Background Although short-term high-fat diet (S-HFD) and long-term high-fat diet (L-HFD) induce metabolic disorder, the underlying epigenetic mechanism is still unclear. Results Here, we found that both 4 days of S-HFD and 10 weeks of L-HFD increased mRNA m 6 A level in epididymal white adipose tissue (eWAT) and impaired metabolic health. Interestingly, S-HFD activated transposable elements (TEs), especially endogenous retroviruses (ERVs) in eWAT, while L-HFD activated long interspersed elements (LINEs). Subsequently, we demonstrated that both S-HFD and L-HFD increased m 6 A level of Ehmt2 and decreased EHMT2 protein expression and H3K9me2 level, accounting for activation of ERVs and LINEs. Overexpression of EHMT2 in eWAT or inhibition of ERVs and LINEs by antiviral therapy improved metabolic health under HFD feeding. Notably, we found that both short-term and long-term HFD feeding increased Fimicutes/Bacteroidota ratio and decreased the gut microbiome health index. Fecal microbiota transplantation (FMT) experiments demonstrated that gut microbiota from S-HFD and L-HFD was responsible for increased m 6 A level in eWAT, resulting in glucose intolerance and insulin insensitivity. Furthermore, we identified that both S-HFD and L-HFD increased the abundance of the gut microbial metabolite homogentisic acid (HGA), and HGA level was positively correlated with unclassified_f__Lachnospiraceae which was both increased in S-HFD and L-HFD feeding mice. Administration of HGA increased the m 6 A level of Ehmt2 and decreased the EHMT2 protein expression and H3K9me2 level in eWAT, leading to metabolic disorder in mice. Conclusions Together, this study reveals a novel mechanism that S-HFD and L-HFD induce metabolism disorder through gut microbiota-HGA-m 6 A- Ehmt2 -ERV/LINE signaling. These findings may provide a novel insight for prevention and treatment of metabolism disorder upon short-term or long-term dietary fat intake.

Background Compared to that of bacteria, the role of gut fungi in obesity development remains unknown. Results Here, alterations in gut fungal biodiversity and composition were confirmed in obese pig models and high-fat diet (HFD)-fed mice. Antifungal drugs improved diet-induced obesity, while fungal reconstruction by cohousing or fecal microbiota transplantation maintained the obese phenotype in HFD-fed mice. Fungal profiling identified 5 fungal species associated with obesity. Specifically, Ascomycota _sp. and Microascaceae_ sp. were reduced in obese mice and negatively correlated with fat content. Oral supplementation with fungi was sufficient to prevent and treat diet-induced obesity. Clec7a , which is involved in fungal recognition, was highly expressed in HFD-fed mice. The Clec7a agonist accelerated diet-induced obesity, while Clec7a deficieny in mice resulted in resistance to diet-induced obesity and blocked the anti-obese effect of antifungal drugs and fungi. Conclusions Taken together, these results indicate that gut fungi/ Clec7a signaling is involved in diet-induced obesity and may have therapeutic implications as a biomarker for metabolic dysregulation in humans.

Epidemiological studies indicate that exposure to persistent organic pollutants is positively associated with the prevalence of obesity. To delineate the potential role of technical-grade chlordane in obesity development, chlordane metabolism and chlordane-induced metabolic changes were investigated in mice fed high-fat diet (HFD) over a 6-week period. Gas chromatography–electron capture detector analysis showed that HFD induced more accumulation of technical chlordane in the liver, muscle and adipose tissue . The enantioselectivities of oxychlordane in selected tissues were also influenced by HFD. 1 H NMR-based liver metabolome indicated that technical chlordane can enhance the metabolic alterations induced by HFD. Compared with the low-fat diet (LFD) group, no differences were observed in the LFD + chlordane group. However, as many as 16 metabolites were significantly different between the HFD group and HFD + chlordane group. Moreover, compared to the LFD + chlordane group, the abundances of 24 metabolites significantly increased or decreased in the HFD + chlordane group. Twenty metabolites were altered in the HFD group compared to the LFD group. Tryptophan profiling suggested that both chlordane and HFD can disturb tryptophan catabolism. These interactions between technical chlordane and HFD suggest that technical chlordane is a candidate obesogen.

