Biomimetic Self-Renewal Polymer Brushes with Protein Resistance Inspired by Fish Skin

Zhonglin Cao,Tiansheng Gan,Gaixia Xu,Chunfeng Ma

Inspired by fish skin, biomimetic self-renewal poly[(ethylene oxide)-co-(ethylene carbonate)] (PEOC) brushes with protein resistance had been prepared via surface-initiated ring-opening polymerization (ROP). The results of hydrolytic degradation indicated that the PEOC brushes could degrade in artificial seawater. Ellipsometry, X-ray photoelectron spectrometry, and contact angle results demonstrated that the PEOC brushes degrade uniformly. By using a quartz crystal microbalance with dissipation, we studied the protein adsorption on the surfaces in artificial seawater at different degradation times. After 24, 48, 96, and 168 h of degradation, the PEOC surfaces showed nearly zero Δf and ΔD for bovine serum albumin, lysozyme, and fibrinogen. More importantly, there was a notably lower density of microorganisms adhered to the surface modified with PEOC compared with that of the surface without PEOC in natural seawater. The current study showed that the PEOC brushes exhibit a self-renewal property with persistent protein resistance and prevent the adhesion of microorganisms. Such a biomimetic polymer had a great potential in marine antibiofouling.

Probing PFOA-induced Metabolic Disorders in Tilapia Through Fish Skin Mucus

Dong Jiaxin,Su Rui,Wu Xiaokang,Dong Deming,Wang Yining,Huang Chenni,Qiu Tao,Gu Yu,Hua Xiuyi,Liang Dapeng

The study of the metabolic toxicity of perfluorooctanoic acid (PFOA) is of great importance in assessing its potential impact on aquatic organisms and the aquatic environment, and provides a fundamental scientific basis for environmental protection and sustainable development. In this study, an electrospray ionization (ESI) source capable of direct analysis of solution samples was combined with a high-resolution Orbitrap Fusion Tribrid mass spectrometer to investigate the metabolic perturbations induced by different concentrations of perfluorooctanoic acid (PFOA) and their correlations in skin mucus and liver tissue of tilapia. Seven common metabolites were found in mucus and liver, and nitrogen metabolism pathways were disturbed in all of them. The up-regulation of glutamate in mucus may reflect an increased demand for amino acids and energy, whereas the down-regulation of glutamate in the liver may lead to dysregulation of nitrogen metabolism, protein synthesis and amino acid metabolism. Enrichment analyses suggest a dysregulation of nitrogen metabolism pathways, which may lead to impaired maintenance of tissue function and dysregulated energy metabolism in tilapia.

Preparation, characterization, and potential biomedical application of composite sponges based on collagen from silver carp skin

Ming-Mao Chen,Yu-Qing Huang,Hao Guo,Yan Liu,Jian-Hua Wang,Jiu-Lin Wu,Qi-Qing Zhang

To reuse fish processing waste for biomedical materials, collagen (Col) was extracted from silver carp skin, and Col–chitosan (Ch) composite sponges were prepared by a freeze-drying method. The atomic force microscopy and electrophoresis results suggest the Col might have been type I. To obtain the optimum conditions for the manufacture of the Col–Ch sponges, the characteristics of sponges composed of different ratios of Col to Ch with different crosslinkers were evaluated. Scanning electron microscopy showed that the sponges had an interconnected network structure with porosity. Infrared spectroscopy demonstrated that intermolecular crosslinkages between Col and Ch occurred. The swelling measurements implied that all of the sponges could bind an 18- to 36-fold amount of distilled water and still maintain their form and stability. When the ratio of Col to Ch was higher than 1:0.25, the swelling and degradation rate decreased with increasing Ch. Cell proliferation, hemolysis, and hemostasis assay indicated that the sponges exhibited noncytotoxicity, biocompatibility, nonhemolysis, and hemostatic efficacy. Overall, we concluded that the optimal ratio of Col and Ch for the sponges was 1:0.25, and glutaraldehyde crosslinking was more suitable than 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride. These results demonstrate the potential application of silver carp skin Col–Ch sponges for tissue engineering and wound dressing in non-weight-bearing tissue. © 2014 Wiley Periodicals, Inc. J. Appl. Carrageenan addition improves the melting temperature and 3D printing performance of the gelatin system

