Physiologically related electrical microenvironments play an indispensable role in nerve regeneration and repair. Simulating the electrophysiological characteristics of nerves through bionic construction of bioelectric fields has attracted wide attention, as it can regulate the behavior of nerve cells and promote the repair of nerve injuries. Electret is a kind of material that can maintain permanent polarization state without external electric field, and can be used as a new self-powered electrical stimulation method for nerve regeneration and repair. In this work, a novel self-powered conductive nerve conduit with both electret electrical effects and conduction activity was constructed to promote peripheral bioelectret nerve regeneration and repair. The microsphere electret is embedded on the bacterial cellulose structure through the self-polymerization reaction of dopamine, wherein the microsphere electret can provide micro-power and release electrical signals through the conducting layer of polydopamine. The maximum conductivity of the composite bioelectret can reach 6.2 × 10 −3 S/cm, and the output charge can reach 264 pC at a bending strain of 50 %. In vitro, composite bioelectret provided a good electrophysiological microenvironment for cells and enhance their adhesion, proliferation and migration capabilities. In the repair of sciatic nerve in rats, the electret effect and conductivity of composite bioelectret provide continuous electrical stimulation for repairing microenvironment, strengthen the communication among cells, and promote axon growth and myelin regeneration. Conductive composite bioelectret provides a new non-contact electrical stimulation strategy for nerve repair by combining the dual synergistic effects of conductivity and electret self-powered.

Motor functional improvement represents a paramount treatment objective in the post-spinal cord injury (SCI) recovery process. However, neuronal cell death and axonal degeneration following SCI disrupt neural signaling, impeding the motor functional recovery. In this study, we developed a multifunctional decellularized spinal cord-derived extracellular matrix (dSECM), crosslinked with glial cell-derived neurotrophic factor (GDNF), to promote differentiation of stem cells into neural-like cells and facilitate axonogenesis and remyelination. After decellularization, the immunogenic cellular components were effectively removed in dSECM, while the crucial protein components were retained which supports stem cells proliferation and differentiation. Furthermore, sustained release of GDNF from the dSECM facilitated axonogenesis and remyelination by activating the PI3K/Akt and MEK/Erk pathways. Our findings demonstrate that the dSECM-GDNF platform promotes neurogenesis, axonogenesis, and remyelination to enhance neural signaling, thereby yielding promising therapeutic effects for motor functional improvement after SCI. Statement of significance The dSECM promotes the proliferation and differentiation of MSCs or NSCs by retaining proteins associated with positive regulation of neurogenesis and neuronal differentiation, while eliminating proteins related to negative regulation of neurogenesis. After crosslinking, GDNF can be gradually released from the platform, thereby promoting neural differentiation, axonogenesis, and remyelination to enhance neural signaling through activation of the PI3K/Akt and MEK/Erk pathways. In vivo experiments demonstrated that dSECM-GDNF/MSC@GelMA hydrogel exhibited the ability to facilitate neuronal regeneration at 4 weeks post-surgery, while promoting axonogenesis and remyelination at 8 weeks post-surgery, ultimately leading to enhanced motor functional recovery. This study elucidates the ability of neural regeneration strategy to promote motor functional recovery and provides a promising approach for designing multifunctional tissue for SCI treatment.

The massive death of nerve cells and the inflammatory microenvironment resulting from the release of reactive oxygen species after spinal cord injury (SCI) hinders the subsequent reconstruction of motor function. Owing to complex pathological changes, combination therapy is a potential new way to treat SCI. In this study, a novel multifunctional hydrogel containing an MXene-Au complex was constructed, and neural stem cells (NSCs) loaded in the hydrogel were combined with electrical stimulation to promote the recovery of motor function after SCI in rats. The constructed MAu-GelMA hydrogel had good reactive oxygen species-scavenging ability, electrical conductivity , and antibacterial ability. Simultaneously, electrical stimulation promoted the proliferation and differentiation of NSCs into nerve cells and promoted the establishment of synaptic connections between neurons. In the rat SCI model, combined treatment using MAu-GelMA hydrogel carrying NSCs and electrical stimulation reduced the formation of cavities and glial scars, enhanced neuronal differentiation and myelin regeneration of NSCs in the injured area, and promoted the recovery of motor function after SCI. In conclusion, in this study, we constructed a novel multifunctional hydrogel capable of carrying NSCs, which upon combining with electrical stimulation during treatment, effectively promoted the recovery of motor function in rats with SCI, thereby providing a new combination therapy strategy for SCI.