Introduction: Obesity, a growing public health issue, is closely associated with excessive energy intake and gut microbiota imbalances. Despite the increasing attention given to black jujube as a blackened fermented food in recent years, the role of its melanoidins (MLD) in obesity-related biological mechanisms has yet to be thoroughly investigated. Methods: This study investigated the effects of black jujube MLD on obesity induced by a high-fat diet in C57BL/6J mice. We hypothesize that MLD exerts an anti-obesity effect, potentially mediated by alterations in gut microbiota composition and the modulation of metabolic responses. Results: The results demonstrate that MLD administration mitigated HFD-induced weight gain, improved insulin resistance, and enhanced glucose metabolism by reducing blood glucose and insulin levels (p < 0.05). MLD also protected the liver, as evidenced by a 16.4% decrease in alanine aminotransferase (ALT) levels and a 29.6% decrease in aspartate aminotransferase (AST) levels (p < 0.05). Furthermore, MLD treatment restored the compromised intestinal microbiota to a healthier state at the genus level by lowering the Firmicutes/Bacteroidetes ratio (38.9%), suppressing the abundance of pathogenic bacteria, such as Erysipelatoclostridium and Bacteroides descendants, and promoting the growth of beneficial bacteria, including Bifidobacterium and norank_f_Muribaculaceae. Metabolomic profiling revealed that MLD can alleviate metabolic disorders by downregulating arginine metabolism and sphingolipid metabolism. Its metabolites are significantly correlated with several bacterial genera, including Bacteroides, Roseburia, Erysipelatoclostridium, Bacteroides, norank_f_Muribaculaceae, and Lachnospiraceae NK4A136 group.Discussion: Based on these findings, MLD may mitigate obesity and other associated metabolic disorders by modulating the gut microbiome–metabolism axis.

The investigation was to validate the alleviating effect of sodium butyrate on high-fat diet-induced gut injury and growth inhibition in largemouth bass ( Micropterus salmoides ). Five diets were designed: four high-fat diets (18 % crude lipid) supplemented with sodium butyrate at 0 % (HF), 0.05 % (HFS1), 0.1 % (HFS2), and 0.2 % (HFS3), as well as a control diet (C, 10 % crude lipid). Three tanks (30 fish each tank) were randomly allocated to each diet. Fish (4.0 g) were fed to apparent satiation for sixty days. Results indicated that HF dramatically reduced fish growth and induced oxidative stress, mucosal barrier dysfunction, gut inflammation, and gut dysbiosis compared to C. However, in comparison to HF, HFS1, HFS2, and HFS3 diets significantly promoted feed intake and weight gain rate, relieved oxidative stress by increasing antioxidant defense (e.g., total superoxide dismutase, catalase, glutathione peroxidase, reduced glutathione, and nuclear factor E2-related factor 2) and reducing malondialdehyde content, enhanced mucosal barrier by upregulating the expression of occludin, mucin3a, zona occluding-1, hypoxia-inducible factor-1α, and lysozyme, and reduced inflammation by downregulating the expression levels of interleukin 1β and tumor necrosis factor α ( P < 0.05). Moreover, HFS1 and HFS3 dramatically reduced Tenericutes and Mesomycoplasma abundance, whereas HFS1 increased Lactococcus abundance when compared to HF ( P < 0.05). Correlation analysis revealed a significant relationship between fish growth and gut injury with mucosal barrier proteins and gut microbiota (e.g., Tenericutes, Mesomycoplasam , and Lactococcus ) ( P < 0.05). These findings suggested that sodium butyrate can help mitigate the detrimental effects of high-fat diets on fish growth and gut health.

A low fasting blood glucose level is a common symptom in diabetes patients and can be induced by high-fat diet (HFD) feeding at an early stage, which may play important roles in the development of diabetes, but has received little attention. In this study, five polysaccharides were prepared fromSargassumfusiformeand their effects on HFD-induced fasting hypoglycemia and gut microbiota dysbiosis were investigated. The results indicated that C57BL/6J male mice fed an HFD for 4 weeks developed severe hypoglycemia and fourSargassumfusiformepolysaccharides (SFPs), consisting of Sf-2, Sf-3, Sf-3-1, and Sf-A, significantly prevented early fasting hypoglycemia without inducing hyperglycemia. Sf-1 and Sf-A could also significantly prevent HFD-induced weight gain. Sf-2, Sf-3, Sf-3-1, and Sf-A mainly attenuated the HFD-induced decrease in Bacteroidetes, and all five SFPs had a considerable influence on the relative abundance ofOscillospira,Mucispirillum, andClostridiales. Correlation analysis revealed that the fasting blood glucose level was associated with the relative abundance ofMucispinllumandOscillospira. Receiver operating characteristic analysis indicated thatMucispinllumandOscillospiraexhibited good discriminatory power (AUC = 0.745–0.833) in the prediction of fasting hypoglycemia. Our findings highlight the novel application of SFPs (especially Sf-A) in glucose homeostasis and the potential roles ofMucispinllumandOscillospirain the biological activity of SFPs.Keywords:Sargassum fusiformepolysaccharides;high-fat diet;gut microbiota;early fasting hypoglycemia

