Ameliorative potential of ellagic acid via PPARγ against hyperlipidemia: Insights from mice and zebrafish

Junfeng Li,Min Guo,Cuiping Yuan,Tiezhu Li,Jie Zhang,Li Ren

ABSTRACT Peroxisome proliferator-activated receptor γ (PPARγ) was a major transcription factor and performed an essential function in the regulation of lipid metabolism. Ellagic acid (EA) was a natural phytochemical extracted from berries and pomegranates, and research had demonstrated its ameliorative effects on lipid metabolism disorders. We intended to explore the interventional effect and mechanism of action of EA on hyperlipidemia. In vitro experiments revealed that EA could reduce lipid accumulation in HepG2 cells. EA was found to bind stably to PPARγ, suppressing its nuclear translocation as well as its luciferase activity. It could also down-regulate the expression of genes and proteins related to lipid metabolism upstream and downstream of PPARγ. To explore the impact of EA on hyperlipidemia in vivo from different levels and perspectives, we established hyperlipidemia mice and hyperlipidemia zebrafish models. Consistent with the in vitro results, in vivo experiments revealed that EA attenuated lipid accumulation in hyperlipidemia mice and reduced the levels of related inflammatory factors in their serum. EA also inhibited lipid accumulation by decreasing the expression of upstream and downstream lipid-associated genes of pparg in hyperlipidemia zebrafish. In conclusion, EA could effectively improve hyperlipidemia by regulating PPARγ.

An Optimized Aerogel-Based Apheresis Device for Targeted Lipid Clearance in Elderly Hyperlipidemia Patients

Hao Zheng,Jiangbangrui Chu,Shikang Li,Xun Tang,Shicheng Bi,Kerong Chen,Anwei Zhou,Youwen Yang,Xinghai Ning,Yurui Xu

Elderly patients with hyperlipidemia often exhibit resistance to conventional hypolipidemic treatments, underscoring the need for more effective strategies to address lipid imbalances in this high-risk group. This study introduces LipClean, an aerogel-based apheresis device specifically designed to remove harmful plasma lipids. LipClean is constructed using hydrophilic cellulose fibers, which serve as a supramolecular platform for synthesizing hydrophobic conjugated polymers through a Sonogashira-Hagihara reaction. These conjugated polymers are then cross-linked with the cellulose fibers via phosphorylation, generating an aerogel monolith with an interpenetrating network of hydrophilic fibers and hydrophobic polymers. Unlike bilayer aerogels that separate hydrophilic and hydrophobic layers, LipClean's interpenetrating structure is precisely engineered through polymer design and gradient cross-linking. This optimization enhances both bodily fluid flow and lipid adsorption while minimizing the removal of essential plasma components and ensuring unobstructed cell passage. In preclinical testing, LipClean significantly reduced triglyceride and cholesterol levels in an elderly rat model of hyperlipidemia and normalized lipid levels in blood samples from hypertensive patients. Importantly, purified blood maintained normal levels of blood cells and physiological and biochemical indicators after apheresis, highlighting LipClean's potential for managing hyperlipidemia-related disorders. This study, therefore, underscores the importance of interdisciplinary collaboration in driving medical device innovation.

Discrimination of aqueous and vinegary extracts of Shixiao San using metabolomics coupled with multivariate data analysis and evaluation of anti-hyperlipidemic effect

