It remains challenging for traditional cartilage replacements to simultaneously have the antifouling ability, self-adhesion, and adequate mechanical properties in the physiological environment. Here we develop a tri-layer hydrogel to address this issue. The precursor hydrogel was prepared through a three-cycle freezing-thawing process involving a blend of poly vinyl alcohol, chitosan and deionized water, followed by 3 h immerse in 25 wt% aqueous solution of grape seed protein. The top layer was modified with methacryloyl chloride and polymerized with sulfobetaine methacrylate (SBMA). Meanwhile, the bottom layer was coated with the adhesive of gelatin and tannin. On account of the multiple linkages (crystalline domains, hydrogen bonds, and ionic interactions), the compressive stress remained 42 MPa and the swelling ratio was 0.1 g/g even after a week immersion in simulated synovial fluid. Because of the introduction of SBMA, the hydrogel could resist non-specific protein adsorption and the coefficient of friction was 0.023 in simulated synovial fluid. The amino acids and polyphenol groups could form noncovalent interactions with various substrates, resulting in excellent underwater adhesion capability. The synergy of biocompatible materials and environmentally friendly progress led to exceptional biocompatibility, having promising applications in cartilage replacement.

Barium titanate (BaTiO 3 ) has been used as a bone implant material because of its piezoelectric properties and the ability to promote cell growth when combined with hydroxyapatite. However, the brittleness of BaTiO 3 inhibits its use as a bone replacement material at load-bearing sites, and the reduction of BaTiO 3 content in the composite reduces its piezoelectric effect on bone growth. In this study, we explored a preparation method, which included directional freeze casting and self-solidification of bone cement, to obtain 1-3-type BaTiO 3 /PMMA bio-piezoelectric composites with a lamellar structure. The lamellar BaTiO 3 layer through the composite from the bottom to the top significantly improved the piezoelectric properties of the composite. In addition, the dendritic ceramic bridges on the BaTiO 3 pore walls can improve the compressive strength and elastic modulus of BaTiO 3 /PMMA bio-piezoelectric composites with a lamellar structure. More importantly, it was found that polarized lamellar BaTiO 3 could induce osteoblasts to grow in the direction of the BaTiO 3 layers. When the width of the BaTiO 3 layer was in the range of 8–21 μm, osteoblasts along the BaTiO 3 layer showed well growth, which can be of great value for the production of biomimetic bone units.

This study evaluated biological compatibility and antibacterial activity of a bone substitute material consisting of silver-doped hydroxyapatite (AgHA) and Bio-Oss ® with different mixture ratios in vitro and investigated its antibacterial mechanism. AgHA was synthesized by a chemical precipitation method. After characterization, AgHA was mixed with Bio-Oss ® at three ratios: 1:1, 1:2, and 1:4 by weight. Then, Porphyromonas gingivalis (Pg) and Fusobacterium nucleatum (Fn) were used to test the antibacterial activity of the mixture. Human periodontal ligament fibroblasts and rat bone marrow stromal cells were selected for cytocompatibility experiments. According to results, the peak value of the size of the AgHA was concentrated in the 100–200 nm range, and AgHA particles consisted of short rods. It was confirmed that the structure of AgHA was similar to that of standard hydroxyapatite. All three mixture ratios exhibited obvious antimicrobial properties, which increased with increasing AgHA. According to the effects on the expression of bacterial virulence genes, groups 1:1 and 1:2 both negatively affected Pg and Fn more significantly than group 1:4. Cytotoxicity experiments showed that 1:1 caused little cytotoxicity, while groups 1:2 and 1:4 exerted no significant cytotoxicity. Considering its biological compatibility and antibacterial activity, group 1:2 is the most recommended. © 2017 Wiley Periodicals, Inc. J Biomed Mater Res Part B: Appl Biomater, 106B: 410–420, 2018.

