Simple SummaryDue to the environment and human health concerns of synthetic repellents, essential oils (EOs) as natural alternatives have received increased attention. In this study, the repellent activity of 60 commercial EOs againstAedes albopictuswas evaluated. In the initial screening, 8 active EOs including cinnamon, marjoram, lemongrass, bay, chamomile, jasmine, peppermint2, and thyme were selected. Twenty-one major constituents (>5% relative area) in the 8 active EOs were identified via gas chromatography-mass spectrometry (GC-MS) analysis. Cinnamaldehyde, citral and terpinene-4-ol displayed the highest repellent activity with more than 60% RR, which were more active thanN,N-diethyl-3-methyl benzoyl amide (DEET). Next, their nanoemulsions were prepared and characterized. In the arm-in-cage assay, cinnamaldehyde- and citral-based nanoemulsions have prolonged mosquito protection time compared with their normal solutions.AbstractMosquitoes are one of the most important disease vectors from a medical viewpoint in that they transmit several diseases such as malaria, filariasis, yellow and Dengue fever. Mosquito vector control and personal protection from mosquito bites are currently the most efficient ways to prevent these diseases. Several synthetic repellents such as DEET, ethyl butylacetylaminopropionate (IR3535) and 1-(1-methylpropoxycarbonyl)-2-(2-hydroxyethyl)piperidine) (Picaridin), have been widely used to prevent humans from receiving mosquito bites. However, the use of synthetic repellents has raised several environment and health concerns. Therefore, essential oils (EOs) as natural alternatives receive our attention. In order to discover highly effective mosquito repellents from natural sources, the repellent activity of 60 commercial EOs againstAe. albopictuswas screened in this study. Eight EOs including cinnamon, marjoram, lemongrass, bay, chamomile, jasmine, peppermint2, and thyme, showed a suitable repellent rate (>40%) at the tested dose of 10 μg/cm2. Then, their main constituents were analyzed by GC-MS, and the active constituents were identified. The most active compounds including cinnamaldehyde, citral and terpinen-4-ol, exhibited an 82%, 65% and 60% repellent rate, respectively. Moreover, the nanoemulsions of the three active compounds were prepared and characterized. In the arm-in-cage assay, the protection times of the nanoemulsions of cinnamaldehyde and citral were significantly extended compared with their normal solutions. This study provides several lead compounds to develop new mosquito repellents, and it suggests that nanoemulsification is an effective method for improving the duration of the activity of natural repellents.

Aphis gossypii Glover (Homoptera: Aphidinae), a major pest of Chinese pepper (Zanthoxylum bungeanum Maxim), causes significant agricultural damage. Ginger (Zingiber officinale Roscoe) has shown potential as a source for developing botanical pesticides due to its strong bacteriostatic and insecticidal properties; however, the underlying mechanisms remain poorly understood. This study evaluated the repellent activity of ginger shoot extract (GSE) across four solvent phases—petroleum ether, trichloromethane, ethyl acetate, and methanol—against A. gossypii. The results demonstrated that GSE exhibited significant repellent effects, with the methanol phase showing the most pronounced activity. Twelve fractions were chromatographically separated from the methanol phase, and electroantennography (EAG) analysis revealed that fraction 4 induced strong EAG responses in both winged and wingless aphids. Further identification of active compounds in fraction 4 by gas chromatography–mass spectrometry (GC–MS) indicated the presence of terpenes, aromatics, alkanes, esters, and phenols as major constituents. Subsequent EAG analysis identified several key compounds—octahydro-pentalene (C1), (Z)-cyclooctene (C2), dimethylstyrene (C3), tetramethyl-heptadecane (C5), tetrahydro-naphthalene (C6), and heptacosane (C9)—as responsible for eliciting EAG responses in both aphid forms. Additionally, results from Y-tube olfactometer assays showed that (Z)-cyclooctene and heptacosane were significantly attractive, while octahydro-pentalene acted as a strong repellent to both winged and wingless aphids. These findings offer valuable insights for the development of synthetic attractants and repellents for A. gossypii and provide a theoretical foundation for utilizing ginger in the creation of botanical pesticides targeting this pest.