Xiaofan Wang,Xu Zhao,Liqiang Gu,Yuanyuan Zhang,Kaishun Bi,Xiaohui Chen

A novel study using LC–MS (Liquid chromatography tandem mass spectrometry) coupled with multivariate data analysis and bioactivity evaluation was established for discrimination of aqueous extract and vinegar extract of Shixiao San. Batches of these two kinds of samples were subjected to analysis, and the datasets of sample codes, t R - m / z pairs and ion intensities were processed with principal component analysis (PCA). The result of score plot showed a clear classification of the aqueous and vinegar groups. And the chemical markers having great contributions to the differentiation were screened out on the loading plot. The identities of the chemical markers were performed by comparing the mass fragments and retention times with those of reference compounds and/or the known compounds published in the literatures. Based on the proposed strategy, quercetin-3-O-neohesperidoside, isorhamnetin-3-O-neohespeeridoside, kaempferol-3-O-neohesperidoside, isorhamnetin-3-O-rutinoside and isorhamnetin-3-O-(2G-α-l-rhamnosyl)-rutinoside were explored as representative markers in distinguishing the vinegar extract from the aqueous extract. The anti-hyperlipidemic activities of two processed extracts of Shixiao San were examined on serum levels of lipids, lipoprotein and blood antioxidant enzymes in a rat hyperlipidemia model, and the vinegary extract, exerting strong lipid-lowering and antioxidative effects, was superior to the aqueous extract. Therefore, boiling with vinegary was predicted as the greatest processing procedure for anti-hyperlipidemic effect of Shixiao San. Furthermore, combining the changes in the metabolic profiling and bioactivity evaluation, the five representative markers may be related to the observed anti-hyperlipidemic effect.

Hypolipidaemic and antioxidant effects of various Chinese dark tea extracts obtained from the same raw material and their main chemical components

Wanjun Ma,Yali Shi,Gaozhong Yang,Jiang Shi,Junpeng Ji,Yue Zhang,Jiatong Wang,Qunhua Peng,Zhi Lin,Haipeng Lv

In order to investigate the hypolipidaemic and antioxidant effects of various dark teas produced from different post-fermentation using the same raw material, a hyperlipidaemia zebrafish model combined with binding bile salts assay and antioxidant assays were performed in this study. Results showed that the hypolipidaemic effect of dark tea extracts increased significantly ( p  < 0.05) while the antioxidant ability decreased sharply compared with raw material. Particularly, Liupao tea (50%) and Pu-erh tea (48%) showed promising hypolipidaemic potential; however, the antioxidant capacity of Pu-erh tea decreased (31–49%) most dramatically. Besides, the levels of total polyphenols and catechins decreased sharply, but theabrownin, gallic acid, and caffeine increased significantly after post-fermentation. Moreover, the potential mechanisms of regulating hyperlipidaemia by dark tea extracts were discussed. These results suggest that microbial fermentation significantly affects the bioactivity of dark teas, and provide theoretical basis for processing and improving of dark tea products for hyperlipidaemia therapy.

Rational construction of AIEgens with wide color tunability and their specific lipid droplet imaging applications

Fei Zhang,Zhe Li,Yaoming Liu,Binsheng Yang,Hui Qiao,Jie Chai,Guangming Wen,Bin Liu

Organic light-emitting materials with aggregation-induced emission (AIE) character have experienced a rapid development in imaging, visualization and sensing. In this paper, by installing rotors to an aggregation-caused quenching (ACQ) compound, we constructed an AIE luminogen (AIEgens) system (A2–A4, B1–B3) with wide emission tunability, which covers almost all regions of visible light (400–780 nm). The calculated energy gap of the compounds is consistent with the value of the absorption transition. In the biological experiments, B3 showed excellent lipid droplet (LD) targeting ability in PC12 cells, and higher photostability compared with the commercial LDs bioprobe. Moreover, as a hydrophobic compound, B3 can distinguish blood from a hyperlipidemia patient and normal people. Thus, this study provides a strategy to construct red/NIR fluorescent materials with AIE character, and further apply it to identify the blood of people with hyperlipidemia.