Yue Wang,Lin Han,Wanying Yu,Weiping Zhao,Jinfeng Pan,Sangeeta Prakash,Xiuping Dong

The sturgeon fish skin gelatin (FG) could not form gelation for printing at room temperature and must be hold its shape at low temperature after printing due to low melting and solidifying temperature, causing certain energy loss. The purpose of this study was to investigate the effects of various types (κ-carrageenan: KG; λ-carrageenan: LG; ι-carrageenan: IG) and proportions of carrageenan on the gelation properties and melting temperatures of sturgeon skin gelatin blended system and realize 3D printability at room temperature. The results showed that the turbidity of the blended systems increased significantly with adding 0.4%–0.8% carrageenan. Meanwhile, when the additive concentration of KG was 0.4%, the highest gel strength was 104.02 g. And the hardness and adhesive of gel were 5.44 times and 5.37 times of that single gelatin sample when the concentration of KG reached 0.8%. When IG was added, the melting temperature of the blended system could be improved most obviously, followed by KG, while LG could improve least. In addition, the blended system with IG could form a more dense and uniform network structure. The results of thixotropy showed the gel formed by the blended system had certain thixotropy, but the lag circle area is small resulted in the insufficient shape stability after printing. Among them, the blended systems with 0.8% KG and IG received successfully 3D printing at room temperature. In short, carrageenan has a positive effect on improving the gelation properties and applying in 3D printing of the sturgeon skin gelatin system at room temperature.

Commercial cold-water fish skin gelatin and bovine bone gelatin: Structural, functional, and emulsion stability differences

Ting Zhang,Rui Sun,Mengzhen Ding,Li Li,Ningping Tao,Xichang Wang,Jian Zhong

Both fish gelatins and mammalian gelatins have been explored in the preparation and application of fish oil-loaded emulsion. However, the comprehensive comparisons and relationships of the structural, functional, and emulsions stability differences between these two gelatins remain unclear. In this work, structural, functional, and the emulsion application differences between commercial cold-water fish skin gelatin (CFG) and bovine bone gelatin (BBG) are explored. The results demonstrated the BBG-stabilized emulsion was more stable than the CFG-stabilized emulsion at 4 °C and higher temperatures (23 °C and 37 °C) decreased the creaming stability differences. The creaming index differences between CFG-stabilized and BBG-stabilized emulsions were mainly dependent on emulsion droplet size and interfacial layer thickness consisted of gelatins. Further, the possible reason for CFG's lower emulsion stability at 4 °C might be mainly dependent on its higher β-antiparallel percentage in the secondary structures and its lower thickness of gelatin film-like nanostructures. This work will provide useful information to understand the relationship between structure and function of protein. It also will provide a guide for the application of fish oil-loaded gelatin-stabilized emulsions in food beverages and other foods such as surimi .

Effects of Spray Drying and Freeze Drying on Physicochemical Properties, Antioxidant and ACE Inhibitory Activities of Bighead Carp (Aristichthys nobilis) Skin Hydrolysates

Ye Dong,Wen Yan,Yi-Qi Zhang

The physicochemical, structural properties, antioxidant, and angiotensin I-converting enzyme (ACE) inhibitory activities of fish skin protein hydrolysate (SPH) that were freeze-dried (SPH-FD) and spray-dried (SPH-SD) were investigated. SPH-SD showed abundant volatile compounds, higher DPPH radical scavenging activity and ferrous iron chelating activity than SPH-FD, while the ABTS radical scavenging activity and ACE inhibitory activity were not influenced by the drying method. Amino acid compositions showed a higher proportion of proline and hydroxyproline residues in SPH-FD. The major molecular weights were both distributed below 1000 Da. SPH-SD had spherical structures, while SPH-FD had glass shard-like structures. The results indicated that the drying method could affect the physicochemical properties of hydrolysates, and SPH-SD showed potential prospects in developing functional fortified foods. Keywords:fish skin hydrolysate;freeze drying;spray drying;antioxidant;ACE inhibitory activity

