When Aristolochia plants wilt and decay, aristolochic acids (AAs) are released into the soil, causing soil contamination. It has been demonstrated that aristolochic acid can be accumulated and enriched in crops through plant uptake. However, there is a lack of systematic studies on the migration and accumulation of AAs in a realistic simulated soil environment. In this study, Aristolochia herbal extracts were mixed with soil for growing three typical vegetables: lettuce, celery, and tomato. The contents of AAs in the above-mentioned plants were determined by an established highly sensitive LC-MS/MS method to study the migration and accumulation of AAs. We found that AAs in the soil can be transferred and accumulated in plants. AAs first entered the roots, which were more likely to accumulate AAs, and partially entered the above-ground parts. This further confirms that AAs can enter the food chain through plants and can have serious effects on human health. It was also shown that plants with vigorous growth and a large size absorbed AAs from the soil at a faster rate. The more AAs present in the soil, the more they accumulated in the plant. Keywords:aristolochic acid;vegetables;uptake;translocation;fate

Aristolochic acids (AAs) naturally occurring in the herbal genus Aristolochia are associated with a high risk of kidney failure, multiple tumors and cancers. However, approaches with high selectivity and rapidity for measuring AAs in biological samples are still inadequate. Inspired by the mechanism of AAs-induced nephrotoxicity, we designed a hybrid magnetic polymer-porous agarose (denoted as MNs@SiO 2 M@DNV-A), mimicking the effect of basic and aromatic residues of organic anion transporter 1 (OAT1) for efficient enriching aristolochic acid I (AA I) and aristolochic acid II (AA II) in the plasma. The monomers of vinylbenzyl trimethylammonium chloride (VBTAC), N-vinyl-2-pyrrolidinone (NVP) and divinylbenzene (DVB) were employed to construct the polymer layer, which provided a selective adsorption for AAs by multiple interactions. The porous agarose shell contributed to remove interfering proteins in the plasma samples. A magnetic solid-phase extraction (MSPE) based on the proposed composite enhanced the selectivity toward AA I and AA II in the plasma samples. In combination of HPLC analysis, the proposed method was proved to be applicable to fast and specific quantification of AAs in blood samples, which was characterized by a good linearity, high sensitivity, acceptable recovery, excellent repeatability and satisfactory reusability.

A convenient, and efficient multi-phase extraction method was used to isolate aristolochic acid from real samples. To increase the efficiency, a new dual ionic liquid-immobilized silica was used as a sorbent and the effects of the adsorption conditions were investigated. IM-BIM@Sil in a methanol/water (60:40, v/v) solution at 60 °C extracted the highest amount of aristolochic acid (16.69 mg/g) compared to the other sorbents examined. The sorbent was then applied to the multi-phase extraction of aristolochic acid. After extraction eight times, 2.4–70.9 × 10 −3 mg/g of aristolochic acid was isolated from nine real samples after several washing and elution steps. The recoveries of aristolochic acid ranged from 70.0% to 110.6% with relative standard deviations of 3.5%–9.1%, highlighting the accuracy of this method.

Aristolochic acid I (AAI) is a carcinogen associated with various human cancers. However, its causal relationship with hepatocellular carcinoma remains controversial, and inconsistent results from rodent studies have suggested species-specific differences. Here we evaluated AAI genotoxicity using functional human-induced hepatocyte-like cells (hiHep cells), a model that closely mimics primary human hepatocytes in gene expression and function, thereby shedding light on its potential hepatocarcinogenic risk in humans. First, we assessed AAI genotoxicity by evaluating AAI-DNA adducts and micronucleus frequency. In hiHep cells, AAI (0.7–2.5 µM) induced up to 105 adducts per 108 nucleotides, indicating high metabolic activation of AAI. A concentration-dependent increase in micronucleus frequency indicated a significant increase in chromosomal aberrations in hiHep cells. Considering the evidence of AAI inducing oxidative stress, we assessed 8-hydroxy-2′-deoxyguanosine and reactive oxygen species levels to evaluate DNA oxidative damage. For both indicators, significantly elevated levels were observed. A mechanism involving oxidative damage was further supported by observations of mitochondrial dysfunction, including changes in mitochondrial membrane potential and mitochondrial complex activity. Ascorbate treatment decreased AAI-induced oxidative DNA damage and DNA adduct formation, providing direct cellular evidence for free radical intermediates in AAI metabolic activation—a mechanism previously hypothesized but not experimentally validated in a human-relevant hepatocyte model. Our study findings revealed the genotoxic effects of AAI on hiHep cells and implicated oxidative stress as the key mechanism. These findings strengthen the association between AAI exposure and liver disease and highlight the potential role of antioxidant therapies in mitigating AAI-associated carcinogenesis.

