Effect of Fe and EDTA on Freshwater Cyanobacteria Bloom Formation

Ting Zhang,Jian He,Xingzhang Luo

Due to the fact that not all eutrophic lakes have cyanobacteria blooms, we hypothesized Fe may be another important limiting factor which regulates cyanobacteria bloom formation. We tested the hypothesis by batch cultures of bloom-forming Cyanobacterium,Microcystis aeruginosawith different ethylenediaminetetraacetic acid (EDTA)-Fe concentrations (0.5–6.0 mg/L), three levels of initial biomass, and excessive N and P (N = 4.2 mg/L, P = 0.186 mg/L) to simulate dynamically a cyanobacteria bloom in eutrophic conditions. The effect of EDTA and Fe uptake kinetics byM. aeruginosawere also examined. Results showedM. aeruginosagrowth rate positively correlated with EDTA-Fe concentration and negatively correlated with biomass. Maximal biomass ofM. aeruginosawas determined by Fe availability and initial biomass. EDTA could decrease both Fe availability and toxicity. Based on experimental results, a conceptual model of how Fe availability regulates cyanobacterial biomass in eutrophic lakes was developed. This study demonstrated bioavailable Fe is a potential limiting factor in eutrophic lakes that should be included in eutrophication management strategies. Keywords:Microcystis;eutrophication control;freshwater;Fe availability

Study on the cyanobacterial toxin metabolism of Microcystis aeruginosa in nitrogen-starved conditions by a stable isotope labelling method

Zong-Yao Qian,Xiao Chen,He-Te Zhu,Jun-zhe Shi,Ting-Ting Gong,Qi-Ming Xian

In this study, the biosynthesis of microcystins (MCs) was investigated after long-term nitrogen-starved conditions in cyanobacterium Microcystis aeruginosa . The results demonstrated that the algal cells were able to survive in a non-growing state with nitrogen starvation for more than one month. The physiological properties of the algal cells were studied to elucidate the mechanisms of viability under nitrogen-deprivation conditions. After the state of nitrogen chlorosis, new toxins could be resynthesized and tracked using 15 N-stable isotope-labelled nitrogen. Nitrogen starvation of nutritionally replete cells resulted in a significant increase of microcystin-LY (MC-LY), thereby suggesting that MC-LY may undergo catabolism to provide nitrogen or that MC-LY may be produced to play an important role in the cell in response to nitrogen deprivation. The rank order of different types of nitrogen in algal cells assimilation was N-ammonium > N-urea > N-nitrate > N-alanine. The relationship between the production of toxin variants and various environmental conditions is an interesting issue for future research and may help improve the understanding of the ecological role of cyanobacterial toxins .

Regulation of Photosynthesis in Bloom-Forming Cyanobacteria with the Simplest β-Diketone

Mihebai Yilimulati,Jiyuan Jin,Xin Wang,Xiaomeng Wang,Dmitry Shevela,Bing Wu,Kai Wang,Lang Zhou,Yunlu Jia,Bingcai Pan,Govindjee Govindjee,Shujuan Zhang

Selective inhibition of photosynthesis is a fundamental strategy to solve the global challenge caused by harmful cyanobacterial blooms. However, there is a lack of specificity of the currently used cyanocides, because most of them act on cyanobacteria by generating nontargeted oxidative stress. Here, for the first time, we find that the simplest β-diketone, acetylacetone, is a promising specific cyanocide, which acts on Microcystis aeruginosa through targeted binding on bound iron species in the photosynthetic electron transport chain, rather than by oxidizing the components of the photosynthetic apparatus. The targeted binding approach outperforms the general oxidation mechanism in terms of specificity and eco-safety. Given the essential role of photosynthesis in both natural and artificial systems, this finding not only provides a unique solution for the selective control of cyanobacteria but also sheds new light on the ways to modulate photosynthesis.

Toxic effects of disinfection by-products on Pseudokirchneriella subcapitata and co-cultured algae community

Jinjie Li,Tielan Wang,Junzeng Xue

The widespread use of disinfectants in response to the COVID-19 pandemic may increase the concentration of disinfection by-products (DBPs) in surface water, posing ecological risks. This study assessed the acute toxicity of haloacetic acids and halophenols DBPs to Pseudokirchneriella subcapitata , and evaluated the impact of environmental concentrations of DBPs on co-cultured algae communities. The results showed that after exposure to disinfection by-products, the photosynthetic pigments of Pseudokirchneriella subcapitata were significantly reduced, and the MDA content and antioxidant enzyme activity were increased. The results of the ecological risk assessment showed that the RQ values of TBAA, 2,4-DBP, and 2,6-DBP were >0.1, with medium and high ecological risks. Despite this, the three DBPs with the highest risk values had little effect on the community structure of co-cultured algae. In contrast, the disinfection by-products with low ecological risk had a greater impact on the community structure and greatly promote the growth of cyanobacteria, which may cause new ecological risks. Therefore, not only high-risk disinfection by-products need to be paid attention to, but the impact of low-risk disinfection by-products cannot be ignored.

