Quercetin (Qc) is a natural flavonoid recognized for its diverse biological activities but is poorly soluble in water. Hydroxypropyl beta-cyclodextrin (HP-β-CD), a cyclodextrin analog, serves as a safe and efficient drug carrier by forming clathrates with therapeutic agents, enhancing their physicochemical and pharmacokinetic properties. This study reports the synthesis of inclusion complexes of HP-β-CD and Qc using ultrasonic magnetic stirring followed by lyophilization. These complexes were analyzed using X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM) and UV–Vis spectroscopy (UV), Fourier-Transform Infrared spectroscopy (FTIR), and Nuclear Magnetic Resonance (NMR) to elucidate their structural and physicochemical properties. The findings validated the successful formation of clathrates between Qc and HP-β-CD, with the resulting inclusion complexes comprising Qc/HP-β-CD demonstrating markedly distinct spectral characteristics compared to unmodified Qc. Moreover, studies revealed a substantial enhancement in the anti-inflammatory and osteogenic activities of Qc upon encapsulation. These findings demonstrate that the Qc/HP-β-CD inclusion complexes markedly enhanced Qc bioavailability while amplifying its therapeutic efficacy in inflammation and bone regeneration.

Black phosphorous (BP) is recognized as an effective reinforcement for polymer scaffold because of its excellent mechanical property and biocompatibility. Nevertheless, its poor stability in physiological environment limits its application in bone repair. In this work, BP was modified with dopamine by self-polymerization approach (donated as BP@PDA) to improve its stability, and then was introduced into poly-L-lactic acid (PLLA) scaffold fabricated by selective laser sintering technology. Results showed the compressive and tensile strength of PLLA/BP@PDA scaffold were improved by 105% and 50%, respectively. The enhanced strength was ascribed to the increased stability of BP and the improved compatibility of BP@PDA with PLLA matrix after modifying with polydopamine. Simultaneously, the bioactivity of PLLA scaffold was significantly improved. It was attributed to that BP@PDA provided the sustained source of ions which could capture Ca2+ ions from physiological medium to facilitate in situ biomineralization, thereby promoting cell adhesion, proliferation and differentiation. This study demonstrated the great potential of BP@PDA in bone repair.

Bone defect repair is a hot topic in tissue engineering, and the development of economical and effective bone scaffold materials to achieve the goal of repairing bone defects is an important task for researchers. In this experiment, fresh bovine cancellous bone was used as a xenogeneic
bone material. After two high-temperature calcinations, β-tricalcium phosphate (β-TCP) material was obtained. An in-situ co-precipitation method was employed to prepare hydroxyapatite+graphene oxide (HA+GO) composite material. Then, the β-TCP material
was immersed in the HA+GO composite material solution to prepare β-TCP+HA+GO composite material. The surface modification of β-TCP material with HA and GO improved the elastic modulus of the raw material. In vitro cell showed that the composite material was non-toxic,
biocompatible, and promoted the proliferation of MC3T3-E1 cells on the β-TCP+HA+GO material. Finally, it was found that β-TCP+HA+GO composite material effectively stimulated the actin and mineralization behavior of MC3T3-E1 cells, and promoted the expression of bone-related
factors such as Alkaline phosphatase (ALP), Collagen I (COLI), Osteocalcin (OCN), Bone Morphogenetic Protein-2 (BMP-2), and Bone Morphogenetic Protein-7 (BMP-7) in MC3T3-E1 cells In conclusion, β-TCP+HA+GO composite material can be applied in the biomedical field as a bone substitute
or repair material.

It is important to eliminate lipopolysaccharide (LPS) along with killing bacteria in periprosthetic joint infection (PJI) therapy for promoting bone repair due to its effect to regulate macrophages response. Although natural antimicrobial peptides (AMPs) offer a good solution, the unknown toxicity, high cost and exogenetic immune response hamper their applications in clinic. In this work, we fabricated a nanowire-like composite material, named [email protected] , by combining chitosan and puerarin via solid-phase reaction, which can finely mimic the bio-functions of AMPs. Chitosan, serving as the bacteria membrane puncture agent, and puerarin, serving as the LPS target agent, synergistically destroy the bacterial membrane structure and inhibit its recovery, thus endowing [email protected] with good antibacterial property. In addition, [email protected] possesses good osteoimmunomodulation due to its ability of LPS elimination and macrophage differentiation modulation. The in vivo results show that [email protected] can inhibit the LPS induced bone destruction in the Escherichia coli infected rat. [email protected] exhibits superior bone regeneration in Escherichia coli infected rat due to the comprehensive functions of its superior antibacterial property, and its ability of LPS elimination and immunomodulation. [email protected] can well mimic the functions of AMPs, which provides a novel and effective method for treating the PJI in clinic.

