Yueying Jin,Shengyu Cheng,Ruobai Liu,Chenglong Yu,Lingli Zhang,Xi-Ling Li,Guanghai Yan,Mingyu Zheng,Jun Zhe Min

Lurong Dabu decoction (LRDBD) is an effective traditional Chinese Korean ethnic medicine prescription composed of eight herbs, which is used for treating asthma. However, its material basis has not been studied yet. Herein, the use of a new and highly sensitive UHPLC–Q Exactive Orbitrap–HRMS technique is proposed for the high-resolution and accurate identification of the material basis of LRDBD. We identified 122 compounds belonging to different groups in LRDBD. Among these, 23 ingredients produced by decoction were identified and compared with 8 single herb compounds. Moreover, 39 other significantly different compounds were identified. Additionally, 29 absorbed prototype components and 35 metabolites were identified in rat plasma. Half of the prototype components were originated from antler velvet, it has corroborated the compatibility theory of Sasang medicine. To the best of our knowledge, the material basis of LRDBD was characterized for the first time. Our findings provide basic data and a method for further discovering potential drug targets and revealing the action mechanism of LRDBD in asthma treatment.

Cranial repair-promoting effect of oxidised sodium alginate/amino gelatine injectable hydrogel loaded with deer antler blood peptides

Taojing Yu,Qiteng Ding,Ning Wang,Shuai Zhang,Zhiqiang Cheng,Chunli Zhao,Qingjie Li,Chuanbo Ding,Wencong Liu

This study aimed to enhance the treatment of bone defects and increase peptide bioavailability. To achieve this, antioxidant-active peptides (DBPs) were extracted from deer antler blood and incorporated into an oxidised sodium alginate/amino gelatine injectable hydrogel (OSA/N-Gel). This bioscaffold was created through the Schiff base reaction, resulting in the development of an injectable hydrogel comprising OSA, amino gelatine, and deer antler blood peptides (OSA/N-Gel/DBP). OSA/N-Gel/DBP is characterised by a loose and porous structure that enhances nutrient flow and confers good degradability, enabling the gradual release of DBP to meet the long-lasting treatment requirements for bone repair. In vitro , 5-Ethynyl-2′-deoxyuridine (EDU), alkaline phosphatase (ALP), and Alizarin Red S (ARS) staining showed the pro-proliferative and pro-mineralising abilities of OSA/N-Gel and OSA/N-Gel/DBP on osteoblasts (MC3T3). OSA/N-Gel/DBP effectively promoted the expression of osteogenesis-related genes, such as ALP and vascular endothelial growth factor (CD31), and deposition of collagen (COL-1), and activated the wingless-related integration site (Wnt) signalling pathway, thereby promoting bone regeneration. The effect of OSA/N-Gel/DBP was significantly superior to that of the OSA/N-Gel group, indicating that DBP has good osteogenic properties. We successfully repaired bone defects and broadened the application of antler blood, thereby providing a novel approach to treating bone defects.

Pilose Antler Peptide-3.2KD Ameliorates Adriamycin-Induced Myocardial Injury Through TGF-β/SMAD Signaling Pathway

Xu Yan,Qu Xiaobo,Zhou Jia,Lv Guangfu,Han Dong,Liu Jinlong,Liu Yuexin,Chen Ying,Qu Peng,Huang Xiaowei

Adriamycin (ADR)-based combination chemotherapy is the standard treatment for some patients with tumors in clinical, however, long-term application can cause dose-dependent cardiotoxicity. Pilose Antler, as a traditional Chinese medicine, first appeared in the Han Dynasty and has been used to treat heart disease for nearly a thousand years. Previous data revealed pilose antler polypeptide (PAP, 3.2KD) was one of its main active components with multiple biological activities for cardiomyopathy. PAP-3.2KD exerts protective effects againt myocardial fibrosis. The present study demonstrated the protective mechanism of PAP-3.2KD against Adriamycin (ADR)-induced myocardial injury through using animal model with ADR-induced myocardial injury. PAP-3.2KD markedly improved the weight increase and decreased the HW/BW index, heart rate, and ST height in ADR-induced groups. Additionally, PAP-3.2KD reversed histopathological changes (such as disordered muscle bundles, myocardial fibrosis and diffuse myocardial cellular edema) and scores of the heart tissue, ameliorated the myocardial fibrosis and collagen volume fraction through pathological examination, significantly increased the protein level of Bcl-2, and decreased the expression levels of Bax and caspase-3 in myocardial tissue by ELISA, compared to those in ADR-induced group. Furthermore, ADR stimulation induced the increased protein levels of TGF-β1 and SMAD2/3/4, the increased phosphorylation levels of SMAD2/3 and the reduced protein levels of SMAD7. The expression levels of protein above in ADR-induced group were remarkably reversed in PAP-3.2KD-treated groups. PAP-3.2KD ameliorated ADR-induced myocardial injury by regulating the TGF-β/SMAD signaling pathway. Thus, these results provide a strong rationale for the protective effects of PAP against ADR-induced myocardial injury, when ADR is used to treat cancer.