Tissue distribution and enhanced in vivo anti-hyperlipidemic-antioxidant effects of perillaldehyde-loaded liposomal nanoformulation against Poloxamer 407-induced hyperlipidemia

Emmanuel Omari-Siaw,Qilong Wang,Congyong Sun,Zengquan Gu,Yuan Zhu,Xia Cao,Caleb Kesse Firempong,Rita Agyare,Ximing Xu,Jiangnan Yu

An optimized perillaldehyde-loaded liposomal nanoformulation (PAH-LNF) was successfully applied to improve the pharmacological effect of perillaldehyde (PAH) in poloxamer 407-induced hyperlipidemia . Oral administration of PAH-LNF (240   mg/kg per body weight) in rats significantly enhanced solubility and relative bioavailability (270.7%) compared to the free PAH with about 2.7-, 1.5-, 1.3-, 1.3- and 1.5-fold increase in AUC, T 1/2 , MRT, C max and T max , respectively. Tissue distribution study also revealed the accumulation of PAH in the liver, lungs, spleen, kidney, brain and heart in order of decreasing affinity. Moreover, a significant decrease in serum total cholesterol (TC), triglyceride (TG) and low-density lipoprotein cholesterol (LDL-C) with simultaneous increase in high-density lipoprotein cholesterol (HDL-C) level was observed in the chemically-induced hyperlipidemic mice which further confirmed PAH’s anti-hyperlipidemic properties. Additionally, PAH-LNF also significantly increased the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) with a concurrent decrease in malondialdehyde (MDA) to affirm the antioxidant and hepatoprotective effects of PAH. Thus, liposomal nanoformulation promises to be a useful drug delivery system for the development of PAH.

Hawthorn fruit acid consumption attenuates hyperlipidemia-associated oxidative damage in rats.

Feng Y,Gao S,Zhu T,Sun G,Zhang P,Huang Y,Qu S,Du X,Mou D

Context Hyperlipidemia is a highly prevalent risk factor for atherosclerosis and stroke. The currently available medications used to treat Hyperlipidemia cannot improve its oxidative stress damage. Consumption of hawthorn can regulate blood sugar and blood lipids, and its rich fruit acid is a natural antioxidant that can improve oxidative stress damage. Objective The present research aimed to investigate the protective effect of hawthorn fruit acid (HFA) on hyperlipidemia and to determine its potential molecular mechanism. Materials and methods Sprague-Dawley rats were fed a high-fat diet (HFD) to induce hyperlipidemia and treated orally with hawthorn fruit acids (HFA). Serum and liver levels of total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), superoxide dismutase (SOD), hydrogen peroxide (CAT), and malondialdehyde (MDA) were measured. Human hepatocellular carcinoma cell lines (HepG2) cells were treated with 0.1 mM oleic acid and HFA (0.125, 0.25 mg/mL), and intracellular TC, TG, HDL-C, SOD, CAT and MDA were measured. Changes in LDLR, HMGCR, Nrf2, HO-1, NQO1 protein and gene expression were analyzed by Western blot and qPCR. Results This study found that HFA treatment effectively reduced the level of triglyceride, cholesterol, and glucose, and attenuated hepatic steatosis in rats. Additionally, oxidative stress damage of rats was effectively reduced by treatment with HFA. Western blot and qPCR analysis indicated that HFA treatment inhibited fat accumulation in HepG2 cells by upregulating LDLR and downregulating HMGCR gene expression. HFA inhibits oleic acid (OA)-induced oxidative damage to HepG2 by activating the Nrf2/HO-1 signaling pathway. Conclusion HFA administration can provide health benefits by counteracting the effects of hyperlipidemia caused by an HFD in the body, and the underlying mechanism of this event is closely related to the activation of the Nrf2/HO-1 signaling pathway.

Antihyperglycemic and antihyperlipidemic activities of a polysaccharide from Physalis pubescens L. in streptozotocin (STZ)-induced diabetic mice

Xiaoyong Chen,Xiong Li,Xiaobo Zhang,Lijun You,Peter Chi-Keung Cheung,Riming Huang,Jianbo Xiao

