Tumor microenvironment (TME) featured by dense extracellular matrix (ECM), high interstitial pressure, hypoxia, and immunosuppression hinders the infiltration of nanoparticles (NPs) into tumors, impairing the efficacy of chemotherapy or phototherapy. To overcome these obstacles, we propose a novel biomimetic nano-carrier, B16F10 cell membrane camouflaged heparin-3-bromopyruvate conjugate modified graphene oxide (GO [email protected] , GBM), for the targeted delivery of etoposide (EPT). EPT/GBM accumulates in tumor tissue and is internalized into B16F10 cells relying on homologous targeting ability. GO-mediated hyperthermia could accelerate the release of EPT, destruct ECM, dilate blood vessels, increase oxygen supply, and induce immunogenic cell death (ICD). The acylhydrazone bond-conjugated 3BP is released in acidic environment, suppressing glycolysis for starvation therapy, and reducing lactate efflux to alleviate immunosuppression. The released EPT entered the cell nuclei to exert a chemotherapeutic effect and downregulated the expression of programmed death ligand 1 (PD-L1) by inhibiting epithelial-mesenchymal transition (EMT)/β-catenin/STT3/PD-L1 signaling. EPT/GBM plus near infrared (NIR) laser treatment activated T lymphocytes and downregulated M2 tumor-associated macrophage (TAM), showing significant efficacy against primary tumors, abscopal tumors and lung metastases. In summary, this study presented a novel combinational strategy that integrated PTT, chemotherapy, starvation therapy and immunotherapy in one system, providing a new perspective for targeted-therapy of melanoma.

As a common malignant bone tumor in adolescents, osteosarcoma (OS) possesses a very poor prognosis and lacks effective treatments. In this study, we developed a multifunctional nanotherapeutic agent (WSe 2 /AIPH/GOx) that can combine photothermal therapy (PTT) /thermodynamic therapy (TDT) /starvation therapy (ST) against OS. The prepared WSe 2 /AIPH/GOx exhibited a high photothermal conversion efficiency of 37.3% under 808 nm near-infrared laser irradiation, which could effectively eliminate tumor cells. In addition, the local high temperature caused by photothermal heat induces the rapid release and decomposition of AIPH, generating oxygen-independent alkyl radicals that promote tumor cell death in the hypoxic microenvironment. More importantly, the introduction of GOx depletes glucose to starve tumor cells on the one hand, and on the other hand can indirectly increase the thermal sensitivity of cells. In vivo experiments have demonstrated that WSe 2 /AIPH/GOx can effectively inhibit tumor growth in mice due to the combined effects of multiple therapies. Overall, this work provides an effective strategy for potential osteosarcoma treatment based on combination therapies.

Piezoelectrodynamic therapy (PEDT) is compromised by hypoxia dilemma of tumors, while starvation therapy is constrained by insufficient enzyme activities. To address these challenges, Janus piezoelectric nanoparticles (NPs) are proposed to spatially immobilize glucose oxidase (GOx) and catalase (CAT), enabling piezoelectric potential-amplified enzyme activities and synergistic PEDT-starvation tumor therapy. Here hollow barium titanate (hBT) NPs are synthesized using SiO 2 templates, followed by partial Au deposition via the Pickering emulsion-masking method to create Janus hBT@Au NPs, which are then conjugated with GOx and CAT on opposing sides to yield C-hBT@Au-G NPs. The hollow structure of hBT enhances flexibility and deformation under ultrasonication, while Schottky heterojunctions with Au layers promote charge carrier transfer, amplifying piezoelectric effects and free electron transfer to boost GOx activities. Piezoelectric field-enhances selective tumor cell internalization of NPs and PEDT generation of reactive oxygen species (ROS), coupled with self-propagated GOx/CAT cascades, intensify tumor cytotoxicities and deplete intracellular adenosine triphosphate. The Janus architecture, ultrasonic cavitation, and O 2 generation collaboratively drive robust propulsion for efficient NP accumulation and deep ROS penetration into tumor tissues, thereby achieving full tumor suppression with negligible systemic toxicity. This design overcomes delivery barriers of tumor accumulation, intratumoral penetration, and cellular uptake and synergizes PEDT-starvation tumor therapy.

