The effects of a phytic acid/calcium ion conversion coating on the corrosion behavior and osteoinductivity of a magnesium-strontium alloy

Li Liu,Qiuyue Yang,Lei Huang,Xiangmei Liu,Yanqin Liang,Zhenduo Cui,Xianjin Yang,Shengli Zhu,Zhaoyang Li,Yufeng Zheng,Kelvin Wai Kwok Yeung,Shuilin Wu

In this work, a conversion coating of phytic acid (PA)/calcium ion (Ca 2+ ) was prepared on a magnesium-strontium (Mg-Sr) alloy surface through a layer-by-layer self-assembly method. The effect of the pH value on the corrosion resistance of coatings was studied systematically. The addition of a Ca 2+ could enhance the intermolecular chelation of PA and repair the defects of the coating itself. The results of electrochemical and immersion testing confirmed that a PA/Ca 2+ conversion coating could adjust the corrosion rate of Mg-Sr alloys. Compared to a bare Mg-Sr alloy, the corrosion current density of Mg&OH&PA(5.5)&Ca 2+ was decreased by about three orders (2.726 × 10 −4  A/cm 2 to 4.304 × 10 −7  A/cm 2 ). In vitro tests showed that a Ca 2+ in the coatings could not only promote the formation of apatite , but also favored osteoblast proliferation. In addition, this conversion coating could significantly enhance the expression of alkaline phosphatase activity by 50–60% compared to a bare Mg-Sr alloy.

Cross-linked vegetable oil with covalently loaded dexamethasone: linear drug release for osteogenic induction of hBMSCs in vitro

Zhaoxing Liu,Lei Cao,Chunyan Bao,Shanfeng Dong,Kerong Dai,Linyong Zhu

A novel covalently dexamethasone-conjugated drug release system based on cross-linked vegetable oil (DEX-CVs) was designed and prepared by simple curing reaction of modified vegetable oil and dexamethasone phosphomonoester (DEX-P) without any toxic catalyst and initiator. UV and HPLC analysis confirmed that the dexamethasone (DEX) drug could be released linearly with high purity by adjusting the component ratios. The analysis of the model reaction indicated that DEX-P in the optimized DEX-CV (A4) was mainly covalently conjugated in the cross-linked vegetable oil network by cyclic phosphotriester bond, and the selective hydrolysis induced the pure release of dexamethasone drug. Finally, the low cytotoxicity and successful osteogenic differentiation effect of A4 on human bone marrow mesenchymal stem cells (hBMSCs) was corroborated by the detection of MTT analysis, ALP activity, staining experiments and some gene expressions, which provided the promising applications in drug release areas and bone tissue engineering.

3D Printing of Cytocompatible Graphene/Alginate Scaffolds for Mimetic Tissue Constructs

Li Jianfeng,Liu Xiao,Crook Jeremy M.,Wallace Gordon G.

Tissue engineering, based on a combination of 3D printing, biomaterials blending and stem cell technology, offers the potential to establish customized, transplantable autologous implants using a patient‘s own cells. Graphene, as a two-dimensional (2D) version of carbon, has shown great potential for tissue engineering. Here, we describe a novel combination of graphene with 3D printed alginate (Alg)-based scaffolds for human adipose stem cell (ADSC) support and osteogenic induction. Alg printing was enabled through addition of gelatin (Gel) that was removed after printing, and the 3D structure was then coated with graphene oxide (GO). GO was chemically reduced with a biocompatible reductant (ascorbic acid) to provide electrical conductivity and cell affinity sites. The reduced 3D graphene oxide (RGO)/Alg scaffold has good cytocompatibility and can support human ADSC proliferation and osteogenic differentiation. Our finding supports the potential for the printed scaffold’s use for in vitro engineering of bone and other tissues using ADSCs and potentially other human stem cells, as well as in vivo regenerative medicine.

