduction of gold nanoparticles. The as-prepared nanocomposites display excellent surface-enhanced Raman scattering activity with the lowest limit of detection and the maximum Raman enhancement factor of dye molecules up to 10−11 M and 1.18 × 107, respectively. For food-borne bacterial detection, a diatomite microporous array decorated with a metal polyphenol network and gold nanoparticle-based surface-enhanced Raman scattering analysis is capable of distinguishing the biochemical fingerprint information of Staphylococcus aureus and Escherichia coli, indicating the great potential for strain identification.
