The aim of the present study was to investigate the effects of methylthiouracil (MTU) on the proliferation and apoptosis of rat bone marrow stromal cells (BMSCs). Rat BMSCs were isolated, cultured in vitro and treated with various concentrations of MTU. Cell growth curves were determined using the Cell Counting Kit‑8 method and the effect of MTU on BMSCs in a logarithmic growth phase was observed. BMSC apoptosis following MTU treatment was detected by flow cytometry. The experimental results demonstrated that the proliferation‑inhibition effect was gradually enhanced with increasing MTU concentrations and the extension of treatment time. Statistically significant differences were observed between the treatment and the control groups (P<0.05). In addition, the BMSC apoptosis rate gradually increased with increasing drug concentrations and treatment time extension; statistically significant differences were observed between the treatment and the control groups (P<0.05). Therefore, the results of the present study demonstrated that MTU inhibited the proliferation of BMSCs and promoted apoptosis, indicating the cytotoxic effects of MTU on BMSCs.

Graphene (Gr) presents promising applications in regulating the osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs). Light illumination is regarded as a spatiotemporally controllable, easily applicable, and noninvasive mean to modulate material responses. Herein, Gr-transferred silicon (Gr/Si) with a Schottky junction is utilized to evaluate the visible-light-promoted osteogenic differentiation of BMSCs. Under light illumination, light-induced charges, owing to the formation of the Schottky junction at the interface of Gr and Si, accumulated on the surface and then changed the surface potential of Gr/Si. The Schottky junction and surface potential at the interface of Gr and Si was measured by photovoltaic test and scanning Kelvin probe microscopy. Alkaline phosphatase (ALP) activity and quantitative real-time polymerase chain reaction (PCR) measurement showed that such variations of surface improved the osteogenic differentiation of BMSCs, and the activation of the voltage-gated calcium channels through surface potential and accumulation of cytosolic Ca2+ could be the reason. Moreover, X-ray photoelectron spectroscopy characterization showed that surface charge could also affect BMSCs differentiation through the promotion or inhibition of the adsorption of osteogenic growth factors. Such light-promoted osteogenic differentiation of BMSCs on Gr/Si may have huge potential for biomedical materials or devices for bone regeneration application.

Cadmium (Cd), a heavy metal pollutant that accumulates in organisms, is osteotoxic and contributing to the development of osteoporosis. Lycium barbarum polysaccharide (LBP), a polysaccharide obtained from the Chinese medicinal plant Lycium barbarum, exhibits protective effects against oxidative stress and cellular damage. However, it is unclear whether LBP can effectively inhibit Cd-induced osteotoxicity. This study investigated whether LBP alleviates Cd-induced osteogenic suppression in bone marrow mesenchymal stem cells (BMSCs) via autophagy modulation. Cell viability, osteogenic markers (ALP, BMP2, COL1, RUNX2), and mineralization were assessed using CCK-8, qPCR, Western blot, and alizarin red staining. Autophagy flux was evaluated via GFP-mCherry-LC3B transfection and protein markers (LC3-II, P62, BECN). We confirmed that Cd inhibited the formation of cellular autophagosomes while adversely affecting BMSCs. However, LBP mitigated the Cd-induced osteogenic inhibitory effect by stimulating the autophagy processes and facilitating the formation of autophagosomes and autolysosomes. In conclusion, LBP is a potential bone-enhancing agent that can improve cell viability and osteogenic differentiation while mitigating the detrimental effects of Cd on BMSCs through the activation of autophagy.

Infected bone defect is a critical problem in clinical practice of orthopedics. To solve this problem, implants with dual-functional surfaces which possess both antibacterial and osteogenic properties have extensive applications. However, there remains a pressing need to develop a convenient strategy for preparing such surfaces facilely. In this study, we reported a novel one-step polyphenol-based surface functionalization strategy for preparing two fibrous membranes with antibacterial and osteogenic capabilities. The formation of RGD-HHC36 (RH) or RGD-polyhexamethylene guanidine (RP) coating was fabricated by Michael reaction with poly (tannic acid) (PTA) to produce dual-functional fibrous membranes (PLA-RH and PLA-RP, respectively). The surfaces of PLA-RH and PLA-RP exhibited increased roughness and hydrophilicity. In vitro tests revealed that PLA-RH and PLA-RP could eradicate over 99% of clinically common pathogenic bacteria while exhibiting significant improvements in the adhesion, proliferation and osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs). Moreover, RNA-Seq analysis revealed that the up-regulation of Chrdl1 played a key role in promoting ossification through activation of the CHRDL1 - BMP4 pathway. The favourable in vivo anti-infection and osteogenesis-promoting performances were demonstrated in an infected bone defect model. PLA-RH and PLA-RP inhibited bacterial infection in the early stage, and enhanced osteogenesis in the late stage. This study provides a facile and universal strategy for designing and fabricating dual-functional surfaces for antibacterial application and tissue regeneration simultaneously.