ZrO2-CPS/ZrO2 graded composite ceramics with high osteoinductivity for dental implants fabricated by stereolithography

Jiaxin Wu,Zhi Li,Xingliang Meng,Bo Cheng,Jing Yang,Binbin Li,Xinyu Wang

Promoting mechanical strength and osteoinductivity properties is crucial for dental implants materials. Herein, we synthesized silicocarnotite (CPS) powder via a hydrothermal method, and mixed it with zirconia to create a composite material for preparing four slurries: xCPS-(1−x)ZrO 2 (x = 0, 1/6, 1/3, and 1/2 vol%). The optimal printing parameters for each slurry were investigated. Following the printing of the slurries, debinding was performed on them in the temperature range of 25 °C–600 °C in nitrogen atmosphere and 600 °C–1000 °C in air atmosphere, respectively, followed by sintering at 1350 °C for 2 h. The graded zirconia-based composite ceramics prepared using stereolithography (SLA) exhibited excellent mechanical properties, including a flexural strength of 742.37 ± 22.52 MPa, Vickers hardness of 7.29 ± 0.15 GPa, fracture toughness of 5.34 ± 0.29 MPa m 1/2 , and a relative density of 96.3 %. Compared with zirconia ceramics, the graded composite ceramics displayed higher surface roughness. The graded composite ceramics degraded and released Ca, Si, and P ions, promoting MC3T3-E1 cells proliferation, adhesion, and osteogenic differentiation. These findings suggest that ZrO 2 -CPS/ZrO 2 graded composite ceramics synthesized via SLA holds great potential as dental implants with high osteoinductivity.

Black Phosphorus and E7-Functionalized Sulfonated Polyetheretherketone with Effective Osteogenicity and Antibacterial Activity

Xiao Wang,Shuning Zhang,Ai Zhu,Lingyan Cao,Long Xu,Junjie Wang,Fei Zheng,Xiangkai Zhang,Hongyan Chen,Xinquan Jiang

Given its excellent biological properties and the matching of its elastic modulus with that of human bone tissue, medical polyetheretherketone (PEEK) is considered a desirable candidate for bone-implant materials. However, its poor osseointegrative and antibacterial properties greatly limit its clinical application. To address these concerns, a functional PEEK implant is needed. Herein, a novel photo-responsive multifunctional PEEK-based implant material (sPEEK/BP/E7) with both effective osteogenesis and good disinfection properties was constructed via the self-assembly of black phosphorus (BP) nanosheets, mussel-inspired polydopamine (PDA), and bioactive short peptide E7 on sulfonated PEEK (sPEEK). The versatile micro-/nano-structured PEEK surface provides superior hydrophilicity, a favorable osteogenic microenvironment, and excellent photothermal effects under near-infrared (NIR) irradiation. The in vitro results showed that sPEEK/BP/E7 displays enhanced cytocompatibility and osteogenicity in terms of cell adhesion, proliferation, alkaline phosphatase (ALP) activity, matrix mineralization, and osteogenesis-related gene expression, superior to those of the sPEEK and sPEEK/BP samples. In addition to osteogenesis, the multifunctional coating exhibited strong antibacterial activity against both Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ). Furthermore, it was confirmed in a rat femoral infection model that sPEEK/BP/E7 effectively resisted infection caused by S. aureus under NIR light irradiation and promoted osseointegration in vivo . Thus, this work presents a facile strategy to realize improvement of the “functional integration” of new polymer bone-implant materials and provide new ideas for their clinical application.

3D-Printed Carbon Scaffolds With Osteoinductivity via Nanotopography-Mediated Mechanotransduction

Xuan Pei,Haitao Peng,Ya Ren,Lina Wu,Donglin Ma,Changchun Zhou,Li Min,Yujiang Fan,Maling Gou

Osteoinductive biomaterials have emerged as promising platforms for bone regeneration. Nevertheless, conventional calcium phosphate-based systems exhibit suboptimal mechanical properties, particularly under load-bearing conditions, which severely restricts their clinical applicability in critical-sized defect repair. Here, a carbon-based scaffold fabricated through a 3D printing-pyrolysis strategy, featuring osteoinductive coralloid nanostructures on its surface is reported. The scaffold demonstrates a specific strength of 237.5 kN·m kg −1 , surpassing that of commercial orthopedic implants. Notably, the scaffold shows intrinsic osteoinductive properties through successful ectopic bone formation in gluteus maximus, a phenomenon attributed to its nanotopography-mediated activation of mechanotransduction pathways via Yes-associated protein (YAP) nuclear translocation. The scaffold outperformed titanium alloys in repairing both femoral bone defects and cranial bone defects in vivo. Its bone repair efficiency can be further improved by a biomimetic mineralization process. This work highlights the potential of a robust carbon scaffold with a nanostructured surface for osteoinductivity, which will lead to the development of future biomaterials for bone repair.