Through the dual-anchoring effect, hydroxyapatite (HA) molecules were intercalated into graphene oxide (GO) layers to form a GO–PEG–HA hybrid, and PVA/GO–PEG–HA nanocomposite hydrogels were prepared. Compared with GO–HA, enhanced hydrogen bonding formed for GO–PEG–HA hybrid, resulting in a higher intercalation ratio and interlayer spacing of GO and homogeneous distribution of HA particles on GO layers without aggregation. With increasing HA content, the pore size increased, and compressive deformation resistance and the lubrication property of hydrogel were improved. Mouse bone marrow mesenchymal stem cells (mBMSCs) with a spindle shape were distributed uniformly and directionally on the hydrogel, and introduction of HA significantly promoted cell proliferation and improved the cytocompatibility of the hydrogel by coating the sharp edge of GO sheets. When the hydrogel was dipped into simulated body fluid solution, the mass ratio of the hydrogel showed a remarkable rise, and a rough surface with deposited HA and HCA nanoparticles was observed, resulting from the osteoinductive deposition effect and mineralizing capability of the hydrogel. Such PVA hydrogels showed promising potential for cartilage replacement application.

The coral, whose main composition is aragonite-type calcium carbonate, has been widely used as bone substitute in clinic. However, the study on calcium carbonate bioceramic has not been largely reported due to difficulty in sintering calcium carbonate which is liable to evidently decompose at low temperature. In this study, a novel calcium carbonate composite ceramic was fabricated by sintering fast at a low temperature. A degradable, biocompatible phosphate-based glass (PG) which grew liquid at a low temperature was added as sintering agent in the sintering process. The sintering schedule was explored by thermal analysis. The phase composition, microstructure, compressive strength, and biocompatibility of calcium carbonate composite ceramics were evaluated. The results revealed that the optimum holding time at the sintering temperature was 20 min. The obtained calcium carbonate composite ceramics did not produce calcium oxide but new compounds according to phase analysis. The compressive strength of calcium carbonate composite ceramics correspondingly increased with growing addition of PG ranging from 10 to 50 wt%. The cell proliferation on the calcium carbonate composite ceramic was not compromised but augmented compared to the neat calcium carbonate ceramic without adding PG as sintering agent. The novel calcium carbonate composite ceramic is a promising bone substitute for bone defects.

Cartilage, as a load-bearing tissue with high-water content, exhibits excellent elasticity and high strength. However, it is still a grand challenge to develop cartilage-adaptive biomaterials for replacement or regeneration of damaged cartilage tissue. Herein, protein templating and mechanical training is integrated to fabricate crystal-mediated oriented chitosan nanofibrillar hydrogels (O-CN gels) with similar mechanical properties and water content of cartilage. The O-CN gels with an ≈74 wt% water content exhibit high tensile strength (≈15.4 MPa) and Young's modulus (≈24.1 MPa), as well as excellent biocompatibility, antiswelling properties, and antibacterial capabilities. When implanted in the box defect of rat's tails, the O-CN gels seal the cartilage (annulus fibrosus) defect, maintain the intervertebral disc height and finally prevent the nucleus herniation. This synergy strategy of protein templating and mechanical training opens up a new possibility to design highly mechanical hydrogels, especially for the replacement and regeneration of load-bearing tissues.

Designing load-bearing hydrogels with superior toughness and stiffness are crucially important for cartilage replacement and tissue engineering. In this investigation, we report that hybrid hydrogels can simultaneously achieve high stiffness and toughness by introducing covalent and physical interactions. The chemical crosslinking can sustain hydrogel networks as a rigid component and the hydrophobic association is introduced as a dynamic crosslinking center for efficient energy dissipation. The storage modulus G′ can reach nearly 40 kPa and the compression stress was nearly 8 MPa at 95% compression strain. Moreover, the hybrid hydrogel exhibited time-dependent, rapid self-recoverable and fatigue resistant behavior based on the cyclic loading-unloading compression test. The hybrid hydrogels with excellent self-recovery and fatigue resistance would provide a large number of potential applications, such as artificial cartilages and muscles and scaffolds for tissue engineering .