The common existence of hypoxia within the tumor microenvironment severely restricts the efficacy of photodynamic therapy (PDT), which is attributed to the fact that the PDT process is strongly oxygen (O2) dependent. Here, a multifunctional composite (named CPCG), which combines polyethylene glycol (PEG) functionalized cerium oxide nanoparticles (CeO2) with photosensitizer chlorin e6 (Ce6) and glucose oxidase (GOx), is reported for generating O2 within the tumor microenvironment by the dual-path hydrogen peroxide (H2O2)-modulated ways to ameliorate hypoxia, thereby enhancing the PDT efficiency. This process is realized based on the dual enzyme-like activity of CeO2. The first modulated way is to transform the superoxide anion (O2˙−) into H2O2 by the superoxide dismutase-like activity of CeO2. The second modulated way is to decompose glucose into H2O2 through the catalysis of GOx. Subsequently, H2O2 generated from the above dual modulated ways can further produce O2via the catalase-like activity of CeO2. Additionally, the depletion of glucose could impede the nutrient supply to obtain starvation therapy. Both in vitro and in vivo experiments indicate that the CPCG composite could enhance the efficacy of photodynamic/starvation synergistic therapy. Therefore, this strategy offers great potential to modulate the O2 level in the tumor microenvironment for better therapeutic outcomes, and can act as a promising candidate in photodynamic/starvation synergistic therapy.

Sonodynamic therapy (SDT) is a promising ultrasound (US)-triggered therapeutic modality for anticancer treatments due to its high penetration, safety, and noninvasiveness. Its therapeutic efficiency is closely related to the performance of sonosensitizers and the oxygen concentration of the tumor microenvironment (TME). Herein, inspired by the adhesion of cocklebur, a rough-surface sonosensitizer gold platinum encapsulated with magnesium silicate loaded with glucose oxidase (AuPt@MgSiO3@GOx, APMS-GOx) with an enzymatic cascade activity was designed to improve delivery capability, produce reactive oxygen species, and reduce the glucose concentration in tumor tissue. The rough surface of APMS-GOx can improve the ability of nanomaterials to deliver to tumor areas. The enzymatic cascade reaction of APMS-GOx can induce starvation therapy and improve the level of oxygen. Adequate oxygen can not only alleviate hypoxia TME but also further convert to singlet oxygen (1O2) by APMS-GOx under US activation. Moreover, the activity of GOx increases due to the involvement of US, resulting in a positive effect on starvation therapy. Furthermore, APMS-GOx exhibits near-infrared absorption properties, providing a photoacoustic imaging-guided combination breast tumor therapy strategy.

Integrating imaging-guided diagnoses with therapeutic strategies holds significant potential in the treatment of cancer and other diseases. Herein, we developed a dual stimulus-responsive nanoagent that is responsive to two specific stimuli, and demonstrated its application to the diagnosis and photothermal-starvation therapy of colon cancer. The elevated levels of H 2 S and glucose in colon cancer cells are used as targets for a dual stimulus-responsive, self-assembled copper-albumin-glucose oxidase@polyaniline (Cu 2 + -BSA-GO x @PANI) nanoplatform. Upon entering colon cancer cells, GO x oxidizes glucose to produce gluconic acid, which leads to the protonation of PANI and the red-shifting of the wavelength of maximum absorbance of the nano-platform from 680 nm to 780 nm. Concurrently, Cu 2+ reacts with H 2 S to form CuS, which further induces a redshift from 780 nm to 830 nm, and significantly enhancing the near-infrared II region absorption intensity and enabling photoacoustic imaging under 1064 nm laser irradiation. Additionally, the consumption of glucose within cancer cells inhibits the generation of ATP, achieving starvation therapy while reducing the expression of tumor heat-shock protein 90 to weaken tumoral thermo-tolerance, resulting in a synergistic anti-tumor effect. In vitro and in vivo analyses indicate that the Cu 2+ -BSA-GOx@PANI nano-platform possesses specific visual diagnostic and therapeutic capabilities for colon cancer, providing a novel strategy for the design of highly effective nanodiagnosis-treatment agents.

