The study aimed to estimate the feasibility of α-cyclodextrin (α-CD), β-cyclodextrin (β-CD), and γ-cyclodextrin (γ-CD) to encapsulate Mosla chinensis essential oil (EO) by ultrasonic-assisted method. The physical properties variations, stabilization mechanisms, and formation processes of the inclusion complexes (ICs) were investigated using experimental methods, molecular docking, and molecular dynamics (MD) simulation. Scanning electron microscopy, fourier transform infrared spectroscopy, thermogravimetric analysis, and gas chromatography–mass spectrometry showed that the ICs were successfully prepared, which differentially improved the thermal stability and retained the chemical composition of EO. The dissolution profile showed that the Peppas model can be used to describe the diffuse release mechanism of EO. Finally, molecular docking and MD simulation theoretically confirmed the interaction and conformational changes of carvacrol (the main active component of Mosla chinensis EO) inside the cavity of CDs. The results indicate that hydrogen bonding was the primary driving force for the carvacrol spontaneous access to the cavity. Further, a binding dynamic balance occurs between carvacrol and β-CD, whereas a bind and away dynamic balance occurs in the IC between carvacrol and α-CD, γ-CD. The comprehensive results show that the medium cavity size of β-CD is a suitable host molecule for Mosla chinensis EO of encapsulation, release, and stabilization. A combination of experimental and theoretical calculations is useful for the pinpoint targeted design and optimization of CD molecular encapsulation of small entity molecules. β-CD was rationally screened as a better candidate for stabilizing EO, which provides an option for a meaningful path to realistic EO applications.

Introduction Tumor-infiltrating T lymphocytes in the tumor microenvironment are critical factors influencing the prognosis and chemotherapy outcomes. As a Chinese herbal medicine, Marsdenia tenacissima extract (MTE) has been widely used to treat cancer in China. Its immunoregulatory effects on tumor-associated macrophages is well known, but whether it regulates tumor-infiltrating T-cell functions remains unclear. Method We collected 17 tumor samples from MTE-administered colorectal cancer patients, 13 of which showed upregulation of CD3+/CD8+ tumor-infiltrating T cells. Further in vitro and in vivo experiments were performed to investigate the regulatory effects of MTE on tumor-infiltrating T cells and immune escape of tumors. Results Under single and co-culture conditions, MTE inhibited TGF-β1 and PD-L1 expression in the colorectal cancer (CRC) cell lines HCT116 and LoVo. In Jurkat cells, MTE inhibited FOXP3 and IL-10 expression, increased IL-2 expression, but had no effect on PD-1 expression. These findings were confirmed in vitro using subcutaneous and colitis-associated CRC mouse models. MTE also increased the density of CD3+/CD8+ tumor-infiltrating T cells and exhibited considerable tumor-suppressive effects in these two tumor mouse models. Conclusions Our findings suggested that MTE inhibits the immune escape of cancer cells, a precipitating factor increasing the immune response of T lymphocytes.

Depression, a prevalent and debilitating mood disorder, remains a significant challenge in clinical treatment. Although pharmacological approaches targeting monoamine dysfunction have demonstrated efficacy, they are often accompanied by limitations and side effects. In this study, reserpine-induced depressed mouse models were used to investigate the antidepressant potential of five essential oils (EOs) derived from Jasminum sambac (JSEOs), Rosa rugosa , Magnolia denudata , Aloysia citriodora (ACEOs), and Abies balsamea . The chemical compositions of the five EOs were analyzed using gas chromatography-mass spectrometry (GC-MS). Subsequently, experimental mice inhaled the EOs for 1 h per day, followed by behavioral assessments using the forced swim test (FST) and tail suspension test (TST). Nissl staining was employed to assess the abundance of normal neurons in the mouse brain. At the same time, immunohistochemistry (IHC), Western blot, and reverse transcription-quantitative polymerase chain reaction techniques were utilized to analyze the expression of antidepressant-related genes. GC-MS results revealed that the primary components of the five EOs were oxygenated monoterpenes, sesquiterpene hydrocarbons, and oxygenated sesquiterpenoids. All five EOs exhibited antidepressant-like effects, with JSEOs and ACEOs demonstrating the greatest antidepressant efficacy. Behavioral tests showed a reduction in immobility durations in both FST and TST. Nissl staining results indicated an increase in the number of intact neurons in the hippocampus and hypothalamus of mice following EO treatment. Furthermore, the EOs regulated the expression of crucial biomarkers, including 5-hydroxytryptamine 1A receptor (5-HT1A), 5-HT2A, brain-derived neurotrophic factor, corticotropin-releasing factor, and tropomyosin-related kinase B. This study underscores the potential of EO inhalation as an adjunctive therapeutic strategy for depression treatment, provides novel insights into its molecular mechanisms, and supports further exploration of natural remedies in mental health care. The dried flowers and inflorescences of Buddleja officinalis Maxim are used as traditional medicines in China, and aqueous extracts of the flowers have also been used since ancient times as a yellow rice colorant at local festivals. In this study, HPLC-Q-TOF-MS/MS was used to determine the overall chemical composition of this medicine-food plant. A total of 54 compounds, including 23 flavonoids, 19 phenylethanoid glycosides, 7 alkaloids and 5 other compounds, were detected in the methanol extracts of the herb using this method. Among them, 35 compounds were found firstly in this herb. HPLC fingerprints were also developed, together with a method for the simultaneous quantification of 11 constituents that could be used for quality evaluation of B. officinalis . Fingerprint analysis, using 28 characteristic fingerprint peaks, was used to assess the similarities among 12 samples collected from different geographic areas and showed that the similarity was >0.900. Simultaneous quantification of 11 markers in B. officinalis was then performed to determine consistency of quality. Additionally, the total phenolic content and antioxidant capacity of extracts of the 12 samples of B. officinalis flowers were measured using spectroscopic methods. B. officinalis was found to have good antioxidant capacity and to be a potential natural antioxidant. The highest antioxidant capacity was found in the samples from Guizhou, Sichuan and Guangxi Province. Our results provide valuable information for further understanding and exploiting the herb.