Drag reduction through self-texturing compliant bionic materials

Liu Eryong,Li Longyang,Wang Gang,Zeng Zhixiang,Zhao Wenjie,Xue Qunji

Compliant fish skin is effectively in reducing drag, thus the design and application of compliant bionic materials may be a good choice for drag reduction. Here we consider the drag reduction of compliant bionic materials. First, ZnO and PDMS mesh modified with n-octadecane were prepared, the drag reduction of self-texturing compliant n-octadecane were studied. The results show that the mesh modified by ZnO and PDMS possess excellent lipophilic and hydrophobic, thus n-octadecane at solid, semisolid and liquid state all have good adhesion with modified mesh. The states of n-octadecane changed with temperature, thus, the surface contact angle and adhesive force all varies obviously at different state. The contact angle decreases with temperature, the adhesive force shows a lower value at semisolid state. Furthermore, the drag testing results show that the compliant n-octadecane film is more effectively in drag reduction than superhydrophobic ZnO/PDMS film, indicating that the drag reduction mechanism of n-octadecane is significantly different with superhydrophobic film. Further research shows that the water flow leads to self-texturing of semisolid state n-octadecane, which is similar with compliant fish skin. Therefore, the compliant bionic materials of semisolid state n-octadecane with regular bulge plays a major role in the drag reduction.

Preparation of Calcium-Chelating Peptides from Squid Skin and Evaluation of Calcium Absorption Capacity in Caco-2 Cell Monolayer Model

Jihao Zeng,Xue Bai,Yongli Zhang,Qianyu Le,Jinhong Wu,Huiyun Chen

To develop a highly bioavailable calcium supplement, this study utilized Peruvian squid ( Dosidicus gigas ) skin as a raw material. Through alkaline protease hydrolysis and enzymatic membrane reactor separation, three molecular weight fractions of squid skin peptides were obtained, followed by calcium ion chelation to synthesize calcium-chelating peptides (CCPs-SS). Systematic characterization revealed that the less than 1 kDa fraction of CCPs-SS exhibited superior antioxidant capacity (82.18%) and calcium chelation efficiency (77.14%) in cellular models compared to higher molecular weight counterparts. Optimal synthesis conditions were identified as 60 °C, pH 9, and 12 mg/mL calcium chloride concentration. Post-chelation analyses demonstrated significant physicochemical alterations for CCPs-SS: ζ-potential shifted from −18.4 mV to −10.47 mV, while particle size increased from 476.75 nm to 664.4 nm. Notably, membrane separation enhanced phenylalanine and leucine molar concentrations by 25.5% and 57.6%, respectively, suggesting structural modifications that potentiate bioactivity. These findings demonstrate an innovative strategy for converting squid processing byproducts into functional nutraceuticals, which not only addresses calcium deficiency challenges but also promotes resource sustainability by utilizing waste materials.

Design and characterization of edible chitooligosaccharide/fish skin gelatin nanofiber-based hydrogel with antibacterial and antioxidant characteristics

Yanjing Liu,Xiaodong Xia,Xiyue Li,Fuming Wang,Yaping Huang,Botian Zhu,Xuyang Feng,Ying Wang

Antibacterial and active packaging materials have gained significant research attention in response to the growing interest in food packaging. In this investigation, we developed hydrogel packaging materials with antibacterial and antioxidant properties by incorporating chitooligosaccharide (COS) and fish skin gelatin (FSG) nanofiber membranes, which readily absorbed water and exhibited swelling characteristics. The nanofiber membranes were fabricated by electrospinning technology, embedding COS within FSG, and subsequently crosslinked through the Maillard reaction facilitated by the addition of glucose. The behavior of conductivity, viscosity, and surface tension in the spinning solutions was analyzed to understand their variation patterns. Scanning electron microscopy (SEM) results revealed that the crosslinked COS/FSG nanofiber membranes possessed a uniform yet disordered fiber structure, with the diameter of the nanofibers increasing as the COS content increased. Remarkably, when the COS content reached 25 %, the COS/FSG nanofiber membranes (CF-C-25) exhibited a suitable fiber diameter of 437.16 ± 63.20 nm. Furthermore, the thermal crosslinking process involving glucose supplementation enhanced the hydrophobicity of CF-C-25. Upon hydration, the CF-H-25 hydrogel displayed a distinctive porous structure, exhibiting a remarkable swelling rate of 954 %. Notably, the inclusion of COS significantly augmented the antibacterial and antioxidant properties of the hydrogel-based nanofiber membranes. CF-H-25 demonstrated an impressive growth inhibition of 90.56 ± 5.91 % against E. coli , coupled with excellent antioxidant capabilities. In continuation, we performed a comprehensive analysis of the total colony count, pH, TVB-N, and TBA of crucian carp. The CF-H-25 hydrogel proved highly effective in extending the shelf life of crucian carp by 2–4 days, suggesting its potential application as an edible membrane for aquatic product packaging.