BACKGROUND Obesity, a global chronic disease, has been recognized as a severe risk to health. In our study, a novel polysaccharide named ARS was isolated and purified from aerial part of salviae miltiorrhizae radix . Our aim is to investigate the weight-reducing effect of a polysaccharide from salviae miltiorrhizae radix on mice fed a high-fat diet. RESULTS The novel polysaccharide ARS mainly consisted of glucose and galactose with a molar ratio of 0.59:1.00. We found that treatment with ARS could inhibit weight gain of mice fed a high-fat diet via modulating the intestinal bacteria. Moreover, we surveyed its mechanism in mice, and the gut microbiota sequencing results demonstrated that ARS can reverse or resist high-fat-diet-induced significant weight gain or obesity by increasing the diversity of gut microbiota and optimizing the ratio of Firmicutes to Bacteroidetes. Phylum and species analysis of gut microbiota demonstrated that obesity caused by a high-fat diet was accompanied by significant changes in the microbial communities, but ARS could reverse the disturbance of gut microbiota induced by the high-fat diet to maintain homeostasis. CONCLUSION Overall, our findings suggested a new function of ARS in regulating gut microbiota, which provides a theoretical basis for the development of high-quality ARS functional foods and the application of dietary supplements. © 2023 Society of Chemical Industry.

This present study was undertaken to explore the influences of phospholipids (PL) addition in the high-fat diet (HFD) on growth performance, nutritional composition, oxidative stress, lipid and protein metabolism in the muscle tissue of yellow catfish Pelteobagrus fulvidraco. The five experimental diets were formulated, which included the control (10.31 % normal dietary lipid without PL addition), high-fat dietary group (HFD, 14.99 % dietary lipid without PL addition), 0.1 % PL group (PL1,10.83 % dietary lipid with 0.1 % PL addition), high-fat diet +0.1 % PL group (HFD + PL1, 14.93 % dietary lipid with 0.1 % PL addition) and high-fat diet +0.2 % PL group (HFD + PL2, 14.76 % dietary lipid with 0.2 % PL addition). The feeding experiment continued for 10wk, and then the muscle samples were taken. Compared to the control, HFD reduced growth performance, feed utilization, muscle crude protein and total PL content, induced oxidative stress, lipogenesis and caused muscle lipid accumulation, and PL addition alleviated the HFD-induced changes above. HFD group had the smaller (100-249 μm) myofiber areas, while other groups showed a higher proportion of greater myofiber areas (>350 μm). Additionally, HFD suppressed the expression of muscle regulatory factors and protein synthesis, triggered protein degradation, whereas the PL addition in the diet mitigated the suppressive effects induced by HFD. Furthermore, the HFD inhibited the expression of muscle regulatory factors and protein synthesis, induced protein degradation; PL addition alleviated the HFD-induced inhibition of muscle regulatory factors and protein synthesis, and alleviated the HFD-induced protein degradation. Mechanistically, HFD enhanced the MYOG/FOXO3 binding with the atrogin-1/murf-1 promoters and increased their expression, and PL addition alleviated these effects induced by HFD. Taken together, our research demonstrated that dietary phospholipids could alleviate the adverse effects induced by HFD, and accordingly are beneficial additives for aquatic feeds. Our results also provided innovative insights into the regulatory mechanism of muscle development and growth in the vertebrates. Statement of relevance Our results indicated that dietary PL alleviated adverse effects of HFD on growth performance, oxidative stress, lipid and protein metabolism in the muscle of fish, and accordingly was a good additive in the aquafeeds.

Saccharina japonica polysaccharide could modulate gut microbiota composition; however, the composition-activity relationship remains unclear, thus restricting its application. In the current study, we investigated the impact of eight different S. japonica polysaccharide fractions on the gut microbiota after day 2 and day 14 treatments on high-fat diet (HFD) feeding mice. The results showed that a 2 day HFD dramatically altered gut microbiota composition, and the additional 12 day HFD further strengthened the gut microbiota dysbiosis in the HFD group. LjA-1 and LjA-3 could partially alleviate the dysbiosis of gut microbiota composition and significantly alter gut microbiota function. Multiple linear regression analysis revealed that the sulfate content and the molecular weight distributions were the main factors affecting the dominant gut bacterial genera. Our findings reveal that gut microbiota homeostasis could be disordered by HFD at day 2 and provide insights into the quantitative composition-activity relationships of polysaccharides in regulating gut microbiota.