The aim of this research was to determine chemical composition and repellent and insecticidal activities of the essential oil of Kaempferia galanga L. rhizomes against the booklouse, Liposcelis bostrychophila Badonnel, and to isolate insecticidal or repellent constituents from the oil. The essential oil was obtained by hydrodistillation and analyzed by gas chromatography–mass spectrometry. Twenty-eight components of the oil were identified. The major compounds in the oil were ethyl-ρ-methoxycinnamate (38.6%), ethyl cinnamate (23.2%), 1,8-cineole (11.5%), trans-cinnamaldehyde (5.3%), and borneol (5.2%). Based on bioactivity-guided fractionation, four active constituents were isolated from the oil and identified as 1,8-cineole, ethyl cinnamate, ethyl ρ-methoxycinnamate, and trans-cinnamaldehyde. The essential oil exhibited contact toxicity against the booklouse with an LC50 value of 68.6 μg/cm2. Ethyl cinnamate (LC50 = 21.4 μg/cm2) exhibited stronger contact toxicity than ethyl ρ-methoxycinnamate and trans-cinnamaldehyde (LC50 = 44.6 and 43.4 μg/cm2, respectively) while 1,8-cineole showed weak acute toxicity. The essential oil also possessed fumigant toxicity against the booklouse with a LC50 value of 1.5 mg/liter air. 1,8-Cineole and trans-cinnamaldehyde (LC50 = 1.1 and 1.3 mg/liter, respectively) possessed stronger fumigant toxicity against the booklouse than ethyl cinnamate and ethyl ρ-methoxycinnamate (LC50 = 10.2 and 10.2 mg/liter air, respectively). trans-Cinnamaldehyde was strongly repellent to booklice, whereas ethyl cinnamate and ethyl ρ-methoxycinnamate were weakly repellent and 1,8-cineole did not repel booklice. The results indicate that the essential oil and its constituent compounds have potential for development into natural insecticides or fumigants and repellents for control of insects in stored grains.

The red imported fire ant (RIFA), Solenopsis invicta Buren, is an invasive pest that is extremely aggressive and thus poses a strong threat to human health, agricultural production, equipment and the environment. In this study, we evaluated the repellent effects of eight Chinese herbal essential oils (EOs) on RIFAs, and identified the main repellent components in the EOs. Results showed that all eight EOs had outstanding repellency to RIFAs, among which A. argyi and S. album EOs had the strongest repellent effects on RIFAs. GC-MS revealed 34 ingredients in A. argyi EO, among which sesquiterpenoids and their oxygenated derivatives accounted for the greatest proportion (50 %), followed by monoterpenes and their oxygenated derivatives (47.06 %) and unknown components (2.94 %). Dipentene, 1-caryophyllene, p-cymene, α-terpineol, and Δ-cadinene were the most abundant compounds. A total of 61 ingredients were identified in S. album EO, including monoterpenes and their oxygenated derivatives (40.98 %), sesquiterpenoids and their oxygenated derivatives (39.34 %), diterpenoids and their oxygenated derivatives (3.28 %), aromatic compounds (3.28 %), and unknown components (13.12 %). Among them, the most abundant compounds were diethyl phthalate, α-cetone, 2-phenylethanol and sandacanol. The GC-EAD results showed that RIFA workers exhibited strong electroantennographic responses to dipentene, α-terpineol and 1-caryophyllene from A. argyi EO, as well as to linalool, 2-phenylethanol and benzyl acetate from S. album EO. Furthermore, all of the above six ingredients had significant repellent effects against RIFAs based on behavioral experiments. In conclusion, A. argyi and S. album EOs have good repellent effects on RIFAs, and the active ingredients include dipentene, α-terpineol, 1-caryophyllene, linalool, 2-phenylethanol and benzyl acetate. Our findings provide new information for the development of green, safe, and efficient repellents against RIFAs.

Numerous essential oils have been well acknowledged as eco-friendly alternatives to combat insect pests due to synthetic insecticide-induced pest resistance and environment pollution. As a highly commercial essential oil, tea tree oil exhibits excellent insecticidal and repellent activities. However, the molecular mechanism of the olfactory system mediating the tea tree oil-induced repellency against insect pests remains unknown. In our study, mosquito was used as a suitable model to examine the molecular mechanism of tea tree oil-induced repellency against insect pests. The results showed that tea tree oil exhibited excellent spatial and oviposition repellency against Aedes aegypti adults and outstanding repellency against larvae, which were conferred by the main constituent terpinen-4-ol. The reduced repellency in the Orco–/– mutant strain revealed that tea tree oil-induced repellency against mosquitoes was dependent on odorant receptor(s). Moreover, we identified one antenna-biased odorant receptor, AaOr96, that was involved in detecting constituents of tea tree oil to elicit repellency, and the predicted protein–ligand complex indicated that AaOr96 interacted with terpinen-4-ol via van der Waals forces from five key residues. Finally, knocking out AaOr96 resulted in a reduced spatial repellency against A. aegypti by tea tree oil and terpinen-4-ol, and a reduced oviposition repellency by terpinen-4-ol, but not by tea tree oil. Our study not only reveals that tea tree oil has great potential in pest management but also provides more insights into the molecular basis of repellency of essential oils.

