With the increase in population aging, the tendency of osteochondral injury will be accelerated, and repairing materials are increasingly needed for the optimization of the regenerative processes in bone and cartilage recovery. The local environment of the injury sites and the deficiency of Mg2+retards the repairing period via inhibiting the progenitor osteogenesis and chondrogenesis cells’ recruitment, proliferation, and differentiation, which results in the sluggish progress in the osteochondral repairing materials design. In this article, we elucidate the Mg2+-concentration specified effect on the cell proliferation, osteochondral gene expression, and differentiation of modeling chondrocytes (extracted from New Zealand white rabbit) and osteoblasts (MC3T3-E1). The concentration of Mg2+in the culture medium affects the proliferation, chondrogenesis, and osteogenesis: (i) Appropriate concentrations of Mg2+promote the proliferation of chondrocytes (1.25–10.0 mM) and MC3T3-E1 cells (2.5–30.0 mM); (ii) the optimal concentration of Mg2+that promotes the gene expression of noncalcified cartilage is 15 mM, calcified cartilage 10 mM, and subchondral bone 5 mM, respectively; (iii) overdosed Mg2+leads to the inhibition of cell activity for either chondrocytes (>20 mM) or osteoblasts (>30 mM). The biomimetic elucidation for orchestrating the allocation of gradient concentration of Mg2+in accordance of the physiological condition is crucial for designing the accurate microenvironment in osteochondral injury defects for optimization of bone and cartilage repairing materials in the future. Keywords:magnesium;chondrogenesis;osteogenesis;osteochondral defect;osteochondral unit

The distinctive structure and composition of the Haversian canal within osteons play a pivotal role in sustaining the nutritional supply to individual bone cells. Consequently, the osteon's inherent advantages in facilitating bone repair have garnered increasing attention. However, most existing designs emulate only partial aspects of the osteon's architecture, failing to replicate its structure and functionality comprehensively. This study aimed to develop a functional 3D biomimetic osteon to enhance bone regeneration. To achieve this, oriented nanofibers mimicking osteonal lamellae are fabricated via electrospinning, while a biomimetic blood vessel, simulating the Haversian canal, is constructed using microfluidics technology. The biomimetic blood vessels, seeded with rat umbilical vein endothelial cells, secreted bone morphogenetic protein to stimulate osteogenesis and released platelet-derived growth factor to promote angiogenesis, further supporting osteogenesis processes. The nanofibers, composed of type I collagen and nano-hydroxyapatite, released calcium ions, can facilitate the recruitment of bone marrow mesenchymal stem cells, enhance their adhesion to the fibers, and promote osteogenic differentiation. These findings demonstrate that replicating both the natural structure and function of bone tissue provides a superior strategy for bone repair.

The interconnectivity of porous scaffold is vital for cell and tissue infiltration, and vascular networks formation, determining the successful bone regeneration in large bone defects. Here, inspired by the lotus-root and Haversian system of natural bone, a nano-hydroxyapatite/polyurethane (n-HA/PU) lotus root-like scaffold inlaid with micro holes on the wall of the adjacent channel was utilized to mediate cell biomineralization and vascularized bone tissue regeneration. Such a particular lotus-type structure remarkably promoted cells to proliferate and infiltrate into the center of the entire scaffold, serving as a clue to account for regulating cell alignment and differentiation physically. In vitro studies suggested that apatite accumulated dramatically on the channel wall in the lotus-type scaffold, probably promoting specific osteogenic differentiation of cells by the orientated channels, even in the absence of osteogenic factors. In vivo creation of critical bone defects (15-mm segments) was done in the radius of rabbits and implanted with the scaffold of different geometry to assess the structural parameters on the efficacy of new bone regeneration. The more extensive positive staining of BMP-2, more considerable amount of infiltrated capillary, more robust new bone formation, particularly the biomechanical strength of lotus-type scaffold group could reach the level of the control group without surgery, indicating that the lotus-type scaffold was more favorable for new bone tissue formation along tube-like channels. These results highlighted the potential of this biomimetic scaffold for cell and tissue infiltration and thus repair large bony defects.

Repairing large bone defects remains a significant clinical challenge, as they exceed the bone’s innate regenerative capacity. The current gold standard, autologous bone grafting, is constrained by donor site morbidity, limited tissue availability, and additional surgical risks. Bone tissue engineering offers a compelling alternative; yet achieving fully functional, vascularized bone regeneration remains challenging. Inspired by the hierarchical architecture of native osteons, a dual-ring hydrogel scaffold was developed by incorporating the zwitterionic monomer 2-(methacryloyloxy)ethyl choline phosphate (MCP) into a gelatin methacryloyl (GelMA) matrix. This design facilitated spatially programmed encapsulation of human umbilical vein endothelial cells (HUVECs) and bone marrow-derived mesenchymal stem cells (BMSCs), replicating the cellular spatial organization of native osteons. MCP integration imparted the GelMA hydrogel with enhanced hydrophilicity, tunable porosity, and optimized mechanical and degradable properties. Optimized GelMA-MCP formulations supported the cell-specific viability and functional activity of HUEVCs and BMSCs, while the spatial configuration promoted synergistic angiogenesis and osteogenesis. Both in vitro and in vivo evaluations demonstrated that the osteon-like scaffold significantly enhanced angiogenesis and bone regeneration compared to mixed or single-cell controls. This study describes a modular and biomimetic strategy for engineering vascularized bone tissue through the integration of functional monomers and spatially guided cell patterning.