Multifunctional Bone Regeneration Membrane with Flexibility, Electrical Stimulation Activity and Osteoinductive Activity

Shihao Zhang,Lei Huang,Mengxuan Bian,Lan Xiao,Dong Zhou,Ziwei Tao,Zheng Zhao,Jian Zhang,Li-Bo Jiang,Yulin Li

The use of membrane-based guided bone regeneration techniques has great potential for single-stage reconstruction of critical-sized bone defects. Here, a multifunctional bone regeneration membrane combining flexible elasticity, electrical stimulation (ES) and osteoinductive activity is developed by in situ doping of MXene 2D nanomaterials with conductive functionality and β -TCP particles into a Poly(lactic acid-carbonate (PDT) composite nano-absorbable membrane (P/T/MXene) via electrostatic spinning technique. The composite membrane has good feasibility due to its temperature sensitivity, elastic memory capacity, coordinated degradation profile and easy preparation process. In vitro experiments showed the P/T/MXene membrane effectively promoted the recruitment and osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs) under ES and enhanced the angiogenic capacity of endothelial cells, which synergistically promoted bone regeneration through neovascularization. In addition, an in vivo rat model of cranial bone defects further confirmed the bone regeneration efficacy of the P/T/MXene membrane. In conclusion, the developed P/T/MXene membrane can effectively promote bone regeneration through their synergistic multifunctional effects, suggesting the membranes have great potential for guiding tissue regeneration and providing guidance for the biomaterials design.

Synthesis, characterization, and biological evaluation of “Bamboo lamellar” biomimetic hydroxyapatite

Chao Wang,Siyi Xue,Yufeng Huang,Yangyang Li,Xiao Ma

Hydroxyapatite (HA) nanocrystals in bones typically exhibit a layered structure, characterized by extensive exposure of (100) facet and uniaxial orientation with organic collagen fibers. However, the inherent brittleness of traditional hydroxyapatite particles often complicates the achievement of optimal structural and property characteristics in subsequent composite preparations. Researchers found that high aspect ratio and small crystal size can greatly improve the rigidity of hydroxyapatite. In this study, we synthesized "bamboo lamellar"-like hydroxyapatite particles (LHA) that combine the biological advantages of exposed crystal planes with the mechanical benefits of a higher aspect ratio. We investigated the effects of the calcium complexing agent stearic acid and the alkaline regulator urea on the morphology of LHA samples using XRD, SEM, and TEM, with all final samples exhibiting pronounced laminar characteristics. Biocompatibility studies with pre-osteoblast MC3T3-E1 cells demonstrated that the LHA series samples displayed superior biocompatibility compared to commercially available hydroxyapatite. Alkaline phosphatase (ALP) activity assays indicated that LHA significantly outperformed commercial hydroxyapatite in promoting bone induction. Additionally, MC3T3-E1 cells adhered well to the surface of the LHA film. Incorporating LHA into a chitosan matrix significantly enhanced performance compared to both the pure organic matrix and the commercial hydroxyapatite control group. Thus, this " bamboo lamellar " biomimetic hydroxyapatite particle shows great potential as both an organic enhancer and a promoter of pre-osteoblast-induced bone formation, positioning it as a promising material for bone tissue engineering.

Bisphosphonates activated keratin assembled nanoparticle targeting bone fracture for enhanced immunomodulatory and osteoinductivity

Jiaqi Zhong,Xiaodong Hu,Jieyang Dong,Yujiong Chen,Botao Liu,Mingming Hao,Haijiao Xie,Chunhai Ke,Zhaoxiang Peng

Bone fractures pose significant clinical challenges and require innovative solutions for effective treatment and rapid recovery. Nanoscale protein-based materials are prominent in biotechnology and biomedicine, demonstrating versatility in biological processes and drug delivery. Herein, we designed and fabricated self-assembled keratin nanoparticles via a bisphosphonate (BP)-activated conformation-conservative assembly process. We determined the structural properties of the nanoparticles and explored the immunomodulatory and osteoinductivity functions retained from keratin. In addition, the keratin-BP nanoparticles could repair bone fractures. This study on multifunctional self-assembled protein-drug nanoparticles provides useful baseline data for exploring novel therapeutic options for acute bone fracture and other diseases.

