With the increase in population aging, the tendency of osteochondral injury will be accelerated, and repairing materials are increasingly needed for the optimization of the regenerative processes in bone and cartilage recovery. The local environment of the injury sites and the deficiency of Mg2+retards the repairing period via inhibiting the progenitor osteogenesis and chondrogenesis cells’ recruitment, proliferation, and differentiation, which results in the sluggish progress in the osteochondral repairing materials design. In this article, we elucidate the Mg2+-concentration specified effect on the cell proliferation, osteochondral gene expression, and differentiation of modeling chondrocytes (extracted from New Zealand white rabbit) and osteoblasts (MC3T3-E1). The concentration of Mg2+in the culture medium affects the proliferation, chondrogenesis, and osteogenesis: (i) Appropriate concentrations of Mg2+promote the proliferation of chondrocytes (1.25–10.0 mM) and MC3T3-E1 cells (2.5–30.0 mM); (ii) the optimal concentration of Mg2+that promotes the gene expression of noncalcified cartilage is 15 mM, calcified cartilage 10 mM, and subchondral bone 5 mM, respectively; (iii) overdosed Mg2+leads to the inhibition of cell activity for either chondrocytes (>20 mM) or osteoblasts (>30 mM). The biomimetic elucidation for orchestrating the allocation of gradient concentration of Mg2+in accordance of the physiological condition is crucial for designing the accurate microenvironment in osteochondral injury defects for optimization of bone and cartilage repairing materials in the future. Keywords:magnesium;chondrogenesis;osteogenesis;osteochondral defect;osteochondral unit

The function of hedgehog signaling has previously been shown to be crucial for craniofacial development. In this study, we treated C57/BL6J mice with the hedgehog pathway inhibitor vismodegib by oral gavage to establish a stable vismodegib-induced cleft palate model.
At E10.5 and E12.5, mice in the experimental group were treated with 100 mg/kg of vismodegib, whereas mice in the control group were treated with solvent. The treated pregnant mice were sacrificed on E13.5, E14.5, E15.5, and E16.5. Palatal shelf growth was evaluated via histological and
immunohistochemical analyses as well as palatal organ culture. Immunohistochemical staining was performed to examine the expression of osteogenic proteins in the palatal tissue. A high proportion of the mice administered 2 doses of 100 mg/kg of vismodegib displayed a cleft palate. Histologic
examination revealed severely retarded palatal shelf growth and thickened epithelium in the experimental group. Vismodegib exposure induced complete cleft palate, which was attributed to a reduced cell proliferation rate in the palatal mesenchyme along the anterior-posterior axis. Moreover,
this model also showed delayed ossification in the region of palatine bone with downregulation of Indian hedgehog (Ihh) protein. Our results suggest that vismodegib can be used to inhibit hedgehog signaling to affect palatal morphogenesis. Under treatment with this exogenous inhibitor, the
cell proliferation rate of the palatal shelves and the osteogenic potential of the hard palate were decreased, which likely contributed to the complete cleft palate.

Fermented milk has abundant bioactive peptides, and there is a growing interest in seeking osteogenic peptides of dairy origin. This study employed computer-assisted screening to identify potential osteogenic peptides from fermented milk by Lactobacillus helveticus and investigated the effects of these osteogenic peptides on the proliferation, differentiation, and mineralization of MC3T3-E1 cells, along with their underlying mechanisms. Three osteogenic peptides were identified: AF10 (ARHPHPHLSF), AY17 (AAGGPGAPADPGRPTGY), and NE19 (NIPPLTQTPVVVPPFLQPE). Treatment with 0.2 μM of AF10, AY17, and NE19 promoted the proliferation of MC3T3-E1 cells by increasing the proportion of cells in the S phase and G2/M phase. Further molecular docking revealed significant differences in the interaction of the three osteogenic peptides with the critical domain (Asn12-Lys13-Leu14-Thr15-Gln16-Leu17-Gly18) of the epidermal growth factor receptor (EGFR). The three peptides exhibited different degrees of promotion on the differentiation and mineralization of MC3T3-E1 cells, as well as significant differences in the expression of genes and proteins related to ERK1/2, RUNX2, and BMP2. The AF10 peptide was selected for further validation of the effect of the mitogen-activated protein kinase (MAPK) signaling pathway on the proliferation and expression of differentiation and mineralization-associated genes in MC3T3-E1 cells by combining MAPK pathway inhibitors. Molecular dynamics simulation revealed the stability of AF10 when binding to EGFR. These findings provide valuable insights into the screening of osteogenic peptides of various origins and the development of functional dairy products accordingly.

