Detection of stx1 and stx2 and subtyping of Shiga toxin-producing Escherichia coli using asymmetric PCR combined with lateral flow immunoassay

Shan Shan,Yanmei Huang,Zhaohong Huang,Zhonger Long,Chengwei Liu,Xuelong Zhao,Keyu Xing,Xiaoyue Xiao,Jintao Liu,Yunhong Huang,Weihua Lai,Daofeng Liu

Shiga toxin-producing Escherichia coli (STEC) is known as a kind of foodborne pathogens that can produce Shiga toxin type1, type 2, or both, encoded by stx1 and stx2 genes, respectively. Rapid and accurate detection of STEC and stx1 / stx2 subtyping assay in food and environment is essential for foodborne outbreak investigation. In this study, a method for the accurate and rapid subtyping of STEC single colony using asymmetric polymerase chain reaction (asPCR) combined with lateral flow immunoassay (LFIA) was established. AsPCR was conducted using biotinylated stx1 and digoxin-labeled stx2 primers to obtain several single stranded DNA amplicons labeled with biotin or/and digoxin. Subsequently, these amplicons were tagged with specific probes labeled with carboxylate-modified red polystyrene microspheres using sequential hybridization. Finally, STEC with different Shiga toxin gene types could be distinguished depending on the appearance of different test lines on an LFIA strip. Thus, the STEC single colony could be identified and subtyped accurately and conveniently using this method. This method showed high specificity. Twenty-four strains of STEC in milk were detected and subtyped using our method, which combines asPCR with LFIA.

Effect of UVC Light-Emitting Diodes on Pathogenic Bacteria and Quality Attributes of Chicken Breast

Wenwen Wang,Dianbo Zhao,Ke Li,Qisen Xiang,Yanhong Bai

ABSTRACT This study was conducted to investigate the inactivation of foodborne pathogens and the quality characteristics of fresh chicken breasts after UVC light-emitting diode (UVC-LED) treatment. Fresh chicken breasts were separately inoculated with Salmonella Typhimurium, Escherichia coli O157:H7, and Listeria monocytogenes at initial populations of 6.01, 5.80, and 6.22 log CFU/cm 2 , respectively, and then treated with UVC-LED irradiation at 1,000 to 4,000 mJ/cm 2 . UVC-LED irradiation inactivated the test bacteria in a dose-dependent manner. After UVC-LED treatment at 4,000 mJ/cm 2 , the populations of Salmonella Typhimurium, E. coli O157:H7, and L. monocytogenes on chicken breasts were decreased by 1.90, 2.25, and 2.18 log CFU/cm 2 , respectively. No significant changes ( P > 0.05) were found in color, pH, texture, and thiobarbituric acid–reactive substances of chicken breasts following UVC-LED irradiation at doses ≤4,000 mJ/cm 2 . These results indicate that UVC-LED radiation is a promising technology for reducing the level of microorganisms while maintaining the physicochemical characteristics of poultry meat.

Shiyu He,Yanzhi Huang,Yingwei Ma,Haoyan Yu,Bo Pang,Xingxing Liu,Caihong Yin,Xiaomu Wang,Yuan Wei,Yuling Tian,Chao Zhao,Kun Xu,Juan Wang,Chunping Lv,Xiuling Song,Minghua Jin

Foodborne pathogen contamination is a major safety issue for many foods and is causing concern worldwide. In this study, a detection system based on magnetic separation, multiplex PCR (MPCR) and capillary electrophoresis (CE) technologies was developed for the simultaneous detection of four foodborne pathogens. Magnetic separation technology is used to rapidly capture pathogenic bacteria in food samples, and then a combination of MPCR and CE can be used to greatly increase detection sensitivity. The detection limit for bacterial DNA reached 10 −5 –10 −7 ng μL −1 and in the analysis of mocked food samples, the assay showed good sensitivity for bacterial detection ranging from 10 1 to 10 5 CFU mL −1 with excellent specificity. Compared to similar detection methodologies, this technique avoids the need for time-consuming enrichment cultures, is more sensitive, and can be used to assay simultaneously four foodborne pathogens. Graphical and Lay Summary Food-borne bacteria contamination is one of the main problems of food safety, which seriously affects the economic development of many countries. In this study, the author achieved rapid separation and enrichment of multiple pathogens in a complex sample matrix through magnetic nano carbon dots, and constructed a rapid detection system for four foodborne pathogens by combining multiplex PCR and capillary electrophoresis technologies. This work shows great potential and application value in food safety monitoring.