Biocompatible Poly(ε-caprolactone)-based Shape-memory Polyurethane Composite Scaffold with Bone-induced Activity

Luo Kun,Wang Li,Chen Xiaohu,Zeng Xiyang,Zhou Shiyi,Zhang Peicong,Li Junfeng

3D porous scaffold could provide suitable bone-like structure for cell adhesion and proliferation; however, surgical suffering from large volume implantation is a great challenge for patients. In this study, a shape programmable porous poly( ε -caprolactone) (PCL)-based polyurethane scaffold with memory effect was synthesized via gas foaming method, using Citrate modified Amorphous calcium Phosphate (CAP) as bioactive factor. The bending experiments indicated that the scaffolds achieved excellent shape-memory effect, which could be influenced by particle weight content. In vitro mineralization results suggested that the deposition of hydroxyapatite was promoted by scaffolds. Additionally, cell assay showed that composite scaffolds presented good cell toxicity and osteogenicity by the differentiation of rat Mesenchymal Stem Cells (rMSCs) into the osteogenic lineage. In the model of rat cranial implantation, the reparative tissue covered the defect site and bone-like structure deposited on the scaffold due to the formation of new bones. In summary, the porous smart shape-memory composite scaffolds could be a potential candidate in future distinctive bone repair applications.

Photolithographic-based stamp technique for improving the biocompatibility of antibacterial titanium implant by dynamic clearance of antibacterial agents

Yulu Yang,Ju Liu,Maowen Chen,Pengfei Gao,Yi Gong,Rui Chen,Yu Tang,Weihu Yang,Kaiyong Cai

Antibacterial titanium implants used in orthopedic and oral tooth repair have been widely researched for reducing the occurrence of second replacement operation. Although possessing a good antibiosis effect, the traditional methods have poor biocompatibility and even may cause damage to the body. Especially for loading antibiotic, after killing pathogenic bacteria, the residual antibacterial drugs may hinder the implant-tissue binding. Here, we develop a novel strategy for Ti implant based on photolithography, which can achieve the near-infrared (NIR) light-controlled antimicrobial agents loading, releasing, and clearing. A grid-like stamp of antibacterial complex and osteogenic growth peptide (OGP) were successfully prepared onto lanthanide-doped upconverting nanoparticles (UCNPs) decorated Ti substrate by selectively flicking antimicrobial agents away and replacing them with OGP. In vitro antibacterial evaluation and cell experiments proved that the modified Ti substrate (Ti-LA/OGP) exerts excellent antibacterial ability, as well as outstanding osteoinductive abilities for osteoblasts adhesion and differentiation. When implanted in infection models, Ti-LA/OGP possesses high antibacterial property at the early stage of implantation. After the success of antibiosis, irradiated with NIR light to remove the residual antibacterial agents, the osteogenesis of Ti-LA/OGP implants can be improved obviously. Moreover, this work may provide a promising strategy for the dynamic study of orthopedic application.

Multi-functionalized nanofibers with reactive oxygen species scavenging capability and fibrocartilage inductivity for tendon-bone integration

Peixing Chen,Sixiang Wang,Zhi Huang,Yan Gao,Yu Zhang,Chunli Wang,Tingting Xia,Linhao Li,Wanqian Liu,Li Yang

The presence of excessive reactive oxygen species (ROS) after injuries to the enthesis could lead to cellular oxidative damage , high inflammatory response, chronic inflammation, and limited fibrochondral inductivity, making tissue repair and functional recovery difficult. Here, a multifunctional silk fibroin nanofiber modified with polydopamine and kartogenin was designed and fabricated to not only effectively reduce inflammation by scavenging ROS in the early stage of the enthesis healing but also enhance fibrocartilage formation with fibrochondrogenic induction in the later stages. The in vitro results confirmed the antioxidant capability and the fibrochondral inductivity of the functionalized nanofibers. In vivo studies showed that the multifunctional nanofiber can significantly improve the integration of tendon-bone and accelerate the regeneration of interface tissue, resulting in an excellent biomechanical property. Thus, the incorporation of antioxidant and bio-active molecules into extracellular matrix-like biomaterials in interface tissue engineering provides an integrative approach that facilitates damaged tissue regeneration and functional recovery, thereby improving the clinical outcome of the engineered tissue.