BACKGROUND:Inspired by natural bones, many organic components were added to Calcium Phosphate Cements (CPCs) to improve their mechanical strength. However, the strength of these composite CPCs is limited by the low strength of organic components itself and the weak interaction between organic components and CPCs. OBJECTIVE:Firstly, a composite CPC containing mussel-inspired adhesive, Poly-(Dopamine Methacrylamide-co-2-methoxy Ethylacrylate) (pDM) was developed. Secondly, the interactions between pDM and CPC and their effect on mechanical properties were investigated. METHODS:The interactions between pDM and CPC were performed by Nuclear Magnetic Resonance, Laser Raman, X-ray Photoelectron Spectroscopy, Fourier Transform-Infrared Spectroscopy and X-ray Diffraction Analysis. RESULTS:The toughness and compressive strength of pDM-CPC scaffold were both significantly enhanced, because of the enhanced interface binding strength among CPC and pDM due to their interaction and the improved mechanical strength of pDM owing to its self-oxidation cross-linking. The toughness of pDM-CPC scaffolds increased with the increased contents of pDM, while pDM-CPC scaffold containing 35 wt.% pDM had the highest compressive strength of all, which the latter was more than five times compared to that of CPC. CONCLUSION:The mechanically strong pDM-CPC scaffolds has potential application in bone regeneration as well as in craniofacial and orthopedic repair.

Carbon fiber reinforced hydroxyapatite (CF/HA) composites have been considered as a promising candidate for load-bearing bone replacement. However, high-temperature sintering decomposes HA in CF/HA composites, thereby causing oxidation damage in carbon fibers and decreasing the mechanical properties of the composites. In this study, CF/HA composites with excellent mechanical properties and biological activities were fabricated by spark plasma sintering (SPS). The mechanical properties and microstructures of the composites under different sintering temperatures and fiber contents were systematically investigated. Oxidative damage in CF and interfacial problem between fibers and the matrices in the composites were prevented in contrast to that in CF/HA composites prepared through pressureless or hot-pressing sintering. The reinforcing effect of CF was greatly enhanced owing to the absence of oxidative damage. FTIR and XRD results indicates that the HA phases in the composites did not decompose. HA particles size prepared by SPS was smaller than the one prepared by the traditional sintering method. The addition of CF with excellent electrical and thermal conductivities reduced thermal and electrical gradients in the composites during SPS. Accordingly, CF/HA composites with a refined microstructures and improved mechanical properties were obtained. The hardness and bending strength of 0.5 wt% CF-reinforced HA composites (0.5 C F/HA) were 574.0 H V and 110.4 MPa, respectively. The fracture toughness and brittleness index of 0.5 C F/HA were 2.41 MPa m 1/2 and 2.59 μm −1/2 , respectively. Importantly, the fracture toughness and bending strength of 0.5 C F/HA were can match the properties of compact bones. The in vitro behaviour of the CF/HA composites for cell morphology and proliferation were evaluated using MC3T3-E1. MC3T3-E1 cultured on the composites fabricated SPS showed significantly higher attachment and proliferation rates than those cultured on composites prepared through pressureless sintering. The CF/HA prepared by SPS showed potential as load-bearing bone substitutes owing to their excellent properties.