Malignant bone tumor defects are difficult to treat because of the simultaneous need for tumor treatment and bone-repair promotion. This study presents a bioactive composite scaffold (T-rGO@Au) for personalized bone defect repair and bone tumor treatment. The T-rGO@Au scaffold has a porous structure, and its mechanical properties are close to those of human cancellous bone. The T-rGO@Au scaffold can induce upregulation of osteopontin (OPN), RUNX-2, and osteocalcin (OCN) gene expression. In vivo experiments showed that the bone volume/total volume (BV/TV) ratio with the T-rGO@Au scaffold was the highest. The new bone was tightly integrated with the implant, demonstrating effective osseointegration. The T-rGO@Au scaffold locally generated high temperatures and reactive oxygen species under near-infrared excitation, and AuNPs enhanced the photothermal performance of rGO through the “antenna effect.” Furthermore, in vitro experiments showed that the tumor cell nuclei were destroyed, late-stage apoptotic cells increased, and cell morphology was severely damaged. Additionally, RNA-seq revealed that tumor cell destruction was mediated through signaling pathways, such as the MAPK pathway. In vivo antitumor experiments also demonstrated that the T-rGO@Au scaffold significantly inhibited the growth of tumor cells within 2 weeks. Thus, the T-rGO@Au scaffold provides a new treatment strategy for the development of implantable scaffolds for bone tumor defects.

Poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) has a great potential in bone repair, but unfortunately, the poor mechanical properties limit its further application. In this work, zinc oxide (ZnO) nanoparticles were incorporated into PHBV porous scaffold fabricated by selective laser sintering technique. It was because ZnO nanoparticles could provide nucleating sites for the orderly stacking of polymer chains, thereby enhancing the crystallinity of PHBV. It was well known that the mechanical properties of PHBV scaffold could be enhanced with the increase of crystallinity. More significantly, the released Zn 2+ would combine negatively charged cell membranes of bacterial through electrostatic interaction and consequently destructed the protein structure and resulted in the death of bacterial, which was highly desired in reducing the risk of implant infection. Results indicated that the relative crystallinity of scaffold with 3 wt.% ZnO increased remarkably from 38% to 64% compared to pure PHBV scaffold, which effectively enhanced the compression strength and modulus by 56% and 51.5%, respectively. Moreover, the scaffold had a favorable antibacterial activity. Cell culture experiments proved that the scaffold could promote the cell behaviors. The positive results demonstrated the scaffold may serve as a potential replacement in bone repair.

Critical-sized bone defects present a formidable challenge in tissue engineering, necessitating innovative approaches that integrate osteogenesis and angiogenesis for effective repair. Inspired by the hierarchical porous structure of natural bone, this study introduces a novel method for the scalable production of ultra-long, copper-doped hydroxyapatite (Cu-HAp) fibers, utilizing the rapid gelation properties of guar gum (GG) under controlled conditions. These fibers serve as foundational units to fabricate three-dimensional porous scaffolds with a biomimetic hierarchical architecture. The scaffolds exhibit a broad pore size distribution (1–500 μm) and abundant nanoporous features, mimicking the native bone extracellular matrix. Physicochemical characterization and in vitro assays demonstrated that the copper doping significantly enhanced osteogenic and angiogenic activities, with optimized concentrations (0.8 % and 1.2 % Cu) facilitating the upregulation of osteogenesis-related genes and proteins, as well as promoting endothelial cell proliferation. In vivo studies further confirmed the scaffolds' efficacy, with the 1.2Cu-HAp group showing a remarkable increase in bone regeneration (bone volume/total volume ratio: 35.7 ± 1.87 %) within the defect site. This research offers a promising strategy for the rapid fabrication of multifunctional scaffolds that not only support bone tissue repair but also actively accelerate the healing process through enhanced vascularization.

Microspheres (MPs) and porous microspheres (PMPs) are the two most widely used microparticles in tissue engineering and stem cell therapy. However, how stem cells perceive the topological differences between them to regulate cell function remains to be unclear. Here, we systematically studied the changes in stem cell function under the action of MPs and PMPs and elucidated the related mechanisms. Our findings show that the porous structure of PMPs can be sensed by focal adhesions (FAs), which triggers the synthesis of F-actin to inhibit the phosphorylation and degradation of Yes-associated protein (YAP), while also transmitting stress to the nucleus through the contraction of F-actin, thereby enhancing the nuclear translocation of YAP protein. The activation of YAP significantly enhances the proliferation, osteogenesis, paracrine and glucose metabolism of BMSCs, making them exhibit stronger bone repair ability in both in vivo and in vitro experiments. In summary, this study provides a comprehensive and reliable understanding of the behavior of BMSCs in response to MPs and PMPs. It also deepens our understanding of the association between microparticles’ topological cues and biological functions, which will provide valuable guidance for the construction of bone tissue engineering (BTE) scaffolds.