Biodegradable scaffolds have significant potential for directed tissue regeneration in craniofacial and oral applications and bone regeneration. We designed an innovative cerium-altered/collagen-clacked polycaprolactone/poly-lactic-co-glycolic acid (PC/PL) electrospun scaffolds (PC/PL-PD-Ce-CL) to address the deficiencies of existing scaffold materials regarding osteogenic and antimicrobial characteristics. Novel scaffolds were developed by electrospinning a fundamental PCL/PLGA matrix, impregnating in situ reduction with cerium nanoparticles (CeNPs), applying a polydopamine coating, and clacking with collagen. The mechanical tests and scanning electron microscope demonstrated that the distinctive NSs preserved their tensile strength and elasticity modulus post-modification. The cell proliferation CCK-8 assay indicated that the PC/PL-PD-Ce-CL NSs exhibited a superior MG63 cell proliferation and cell adhesion ratio compared to the rest of the NSs. The alkaline phosphatase (ALP) activity and the expression levels of BMP2 and RUNX2 in MG63 cells cultivated on PC/PL-PD-Ce-CL NSs for 7 and 14 days were considerably elevated compared to the controls. The PC/PL-PD-Ce-CL and PC/PL-PD-Ce NSs exhibited a broader antibacterial zone compared to the control NSs, and bacterial fluorescence diminished on the Ce-altered NSs of incubation with S. mutans and S. aureus . Our innovative PC/PL-PD-Ce-CL NSs improved cytocompatibility, osteogenesis, and antibacterial capabilities, demonstrating their promising therapeutic applications for orofacial treatment.

Despite significant advances in osteochondral tissue engineering, it remains challenging to successfully reconstruct native-like complex tissues organized in three-dimension with spatially varying compositional, structural and functional properties. In this contribution, inspired by the gradients in extracellular matrix (ECM) composition and collagen fiber architecture in native osteochondral tissue, we designed and fabricated a tri-layered (superficial cartilage (S), deep cartilage (D) and subchondral bone (B) layer) stratified scaffold in which a mesenchymal stem cell (MSC)-laden gelatin methacrylamide (GelMA) hydrogel with zone-specific growth factor delivery was combined with melt electrowritten triblock polymer of poly(ε-caprolactone) and poly(ethylene glycol) (PCEC) networks with depth-dependent fiber organization. Introducing PCEC fibers into the weak GelMA hydrogel contributed to a significant increase in mechanical strength . In vitro biological experiments indicated that the stratified fiber-reinforced and growth factor-loaded hydrogel construct induced the MSCs to differentiate down both the chondrogenic and osteogenic lineages and that the engineered complex exhibited cellular phenotype and matrix accumulation profiles resembling those of the native tissue. Simultaneous cartilage and subchondral bone regeneration were achieved in vivo by using the tri-layered integrated scaffold. More importantly, the inclusion of the S layer could impart the regenerated cartilage with a more lubricating and wear-resistant surface. These findings suggest that the bioinspired construct mimicking the spatial variations of native osteochondral tissue might serve as a promising candidate to enhance osteochondral regeneration.