In this study, a polysaccharide was extracted from Physalis pubescens L. (named PP). Its antihyperglycemic and antihyperlipidemic activities were evaluated in STZ-induced diabetic mice. Results showed that PP was determined to be composed of rhamnose (Rha), arabinose (Ara), fucose (Fuc), xylose (Xyl), mannose (Man), glucose (Glc) and galactose (Gal) with molar percentages of 4.65%, 17.34%, 1.43%, 6.24%, 5.52%, 45.5%, and 19.31%, respectively. The average molecular weight (Mw) was found to be 20.0 kDa. It had a strong α-glucosidase inhibitory activity in vitro. PP treatment could enhance the oral glucose tolerance, and increase the levels of SOD, GSH, CAT, vitamin C, vitamin E, HDL-c, C-peptide, GCK and hepatic glycogen in diabetic mice. Besides, PP treatment could also decrease the levels of MDA, TG, TC, LDL-c, BUN and G-6-Pase. The regulating effects were stronger in high dose PP treatment than those in the low and medium dose treatments. In short, PP played an important role in protecting STZ-induced diabetic mice, and the effect was closely related to its activities in antioxidation and regulating glucose and lipid metabolism.

Effect of Bergamot and Laoxianghuang Polysaccharides on Gut Microbiota Derived from Patients with Hyperlipidemia: An Integrative Analysis of Microbiome and Metabolome during In Vitro Fermentation

Yang Zheng,Yi Wang,Donghui Luo,Lianzhu Lin,Xingyu Lu,Jie Gao,Chuqiao Xiao,Mouming Zhao

The aim of this study was to investigate the effects of bergamot polysaccharide (BP) and Laoxianghuang polysaccharides (LPs, fermented bergamot) on the microbiome and metabolome during the in vitro fermentation of gut microbiota from patients with hyperlipidemia. Results indicated that both BP and LPs were able to increase the production of acetic acid, propionic acid, and butyric acid. However, only LPs could decrease the content of isobutyric acid and isovaleric acid, which are detrimental to gut health. A 16S rRNA analysis showed that both BP and LPs could reduce the proportion ofFusobacterium, whereas they increased theBacteroidescontent in hyperlipidemia. Untargeted UPLC-MS/MS metabolomic profiling found six bio-markers that were significantly changed after BP and LPs intervention, and four of the down-regulated metabolites were long-chain fatty acids associated with vascular diseases. These findings provide new evidence that BP and LPs have the potential to regulate imbalances in the gut microbiota in patients with hyperlipidemia and ameliorate its metabolic abnormalities. Keywords:polysaccharide;hyperlipidemic;gut microbiota;SCFAs;metabolomics

Extraction of Pithecellobium clypearia Benth polysaccharides by dual-frequency ultrasound-assisted extraction: Structural characterization, antioxidant, hypoglycemic and anti-hyperlipidemic activities

Zihao Chen,Chuanju Wang,Jiarong Su,Guixin Liang,Shaofan Tan,Yongguang Bi,Fansheng Kong,Zhong Wang

In this research, the extraction process of polysaccharides from Pithecellobium clypearia Benth (PCBPs) was optimized using dual-frequency ultrasound-assisted extraction (DUAE). The biological activities of PCBPs were investigated by in vitro antioxidant, hypoglycemic, and anti-hyperlipidemic assay. High-performance anion-exchange chromatography, high-performance gel permeation chromatography, SEM, UV–Vis spectroscopy, and FT-IR spectra were used to analyze the monosaccharide composition, molecular weight, microscopic morphology, and characteristic structure of PCBPs. The results showed that the maximum extraction rate of PCBPs was 9.90 ± 0.16% when the ultrasonic time was 8 min, the liquid-to-material ratio was 32 mL/g, and the ultrasonic power was 510 W. The PCBPs also possessed excellent in vitro antioxidant, hypoglycemic, and anti-hyperlipidemic activities. In addition, the average molecular weight of PCBPs was 15.07 kDa. PCBPs consisted of rhamnose, arabinose, galactose, glucose, xylose, mannose, and glucuronic acid, with the molar ratios of 11.07%, 18.54%, 48.17%, 10.44%, 4.62%, 4.96%, and 2.20%, respectively. Moreover, the results of SEM showed that PCBPs mainly showed a fine spherical mesh structure. The above studies provided a valuable theoretical basis for the subsequent in-depth study of PCBPs.