The inhibition mechanism and death mode of Microcystis aeruginosa induced by the continuous pressure of artemisinin sustained-release microspheres (ASMs)

Cunhao Du,Lixiao Ni,Jakpa Wizi,Wenlu Sang,Shiyi Rong,Xianglan Li,Chu Xu,Yan Li,Xuqing Chen,Shiyin Li

Artemisinin , a natural allelochemical substance extracted from Artemisia annua , has been shown to have strong inhibitory effects on harmful cyanobacteria . However, only a few studies have systematically studied the mechanism of artemisinin in inhibiting algae and the process of algae cell death. In this study, the inhibition mechanism and death mode of M. aeruginosa under artemisinin sustained-release microspheres (ASMs) were investigated using a series of morphological, physiological and biochemical characteristics. The findings revealed that M. aeruginosa was significantly inhibited immediately after exposure to the 0.54 g·L −1 ASMs, with an inhibition rate (IR) exceeding 95 %, and the synthesis of phycobiliprotein and esterase activity were disturbed. In addition, ASMs exposure significantly increased caspase-3(-like) activities of M. aeruginosa . Under the continuous stress of ASMs, M. aeruginosa cells had three cell death modes: the first was necrosis-like cell death, which was characterized by cell wall rupture and leakage of cell contents; the second was apoptosis-like cell death, which was characterized by the disintegration of the cell structure, the integrity of the cell membrane , and the appearance of apoptotic bodies; and the last was autophagy-like cell death, which was characterized by cell swelling and appearance of autophagy vacuoles. These results indicated that ASMs could inhibit photosynthesis of M. aeruginosa cells, interfere with its antioxidant mechanisms and activate caspase-3(-like) activity, resulting in programmed cell death (PCD).

Use of chlorophyll a fluorescence to elucidate the toxicity target of N-phenyl-2-naphthylamine on photosynthetic system of Cylindrospermopsis raciborskii (Cyanobacteria)

Rui Liu,Xiaofei Ran,Fang Bai,Jinzhu Xu,Songqi Yang,Junqiong Shi,Zhongxing Wu

In order to investigate the toxicity targets of the secondary metabolite from aquatic plants, N-phenyl-2-naphthylamine, on Cylindrospermopsis raciborskii photosynthetic system II (PSII), we determined chlorophyll a (Chl a), specific growth rate (μ−d), photosynthetic rapid light curves and polyphasic Chl a fluorescence transients following exposure to N-phenyl-2-naphthylamine. The results showed significant decreases in Chl a following 48 h at high concentration (0.50 mg l−1) exposure to N-phenyl-2-naphthylamine. The PSII electron transfer rate and Chl a fluorescence transients appeared to be affected after the cells were exposed to concentrations of 0.50, 1.00 and 2.00 mg l−1. Exposure to higher concentration of N-phenyl-2-naphthylamine (> 0.50 mg l−1) mainly depressed the density of reaction centers (RC/CS0) and quantum energy flux ratios (ϕP0 and ϕE0) but increased the absorption flux per RC (ABS/RC), dissipated energy flux per RC (DI0/RC) and the average redox state of QA−/QA in the time span from 0 to tFmax (Sm/tFmax). However, exposure to low concentration (≤ 0.5 mg l−1) mainly inhibited the trapping energy fluxes per reaction center (TR0/RC), electron transport energy fluxes per reaction center (ET0/RC) and energy flux ratio (ψ0). This suggested that the donor and the acceptor sides of PSII were targets of N-phenyl-2-naphthylamine in C. raciborskii. Additionally, our results indicated that the toxicity target at low concentrations of N-phenyl-2-naphthylamine was at each step of electron transport between QA− and the Cyt b6/f complex; whereas, the reduction of QA was the potential target at high concentrations. We suggest that N-phenyl-2-naphthylamine may be a potential allelochemical to control harmful cyanobacterial blooms.