Designing nanozyme hybrids with cascade catalytic activities and coupling effects has significant promises in tumor therapy through concerted effects. In this work, a goethite-derived porous Fe 2 O 3 /Au hybrid nanozyme was fabricated for synergistic treatment of triple negative breast cancer. The as-prepared porous Fe 2 O 3 nanoparticle exhibits excellent peroxidase-like activity compared to goethite particles, which can generate hydroxyl radicals in the tumor microenvironment. Furthermore, the Fe 2 O 3 nanoparticle induces ferroptosis through intracellular lipoperoxide accumulation. After decorating glucose oxidase-mimicking gold nanoparticles, the Fe 2 O 3 /Au hybrid nanozyme is able to catalyze glucose into gluconic acid and H 2 O 2 . The glucose consumption deprives the energy of the tumor cell, and the generated H 2 O 2 can be converted to hydroxyl radicals through the biocatalytic reaction. Importantly, the coupling of gold nanoparticles enhances the photothermal conversion of the porous Fe 2 O 3 nanoparticle. Thus, the designed Fe 2 O 3 /Au hybrid nanozyme can be used for tumor therapy through starvation, cascade catalytic reactions, ferroptosis, and photothermal manners. This work presents a derivative method that transforms minerals into nanozyme and couples nanozymes for synergistic tumor therapy, shedding light on the rational design of nanozyme composites for biomedicine.

The current trend of cancer therapy has changed from monotherapy to synergistic or combination therapies. Among the treatment strategies, photodynamic therapy (PDT) and starvation therapy are widely employed together. However, the therapeutic effect of these treatments could lead to strong resistance and poor prognosis due to tumor hypoxia. Therefore, a smart nanoplatform (MONs-GOx@MnO2-Ce6) has been constructed herein by the assembly of glucose oxidase (GOx)-coated mesoporous organosilica nanoparticles (MONs) and MnO2 nanosheets-chlorin e6 (Ce6), which form a nanosystem. Once MONs-GOx@MnO2-Ce6 enter tumor cells, it catalyzes the oxidation of glucose using oxygen (O2) and generates hydrogen peroxide (H2O2) and gluconic acid, the former of which may accelerate the decomposition of MnO2 nanosheets. The released MnO2 nanosheets would regenerate O2 in the presence of H2O2. In this case, MnO2 nanosheets serve as (i) a nanocarrier and fluorescence quencher for the photosensitizer Ce6, (ii) a degradable material that is activated by the tumor microenvironment (TME) for fluorescence recovery, and (iii) an O2-producing carrier that reacts with H2O2 for relieving hypoxia in the tumor, which contributes to the combined starvation/photodynamic cancer therapy since these treatment strategies need O2. MONs-GOx@MnO2-Ce6 could not only realize cancer cell imaging, but also reduce intracellular glucose uptake and Glut1 expression, inhibiting the metabolism of cancer cells. This strategy shows great potential for clinical applications.

Recently, starvation-inducing nutrient deprivation has been regarded as a promising strategy for tumor suppression. As a first-line lipid-lowering drug, atorvastatin (ATV) significantly reduces caloric intake, suggesting its potential in starvation therapy for suppressing tumors. Accordingly, we developed a novel starvation therapy agent (HA-Se-ATV) in this study to suppress tumor growth by using hyaluronic acid (HA)-conjugated chitosan polymer-coated nano-selenium (Se) for loading ATV. HA-Se-ATV targets cancer cells, following which it effectively accumulates in the tumor tissue. The HA-Se-ATV nanoplatform was then activated by inducing a weakly acidic tumor microenvironment and subsequently releasing ATV. ATV and Se synergistically downregulate the levels of cellular adenosine triphosphate while inhibiting the expression of thioredoxin reductase 1. Consequently, the starvation–stress reaction of cancer cells is significantly elevated, leading to cancer cell death. Furthermore, the in vivo results indicate that HA-Se-ATV effectively suppresses tumor growth with a low level of toxicity, demonstrating its great potential for clinical translation.