Aristolochic acid I (AA-I), a known nephrotoxin and carcinogen, is commonly found in certain medicinal plants, posing significant safety concerns. In this work, defect-engineered, molecularly imprinted UiO-66-NH2 was synthesized via a one-pot hydrothermal method using indomethacin (IND) as a green alternative template. The resulting imprinted metal–organic framework effectively mitigates matrix interference, enabling highly selective recognition and the efficient adsorption of AA-I. Structural characterization confirmed that the imprinting process and template removal did not compromise the framework integrity of UiO-66-NH2. Spectroscopic analysis revealed that AA-I recognition was governed by Zr coordination, electrostatic interactions, and spatially confined recognition sites within the porous framework. Density functional theory calculations further elucidated the interaction mechanisms involved in the adsorption process. The imprinted UiO-66-NH2 exhibited a maximum adsorption capacity of 42.74 mg/g and an imprinting factor of 2.48. In real sample analysis, the material achieved recovery rates of 96.7–103.5% and excellent selectivity in Clematis armandii extracts. The adsorption efficiency remained above 80% after five adsorption–desorption cycles, indicating good structural stability and reusability. This work offers a promising and sustainable strategy for improving herbal medicine safety through the rational design of ecofriendly MOF-based adsorbents.

A method is described for the functionalization of magnetic carbon nanotubes to recognize aristolochic acid Ⅰ and Ⅱ. 3-Glycidyloxypropyltrimethoxysilane was used as a coupling agent to immobilize adenine on a solid support. The morphology and structure of adenine-coated magnetic carbon nanotubes was investigated using transmission electron microscopy (TEM), scanning electron microscopy (SEM), X-ray diffraction (XRD) and a vibrating sample magnetometer (VSM). The adsorption performance of the adenine-coated magnetic carbon nanotubes was evaluated via adsorption isotherms, the kinetics and selectivity tests. The adsorption capacity of the adenine-functionalized sorbent for aristolochic acid Ⅰ was determined to be 24.5 μg mg −1 . By combining magnetic solid phase extraction with HPLC detection, a method was developed to enrich and detect aristolochic acids used in traditional Chinese medicine. A satisfactory recovery (92.7 - 97.5% for aristolochic acid Ⅰ and 92.6 - 99.4% for aristolochic acid Ⅱ) and an acceptable relative standard deviation (<4.0%) were obtained.

Aristolochic acids (AAs) contained in herbal plants are implicated in multiple organ injuries and have a high mutational burden in upper tract urothelial cancers. The currently available techniques for monitoring AAs include LC (liquid chromatography) and LC/MS (mass spectrometry), but the application of these approaches are limited due to the complex sample preparation and derivatization steps. Therefore, there is an urgent need to develop efficient methods for identifying and quantifying AAs. Here, we present a new dual-spectroscopic approach for the direct detection of AAs from blood and tissue samples; the detection of aristolochic acid I (AAI) is performed by surface-enhanced Raman spectroscopy (SERS), and its bioproduct, aristololactam (AAT), is detected by fluorescence spectroscopy based on their distinctive spectral response. Furthermore, a graphene assisted enrichment coupled with a magnetic retrieval strategy was developed to enhance SERS sensitivity toward AAI. Our method was successfully applied to directly determine both AAI and AAT from the blood, liver, and kidney of rats. The potential for real-world application was demonstrated by continuously monitoring AAI and AAT in rat blood and tissues after AAI feeding. The results showed that AAI was gradually metabolized to AAT and transported to different organs. It was found that the metabolism of AAI took place in the kidney, but AAT residue was detected in both liver and kidney, which might be related to long-term toxicity and gene mutation. The proposed dual-spectroscopic strategy is applicable to long-term toxicology research and to the direct diagnosis of AAI-induced organ injury.

A novel and efficient pretreatment method is proposed for the enrichment of sanguinarine (SA) from the extraction wastewater of Macleaya cordata (Willd) R. Br. using magnetic molecularly imprinted polymers (MMIPs) as solid phase adsorbent. The obtained MMIPs were synthesized by molecular imprinted technology and were well characterized with TEM, FT-IR, XRD, TGA, and VSM. The adsorption capacity and selectivity of MMIPs were also investigated. The results showed that the obtained MMIPs were core–shell spherical morphologies with an average diameter of 300 nm. The adsorption experiments showed that the MMIPs exhibited an almost three times higher adsorption capacity towards SA ( Q max = 12.24 mg/g) than magnetic molecularly non-imprinted polymers (MNIPs) ( Q max = 4.15 mg/g). The calculated parameters of adsorption isotherms and kinetics suggested that the adsorption process matched the Langmuir adsorption isotherm and pseudo-second-order kinetics model. The thermodynamic parameters were calculated, indicating that the adsorption process was spontaneous (ΔG 298K = −3.24 kJ/mol). The selectivity experiment showed that the MMIPs displayed higher selective adsorption capacity for SA compared to its reference compounds. Moreover, the MMIPs were applied to enrich SA from the extraction wastewater of M. cordata . The proposed method could provide an effective, simple and environmentally friendly way to identify and enrich SA from the extraction wastewater of M. cordata .

