Functionalized BaTiO3 enhances piezoelectric effect towards cell response of bone scaffold

Cijun Shuai,Guofeng Liu,Youwen Yang,Wenjing Yang,Chongxian He,Guoyong Wang,Zheng Liu,Fangwei Qi,Shuping Peng

Piezoelectric effect of polyvinylidene fluoride (PVDF) plays a crucial role in restoring the endogenous electrical microenvironment of bone tissue, whereas more β phase in PVDF leads to higher piezoelectric performance. Nanoparticles can induce the nucleation of the β phase. However, they are prone to aggregate in PVDF matrix, resulting in weakened nucleation ability of β phase. In this work, the hydroxylated BaTiO 3 nanoparticles were functionalized with polydopamine to promote their dispersion in PVDF scaffolds fabricated via selective laser sintering. On one hand, the catechol groups of polydopamine could form hydrogen bonding with the hydroxyl groups of the BaTiO 3 . On the other hand, the amino groups of polydopamine were able to bond with C F group of PVDF. As a result, the functionalized BaTiO 3 nanoparticles homogeneously distributed in PVDF matrix, which significantly increased the β phase fraction from 46% to 59% with an enhanced output voltage by 356%. Cell testing confirmed the enhanced surface electric cues significantly promoted cell adhesion, proliferation and differentiation. Furthermore, the scaffolds exhibited enhanced tensile strength and modulus, which was ascribed to the rigid particle strengthening effect and the improved interfacial adhesion. This study suggested that the piezoelectric scaffolds shown a potential application in bone repair.

Reduced graphene oxide-mediated electron-hole separation using titanium dioxide increases the photocatalytic antibacterial activity of bone scaffolds

Feng Pei,Tian Haifeng,Yang Feng,Peng Shuping,Pan Hao,Shuai Cijun

Fast electron–hole recombination issues during titanium dioxide (TiO 2 ) photocatalysis limit its application in preventing bacterial infection during bone defect repair. In this study, TiO 2 @reduced graphene oxide (rGO) composites were synthesized using a hydrothermal method in which rGO, which possesses very high electrical conductivity, promotes the separation of photoelectron–hole pairs of TiO 2 , thus improving the efficiency of photocatalytic production of reactive oxygen species (ROS). Subsequently, TiO 2 @rGO composites were introduced into poly-L-lactic acid (PLLA) to prepare bone scaffolds with photocatalytic antibacterial function via selective laser sintering. The results showed that TiO 2 grew on the surface of rGO and formed a covalent bond connection (Ti–O–C) with rGO. A decreased electrochemical impedance of TiO 2 @rGO composites was observed, and the transient photocurrent intensity increased from 0.05 to 0.5 µA/cm 2 . Analysis of electron spin resonance found that the photocatalytic products of TiO 2 were ·OH and ·O 2 − , two kinds of ROS capable of killing bacteria via disrupting the structure of the bacterial membrane in vitro. Antibacterial experiments showed that the PLLA/TiO 2 @rGO scaffolds had good antibacterial properties against Escherichia coli and Staphylococcus aureus . Finally, we report that these scaffolds exhibited both enhanced mechanical properties due to the addition of TiO 2 @rGO as a reinforcement material and good biocompatibility during cell proliferation.

O2-self-supplying and glutathione-depleting scaffold for Synergistic Cancer Therapy

Jiajia Zeng,Chongxian He,Xinna Bai,Cijun Shuai,Hao Pan

Photodynamic therapy (PDT) has great potential for tumor therapy because of its non-invasive and high selectivity. However, the therapeutic effect of PDT is critically restricted by hypoxia and glutathione (GSH) overexpression in the tumor microenvironment (TME). In this work, CeO 2 with catalase and peroxidase dual enzyme activities was loaded on Ti 3 C 2 nano-sheet, and the nanosystem was then introduced in poly-L-lactic acid (PLLA) and prepared into bone scaffold using selective laser sintering technology. Specifically, CeO 2 alleviates hypoxia through its catalase enzyme activity, which promotes the continuous photodynamic process. CeO 2 also consumes over-expressed GSH through its redox reaction and amplifies the oxidative stress in TME, thus improving the curative effect of PDT. On the other hand, the photothermal effect of scaffold can not only generate heat to kill tumor cells, but also enhance the activity of nanoenzyme, and the catalase activity enhanced by photothermal effect is fed back to PDT, forming a two-way promotion. The results indicated that the concentration of oxygen in PLLA/Ti 3 C 2 -CeO 2 scaffold group was 9.76 mg/L higher, and the concentration of singlet oxygen ( 1 O 2 ) and hydroxyl radical (·OH) was respectively increased by 60% and 170%, compared with the PLLA/Ti 3 C 2 scaffold. Moreover, the PLLA/Ti 3 C 2 -CeO 2 scaffold showed a higher GSH consumption rate, indicating that it could consume the overexpressed GSH. Therefore, the Ti 3 C 2 nano-sheet loaded with CeO 2 could combine the dual enzyme activities, photodynamic and photothermal effect to enhance the tumor therapy with a tumor growth inhibitory effect of approximately 88.8%. In summary, introducing the Ti 3 C 2 nanosystem loaded with nanozyme into PLLA scaffold is a promising strategy to develop bone scaffold with multimodal anti-tumor function.