Polyetheretherketone (PEEK) is widely used in orthopedic and dental implants due to its excellent mechanical properties, chemical stability, and biocompatibility. However, its inherently bioinert nature makes it present weak osteogenic activity, which greatly restricts its clinical adoption. Herein, strontium (Sr) is incorporated onto the surface of PEEK using mussel-inspired polydopamine coating to improve its osteogenic activity. X-ray photoelectron spectroscopy and ion release assay results confirm that different concentrations of Sr are incorporated onto the PEEK substrate surfaces. The strontium-modified PEEK samples show a stable Sr ion release in 35 days of detection. Better results of MC3T3-E1 pre-osteoblasts adhesion, spreading, and proliferation can be observed in strontium-modified PEEK groups, which demonstrates strontium-modified PEEK samples with the improved MC3T3-E1 pre-osteoblasts compatibility. The boosted osteogenic activity of strontium-modified PEEK samples has been demonstrated by the better performed of ALP activity, extracellular matrix mineralization, collagen secretion, and the remarkable up-regulation of ALP, OCN, OPN, Runx2, Col-I, BSP, and OSX of the MC3T3-E1 pre-osteoblasts. Additionally, the strontium-modified PEEK samples exhibit a dose-dependent enhancement of osteoblasts compatibility and osteogenic activity, and the PEEK-Sr10 group shows the best. These findings indicate that strontium-decorated PEEK implants show promising application in orthopedic and dental implants.

Titanium (Ti) implants have been widely used for the treatment of tooth loss due to their excellent biocompatibility and mechanical properties. However, modifying the biological properties of these implants to increase osteointegration remains a research challenge. Additionally, the continuous release of various metal ions in the oral microenvironment due to fluid corrosion can also lead to implant failure. Therefore, simultaneously improving the bioactivity and corrosion resistance of Ti-based materials is an urgent need. In recent decades, micro-arc oxidation (MAO) has been proposed as a surface modification technology to form a surface protective oxide layer and improve the comprehensive properties of Ti. The present study doped nano silicon nitride (Si 3 N 4 ) particles into the Ti surface by MAO treatment to improve its corrosion resistance and provide excellent osteoinduction by enhancing alkaline phosphatase activity and osteogenic-related gene expression. In addition, due to the presence of silicon, the Si 3 N 4 -doped materials showed excellent angiogenesis properties, including the promotion of cell migration and tubule formation, which play essential roles in early recovery after implantation.

Sandblasted with large grits and acid-etched (SLA) surface topography has been a common method of implant surface modification to promote bone-implant integration in titanium-based dental implant manufacturing. However, the acid etching protocols used are various. The aim of this investigation was to validate the effect of different acid-etched treatments of sandblasted titanium surfaces on enhancing osteogenic potential. Five different titanium surface groups were prepared and tested including: i) smooth (Control), ii) sandblasted (SL), iii) sandblasted and acid-etched with H2SO4/HCl mixed solution (SLA/SOCl), iv) sandblasted and acid-etched with H2SO4 solution (SLA/SO), and v) sandblasted and acid-etched with HF solution (SLA/HF). The surface characterization, protein adsorption, and cell behavior were examined to determine the osteogenic potential. Compared with the control group, other surface treated groups showed rougher surface, better wettability, more protein absorption, and cell viability, especially in the SLA/SOCl surface group. The results indicated that SLA surfaces prepared with H2SO4/HCl mixed acid solution provided significant potential to promote osteogenesis.

Magnesium ions (Mg 2+ ) play an essential role in the metabolism and regeneration of bone tissue. Appropriate amounts of Mg 2+ have been shown to promote osteogenic differentiation of bone-derived cells and angiogenesis of endothelial cells. However, the initial burst release of Mg 2+ may compromise the osteogenic effect, so the controlled release of Mg 2+ is the critical consideration of the magnesium-containing tissue-engineered bone materials. This study proposes a microsphere-hydrogel complex to enhance the sustained-release effect and prolong the release cycle of Mg 2+ . For the initial release of Mg 2+ , polylactic acid (PLA) microspheres containing MgO and MgCO 3 were fabricated with uniform morphology. Further microspheres were incorporated into the chitosan-based hydrogel to form microsphere- hydrogel complex for extended release. The complex demonstrated effective sustained release of Mg 2+ over a period exceeding 28 days. In vitro cell experiments, CS/PLA@MgO-MgCO 3 significantly enhanced migration and osteogenic differentiation of MC3T3-E1. Meanwhile, it can facilitate the generation of blood vessels in HUVECs. In conclusion, the magnesium-loaded microsphere-hydrogel complex achieves excellent dual sustained-release properties with an extended-release cycle while enhancing vascularized osteogenic activity in vitro , showing promising prospects for clinical application in bone defect treatment.