Background: Spinal cord injury (SCI) induces neuronal death and disrupts the nerve fiber bundles, which leads to severe neurological dysfunction and even permanent paralysis. A strategy combining biomimetic nanomaterial scaffolds with neural stem cell (NSC) transplantation holds promise for SCI treatment. Methods: Innovative three-dimensional (3D) nanofibrous sponges (NSs) were designed and developed by a combination of directional electrospinning and subsequent gas-foaming treatment. Immunofluorescence, mRNA sequencing, magnetic resonance imaging, electrophysiological analysis, and behavioral tests were used to investigate the in vitro and in vivo regenerative effects of the 3D NSs. Results: The generated 3D NSs exhibited uniaxially aligned nano-architecture and highly controllable hierarchical structure with super-high porosity (99%), outstanding hydrophilicity, and reasonable mechanical performance. They facilitated cell infiltration, induced cell alignment, promoted neuronal differentiation of NSCs, and enhanced their maturation mediated through cellular adhesion molecule pathways. In vivo , the NSC-seeded 3D NSs efficiently promoted axon reinnervation and remyelination in a rat SCI model, with new “neural relays” developing across the lesion gap. These histological changes were associated with regain of function, including increasing the neurological motor scores of SCI rats, from approximately 2 to 16 (out of 21), and decreasing the sensing time in the tape test from 140 s to 36 s. Additionally, the scaffolds led to restoration of ascending and descending electrophysiological signalling. Conclusion: The as-fabricated 3D NSs effectively regulate NSC fates, and an advanced combination of 3D NS design and transplanted NSCs enables their use as an ideal tissue-engineered scaffold for SCI repair.

Electrical stimulation (ES) can restore motor function after spinal cord injury (SCI). However, traditional intraspinal ES has many disadvantages, such as the need for a complex circuitry device, an external power source, and a second surgery to remove the implant. Piezoelectric materials have received increasing attention due to their potential to convert ambient mechanical energy into ES without an external power source or implantation of electrodes. Herein, a novel polydopamine (PDA) coated barium titanate (BaTiO 3 ) nanoparticles (BaTiO 3 @PDA) incorporated silk fibroin (SF) hydrogel (SFBT) was established. The SFBT hydrogel was crosslinked by horseradish peroxidase (HRP)/hydrogel peroxide (H 2 O 2 ) to integrate the piezoelectric potency of BaTiO 3 with the thermal sensitivity of HRP/H 2 O 2 for SCI recovery. The hydrogel containing 5% (w/v) BaTiO 3 @PDA nanoparticles (SFBT-5), which was selected for in vivo study, exhibited a short gelation time (5 min), suitable storage modulus (925 ± 35 Pa), and wireless ES (average current of 124 nA). Moreover, Basso-Beattie-Bresnahan (BBB) scores test and footprint analysis demonstrated that the SFBT-5 hydrogel successfully enhanced motor functional recovery of SCI. In histopathological assessments, the SFBT-5 hydrogel significantly accelerated spinal cord healing, as indicated by smaller lesion cavities (∼16.7% of the SCI group). Meanwhile, the SFBT-5 hydrogel accelerated neurogenesis, facilitated axon regrowth and synapse formation, and promoted remyelination. Overall, this study highlights the potential of piezoelectric hydrogels for SCI regeneration.