Glucose-regulated protein 78 ( grp78 ) and activating transcription factor 6α ( atf6α ) are considered vital endoplasmic reticulum (ER) molecular chaperones and ER stress (ERS) sensors, respectively. In the present study, the full cDNA sequences of these two ERS-related genes were first cloned and characterized from black seabream ( Acanthopagrus schlegelii ). The grp78 cDNA sequence is 2606 base pair (bp) encoding a protein of 654 amino acids (aa). The atf6α cDNA sequence is 2168 base pair (bp) encoding a protein of 645 aa. The predicted aa sequences of A. schlegelii grp78 and atf6α indicated that the proteins contain all the structural features, which were characteristic of the two genes in other species. Tissues transcript abundance analysis revealed that the mRNAs of grp78 and atf6α were expressed in all measured tissues, but the highest expression of these two genes was all recorded in the gill followed by liver/ brain. Moreover, in vivo experiment found that fish intake of a high lipid diet (HLD) can trigger ERS by activating grp78 /Grp78 and atf6α/ Atf6α. However, it can be alleviated by dietary betaine supplementation, similar results were also obtained by in vitro experiment using primary hepatocytes of A. schlegelii . These findings will be beneficial for us to evaluate the regulator effects of HLD supplemented with betaine on ERS at the molecular level, and thus provide some novel insights into the functions of betaine in marine fish fed with an HLD.

With the increasing prevalence of high-fat diets (HFD) in aquaculture practices, the detrimental effects of HFD on farmed fish have garnered significant attention. Creatine has emerged as a promising green feed additive for aquaculture species; however, its potential role in mitigating the negative impacts of HFD remains poorly understood. To address this knowledge gap, the present study was designed to investigate the protective effects of dietary creatine supplementation on HFD-induced hepatic lipid metabolism disorders and muscle quality deterioration in juvenile grass carp ( Ctenopharyngodon idella ). Three experimental diets were formulated: a control diet (5.20% lipid, control), a HFD (8.11% lipid, HFD), and a HFD supplemented with 2% creatine (HFD + creatine). Juvenile grass carp (initial weight: 4.12 ± 0.02 g) were randomly allocated into nine 300-L indoor tanks and fed the experimental diets for 8 weeks. The key findings of this study revealed that (1) Dietary creatine supplementation significantly ameliorated the adverse effects of HFD on growth performance and feed utilization efficiency in juvenile grass carp. (2) Creatine supplementation improved muscle quality parameters in juvenile grass carp. (3) Dietary creatine attenuated HFD-induced hepatic lipid accumulation through enhanced fatty acid β -oxidation, which was mediated by mfn2-dependent mitochondrial fusion. Notably, this study elucidates a novel molecular mechanism whereby creatine activates mitochondrial fusion through the binding of pparα transcription factor to specific sites on the mitofusin 2 (Mfn2) gene promoter. To our knowledge, this is the first comprehensive investigation from a multi-organ/tissue perspective combined with mitochondrial dynamics analysis, providing valuable insights for developing effective nutritional strategies to counteract HFD-induced adverse effects in farmed fish through creatine supplementation.

Obesity poses a significant risk for metabolic disorders, such as insulin resistance and metabolic-associated fatty liver disease (MAFLD), yet effective treatments remain limited. Cell Death-Inducing DNA Fragmentation Factor-α-Like Effector C (CIDEC), a lipid droplet membrane protein, facilitates lipid droplet fusion and is crucial for adipose tissue expansion, making it a key target for obesity and related metabolic diseases. However, previous research revealed that complete genetic deletion of Cidec in adipose tissues, while reducing fat accumulation, induced severe insulin resistance in high-fat diet (HFD)-fed mice, potentially due to ectopic fat storage in the liver. Given that complete knockout is an extreme approach, partial inhibition holds greater clinical relevance. Therefore, this study aimed to investigate the effects of partial inhibition of CIDEC in adipose tissues on fat accumulation and insulin sensitivity in mice. Using the Cre-LoxP system, we generated adipose Cidec haploinsufficient mice. Under a standard diet, these mice exhibited normal body weight, fat accumulation, and insulin sensitivity. Notably, under HFD conditions, mice with partial Cidec deficiency showed reduced fat accumulation in adipose tissues while hepatic fat accumulation remained unchanged, accompanied by improved insulin sensitivity and increased energy expenditure. Mechanistically, we found partial Cidec deficiency activated thermogenic program in adipocytes in vivo and in vitro through the ATGL-PPARα pathway. In conclusion, adipose CIDEC partial inhibition attenuates HFD-induced obesity and insulin resistance by enhancing ATGL-PPARα-mediated energy expenditure, establishing this approach as a promising therapeutic strategy for obesity and related metabolic diseases.