Transcrystalline growth of PLLA on carbon fiber grafted with nano-SiO2 towards boosting interfacial bonding in bone scaffold

Pei Feng,Jiye Jia,Shuping Peng,Yang Shuai,Hao Pan,Xinna Bai,Cijun Shuai

Background The reinforcement effect of fiber-reinforced polymer composites is usually limited because of the poor interfacial interaction between fiber and polymer, though fiber reinforcement is regarded as an effective method to enhance the mechanical properties of polymer. Methods In this study, nano-SiO 2 particles grafted by 3-Glycidoxypropyltrimethoxysilane (KH560) were introduced onto the surface of 3-Aminopropyltriethoxysilane (KH550) modified carbon fiber (CF) by a self-assembly strategy to improve the interfacial bonding between CF and biopolymer poly (lactic acid) (PLLA). Results The results indicated that PLLA chains preferred to anchor at the surface of nano-SiO 2 particles and then formed high order crystalline structures. Subsequently, PLLA spherulites could epitaxially grow on the surface of functionalized CF, forming a transcrystalline structure at the CF/PLLA interface. Meanwhile, the nano-SiO 2 particles were fixed in the transcrystalline structure, which induced a stronger mechanical locking effect between CF and PLLA matrix. The results of tensile experiments indicated that the PLLA/CF-SiO 2 scaffold with a ratio of CF to SiO 2 of 9:3 possessed the optimal strength and modulus of 10.11 MPa and 1.18 GPa, respectively. In addition, in vitro tests including cell adhesion and fluorescence indicated that the scaffold had no toxicity and could provide a suitable microenvironment for the growth and proliferation of cell. Conclusion In short, the PLLA/CF-SiO 2 scaffold with good mechanical properties and cytocompatibility had great potential in the application of bone tissue engineering.

Oxygen vacancy boosting Fenton reaction in bone scaffold towards fighting bacterial infection

Cijun Shuai,Xiaoxin Shi,Feng Yang,Haifeng Tian,Pei Feng

Bacterial infection is a major issue after artificial bone transplantation due to the absence of antibacterial function of bone scaffold, which seriously causes the transplant failure and even amputation in severe cases. In this study, oxygen vacancy (OV) defects Fe-doped TiO2 (OV-FeTiO2) nanoparticles were synthesized by nano TiO2 and Fe3O4 via high-energy ball milling, which was then incorporated into polycaprolactone/polyglycolic acid (PCLGA) biodegradable polymer matrix to construct composite bone scaffold with good antibacterial activities by selective laser sintering. The results indicated that OV defects were introduced into the core/shell-structured OV-FeTiO2 nanoparticles through multiple welding and breaking during the high-energy ball milling, which facilitated the adsorption of hydrogen peroxide (H2O2) in the bacterial infection microenvironment at the bone transplant site. The accumulated H2O2 could amplify the Fenton reaction efficiency to induce more hydroxyl radicals (·OH), thereby resulting in more bacterial deaths through ·OH-mediated oxidative damage. This antibacterial strategy had more effective broad-spectrum antibacterial properties against Gram-negative Escherichia coli (E. coli) and Gram-positive Staphylococcus aureus (S. aureus). In addition, the PCLGA/OV-FeTiO2 scaffold possessed mechanical properties that match those of human cancellous bone and good biocompatibility including cell attachment, proliferation and osteogenic differentiation.