Plutella xylostella Linnaeus (Lepidoptera: Plutellidae) is one of the most important pests of cruciferous vegetables. Mechanically damaged Mentha spicata Linnaeus (Lamiales: Lamiaceae) strongly repels P. xylostella adults; however, the repellent mechanism remains unclear. In this study, the repellent mechanism was explored based on the electroantennogram (EAG) and behavioural responses of P. xylostella to volatiles from the damaged M. spicata . First, the active volatile compounds in the headspace volatiles of damaged M. spicata were identified by gas chromatography-electroantennographic detection (GC-EAD) and gas chromatography–mass spectrometry (GC–MS). Furthermore, the EAG and behavioural responses of P. xylostella to these compounds were investigated. The GC-EAD results indicated that four compounds, 2,4-dimethylheptane, 4-methyloctane, 2-methylnonane, and myrcene induced EAG responses in female antennae. 2-methylnonane and myrcene elicited strong dose-dependent EAG responses starting at a dose of 1 μg/μL. Choice tests in a four-arm olfactometer showed significant repellent effects of the four compounds on P. xylostella adults at 100 μg/μL, and the repellent indices of 2,4-dimethylheptane, 4-methyloctane, 2-methylnonane, and myrcene were 70.0%, 76.7%, 66.7%, and 73.3% to mated females, respectively. Finally, oviposition repellence assays showed that female adults were significantly repelled by these compounds, and the oviposition repellent index for myrcene was 23.9%. 2,4-dimethylheptane, 4-methyloctane, 2-methylnonane, and myrcene mediate the repellence of wounded M. spicata to P. xylostella adults, suggesting that the four compounds contribute towards developing behavioural regulators of this important pest.

The diamondback moth (DBM), Plutella xylostella (L.), is a cosmopolitan obligate pest of cruciferous crops. Currently, DBMs management is largely dependent on broad-spectrum synthetic pesticides, which has led to the adverse effects on the environment, human health, and nontargeted organisms. Thus, alternative control methods are needed. The essential oils (EOs) derived from bioactive species may be developed as effective alternatives for the management of DBMs that is safe and eco-sustainable than conventional pesticides. In this study, we investigated electrophysiological (EAG) and behavioral responses of DBM adults to the thirteen non-host EOs of Acorus calamus , Ageratum conyzoides , Artemisia argyi , Chenopodium ambrosioides , Cymbopogon citratus , Foeniculum vulgare , Ocimum basilicum , Pelargonium hortorum , Perilla frutescens , Pogostemon cablin , Polygonum hydropiper , Rosmarinus officinalis , and Ruta graveolens in order to contribute to the development of low environmental impact management alternatives. EAG recordings revealed all the tested EOs significantly elicited EAG responses of DBM females, and the males were sensitive to the eleven EOs except A. conyzoides and O. basilicum . Meanwhile, EAG recordings of DBMs exhibited dose-dependent responses to these tested EOs in most cases. In Y-tube olfactometer bioassays, the EOs of A. calamus , C. citratus , P. hortorum , P. hydropiper , and R. graveolen demonstrated prominent repellent effects on DBM females at concentrations of 10 μg/μL or higher. The results of oviposition bioassays found the number of eggs on the two plants with the five tested EOs were less than that on the two plants with paraffin oil. And the oviposition indexes of the plants that fixed with the EOs of P. hortorum , R. graveolen , A. calamus , P. hydropiper , C. citratus , and paraffin oil were –71.81%, –59.43%, –57.46%, –51.11%, –35.41%, and 0.09%, respectively. These findings indicate that the five EOs, especially P. hydropiper EO, provide active and safe tools for natural repellents to prevent and control DBMs infestations of cruciferous vegetables.

Rosemary (Rosmarinus officinalis L) essential oil (REO) has an inhibitory effect against pests and is regarded as a safe alternative to synthetic acaricides. This study investigated the acaricidal and repellent activity of REO and underlying mechanisms against Aleuroglyphus ovatus, an important mite pest of stored products. Gas Chromatography-Mass Spectrometry analysis of REO indicated that the compounds of 1,8-cineole (25.20%) was the highest. REO at 13% concentration provided the highest repellent effect (92.67%) on A. ovatus, and the lowest repellent activity (48.67%) was at 0.8% concentration. Microscopic observations revealed that the mite epidermis started to shrink after treatment with REO at 6 h post-treatment. The enzyme level of catalase (CAT), acetylcholinesterase (AChE) and superoxide dismutase (SOD) decreased significantly (P < 0.05), whereas there was a significant increase (P < 0.05) in nitric oxide synthase (NOS) and glutathione-s-transferases (GST) activities in A. ovatus treated with REO. In the molecular docking study, camphor and 1,8-cineole, the main monoterpenes of REO, bind more closely with CAT and AChE, and PI-conjugated interaction, hydrogen bonding, and hydrophobic were found by molecular docking 3D model. Overall, our results indicate the potential of REO as an effective botanical acaricide to manage A. ovatus.