Injectable polymethylmethacrylate (PMMA) bone cement is a widely used bone substitute in cemented arthroplasty, vertebroplasty and osteoporosis fractures. However, due to the inappropriate stiffness, poor bioactivity and high polymerization temperature of PMMA, aseptic loosening of the implanted cement at the bone–cement interface still could be observed in a high rate of patients. To improve the performance of PMMA, artificial extracellular matrices like chitosan–glycerophosphate (CS–GP) thermosensitive hydrogel was introduced into PMMA acting as a pore forming agent and osteoconductive nano-sized hydroxyapatite (nano-HA)/antibiotic gentamicin (GM) as a carrier. It is shown that CS–GP thermosensitive hydrogel can effectively create open pores at the surface of the PMMA cement, which is believed to facilitate bone tissue ingrowth and improve the cement anchorage at the bone–cement interface in future clinical applications. Meanwhile, such a hydrogel effectively decreases the maximum polymerization temperature to below 30 °C, prolongs the working time to more than 720 s and produces cement with a proper modulus of elasticity and a compressive yield strength ranging from 402 to 584 MPa and from 3.1 to 5.9 MPa, respectively. Furthermore, the incorporated nano-HA particles sufficiently increase the mineralization capacity of the cement without compromising its mechanical properties and the incorporated GM remarkably enhances the anti-bacterial activity of the cement. More importantly, nano-HA and GM enriched CS–GP thermosensitive hydrogel effectively improve the overall performance of PMMA cement without influencing the cell survival, suggesting the injectable p-PMMA/CS–GP/nano-HA/GM cement will hold strong promise for future bone reconstruction applications.

Poly(vinyl alcohol) (PVA) hydrogels with high water content, good load-bearing property, low frictional behavior as well as excellent biocompatibility have been considered as promising cartilage replacement materials. However, the lack of sufficient mechanical properties and cell adhesion are two critical barriers for their application as cartilage substitutes. To address these problems, herein, methacrylated PVA with low degree of substitution of methacryloyl group has been synthesized first. Then, methacrylated PVA-glycidyl methacrylate/hydroxyapatite (PVA-GMA/Hap) nanocomposite hydrogels have been developed by the photopolymerization approach subsequently. Markedly, both pure PVA-GMA hydrogel and PVA-GMA/Hap nanocomposite hydrogels exhibit excellent performance in compressive tests, and they are undamaged during compressive stress–strain tests. Moreover, compared to pure PVA-GMA hydrogels, 8.5-fold, 7.4-fold, and 14.2-fold increase in fracture stress, Young's modulus and toughness, respectively, can be obtained for PVA-GMA/Hap nanocomposite hydrogels with 10 wt % Hap nanoparticles. These enhancements can be ascribed to the intrinsic property of PVA-GMA and strong hydrogen bonding interactions between PVA-GMA chain and Hap nanoparticles. More interestingly, significant improvement in the cell adhesion can also be successfully achieved by incorporation of Hap nanoparticles. These biocompatible nanocomposite hydrogels have great potential to be used as cartilage substitutes. © 2018 Wiley Periodicals, Inc. J. Polym. Sci., Part A: Polym. Chem. 2019, 57, 1882–1889

The manufacturing of biomimetic bone characterized by an organic–inorganic combination and fibrous structure has garnered significant attention. Inspired by the formation of multi-layered fibrous structures in bone tissue, this study is based on the fibril assembled from poly(γ-benzyl-L-glutamate) (PBLG) in helicogenic solvent, proposing a non-solvent-assisted 3D printing method for realizing the PBLG 3D printing while generating biomimetic fiber structures in One-Step to mimic the formation of collagen fiber bundles. The unprintable mixture of PBLG and hydroxyapatite nanoparticles (nHA) in 1,4-dioxane exhibits extrudability, self-supporting properties, and plasticity in ethanol. Meanwhile, ethanol-assisted printing leads to the spontaneous growth of PBLG-fibrils into submicron-fibers. Moreover, the integration of nHA with PBLG-fibers through hydrogen bonding contributes to the improvement of printability and mechanical properties. This method of ethanol-assisted fiber generation is successful with concentrated PBLG solutions, overcoming the limitation of previous research that focused only on dilute solutions. To expand the printable window, an ethanol-gel is developed as a support to achieve omnidirectional printing, resolving the issue of interlayer collapse caused by gravity and the conflict between printability and biomimetic fibers generation, optimizing the biomimetic bone manufacturing, leading to the precise biomimetic design of bone structures.