Glucose is used as an important nutrient to support cell growth. The glucose oxidase (GOx) can transform glucose into gluconic acid and toxic H 2 O 2 , which can be used for tumor starvation therapy. However, the leakage of GOx may cause severe side effects to the normal tissue. To prevent the accidental leakage of GOx, this study proposes the chemical modification of GOx on the photothermal transducing agent surface, to realize the safe and combined starvation and photothermal therapy of colorectal tumors . Polyvinylpyrrolidone (PVP)-modified WS 2 nanobowls (WS 2 -PVP) as a photothermal transducing agent were produced using a one-pot preparation method. Then, α-lipoic acid (LA) molecules were immobilized at the sulfur-deficient sites on the surface of WS 2 nanobowls to afford the chemical loading of GOx through amide bonds. Under the irradiation of a near-infrared laser (808 nm), thermal energy is generated by WS 2 to kill colorectal cancer cells locally. The photothermal conversion efficiency of WS 2 -PVP-LA was 27.2%. This study is anticipated to open up an alternative avenue for the rational design of multifunctional nanotherapeutics for tumor therapy.

Phototherapy and sonodynamic therapy (SDT) are widely used for the synergistic treatment of tumors and have received considerable attention. However, an inappropriate tumor microenvironment, including pH, H 2 O 2 , oxygen, and glutathione levels, can reduce the therapeutic effects of synergistic phototherapy and SDT. Here, a novel Bi-based soluble microneedle (MN) is designed for the CT imaging of breast tumors and starvation therapy/gas therapy-enhanced phototherapy/SDT. The optimized Bi/BiVO 4 Schottky heterojunction serves as the tip of the MN, which not only has excellent photothermal conversion ability and CT contrast properties, but its heterojunction can also avoid the rapid combination of electrons and hole pairs, thereby enhancing the photodynamic/sonodynamic effects. A degradable MN with excellent mechanical properties is fabricated by optimizing the ratios of poly(vinyl alcohol), poly(vinyl pyrrolidone), and sodium hyaluronate. Glucose oxidase (GO x ) and diallyl trisulfide are loaded into the MN to achieve tumor starvation and gas therapy, respectively; And the controlled release of GO x and H 2 S can be achieved under ultrasound or near-infrared laser irradiation. The in vitro and in vivo results demonstrate that this multifunctional MN can achieve high therapeutic efficacy through starvation therapy/gas therapy-enhanced phototherapy/SDT. The designed multifunctional MN provides a prospective approach for synergistic phototherapy and SDT.

The combination of photodynamic therapy (PDT) and chemodynamic therapy (CDT) in tumor treatment has attracted considerable attention. However, tumor hypoxia and glutathione (GSH) overproduction in the tumor tissue restricted the progress of their applications. Herein, a Mn-porphyrinic metal–organic framework (Mn-TCPP) was constructed by the one-pot method and further used for immobilizing glucose oxidase (GOx) to obtain GOx@Mn-TCPP. GOx would rapidly exhaust endogenous glucose into hydrogen peroxide (H 2 O 2 ) and gluconic acid, thus shutting off the energy supply of tumor cells for starvation treatment. Mn-TCPP catalyzed H 2 O 2 to produce oxygen, regulating the hypoxic tumor microenvironment and in turn improving 1 O 2 generation under laser irradiation. Interestingly, Mn-TCPP can reduce reactive oxygen species consumption owing to the redox reaction between Mn 3+ and GSH, thus greatly enhancing PDT. Furthermore, benefiting from GOx-mediated starvation therapy, Mn 2+ produced by Mn 3+ reduction can react with sufficient intracellular H 2 O 2 to generate ·OH with high cytotoxicity through a Fenton-like reaction. After treatment by GOx@Mn-TCPP under laser irradiation in vitro, the cell viability of 4T1 and A549 tumor cells reached to 20%, reflecting GOx@Mn-TCPP could give full play to the advantages of PDT/CDT/starvation therapy. The results in vivo demonstrated that GOx@Mn-TCPP mediated synergistic cascade therapy could significantly inhibit tumor growth and improve the therapeutic effect.