Tribolium confusum is a major stored-product pest that exhibits resistance to chemically synthesized pest repellents. This study investigated the potential of essential oil (EO) extracted from the roots of Cyperus esculentus as a natural alternative for pest management. The EO was obtained through steam distillation, and its chemical composition was elucidated using gas chromatography–mass spectrometry. The primary compounds, cyperotundone and cyperene, were further isolated from the EO through silica gel column chromatography. The efficacy of the EO and its isolated compounds as pest repellents was evaluated against a flaxseed pest, which was identified as T. confusum through DNA sequence analysis. The results demonstrated that at 86.12 μg/cm 2 , the EO and its two main components maintained significant repellent activity for up to 24 h. In contrast, the effectiveness of the positive control, N, N-diethyl-3-methylbenzamide (DEET) declined rapidly after 8 h. At 16 h, the repellent activity of the EO and one of its main components, cyperotundone, was significantly greater than that of DEET. Furthermore, at a lower concentration of 43.06 μg/cm 2 , cyperotundone’s repellent activity was significantly stronger than DEET’s at 16 h. Additionally, cyperotundone outperformed DEET significantly from 4 to 16 h at 21.53 μg/cm 2 and at 16 h at 10.76 μg/cm 2 . Among the two compounds, cyperotundone exhibited a longer-lasting repellent effect compared to cyperene, which is consistent with the lower evaporation rate of cyperotundone. Biochemical assays revealed that exposure to the EO of C. esculentus and its major compounds significantly reduced ( p < 0.05) the activities of acetylcholinesterase and glutathione-S-transferase in T. confusum . Molecular docking experiments indicated that the compounds could bind to olfactory receptors with low binding energies. qRT-PCR analysis revealed that the EO and its two compounds significantly altered ( p < 0.05) the expression levels of odorant receptor genes in the pest. These findings suggest that the repellent action of C. esculentus EO and its major compounds on T. confusum may be mediated through the modulation of the pest’s olfactory system, as well as by inhibiting essential enzymatic activities in the pests. This research contributes valuable insights into the development of sustainable, long-lasting, and eco-friendly pest repellents, harnessing the potential of the rich botanical resource C. esculentus .

Using plant essential oil instead of chemical pesticides to control melon fly ( Bactrocera cucurbitae (Coquillett)) is still a great challenge in agricultural practices, which must effectively slow down the volatility rate of plant essential oil. In this study, citral as a natural plant essential oil was blended with environmentally friendly castor oil based waterborne polyurethane (PU). The interaction, chemical resistance, adhesion property and sustained release behavior of the composites with different citral content were clarified, and the efficacies against the melon fly under field conditions were explored. The results indicated that the composites prepared exhibited excellent adhesion behavior on bitter gourds ( Momordica charantia L.) even under rainfall washing, meanwhile the chemical resistance of composites could be tailored via controlling citral content. Low citral content composite films could be washed off by alcohol without worrying about the safety of that on crops. The composite prepared exhibited long-lasting repellent behavior against melon fly. The composite can be prepared via a simple technology and low cost, which has great value for plant essential oil applied in agriculture and can provide a new approach for melon fly control.

The diamondback moth (DBM), Plutella xylostella (Lepidoptera: Plutellidae), is a notorious cruciferous vegetable pest globally. Mechanically damaged geranium (Pelargonium × hortorum) can strongly repel DBM, but specific plant bioactive compounds responsible for such effects have not been identified. The headspace volatiles from wounded geranium were analyzed using gas chromatography–electroantennographic detection and gas chromatography–mass spectrometry. The electrophysiological and behavioral responses of DBM females to these chemicals and two blends were investigated. The results showed that five components myrcene, γ-terpinene, linalool, camphor, and terpinen-4-ol in geranium plants mediated the repellence of DBM mated females. These substances significantly repelled the oviposition of females, and blend-2 (a mixture of the five components with a ratio 1:5:3:4:3) was most effective. The slow-release blend-2 had a repellent range of 0.9 m and a repellent longevity of 26 days. These five substances are promising behavioral regulators of the destructive moths and could be potential candidates for “push” components in plant-based “push–pull” strategies.