Effects of different metal ions (Ca, Cu, Pb, Cd) on formation of cyanobacterial blooms

Peng Gu,Qi Li,Weizhen Zhang,Zheng Zheng,Xingzhang Luo

Eastern China is a typical region that suffers from harmful cyanobacterial blooms. Numerous studies have focused on bloom formation mechanisms; however, the detailed mechanisms remained unclear. Our study explored the influence of four metal ions (Ca, Cu, Pb and Cd) on Microcystis aeruginosa to determine their effects on bloom formation. We found that Ca concentrations higher than 100 mg L −1 contributed to cyanobacterial bloom formation. The presence of Ca triggered the anti-oxidation process and promoted the secretion of extracellular polysaccharides , thus inducing aggregation of algal cells and enhancing their buoyancy 2.1-fold more than the control ( p <0.05). The reverse regulation of dissolved CO 2 to bicarbonate by carbonic anhydrase formed a large amount of carbonate and decreased the growth rate by 38–56%. Cu (>0.1 mg L −1 ) presented significant toxicity to algal cells while Pb (>1 mg L −1 ) suppressed the algal growth rate due to the acidic condition. Cd (<0.1 mg L −1 ) exhibited no apparent toxicity to the algae. Furthermore, as the buoyancy increased, Cd was likely to facilitate the formation of cyanobacterial blooms, which needs further research. These findings can provide a theoretical basis for eutrophic lake management and contribute to the development of water quality and wastewater discharge standards.

Biodegradation of polychlorinated biphenyls (PCBs) by the novel identified cyanobacterium Anabaena PD-1

Hangjun Zhang,Xiaojun Jiang,Liping Lu,Wenfeng Xiao

Polychlorinated biphenyls (PCBs), a class of hazardous pollutants, are difficult to dissipate in the natural environment. In this study, a cyanobacterial strain Anabaena PD-1 showed good resistance against PCB congeners. Compared to a control group, chlorophyll a content decreased 3.7% and 11.7% when Anabaena PD-1 was exposed to 2 and 5 mg/L PCBs for 7 d. This cyanobacterial strain was capable of decomposing PCB congeners which was conclusively proved by determination of chloride ion concentrations in chlorine-free medium. After 7 d, the chloride ion concentrations in PCB-treated groups (1, 2, 5 mg/L) were 3.55, 3.05, and 2.25 mg/L, respectively. The genetic information of strain PD-1 was obtained through 16S rRNA sequencing analysis. The GenBank accession number of 16S rRNA of Anabaena PD-1 was KF201693.1. Phylogenetic tree analysis clearly indicated that Anabaena PD-1 belonged to the genus Anabaena . The degradation half-life of Aroclor 1254 by Anabaena PD-1 was 11.36 d; the total degradation rate for Aroclor 1254 was 84.4% after 25 d. Less chlorinated PCB congeners were more likely to be degraded by Anabaena PD-1 in comparison with highly chlorinated congeners. Meta - and para -chlorines in trichlorodiphenyls and tetrachlorobiphenyls were more susceptible to dechlorination than ortho -chlorines during the PCB-degradation process by Anabaena PD-1. Furthermore, Anabaena PD-1 can decompose dioxin-like PCBs. The percent biodegradation of 12 dioxin-like PCBs by strain PD-1 ranged from 37.4% to 68.4% after 25 days. Results above demonstrate that Anabaena PD-1 is a PCB-degrader with great potential for the in situ bioremediation of PCB-contaminated paddy soils.

Contrasting effects of phytoplankton aging on microplastic antibiotic adsorption depending on species tolerance, and biofouling level

Claude Kiki,Adénikè Bernice Eloise Adéoyé,Xi Li,Xiaopeng Yan,Jinlu Feng,Chang-Ping Yu,Qian Sun

Recent studies have reported conflicting results on the effects of biofouling on the adsorption behavior of microplastics (MPs). However, the underlying mechanisms driving the adsorption of MPs undergoing biofouling in aquatic environments remain unclear. This study examined the interactions between polyamide (PA), polyvinyl chloride (PVC) and polyethylene (PE) with two phytoplankton, namely cyanobacteria Microcystis aeruginosa and microalgae Chlorella vulgaris . Results indicated that MP effects on phytoplankton were dose- and crystalline-type dependent, with M. aeruginosa being more sensitive to MP exposure than C. vulgaris in the inhibitory order PA > PE > PVC. Analysis of antibiotic adsorption of the MPs showed significant contributions from CH/π interactions on PE and PVC and hydrogen bonding on PA, which decreased with phytoplankton biofouling and aging. Meanwhile, higher levels of extracellular polymeric substances on microalgae-aged MPs compared to cyanobacteria-aged MPs were conducive to adsorption of antibiotics, mainly through hydrophobic interactions. Overall, promotional and anti-promotional adsorption of antibiotics on MPs was induced by biofouling and aging of microalgae and cyanobacteria, respectively. This study provides deep insights into the specific mechanisms by which biofouling affects MP adsorption in aquatic environments, thus advancing our understanding of this critical environmental issue.