Hypolipidemic and Antithrombotic Effect of 6′-O-Caffeoylarbutin from Vaccinium dunalianum Based on Zebrafish Model, Network Pharmacology, and Molecular Docking

Boxiao Wu,Churan Li,Huan Kan,Yingjun Zhang,Xiaoping Rao,Yun Liu,Ping Zhao

Vaccinium dunalianumleaf buds make one of the most commonly used herbal teas of the Yi people in China, which is used to treat articular rheumatism, relax tendons, and stimulates blood circulation in the body. In addition, 6′-O-caffeoylarbutin (CA) is a standardized extract ofV. dunalianum, which has been found in dried leaf buds, reaching levels of up to 31.76%. Because of the uncommon phenomenon, it is suggested that CA may have a potential therapeutic role in hyperlipidemia and thrombosis. This study was designed to study the efficacy of CA on treating hyperlipidemia and thrombosis and the possible mechanisms behind these effects. Hyperlipidemia and thrombosis zebrafish models were treated with CA to observe variations of the integrated optical density within the vessels and the intensity of erythrocyte staining within the hearts. The possible mechanisms were explored using network pharmacology and molecular docking. The results demonstrate that CA exhibits an excellent hypolipidemic effect on zebrafish at concentrations ranging from 3.0 to 30.0 μg/mL and shows thrombosis inhibitory activity in zebrafish at a concentration of 30.0 μg/mL, with an inhibition rate of 44%. Moreover, network pharmacological research shows that MMP9, RELA, MMP2, PRKCA, HSP90AA1, and APP are major targets of CA for therapy of hyperlipidemia and thrombosis, and may relate to pathways in cancer, chemical carcinogenesis-receptor activation, estrogen signaling pathway, and the AGE–RAGE signaling pathway in diabetic complications.

Fuzhuan brick tea supplemented with areca nuts: Effects on serum and gut microbiota in mice

Ming-Xi Zhou,Xing Tian,Zhong-Qin Wu,Ke Li,Zong-Jun Li

Areca nut and Fuzhuan brick tea, a type of natural plant products, have obvious effects of fat reduction and weight loss; however, there is no report on their synergistic effect. This study investigated the effects of Fuzhuan brick tea supplemented with different concentrations of areca nut (5% (LAF), 10% (MAF), and 20% (HAF)) on serum and gut microbiota in Kunming (KM) mice. The results showed that Fuzhuan brick tea supplemented with areca nuts (AFTs) could reduce weight, prevent the accumulation of fat, inhibit the increase in the levels of serum triglyceride, total cholesterol, low-density lipoprotein cholesterol, blood glucose, free fatty acid, insulin, and total bile acid, alleviate the decrease in high-density lipoprotein cholesterol level, and regulate the composition of gut microbiota by high-fat diet intervention. The HAF group with 20% areca nut content showed the best effect. These results could provide a novel approach to prevent obesity and hyperlipidemia. Practical applications Consumption of areca nut and tea is widespread in Asia and other regions. As a controversial raw material, the damage due to areca nut to oral mucosa health has often aroused public concern and heated discussion; however, its medicinal value has been confirmed in terms of its pharmacological effects in various aspects. Fuzhuan brick tea, a type of traditional postfermented dark tea, has been confirmed to exert effects of antiobesity. Therefore, the areca nut and Fuzhuan brick tea, as a type of natural plant products, have obvious effects of fat reduction and weight loss; however, their synergistic effect has not been reported. To our knowledge, this study is the first to explore the effects of the Fuzhuan brick tea supplemented with areca nuts (AFTs) on serum and gut microbiota in mice. On the premise of exerting their beneficial effects (especially in terms of easing food stagnation and eliminating indigestion) and reducing their toxic and side effects, the effects of AFTs on health were further clarified, which could provide a novel direction for the development and utilization of areca nut. Moreover, our research would increase public understanding of areca nut and provide guidance to the Fuzhuan brick tea processing industry.