As the most promising stem cell, bone marrow-derived mesenchymal stem cells (BMSCs) has attracted many attentions and applied widely in regenerative medicine. A biodegradable polyester with tunable affinity to BMSCs plays critical role in determining the properties of the BMSCs-based constructs. In this study, maleimide functionalized biodegradable polyester (P(MTMC-LA)) was synthesized through ring-opening copolymerization between l -lactide (LA) and furan-maleimide functionalized trimethylene carbonate (FMTMC) and a subsequent retro Diels-Alder reaction. P(MTMC-LA) was modified by different amounts of BMSCs specific affinity peptide (EPLQLKM, E7) through click-chemistry to investigate the effect on BMSCs. The E7 peptide modified P(MTMC-LA) was casted into films on glass slides and BMSCs were seeded onto the films. In vitro study showed that E7 peptide modified P(MTMC-LA) films supported BMSCs adhesion and proliferation compared to unmodified P(MTMC-LA) film. Besides, the adhesion and proliferation were enhanced by the increasing peptide grafting ratio. These results indicated that the novel biodegradable polyester can serve as a biomaterial with great potential application in tissue engineering and regenerative medicine.

Self-repair of articular cartilage defects is a significant challenge that can be addressed using drug-infused hydrogels, which improve injection convenience and provide immediate in situ adhesion. In this study, we developed a hydrogel incorporating Lipo@Kartogenin (KGN) and the cationic functional peptide SKPPGTSS (SKP) linked to aldehyde-based methacrylated hyaluronic acid (AHAMA). The innovative injectable hydrogel responded to visible light, allowing cross-linking under white light (~30 s) and effective adhesion to cartilage tissue. The hydrogel facilitated the sustained release of KGN and SKP over approximately 28 days as it degraded, thereby promoting the homing and differentiation of endogenous bone marrow-derived mesenchymal stem cells (BMSCs). Transcriptome sequencing showed that Smad4 expression and activation of the TGF-β signaling pathway are fundamental to these processes. In vivo studies in Sprague-Dawley (SD) rats showed that this hydrogel supports optimal hyaline cartilage regeneration within 8 weeks. In conclusion, our visible light-responsive adhesive co-delivery hydrogel effectively recruited native BMSCs to cartilage lesion sites and provided an environment conducive to their differentiation into cartilage, thereby facilitating effective cartilage regeneration. This innovation represents a novel approach to the clinical repair of cartilage defects.

Recently, osteoporosis has been a significant clinical challenge for the failed bone regeneration and vascularization. In this study, we developed a multifunctional nanoparticle system to stimulate bone regeneration and vascularization. Poly(D, L-lactide) (PLA)-based nanoparticles were fabricated to encapsulate icariin, which is renowned for its osteogenic potential. Subsequently, these nanoparticles were coated with red blood cell membranes to enhance biocompatibility. To further improve targeting specificity, a bone-targeted polymer-lipid consisting of alendronate-modified PEG-DSPE was synthesized and incorporated into the cell membrane coating. The delivery systems were designed to modulate bone marrow mesenchymal stem cells (BMSCs), thereby enhancing both osteogenesis and angiogenesis. Our findings demonstrate that the therapeutic system could enhance bone formation and vascularization in vitro , indicating its potential as an advanced treatment strategy for osteoporosis. This study provides a promising nanoplatform for the development of multifunctional nanomedicines aimed at improving bone health and vascular integration.

Background The repair of bone defects remains a significant clinical challenge. Although magnesium (Mg)-based biomimetic scaffolds are widely utilized for bone defect repair, the release of Mg²⁺ ions often leads to an alkaline microenvironment, thereby adversely affecting bone regeneration. Regenerative medicine strategies that leverage the recruitment of endogenous bone marrow mesenchymal stem cells (BMSCs) offer a novel approach to treating bone defects. Methods In this study, we employed poly(L-lactic acid) (PLLA) and polyethylene glycol (PEG) as shell materials and nanomagnesium oxide (nMgO) combined with gelatin (G) as core materials to fabricate coaxial fibre membranes with a “core‒shell” structure via coaxial electrospinning technology. Additionally, we grafted the BMSC-affinitive peptide E7 (EPLQLKM) onto the fibres to achieve specific recruitment of endogenous BMSCs.Results Morphological and structural analyses confirmed the successful formation of the “core‒shell” structure of the fibre membranes. Grafting E7 peptides enhanced the hydrophilicity and mechanical properties of the fibre membranes and maintained pH stability in vitro. In vitro experiments demonstrated that the functionalized fibre membranes significantly promoted BMSC proliferation, migration, and osteogenic differentiation. When implanted into a rat cranial defect model, we observed the formation of new bone tissue and the repair of the bone defect. Conclusion sE7 peptide-functionalized coaxial fibre membranes effectively facilitated bone defect repair by promoting the recruitment and osteogenic differentiation of BMSCs, demonstrating substantial potential for tissue engineering applications.