The repair of large bone defects remains a significant challenge in the field of orthopedics, as traditional bone substitutes face issues such as limited supply and structural incompatibility. Therefore, the development of novel biomaterials to promote bone repair is of great significance. This study proposes a composite material based on a gelatin/alginate dual-network hydrogel scaffold (Gel/AlgMA), which regulates oxidative stress in the local microenvironment of bone defects by loading cerium metal–organic frameworks (Ce-UiO-66) and resveratrol (Res). The cerium ions in Ce-UiO-66 exhibit excellent antioxidant properties in their multivalent states, capable of scavenging excess reactive oxygen species (ROS), improving mitochondrial function, and enhancing stability through a polydopamine (PDA) coating. The Gel/Alg@Ce-Res/PDA composite scaffold was prepared via photopolymerization and its in vitro biocompatibility, antioxidant properties, and osteogenic potential were evaluated. The results indicated that the composite scaffold effectively scavenged ROS, alleviated oxidative stress, and promoted the proliferation and differentiation of osteoblasts. Moreover, in vivo experiments further confirmed the favorable effects of the Gel/Alg@Ce-Res/PDA scaffold in bone defect repair. This study provides an innovative biomaterial design for bone defect treatment, with promising clinical application prospects.

Hydroxyapatite (HA) has been broadly used for the repair of human hard tissues due to its bioactivity and similarity to the mineral of bones and teeth. We prepared a fluorine (F), ytterbium (Yb) and holmium (Ho) ternary doped HA (HYH-F) material and investigated its dual functions of bone repair and multimodal tracking. The results showed that the HYH-F material could display clear X-ray micro-computed tomography (Micro-CT) images based on Yb ions, and Ho ions due to its superparamagnetic properties for magnetic resonance imaging (MRI), while the F ions could enhance the upconversion (UC) fluorescence of the Yb/Ho combination. The ternary doped HYH-F material shows good compatibility with cells and bone tissue. A joint usage of Micro-CT imaging and UC fluorescence imaging distinctly and synergistically demonstrated the distribution state and degradation of the HYH-F material during new bone reconstruction. The single matrix HA material with ternary doping will be beneficial for future biomedical investigation and applications, and it can not only be a bone repair biomaterial, but also provide lifelong multimodal tracking efficacy.

Cartilage repairing grafts have been widely studied, and osteochondral replacement hydrogels have proven to be an excellent method in research and clinical fields. However, it has been difficult to simultaneously solve three main issues in osteochondral replacement preparation: surface lubrication, overall mechanical support and good simulations of cell regeneration. A novel integrated bilayered hydrogel osteochondral replacement was constructed by blending polyvinyl alcohol (PVA) and β-tricalcium phosphate (β-TCP) in this study. Separated nano-ball milling with ultrasound dispersion prepared β-TCP demonstrated suitable properties of tiny particle size, high purity and ideal distribution, improving the mechanical properties of the novel integrated hydrogel, and providing a cartilage-like lubrication effect and high biocompatibility, including cytocompatibility and osteogenesis. The reinforcement of β-TCP and integrated molding technology enabled the hydrogel to demonstrate excellent component compatibility and good bonding between the two layers, which promoted the strengthening of the compression modulus and tensile modulus up to three times by mechanical testing. The surface lubrication properties of the novel osteochondral hydrogel were similar to the natural cartilage by friction coefficient characterization. The two layers of the novel integrated graft provided a considerable bio-function by co-culturing with chondrocytes and synovium mesenchymal stem cells: chondrocytes promoted adherence achieved by the upper density layer and better osteogenesis performance of the porous lower layer. The design of the bilayered β-TCP/PVA osteochondral hydrogel is promising for use in articular cartilage repair.

Current approaches for bone repair predominantly target localized delivery of growth factors that are aimed at the coupling of angiogenesis and osteogenesis. However, delayed revascularization and regeneration of critical-sized bone defects are still challenging. In this study, we engineer an ossification center-like organoid (OCO) that consist of an inner-core bone morphogenetic and neurotrophic spheroid generated via MSCs-loaded 3D printing, alongside the interstitially distributed outer-shell proangiogenic neurotrophic phase. Our results demonstrate that collective implantation of OCOs achieves rapid bone bridging with successive OC-like bone ossicles formation across the bone defect in a “divide-and-conquer” way. Single-cell RNA sequencing analysis unveils a developmentally mimicking stem cell community that dominated with Krt8 + skeletal stem cells (SSCs) is uniquely recruited by the pro-regenerative in-situ organoid fusion and maturation. Particularly noteworthy is the specific expansion of Krt8 + SSCs concomitant with the simultaneous reduction of Has1 + migratory fibroblasts (MFs) post-OCO implantation. Furthermore, cross-species comparisons employing machine learning reveal high resemblance of the relative Krt8 + SSCs/Has1 + MFs composition in bone regeneration with that in public data from developmental bone tissues. Our findings advocate an approach akin to “divide-and-conquer” utilizing engineered OC-like organoids for prompt regeneration of large-sized bone defects.