Functional improvement and characterization of protein hydrolysates prepared by the fermentation of irradiated tilapia skin

Litong Liu,Junwei Qin,Bifeng Lan,Xi Hu,Tao Liao,Xiaofei Tian,Zhenqiang Wu

Gamma irradiation (γ-irradiation) sterilizes and modifies proteins. This study aimed to enhance the performance of protein hydrolysates of fermented tilapia fish skin using irradiation. The results showed that γ-irradiation could effectively kill harmful microorganisms in fish skin at 6 kGy. Irradiation at 1–6 kGy dose disintegrated the internal structure of fish skin proteins, thus increasing the degree of hydrolysis after fermentation by Bacillus subtilis L4; however, irradiation at 9–12 kGy caused cross-linking and aggregation of fish skin proteins. The solubility, and water and oil holding capacity of the protein hydrolysates were significantly improved compared to those of the non-irradiated sample. The maximum emulsifying activity (164.61±16.28 m 2 /g), foaming activity (91.13±2.30 mL), and surface hydrophobicity index (5766.43±770.56) were observed in the protein hydrolysates fermented from fish skin irradiated at 12 kGy. The antioxidant activity of the protein hydrolysates increased with increasing irradiation doses. This suggested that the functional properties and antioxidant activity of protein hydrolysates from fish skin fermentation can be improved by appropriate irradiation pretreatment, providing a feasible method for improving the utilization and quality of proteins in fishery waste.

Tribological behavior of ZrO2/WS2 coating surfaces with biomimetic shark-skin structure

Xuemu Li,Jianxin Deng,Yang Lu,Liangliang Zhang,Jie Sun,Fengfang Wu

This study demonstrated the dry friction and wear behavior of sol-gel ZrO 2 /WS 2 coatings with biomimetic shark-skin textured surfaces. The effect of added WS 2 on the microstructure and mechanical properties of ZrO 2 coatings was examined and the dry sliding wear and friction behaviors against commercial AISI1045 steel balls were investigated. After adding WS 2 , the micro hardness of ZrO 2 coating decreased by ~39% (from 23.7 GPa to 14.5 GPa), and the average steady state friction coefficient was decreased from ~0.43 to ~ 0.2. When the ZrO 2 /WS 2 coating surface was textured biomimetic shark skin, nearly 66% decrease of the average steady state friction coefficient and 82% decrease of ball wear rate were obtained as compared with the pure ZrO 2 coatings. The possible frictional mechanism of the biomimetic shark-skin structures was discussed.

Structural and emulsion stabilization comparison of four gelatins from two freshwater and two marine fish skins

Lili Yang,Mengyang Yang,Jiamin Xu,Yinghua Nie,Wenjuan Wu,Ting Zhang,Xichang Wang,Jian Zhong

This study analyzed the structural and emulsion stabilization properties of two freshwater and two marine fish skin gelatins: Chinese longsnout catfish skin gelatin (CLCSG), silver carp skin gelatin (SCSG), salmon skin gelatin (SSG), and Alaska pollack skin gelatin (APSG). Their gel strengths (Bloom values) were: 361 ± 1 (SCSG) > 253 ± 4 (CLCSG) > 69 ± 1 (SSG) > 36 ± 2 (APSG). Higher molecular weights and α/β subunit contents of gelatins might induce higher gel strengths. Both creaming and droplet stability were completely the same to the contents of imino acids, β-sheet percentages, and β-turn percentages, whereas they were completely the opposite to random coil percentages. The emulsion stabilization mechanisms involved an “fish skin source – protein chemical composition – protein secondary structure – protein functional properties – emulsion stability” route. This study provided useful knowledges for gelatin science and for the comprehensive utilization of aquatic by-products in gelatin industry.