Construction of S-Scheme Heterojunction by TiO2/rGO Assembled With g-C3N4 for Efficient Visible-Light-Driven Antibacterial Activity of Bone Scaffold

Jiye Jia,Shijie Chen,Haifeng Tian,Dong Wang,Feng Yang,Pei Feng,Cijun Shuai

The low photogenerated charge carriers and quick complex annihilation of titanium dioxide (TiO 2 )/rGO composite leading to an unsatisfactory photocatalysis antibacterial effect limit its application in the bone scaffold. Herein, graphitic carbon nitride (g-C 3 N 4 ) was assembled with TiO 2 /rGO via a solvothermal method for enhancing the photocatalysis antibacterial properties of poly (lactic acid) (PLLA) bone scaffold prepared via selective laser sintering. The results revealed that g-C 3 N 4 expanded the light absorption range of TiO 2 /rGO from blue-violet light to visible light, resulting in the composites having a narrower band gap and producing more photoinduced charge carriers. More importantly, an S-scheme heterojunction was formed between g-C 3 N 4 and TiO 2 , which promoted the recombination of the electrons in the conduction band of TiO 2 and the holes in the valence band of g-C 3 N 4 . Meanwhile, rGO with excellent electrical conductivity reduced the interfacial charge transfer resistance and enhanced charge transfer efficiency, thus further avoiding the recombination rate of electrons and holes, whether TiO 2 or g-C 3 N 4 , and enhancing the photocatalytic properties. Consequently, the holes in the valence band of TiO 2 and the electrons in the conduction band of g-C 3 N 4 possessed high oxidation and reduction capacity, which oxidized H 2 O and reduced O 2 in the microenvironment, respectively, and generated lots of reactive oxygen species. In vitro antibacterial experiments indicated that the PLLA/TiO 2 /g-C 3 N 4 /rGO scaffold showed a better antibacterial effect, with the bacteriostasis rates of E. coli and S. aureus being 99.1% and 98.9%, respectively. In addition, cell adhesion and fluorescence results indicated that the scaffold had good biocompatibility and could provide a suitable microenvironment for the growth and proliferation of cells.

Tip-Viscid Electrohydrodynamic Jet 3D Printing of Composite Osteochondral Scaffold

Kai Li,Dazhi Wang,Fangyuan Zhang,Xiaoying Wang,Hairong Chen,Aibing Yu,Yuguo Cui,Chuanhe Dong

A novel method called tip-viscid electrohydrodynamic jet printing (TVEJ), which produces a viscous needle tip jet, was presented to fabricate a 3D composite osteochondral scaffold with controllability of fiber size and space to promote cartilage regeneration. The tip-viscid process, by harnessing the combined effects of thermal, flow, and electric fields, was first systematically investigated by simulation analysis. The influences of process parameters on printing modes and resolutions were investigated to quantitatively guide the fabrication of various structures. 3D architectures with high aspect ratio and good interlaminar bonding were printed, thanks to the stable fine jet and its predictable viscosity. 3D composite osteochondral scaffolds with controllability of architectural features were fabricated, facilitating ingrowth of cells, and eventually inducing homogeneous cell proliferation. The scaffold’s properties, which included chemical composition, wettability, and durability, were also investigated. Feasibility of the 3D scaffold for cartilage tissue regeneration was also proven by in vitro cellular activities.Keywords:3D printing;electrohydrodynamic jet;thermal field;fluid viscosity;osteochondral scaffold

Silver-modified graphene oxide nanosheets for antibacterial performance of bone scaffold

Yu Zhiheng,Xu Yang,Tian Xiu

Endowing scaffold with antibacterial activity is an effective countermeasure to prevent bacterial infection in bone repair. Silver nanoparticles (Ag NPs) possess broad-spectrum antibacterial efficiency, whereas the agglomeration and burst releasing of Ag NPs hindered their clinic application in bone repair. In this work, Ag NPs were in situ grown on graphene oxide (GO) to construct Ag@GO nanohybrids and then were introduced into polymer scaffold. GO could efficiently load Ag NPs thereby improving their agglomeration in a scaffold, owing to their abundant active groups and large surface areas. Furthermore, GO could realize the sustained release of Ag ions from the scaffold. The results demonstrated the antibacterial scaffold exhibited robust antibacterial performance with an antibacterial rate of 95% against Staphylococcus aureus . On one hand, GO with honeycomb nanostructure and sharp edge could capture and pierce bacteria membrane, which results in physical damage of bacteria. On the other hand, the released Ag NPs from Ag@GO nanohybrids could promote the generation of reactive oxygen species, which causes the inactivation of bacteria. Encouragingly, the antibacterial scaffold also exhibited good cytocompatibility. This work developed an efficient antibacterial material for the scaffold in bone repair.