Effects of UVC light-emitting diodes on microbial safety and quality attributes of raw tuna fillets

Liumin Fan,Xiao Liu,Xiuping Dong,Shanshan Dong,Qisen Xiang,Yanhong Bai

Ultraviolet light-emitting diodes (UV-LEDs) have become widespread for decontaminating the surfaces of food products and their packaging as a safer alternative to traditional mercury-vapor based UV lamps. This study aimed to investigate the effect of UVC- LEDs (275 nm) on the inactivation of foodborne pathogens and quality attributes of raw tuna fillets. Raw tuna fillets inoculated with cocktails of Salmonella Typhimurium, Listeria monocytogenes , and Escherichia coli O157:H7 were exposed to UVC-LEDs at 500-4000 mJ/cm 2 . UVC-LED treatments of single-layer tuna fillets at a dose of 4000 mJ/cm 2 resulted in 1.31-, 1.86-, and 1.77- log reduction of S . Typhimurium, L . monocytogenes , and E . coli O157:H7, respectively. The pH value, water-holding capacity, metmyoglobin, texture, and color characteristics of tuna fillets were not significantly ( p  > 0.05) changed following the UVC-LED treatment at radiation doses up to 4000 mJ/cm 2 . Nonetheless, the UVC-LED irradiation led to increased levels of lipid oxidation in tuna fillets. Overall, the present study demonstrates that UVC-LEDs are a promising technology to reduce microbial flora and maintain the physicochemical properties of fish products.

Exploration of Novel Antimicrobial Agents against Foodborne Pathogens via a Deep Learning Approach

Huixi Zhang,Shanxue Jiang,Haishu Sun,Yushuang Li,Zhiliang Yao

The emergence of antibiotic-resistant bacteria poses a severe threat to food safety and human health, necessitating an urgent search for novel antimicrobial agents that can be applied in the food industry. This study utilizes a deep learning approach to establish the optimal models for antibacterial activity against foodborne pathogens, particularly Escherichia coli and Staphylococcus aureus, as well as for predicting carcinogenicity. These optimal models are applied to screen natural products from the COCONUT database, resulting in the identification of 130 compounds with both antibacterial activity and noncarcinogenic properties. Two natural products, bis(hexamethylene)triamine and N-phenethylbiguanide, are selected for experimental validation of their antibacterial activity. The confirmation of antimicrobial properties validates the reliability of the models developed in this study. By providing an innovative approach for identifying antimicrobial agents for foodborne pathogens, this research offers new insights for discovering effective antimicrobials in an efficient manner.

Imprinted membrane-covalent organic framework platform for efficient label-free visual detection of Listeria monocytogenes and Salmonella typhimurium in food samples

Yuanyuan Guo,Hongqian Cao,Wenli Di,Xibao Gao

Background Rapid and sensitive detection of foodborne pathogens in food plays a crucial role in controlling outbreaks of foodborne diseases, of which Listeria monocytogenes and Salmonella typhimurium are representative and notable pathogens. Thus, it's of great importance to achieve the effective detection of these pathogens. However, the most common detection methods (culture-based technique, Polymerase Chain Reaction and immunological methods) have disadvantages that cannot be ignored, such as time-consuming, laborious, complex sample preparation process, and the possibility of cross-reaction. Hence, it is essential to develop a facile detection method for the pathogens with high sensitivity and specificity to avoid the above-mentioned disadvantages. Results We report a label-free visual platform for the simultaneous capture and detection of Listeria monocytogenes and Salmonella typhimurium . For the first time, we have prepared polydimethylsiloxane-Chromotrope 2R membrane which serves as the substrate for bacterial capture and enrichment through the formation of specific recognition sites. The positively charged Pt-covalent organic framework combines with the pathogens through surface charge interaction, thereby the label-free sandwich platform is formed. Remarkable peroxidase activity of Pt-covalent organic framework converts the conversion of bacterial quantity into amplified color signal by catalyzing 3,3′,5,5′-Tetramethylbenzidine to oxidized 3,3′,5,5′-Tetramethylbenzidine. The platform demonstrates the capability to identify two representative food-borne pathogens within a time frame of 100 min, exhibiting high sensitivity and excellent specificity without the interference from non-target bacteria. The limit of detection of the visual platform toward Listeria monocytogenes and Salmonella typhimurium was 1.61 CFU mL −1 and 1.31 CFU mL −1 , respectively. And the limit of quantification toward Listeria monocytogenes and Salmonella typhimurium was 4.94 CFU mL −1 and 2.47 CFU mL −1 , respectively. The relative standard derivations of the visual platform for both bacteria were lower than 4.9 %. Furthermore, our proposed platform has obtained reliable and satisfactory results on analyzing diverse food samples. Significance This research expands the application of a label-free platform combined with unlabeled nanocomponents in the rapid isolation and detection of diverse of food-borne pathogens. The platform possesses the advantages of simple operation and real-time monitoring, without complicated sample pretreatment process. The whole detection process can realize the simultaneous monitoring of Listeria monocytogenes and Salmonella typhimurium within 100 min. Furthermore, it is also of reference significance for the detection of other common pathogens.

