Changes in Physicochemical Properties, Metabolites and Antioxidant Activity of Edible Grass during Spontaneous Fermentation

Xianxiu Li,Tao He,Yangchen Mao,Jianwei Mao,Xiaojin Lai,Hangjia Tu,Yi Zhou,Ruyi Sha

Fermentation is a crucial technology to improve the nutritional and functional properties of food materials. In this study, edible grass was processed by spontaneous fermentation. Changes in physicochemical properties, metabolites, and antioxidant activity of edible grass were investigated by colorimetric method and chromatography mass spectrometry-based non-targeted metabolomics approach during fermentation. The highest total polyphenol and total flavonoid contents, and free radical scavenging abilities were observed on the 17th day of fermentation. The maximum activity of superoxide dismutase was maintained stable in the fermentation time range of 7–70 days. In total, 16 differential metabolites were identified with fermentation duration up to 124 days. Fermented edible grass exerted protection from H2O2-induced cytotoxicity on HepG2 cells, regulating by the reduction in reactive oxygen species level and the increase in antioxidant enzyme activities. Overall, this study confirms that fermented edible grass obtained by spontaneous fermentation presented favorable nutritional and functional quality, and is expected to be a kind of food with antioxidant function. Keywords:edible grass;fermentation;metabolite;antioxidant activity;HepG2 cells

Studies on the Changes of Fermentation Metabolites and the Protective Effect of Fermented Edible Grass on Stress Injury Induced by Acetaminophen in HepG2 Cells

Tao He,Xianxiu Li,Zhenzhen Wang,Jianwei Mao,Yangchen Mao,Ruyi Sha

In this study, gas chromatography-mass spectrometry (GC-MS) based untargeted metabolomics was used to describe the changes of metabolites in edible grass withLactobacillus plantarum(Lp) fermentation durations of 0 and 7 days, and subsequently to investigate the protective effect of fermented edible grass on acetaminophen-induced stress injury in HepG2 cells. Results showed that 53 differential metabolites were identified, including 31 significantly increased and 22 significantly decreased metabolites in fermented edible grass. Fermented edible grass protected HepG2 cells against acetaminophen-induced stress injury, which profited from the reduction in lactate dehydrogenase (LDH) and malondialdehyde (MDA) levels and the enhancement in superoxide dismutase (SOD) activity. Cell metabolomics analysis revealed that a total of 13 intracellular and 20 extracellular differential metabolites were detected. Fermented edible grass could regulate multiple cell metabolic pathways to exhibit protective effects on HepG2 cells. These findings provided theoretical guidance for the formation and regulation of bioactive metabolites in fermented edible grass and preliminarily confirmed the protective effects of fermented edible grass on drug-induced liver damage. Keywords:edible grass;fermentation;metabolomics;HepG2 cells;acetaminophen-induced stress injury

Development of broad-specific monoclonal antibody-based immunoassays for simultaneous ochratoxin screening in medicinal and edible herbs

Mengyue Guo,Jing zhang,Jianxin Lv,Tongwei Ke,Jiao Tian,Kun Miao,Yudan Wang,Dandan Kong,Haonan Ruan,Jiaoyang Luo,Meihua Yang

The frequent occurrence of ochratoxins in food and herbs poses a considerable threat to consumer and animal health. Here, we generated a broad-specificity monoclonal antibody against three ochratoxins. Concentrations for 50% inhibition (IC 50 ) for OTA, OTB, and OTC were 0.37, 0.23, and 2.24 ng/mL, respectively. In the two matrices, the proposed indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) exhibited a limit of detection (LOD) of 1.5 μg/kg for OTA and 1.0 μg/kg for OTB, respectively, whereas the cut-off levels of lateral flow immunoassay (LFIA) for OTA and OTB were respectively determined to be 1 and 0.25 ng/mL. As for OTC, the LOD of ic-ELISA and cut-off concentration of LFIA were 15.5 μg/kg and 8 ng/mL for milkvetch root and 37.5 μg/kg and 16 ng/mL for ginger, respectively. The real sample detection results by ic-ELISA and LFIA methods were validated via LC-MS/MS analysis. Therefore, the developed immunoassays are useful and effective tools for high-throughput screening and rapid on-site determination of ochratoxins in complex matrices.

Effects of Edible Grass (Rumex patientia L. × Rumex tianschanicus A. LOS) Leaf Powder on Growth Performance, Antioxidant Properties, Cecal Short-Chain Fatty Acids, and Microbial Community Levels in Broilers

Xinyao Li,Hao Ling,Zengyang He,Zihui Yang,Tao Jiang,Peng Huang,Jianguo Zeng

The hybrid plant edible grass (Rumex patientiaL. ×Rumex tianschanicusA. LOS), a member of the Rumex genus, presents a novel food source with a protein content of approximately 30–40%. By incorporating non-traditional feed sources, such as edible grass leaf powder (EGLP), into broiler production, costs could be reduced. The experimental subjects of this study were Arbor Acres (AA) broilers. A total of 300 newly hatched broilers were randomly divided into to five groups, with each group consisting of six cages, housing 10 birds per cage. The control group was fed a basal diet composed of maize and soybean meal. In the experimental groups, varying percentages (3%, 6%, 9%, and 12%) of EGLP were substituted for the corn–soybean meal in the basal diet. In the diet, for days 1–21, the corn content was reduced from 1.90% to 8.20%, and the soybean meal content was lowered from 1.50% to 6.00%. Similarly, in the diet, for days 22–42, the corn content was decreased from 1.17% to 7.00%, while the soybean meal content was reduced by 1.50% to 6.00%. The experiment lasted 42 days and was divided into two phases: the brooding phase (days 1 to 21) and the finishing phase (days 22 to 42). The results show that substituting 3% and 6% EGLP for corn and soybean meal significantly increased the average daily gain (ADG) during the brooding period compared to the control group (p< 0.01). Additionally, the group with a 3% substitution rate exhibited a significant increase in the average daily feed intake (ADFI) during the brooding phase (p< 0.01). Throughout the 42-day experiment, substituting 3% and 6% of EGLP for maize and soybean meal did not significantly affect the overall growth performance of broilers (p> 0.05). However, a 6% supplementation notably reduced the feed conversion ratio (FCR) (p< 0.01). Both the 3% (EG3) EGLP and 6% (EG6) EGLP replacement meals significantly enhanced the antioxidant capacity of broiler chickens, as indicated by the increased levels of the total antioxidant capacity (T-AOC), superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) (p< 0.01). Additionally, broilers supplemented with 3–12% showed a marked increase in cecal short-chain fatty acids (SCFAs) compared to the control group (p< 0.01). The 3% EGLP replacement diet also significantly boosted the prevalence ofLactobacillusin the cecum (p< 0.01). Furthermore, after EGLP supplementation, there was a higher abundance of SCFA-metabolizing bacteria, particularlyAlistipesandChristensenellaceae_R-7, compared to pre-treatment (p< 0.05). The prevalence ofClostridiumwas significantly greater in the groups receiving 9% and 12% EGLP (p< 0.05), whileButyrivibriolevels were notably higher after supplementation with 12% EGLP (p< 0.05).