Improvement of the mechanical properties and osteogenic activity of 3D-printed polylactic acid porous scaffolds by nano-hydroxyapatite and nano-magnesium oxide

Dian Xu,Zexian Xu,Lidi Cheng,Xiaohan Gao,Jian Sun,Liqiang Chen

Porous bone scaffolds based on high-precision 3D printing technology gave recently been developed for use in bone defect repair. However, conventional scaffold materials have poor mechanical properties and low osteogenic activity, limiting their clinical use. In this study, a porous composite tissue-engineered bone scaffold was prepared using polylactic acid, nano-hydroxyapatite, and nano-magnesium oxide as raw materials for high-precision 3D printing. The composite scaffold takes full advantage of the personalized manufacturing features of 3D printers and can be used to repair complex bone defects in clinical settings. The composite scaffold combines the advantages of nano-hydroxyapatite, which improves the formability of scaffold printing, and of nano-magnesium oxide, which regulates pH during degradation and provide a good environment for cell growth. Additionally, nano-magnesium oxide and nano-hydroxyapatite have a bidirectional effect on promoting the compressive strength and osteogenic activity of the scaffolds. The prepared composite porous scaffolds based on 3D printing technology show promise for bone defect repair.

Surface modification enhances interfacial bonding in PLLA/MgO bone scaffold

Cijun Shuai,Jun Zan,Youwen Yang,Shuping Peng,Wenjing Yang,Fangwei Qi,Lida Shen,Zongjun Tian

The poor interfacial bonding and resultant agglomeration of nanoparticles in polymer-based composite severely deteriorated their reinforcement effect. In this work, MgO nanoparticles (MgO-NPs) were surface modified with Poly (L-lactic acid- co -malic acid) (PLMA) to improve the interfacial compatibility in Poly-l-lactic acid (PLLA) scaffold manufactured by selective laser sintering. PLMA possess a hydrophilic end with carboxyl group (comes from the malic acid) and an l-lactic acid chain. On one hand, the carboxyl group was able to form hydrogen bonding with the hydroxyl groups of MgO-NPs. On the other hand, the l-lactic acid chain containing the hydroxyl groups could react with the carboxyl group of PLLA. Results revealed that the scaffold exhibited significantly enhanced compressive strength and modulus by 47.1% and 237.7%, respectively, which could be ascribed to the enhanced interfacial bonding between PLLA and MgO-NPs, as well as the rigid particle reinforcement. In addition, the scaffold was favorable for cell adhesion, proliferation and differentiation, owing to the improved hydrophilic and suitable pH environment. It was suggested the scaffold was a promising material for bone repair application.

Surface-Modified Graphene Oxide with Compatible Interface Enhances Poly-L-Lactic Acid Bone Scaffold

Guoyong Wang,Chongxian He,Wengjing Yang,Fangwei Qi,Guowen Qian,Shuping Peng,Cijun Shuai

Graphene oxide (GO) usually serves as a reinforce phase in polymer because of its superior mechanical strength and high specific surface area. In this work, GO was grafted with L-lactic acid monomer (denoted as GO@PLLA) to overcome the aggregation in matrix and then incorporated into the poly-L-lactic acid (PLLA) scaffold fabricated by selective laser sintering. In hybrid scaffold, GO@PLLA exhibited uniform dispersion in the matrix. Furthermore, mechanical interlock between GO@PLLA and PLLA matrix formed and reinforced the interface bonding. On the other hand, the heterogeneous distributed GO acted as effective nucleating agent and resultantly enhanced the crystallization. Results showed that the tensile and compressive strength of scaffolds increased by 143.3% and 127.6%, respectively. Meanwhile, the scaffold exhibited an increased degradation rate of 37.9%, which could be attributed to the abundant hydrophilic functional groups on GO. Moreover, the scaffold exhibited favorable bioactivity and biocompatibility. Herein, the developed hybrid scaffold showed potential capacity for bone tissue engineering.

Bioactive porous ZrO2-based ceramics with a hierarchical porosity for artificial bone scaffolds

Bohan Wang,Wenjun Yu,Le Fu

Given the pressing clinical need, the market for artificial bone scaffolds in orthopedics is growing at a rapid rate. In the past, ZrO 2 -based biomaterials intended for implantation were dense and ‘bio-inert’. Material scientists have now shifted toward the design of deliberately porous and ‘bioactive’ ZrO 2 -based biomaterials that integrate with human cells and tissues. In this study, we designed and prepared porous and bioactive ZrO 2 –SiO 2 nanocrystalline ceramics. The phase composition, microstructure, pore formation mechanism, and ion release behaviors of the ceramics were explored. The ceramics showed a hierarchical porosity , consisting of connected macropores (∼100 μm or larger) and micropores (∼1 μm). The spherical macropores were formed by a pore-forming agent method, and the irregular micropores were formed because of incomplete densification. The bioactive dopants (Ca and Sr) acted as a destabilizer of tetragonal ZrO 2 . Ca, Si, and Sr ions continuously released from the surface of the samples, providing good bioactivity for the ceramics. The porous and bioactive ceramics show great potential to be used as artificial bone scaffolds.