Construction of a Self-Enhanced Fluorescent Aptasensor Based on Lanthanide Ligand Self-Assembly and Catalytic Hairpin Assembly for Highly-Sensitive Detection of Anatoxin-a

Alina Bari,Guiying Jin,Yinbo Ban,Fuqiang Zhang,Guangfeng Wang

Anatoxin-a (ATX-a) is a potent neurotoxin produced by cyanobacteria, with growing evidence unequivocally linking it to acute aquatic toxicity, neurological impairment, and increased mortality in wildlife and livestock, highlighting its significant threat to ecosystem stability and public health. Here, we report a novel fluorescent aptasensor system (FAS) that integrates novel DNA-functionalized Eu nanoparticles (DNA-Eu NPs) with a catalytic hairpin assembly (CHA) strategy for ultrasensitive ATX-a detection. We used single-stranded DNA C16T20A16 (ssDNA) oligomers rich in cytosine, thymine, and adenine as effective antenna ligands to sensitize the luminescence of Eu. The Black Hole Quencher (BHQ) labeled hairpin DNA (H3) hybridizes with the exposed DNA sequence on the surface of NPs through complementary base pairing, promoting the fluorescence energy transfer between the quenching group BHQ and DNA-Eu NPs, leading to the formation of DNA-Eu-H3 and providing stable “on–off” fluorescence. In the presence of ATX-a, it specifically binds to the aptamer (B) to release trigger DNA (T) from duplex DNA (B-T). T initiates a CHA-driven cascade amplification between H1, H2, and DNA-Eu-H3, resulting in fluorescence recovery and signal amplification of DNA-Eu NPs. This FAS exhibits good stability, excellent specificity, and high sensitivity, with a detection limit (LOD) of 0.30 pg/mL and a linear range of 0.001–500 ng/mL. This strategy provides an innovative, highly sensitive, label-free assay and is expected to be a powerful tool for environmental toxin analysis.

Effect of erythromycin exposure on the growth, antioxidant system and photosynthesis of Microcystis flos-aquae

Jinjin Wan,Peiyong Guo,Xiaofang Peng,Keqi Wen

Erythromycin, a macrolide antibiotic, is commonly used in human life. This compound and its derivatives have been detected in various aquatic compartments and may pose a serious threat to aquatic organisms. This study investigated the effects of erythromycin on the growth, antioxidant system and photosynthesis of Microcystis flos-aquae . The results showed that at 0.001–0.1   μg   L −1 , erythromycin could stimulate the growth of M. flos-aquae and increase its photosynthetic activity; however, it did not significantly increase the activities of superoxide dismutase (SOD) and catalase (CAT) or the levels of malondialdehyde (MDA) and reactive oxygen species (ROS). In contrast, the growth of M. flos-aquae was significantly inhibited ( p   <   0.01) at high levels of erythromycin, reaching an inhibition rate of 81.6% at 40   μg   L −1 erythromycin. At the same time, the activities of SOD and CAT along with MDA content also increased significantly ( p   <   0.01), indicating that the high concentrations of erythromycin caused a severe oxidative stress on algae. However, the balance between oxidants and antioxidant enzymes were disrupted because ROS content simultaneously increased. In addition, the fluorescence parameters of M. flos-aquae decreased significantly with both exposure time and increasing concentration of erythromycin, indicating that photosynthesis was inhibited.

Low-frequency ultrasound assisted contact-electro-catalysis for efficient inactivation of Microcystis aeruginosa

Peiyun Wei,Mengxia Tang,Yao Wang,Baowei Hu,Xiaolei Qu,Yanfeng Wang,Guandao Gao

Frequent cyanobacterial blooms pose a serious threat to the aquatic ecosystem and human health, so developing an efficient algae removal method is a long-term goal for bloom management. Current technologies for algal bloom control need urgent improvement in terms of algicide recovery, eco-friendliness and cost. Here we propose a contact-electro-catalytic method, using polytetrafluoroethylene (PTFE) film as a reusable catalyst. This contact-electro-catalytic approach involves the generation of reactive oxygen species (e.g., O 2 • – , HO•, 1 O 2 and H 2 O 2 ) through water-PTFE contact electrification under the low-frequency ultrasonic waves, facilitating the inactivation of algae. The removal rate of the cyanobacterium Microcystis aeruginosa ( M. aeruginosa ) exposured to the water-PTFE contact-electro-catalytic system is almost five times greater than that of ultrasound alone after 5 h. A mechanistic investigation revealed that the contact-electro-catalytic system damaged the photosynthetic activity, antioxidant system and membrane integrity of the cells. Additionally, LC-MS metabolomic analysis indicated that this system caused substantial significant disruptions in the TCA cycle, amino acid metabolism, purine metabolism and phospholipid metabolism. Three-dimensional fluorescence spectroscopy suggested contact-electro-catalysis could further availably degrade the organic matter. We anticipate that this method can provide an eco-friendly, highly efficient and economic approach for effective control of harmful algal blooms.