A novel type of magnetic molecularly imprinted polymer was prepared for the selective enrichment and isolation of chelerythrine from Macleaya cordata (Willd) R. Br . The magnetic molecularly imprinted polymers were prepared using functional Fe 3 O 4 @SiO 2 as a magnetic support, chelerythrine as template, methacrylic acid as functional monomer, and ethylene glycol dimethacrylate as cross-linker. Density functional theory at the B3LYP/6-31G (d, p) level with Gaussian 09 software was applied to calculate the interaction energies of chelerythrine, methacrylic acid and the complexes formed from chelerythrine and methacrylic acid in different ratios. The structural features and morphology of the synthesized polymers were characterized by using Fourier transform infrared spectroscopy, X-ray diffraction, transmission electron microscopy, and vibration sample magnetometry. Adsorption experiments revealed that the magnetic molecularly imprinted polymers possessed rapid kinetics, high selectivity, and a higher binding capacity (7.96 mg/g) to chelerythrine than magnetic molecularly non-imprinted polymers (2.36 mg/g). The adsorption process was in good agreement with the Langmuir adsorption isotherm and pseudo-second-order kinetics models. Furthermore, the magnetic molecularly imprinted polymers were successfully employed as adsorbents for the extraction and enrichment of chelerythrine from Macleaya cordata (Willd) R. Br . The results indicated that the magnetic molecularly imprinted polymers were suitable for the selective adsorption of chelerythrine from complex samples such as natural medical plants.

In this study, a novel berberine based anion-exchanged hypercrosslinked polymer (BBR-HCPs) was synthesized via Friedel-Crafts alkylation using dimethoxymethane as a cross-linking agent. The proposed BBR-HCPs polymer exhibited excellent adsorption capacities for aristolochic acids (AAs). Moreover, there are various interactions such as hydrophobic, π-π stacking, hydrogen bonding, and electrostatic interactions between BBR-HCPs and AAs. A solid phase microextraction (SPME) coating, prepared with BBR-HCPs as the sorbent and polyacrylonitrile (PAN) as the binder, was applied for the determination of AAs in herbal plants using a 24-well plate for high-throughput, with instrumental analysis carried out via high performance liquid chromatography with diode array detector (HPLC-DAD). The optimized and validated BBR-HCPs-SPME/HPLC-DAD method exhibited excellent linearity over range of 5–400 ng mL −1 with R 2 > 0.9974. Detection limits (S/N = 3) for AAs were found to be 0.03–0.150 ng mL −1 , with recoveries ranging from 82.6 to 117.8 % and relative standard deviations (RSDs) less than 10.49 %. This study underscores the potential of BBR-HCPs as a promising adsorbent for selective extraction and enrichment of AAs while introducing a reliable novel strategy for monitoring trace amounts of AAs in water and herbal plant samples.

Aristolochic acid (AA) I and AA II, which have been classified as carcinogenic to human and have been proven to be nephrotoxic, are bioactive ingredients of many traditional Chinese medicines (TCMs). Thus, development of an efficient approach for separation and determination of AA I and AA II in biological samples and herbal plants is of significance. Herein, electromembrane extraction (EME) was for the first time used to separate AA I and AA II. It is noted that also for the first time 1-decanol was discovered and used as an efficient SLM solvent for EME of acidic compounds. The proposed EME system was used to extract AA I and AA II from urine samples (recovery≥68%). The approach of EME combined with LC–MS (EME-LC/MS) was evaluated using urine samples. The linear range for AA I and AA II was 10–1000 ng mL −1 (R 2 ≥0.9970), and the limits of detection (LOD, S/N = 3) for AA I and AA II were 2.7 and 2.9 ng mL −1 , respectively. Finally, this EME-LC/MS approach was employed to discover AA I and AA II in the herbal plants. In addition, using standard addition method, AA I in Aristolochicaceae-Liao Asarum (ALA) and Radix Aristolochice (RA) were 0.23 and 2044.13 μg g -1 , and AA II in ALA and RA were 0 and 338.48 μg g -1 , respectively. The repeatability of EME-LC/MS at all cases for both urine samples and herbal plants was below 15% (RSD-value). We believe that EME would be a useful tool to isolate bioactive ingredients of TCMs from complex samples for different purposes.