A label-free and naked-eye fluorescence turn-on assay for one-pot LAMP detection of foodborne pathogens using AuNCs-Cu2+ complex

Fuyuan Huang,Paner Jiang,Yiliang Chen,Weiwei Wang,Yaxin Han,Chenlu Chen,Guangxin Xiang,Sheng Ye,Laibao Zheng

Loop-mediated isothermal amplification (LAMP) is a rapid and sensitive approach for foodborne pathogen detection; however, a simple, user-friendly, and naked-eye readable readout method remains a challenge. Herein, we synthesized bovine serum albumin stabilized gold nanoclusters (BSA-AuNCs), which exhibit fluorescence quenching upon copper ions (Cu 2+ ) binding and fluorescence recovery in the presence of phosphate ions (PPi). Leveraging this property, we developed a label-free fluorescence turn-on assay for naked-eye LAMP detection by monitoring PPi accumulation under UV light. To mitigate potential cross-contamination risks inherent in LAMP workflows, we integrated the BSA-AuNCs-Cu 2+ complex into the reaction lid, enabling a closed, one-pot detection system. This method demonstrated high sensitivity, detecting Salmonella enterica at concentrations as low as 10 1  CFU/mL within 40 min (excluding nucleic acid extraction process). With its customizable primers and naked-eye visual detection capability, this assay offers a versatile and practical tool for rapid pathogen screening in diverse applications.

Phage/aptamer dual-encoded hydrogel arrays integrated with microfluidic platforms for simultaneous detection of multiple food pathogens

Jie Xu,Siwei Zhou,Yuqi Zhang,Huihui Wu,Tianhua Li,Lei Tan,Ning Gan,Jincan He

Food pathogens pose significant threats to public health. This study presents a universal, sensitive and time-efficient microfluidic chip electrophoresis (MCE) platform for simultaneously detecting multiple food pathogens based on phage/aptamer dual-encoded hydrogel arrays. The arrays are comprised of “all-in-one” hydrogel slices embedded with the capturing probe of specific phages and sensing probe of ATP aptamer/complementary chain (A-Apt/cDNA) hybrid structures. The live bacteria are specifically captured and lysed by the corresponding phages to generate copious amounts of ATP. The ATP further combines with the A-Apt/cDNA hybrids to release the cDNA strands with various lengths from the hydrogel pores into the supernatant. Therefore, cDNA strands whose lengths are specific for each bacteria function as markers for ATP, the level of which represents the number of living bacteria. The qualitation and quantification of multiple bacteria is realized by isolating and analyzing various cDNA using MCE. Taking three food pathogens (i.e., E . coli , Salmonella typhimurium , Staphylococcus aureus ) as target models, the platform were performed within 60 min, with detection limits of 20 CFU/mL, 30 CFU/mL and 15 CFU/mL, respectively. This study offers a universal and rapid strategy for multiple food pathogens analysis.

Morphophysiological Changes in Staphylococcus aureus Biofilms Treated with Plasma-Activated Hydrogen Peroxide Solution

Jianying Zhao,Jing Qian,Ji Luo,Mingming Huang,Wenjing Yan,Jianhao Zhang

Featured ApplicationDuring food processing, the surface of food and processing equipment is easily contaminated by microorganisms, which will cause food spoilage or the spread of disease, threatening food safety. Plasma-activated solution can be used as a novel food disinfectant, enhancing food quality. Plasma-activated solution will not leave chemical residues or cause environmental pollution. It is a green and environmentally friendly disinfectant and promises to replace traditional chemical disinfectants.AbstractPlasma-activated solution has attracted more attention in the food industry due to no chemical residue and good bacteriostatic properties. This study aimed to evaluate the effects of plasma-activated hydrogen peroxide solution (PAH) on the morphophysiology ofStaphylococcus aureusbiofilms. PAH was prepared using dielectric-barrier-discharge plasma and incubated withS. aureusbiofilms for 0–40 min. Changes in biofilm morphophysiology were evaluated with laser scanning confocal microscopy, electron microscopic images, reactive oxygen species (ROS) content, metabolic capacity, and 1% agarose gel. Results indicated that the population ofS. aureusin the biofilms was reduced by 4.04-log after incubation with PAH for 30 min. The thickness and metabolic capacity of biofilms were decreased, the ROS content and DNA fragments of bacteria increased after PAH treatments. Data suggested that PAH treatments significantly destroyed the morphophysiology ofS. aureus(ATCC 6538) biofilms and could be considered as a valuable anti-biofilm technology to reduce foodborne pathogens on food and/or in food facilities. Keywords:foodborne pathogen;cold plasma;nonthermal antimicrobial treatment;metabolic capacity;DNA damage