Repositioning and securing the cranial bone flap after craniotomy remain significant challenges in neurosurgery. Traditional fixation methods often suffer from weak mechanical strength, bioinertness, limited osteogenic capacity, and a lack of antibacterial properties, complicating clinical outcomes. Recent medical adhesives offer superior fixation but face significant limitations in cranial bone applications. In this study, we explored the application of PAH (Poly (allylamine) hydrochloride)-TPP (Tripolyphosphate) coacervate (PT) as a bone adhesive. The PT coacervate demonstrated excellent anti-swelling (anti-swelling ratio less than 1 %), self-healing, and injectable properties, as well as exceptional shape adaptability and cytocompatibility. Adhesion tests revealed its outstanding adhesion (99.06 ± 11.76 kPa for lap shear and 121.42 ± 16.73 kPa for end to end), long-term durability, and tunable adhesion strength. Furthermore, the coacervate demonstrated broad-spectrum antibacterial activity against both Gram-positive and Gram-negative bacteria (antibacterial rate more than 90 %), with mechanistic studies revealing promising strategies to address localized and systemic drug-resistant infections. Additionally, the coacervate's self-mineralizing properties significantly enhanced its osteogenic performance. In vivo studies confirmed its effective fixation, robust antibacterial activity, and improved osteogenesis, underscoring its suitability for cranial bone flap repositioning and fixation after craniotomy. In summary, this coacervate adhesive offers a promising therapeutic solution for addressing the challenges of cranial flap fixation in neurosurgery.

Developing biomaterials capable of promoting bone regeneration in bacteria-infected sites is of utmost urgency for periodontal disease therapies. Here we produce a hybrid hydrogel by integrating CuS nanoparticles (CuSNPs), which could kill bacteria through photothermal therapy (PTT) triggered by a near infrared (NIR) light, and a gelatin methacryloyl (GelMA) hydrogel, which is injectable and biocompatible. Specifically, CuSNPs were precipitated by chitosan (CS) firstly, then grafted with methacrylic anhydride (MA) to form CuSNP@CS-MA, which was photo-crosslinked with GelMA to synthesize hybrid hydrogels (GelMA/CuSNP). The hybrid hydrogels exhibited a broad-spectrum antibacterial property that could be spatiotemprorally manipulated through applying a NIR light. Their mechanical properties were adjustable by controlling the concentration of CuSNPs, enabling the hydrogels to become more adapted to the oral diseases. Meanwhile, the hybrid hydrogels showed good cytocompatibility in vitro and improved hemostasis in vivo. Moreover, they accelerated alveolar osteogenesis and vascular genesis, successfully treating periodontis in four weeks in a rat model. GelMA/CuSNP hydrogels showed a broad-spectrum sterilization ability via PTT in vitro and outstanding antibacterial property in vivo, suggesting that the hybrid hydrogels could function in the challenging, bacteria-rich, oral environment. Such injectable hybrid hydrogels, capable of achieving both facilitated osteogenesis and NIR-inducible sterilization, represent a new biomaterial for treating periodontitis.

Osteoporosis is one of the most disabling consequences of aging, and osteoporotic fractures and a higher risk of subsequent fractures lead to substantial disability and deaths, indicating that both local fracture healing and early anti-osteoporosis therapy are of great significance. However, combining simple clinically approved materials to achieve good injection and subsequent molding and provide good mechanical support remains a challenge. To meet this challenge, bioinspired by natural bone components, we develop appropriate interactions between inorganic biological scaffolds and organic osteogenic molecules, achieving a tough hydrogel that is both firmly loaded with calcium phosphate cement (CPC) and injectable. Here, the inorganic component CPC composed of biomimetic bone composition and the organic precursor, incorporating gelatin methacryloyl (GelMA) and N-Hydroxyethyl acrylamide (HEAA), endow the system with fast polymerization and crosslinking through ultraviolet (UV) photo-initiation. The GelMA-poly (N-Hydroxyethyl acrylamide) (GelMA-PHEAA) chemical and physical network formed in situ enhances the mechanical performances and maintains the bioactive characteristics of CPC. This tough biomimetic hydrogel combined with bioactive CPC is a new promising candidate for a commercial clinical material to help patients to survive osteoporotic fracture. Keywords:injectable hydrogels;osteogenesis;bone cement;biocompatible polymers
