Nitrogen Deposition Influences the Allelopathic Effect of an Invasive Plant on the Reproduction of a Native Plant: Solidago canadensis versus Pterocypsela laciniata

Congyan Wang,Jun Liu,Hongguang Xiao,Jiawei Zhou,Daolin Du

This study aims to gain insights into the allelopathic effects (by using leaf extracts) of the notorious invasive Solidago canadensis L. on seed germination of the associated Pterocypsela laciniata (Houtt.) C. Shih under different nitrogen (N) forms added: inorganic (NO3--N and NH4 -N), organic (urea-N), and mixed N (a mixture of the three N forms at 1:1:1 ratio). Among the two used concentrations of S. canadensis leaf extracts the higher exhibited inhibitory allelopathic effects on seedling height and biomass, germination potential, germination index, and vigor index of P. laciniata. N demonstrated positive effects on seed germination of P. laciniata. The effects of mixed and organic N on the seedling biomass of P. laciniata were more pronounced than those of inorganic N and control treatment. The vigor index of P. laciniata under mixed N was significantly higher than those under single N form and control treatment. Thus, organic and mixed N showed higher ecological effects on seed germination of P. laciniata than inorganic and single N form. All N forms could alleviate the inhibitory allelopathic effects of S. canadensis on seed germination of P. laciniata. Overall, the inhibitory allelopathic impact of S. canadensis on seed germination of native species may be attenuated under increased and diverse N deposition, thus it could prevent its further invasion.

Anti-tumor effect of flavonoids isolated from Bidens Pilosa L. by regulating the activity of myeloid-derived suppressor cells within the tumor microenvironment in mice

Yang Tao,Maoxin Du,Meihua Zhu,Weiqing Sun,Guiyuan Zeng,Jiayan Xiong,Jinmin Li,Ziyi Yang,Baomin Fan,Ruyi Zhang,Guangzhi Zeng

Ethnopharmacological relevance Colon cancer is one of the most common malignant tumors worldwide. Bidens pilosa L., an annual herb of the Asteraceae, has long been used to treat inflammatory-related illnesses, including cancer. As a population of immunosuppressive cells in the TME, MDSCs play a pivotal role in tumorigenesis and progression, and the effect of B. pilosa on MDSCs has rarely been reported. Aim of study To investigate the anti-tumor effect of a mixture of two flavonoids, MTF, isolated from B. pilosa, which showed immunotherapeutic activity in regulating the function of MDSCs in colon cancer. Materials and methods The regulatory effects of the flavonoid MTF on MDSCs differentiation and immune function were tested by qRT-PCR and flow cytometry. Its underlying immunotherapeutic mechanism, cytotoxicity, and anti-angiogenic activity were investigated using SIE luciferase/western blot, CCK-8/apoptosis, and MDSC-HUVEC co-culture assays, respectively. The in vivo anti-tumor activity of MTF was subsequently investigated in both CT26.WT and CT26.WT/MDSC syngeneic models. Results MTF and its components effectively depleted MDSCs by inhibiting their differentiation and inducing apoptosis, thereby restoring suppressed CD4 + T cell function. In vivo, MTF not only reduced intratumoral MDSCs but also counteracted MDSC-driven angiogenesis, leading to inhibited tumor growth and enhanced sensitivity to 5-FU treatment. Conclusion Flavonoid MTF showed a good antitumor effect in mice by regulating MDSCs activity in the TME, which contributes to the clinical use of this traditional herb.

Screening of high-efficiency and low-toxicity antitumor active components in Macleaya cordata seeds based on the competitive effect of drugs on double targets by a new laminar flow chip

Yan Gao,Huaidong Peng,Lisi Li,Feng Wang,Jiang Meng,Hongling Huang,Shumei Wang,Paul C. H. Li,Yue Sun

It is urgent to obtain targeted drugs that selectively bind to pathological targets rather than physiological targets in the early stage of drug screening. G-Quadruplex has become one of the important targets in the development of anti-tumor drugs. However, drugs that target quadruplexes may also bind to dsDNA, which may lead to adverse reactions. In this study, a new three-phase laminar flow chip was constructed to enable the multi-components of a traditional Chinese medicine extract to dynamically and competitively bind with G-quadruplex DNA (on target) and double-stranded DNA (off target), so as to select high-efficiency and low-toxicity anti-tumor drugs. The results showed that there were five compounds in the extracts of Macleaya cordata seeds that exhibited obvious differences in binding to the two targets. Furthermore, the binding constants and modes of four identified alkaloids as they bound to two DNA targets were verified by fluorescence spectra and molecular docking methods. The toxicity to HepG2 and LO2 cells from the four alkaloids was also compared. The results showed that sanguinarine and chelerythrine could be used as candidate drugs with stronger binding to HT24 than DNA26. The chip can also be used for other types of double-target screening of other traditional Chinese medicine extracts or compound libraries.

Allelopathic Molecular Mechanisms of the Two Main Allelochemicals in Sweet Potato

Ruiguo Shi,Guimei Jin,Shicai Shen,Gaofeng Xu,Fengping Zheng,David Roy Clements,Yunhai Yang,Shaosong Yang,Fanghao Wan,Fudou Zhang,Bo Liu

Sweet potato (Ipomoea batatasL.) is one of the most important global food crops. This crop exhibits excellent allelopathic potential against various weeds, but its allelopathic mechanism at the molecular level is unclear. Therefore, metabolomic and transcriptomic analyses were performed to explore the allelopathic effects, metabolic pathway, and associated genes for two major compounds with allelopathic activity, palmitic acid and linoleic acid. The sweet potato variety Ningshu 25 was employed in the current study. The results showed that palmitic acid and linoleic acid had strong allelopathic effects on seed germination, plant growth, antioxidant enzyme activity, and chlorophyll content of two weedsDigitaria sanguinalisandBidens pilosa. The content of the two targeted metabolites was affected by different environmental conditions and was significantly increased under low temperature (15 °C). Five metabolic pathways involved in the two targeted metabolites of fatty acids were found: fatty acid biosynthesis, fatty acid elongation, fatty acid degradation, biosynthesis of cutin, suberine, and wax, and the linoleic acid metabolism pathway. The synthesis of palmitic acid is significantly enriched in the biosynthesis pathways of fatty acids, cutin, suberine, and wax, and the synthesis of linoleic acid is significantly enriched in the linoleic acid metabolism pathway. Under different environmental conditions, there were three key genes expressed—g4988, g11881, and g19673—located in the biosynthesis pathways of cutin, suberine, and wax; four key genes expressed—g31191, g60956, g49811, and g59542—located in the biosynthesis pathway of fatty acids; and six key expressed genes—g26575, g24787, g23517, g57649, g58562, and g4314—located in biosynthesis pathway of linoleic acid, respectively. Our study advances understanding of the molecular mechanisms behind allelopathic traits in sweet potato and provides a set of candidate genes for use in improving allelopathic potential in sweet potato germplasm resources.