Antibacterial activities of R-(+)-Limonene emulsion stabilized by Ulva fasciata polysaccharide for fruit preservation

Ping Shao,Haoya Zhang,Ben Niu,Ligang Jiang

It was previously found that R-(+)-Limonene emulsion could be stabilized by Ulva fasciata polysaccharide . In the present study, emulsions of R-(+)-Limonene were developed for coating of strawberries to improve the shelf life and microbiological safety. Edible coatings with at least 0.15% w/w of R-(+)-Limonene improved the microbial stability of the strawberries, resulted effective in the decontamination of external pathogens such as Escherichia coli , Bacillus subtilis and Staphylococcus aureus . Changes in weight loss, total soluble solid (TSS), pH, total phenolic, ascorbic acid , textural properties and surface color of strawberries during storage were evaluated. R-(+)-Limonene emulsions were effective for decreasing water loss of all the fruit samples tested. The decrease in TSS values was higher in the control strawberries. Initial decreases followed by increases of pH during storage of strawberries. The amount of total phenols detected in control fruit sharply increased during the first 6 days then remained steady, whereas the total soluble phenols content of treated strawberries gradually increased during storage. Strawberries coated with emulsion had higher L* and a* values as compared to control fruits. The R-(+)-Limonene coatings have demonstrated the potential to inhibit foodborne pathogen contamination of strawberries, and prolong their shelf life.

Low-background and label-free loop-mediated isothermal amplification for rapid detection of foodborne pathogens using amplicon-templated copper nanoclusters

Wenyu Du,Shuanglong Wang,Yining Luo,Xiaoyu Tong,Yin Chen,Zhipeng Fang,Junzhuo Shan,Daofeng Liu,Haiyan Ni,Weihua Lai,Shan Shan

Background Loop-mediated isothermal amplification (LAMP) is a simple, low-cost, and instrument-free assay, which is highly adaptable for the detection of foodborne pathogens. However, current methods have a high background, making it difficult to achieve sensitive on-site detection. Results Herein, LAMP-CuNCs assay with a low background signal was established. LAMP amplicons were used as the templates for the synthesis of copper nanoclusters (CuNCs), and LAMP results were determined by the fluorescence intensity of the amplicons-CuNCs. The stability of LAMP amplicons templated CuNCs was improved by the addition of 1.5 g/mL fructose, which effectively prolonged the fluorescence lifetime. The detection limit of Salmonella DNA was 756 pg/μL, and the sensitivity and specificity of this method were 12/12 and 12/12, respectively. This method could detect Salmonella at a minimum concentration of 1910 CFU/mL in spiked salmon samples. Significance The study provided a method that can quickly obtain results within 16 min for detection of genomic DNA and visual inspection under a 365 nm UV flashlight. In addition, this method could also be used to detect Listeria monocytogenes , Vibrio parahaemolyticus , and Escherichia coli .

Amplification-Free, Single-Microbead-Based Cas12a Assay for One-Step DNA Detection at the Single-Molecule Level

Xueping Yang,Jingyun Li,Suixin Zhang,Chao Li,Jiehua Ma

CRISPR/Cas-based systems are highly attractive for developing next-generation diagnostic technologies because of their intrinsic merits such as simplicity, sensitivity, and specificity. However, currently, nucleic acid amplification procedures are still needed to achieve attomolar sensitivity in most CRISPR/Cas-based assays, which causes high cost, operation difficulty, and low efficiency. Herein, we combine the CRISPR/Cas12a-based assay and a single-microbead detection platform for one-step and amplification-free detection of DNA at the single-molecule level. By modifying DNA reporters on a biomimetic membrane-coated microbead, the activated Cas12a by targets will cleave these reporters and lighten the bead within 10 min. The method allows the detection of the target down to three copies in a 5 μL sample. Furthermore, we successfully apply this method for the specific identification of viral infection, foodborne bacteria, and DNA mutation in real samples without extra nucleic acid amplification. We believe that this approach offers new insights for developing CRISPR/Cas-based DNA assays in biomedical applications.