This study aimed to develop a novel artificial joint prosthesis material with osteogenic properties. Deferoxamine mesylate (DFO) was immobilized on the porous surface of sulfonated polyetheretherketone (SPEEK) through polydopamine (PDA), resulting in a novel material designated as DFO-PDA@SPEEK (Abbrev. DFO-PS). DFO-PS induced macrophage M2 phenotype polarization, reduced inflammatory factor expression, promoted osteogenic differentiation of bone marrow mesenchymal stem cells (BMSCs), and enhanced implant osseointegration and osteogenic capacity. In vitro evaluation demonstrated that DFO-PS significantly modulated immune and inflammatory responses, promoted angiogenesis, and enhanced osteogenic differentiation. In the rat model with femoral bone defect, in comparison to the control group, the DFO-PS group exhibited a 1.22-fold increase in trabecular thickness and a 1.51-fold enhancement in maximum pull-out force. This work demonstrates that DFO-PS represents a promising strategy for constructing multifunctional implants with biomineralization and immunomodulation properties for bone joint replacement.

This study explored the role of MEG3 in the cartilage differentiation of bone marrow mesenchymal stem cells (BMSCs). We investigated the effects of over-expression and knockdown of MEG3 on cell viability, cell differentiation, and the expressions of MEG3, miR-129-5p, COL2, chondrocyte differentiation-related genes (sry-type high-mobility-group box 9 (SOX9), SOX5, Aggrecan, silent information regulator 1 (SIRT1), and Cartilage oligomeric matrix protein (COMP)). The targeting relationship between MEG3 and miR-129-5p and the target gene of miR-129-5p was confirmed through Starbase, TargetScan and luciferase experiments. Finally, a series of rescue experiments were conducted to study the regulatory effects of MEG3 and miR-129-5p. BMSCs were identified as CD29+ and CD44+ positive, and their differentiation was time-dependent. As BMSCs differentiated, MEG3 expression was up-regulated, but miR-129-5p was down-regulated. Over-expressed MEG3 promoted the viability and differentiation of BMSCs, up-regulated the expressions of COL2 and chondrocyte differentiation-related genes, and inhibited miR-129-5p. Runt-related transcription factor 1 (RUNX1) was negatively regulated as a target gene of miR-129-5p. Results of rescue experiments showed that the inhibitory effect of miR-129-5p mimic on BMSCs could be partially reversed by MEG3. Over-expression of MEG3 regulated the miR-129-5p/RUNX1 axis to promote the differentiation of BMSCs into chondrocytes. This study provides a reliable basis for the application of lncRNA in articular cartilage injury.

Understanding the shape effect of hydroxyapatite (HAp) microparticles on cellular behavior is important for enabling new kinds of biological and biomedical applications. However, it is still a challenge to prepare HAp microparticles with different shapes but similar physicochemical properties, and then to investigate their relationships with cellular behavior. Herein, we developed a novel, facile route to regulate the morphology of HAp microparticles, and investigated the interaction between the particles and bone marrow mesenchymal stem cells (BMSCs). Our results revealed that the shape of HAp has a strong influence on cellular behavior, and that the sphere-like particles performed better than the rod-like particles. These findings highlight the importance of the shape characteristics of HAp microparticles, and may provide new insights for the utility of HAp-based materials.

Osteochondral defect refers to the damage of cartilage as well as subchondral bone. Cartilage tissue engineering focusing on the regeneration of cartilage and disregarding the subchondral bone always leads to partial regeneration of the damage, resulting in poor mechanical and physiological properties. A scaffold suitable for in situ inductive regeneration of both types of tissues is urgently needed. In this study, a biphasic scaffold integrated by macro-porous fibrin and 3D-printed wollastonite (containing 8% MgSiO3 (CS-Mg8)) scaffolds, either preloaded with rabbit bone marrow mesenchymal stem cells (BMSCs) or not, were fabricated and used to repair osteochondral defects in vivo (full thickness osteochondral defects in rabbits, 4 mm in diameter and 4 mm in depth with bone marrow blood effusion). The fibrin scaffold had a pore size of 100–200 μm, and was degraded gradually and reached weight loss over 80% at 28 days. The presence of BMSCs could accelerate the degradation rate. BMSCs could well proliferate in the fibrin scaffold along with time prolongation. The CS-Mg8 scaffold possessed a regular structure of cross stacked CS-Mg8 rods, and was degraded rather slowly with a mass loss of 8.5% at 28 days. BMSCs adhered and showed well spreading on the CS-Mg8 scaffold, without apparent proliferation in vitro. In vivo transplantation of the biphasic scaffolds, either preloaded with BMSCs or not, could induce the regeneration of both cartilage and subchondral bone to a great extent. Loading of BMSCs enabled better regeneration of cartilage layer, leading to smoother macroscopic appearance, good integrity with surrounding tissue and tide mark formation. However, no significant difference in bone formation and in gene expression was found with and without BMSCs loading.