Functional characterization of triterpene synthases in Cibotium barometz

Zhongju Ji,Baolian Fan,Yidu Chen,Jingyang Yue,Jiabo Chen,Rongrong Zhang,Yi Tong,Zhongqiu Liu,Jincai Liang,Lixin Duan

Cibotium barometz (Linn.) J. Sm., a tree fern in the Dicksoniaceae family, is an economically important industrial exported plant in China and widely used in Traditional Chinese Medicine. C. barometz produces a range of bioactive triterpenes and their metabolites. However, the biosynthetic pathway of triterpenes in C. barometz remains unknown. To clarify the origin of diverse triterpenes in C. barometz , we conducted de novo transcriptome sequencing and analysis of C. barometz rhizomes and leaves to identify the candidate genes involved in C. barometz triterpene biosynthesis. Three C. barometz triterpene synthases (CbTSs) candidate genes were obtained. All of them were highly expressed in C. barometz rhizomes, consisting of the accumulation pattern of triterpenes in C. barometz . To characterize the function of these CbTSs, we constructed a squalene- and oxidosqualene-overproducing yeast chassis by overexpressing all the enzymes in the MVA pathway under the control of GAL-regulated promoter and disrupted the GAL80 gene in Saccharomyces cerevisiae simultaneously. Heterologous expressing CbTS1, CbTS2, and CbTS3 in engineering yeast strain produced cycloartenol, dammaradiene, and diploptene, respectively. Phylogenetic analysis revealed that CbTS1 belongs to oxidosqualene cyclase, while CbTS2 and CbTS3 belong to squalene cyclase. These results decipher enzymatic mechanisms underlying the origin of diverse triterpene in C. barometz .

Quaternized oxidized sodium alginate injectable hydrogel with high antimicrobial and hemostatic efficacy promotes diabetic wound healing

Shuang Ma,Kecheng Chen,Qiteng Ding,Shuai Zhang,Yang lu,Taojing Yu,Chuanbo Ding,Wencong Liu,Shuang liu

In this paper, a hydrogel material with efficient antibacterial, hemostatic, self-healing, and injectable properties was designed for the treatment of diabetic wounds. Firstly, quaternary ammonium salts were grafted with oxidized sodium alginate, and quaternized oxidized sodium alginate (QOSA) was synthesized. Due to the introduction of quaternary ammonium group it has antibacterial and hemostatic effects, at the same time, due to the presence of aldehyde group it can be reacted with carboxymethyl chitosan (CMCS) to form a hydrogel through the Schiff base reaction. Furthermore, deer antler blood polypeptide (DABP) was loaded into the hydrogel (QOSA&CMCS&DABP), showing good swelling ratio and bacteriostatic effect. In vitro and in vivo experiments demonstrated that the hydrogel not only quickly inhibited hepatic hemorrhage in mice and reduced coagulation index and clotting time in vitro, but also significantly enhanced collagen deposition at the wound site, accelerating wound healing . This demonstrates that the multifunctional hydrogel materials (QOSA&CMCS&DABP) have promising applications in the acceleration of skin wound healing and antibacterial promotion.

Multifunctional biomaterial hydrogel loaded with antler blood peptide effectively promotes wound repair

Shuang Ma,Qiteng Ding,Guofeng Xia,Anning Li,Jianguo Li,Pingping Sun,Chuanbo Ding,Wencong Liu

Diabetes is an epidemic in contemporary society, which seriously affects people's health. Therefore, it is imperative to develop a multifunctional wound dressing that can expedite the healing of diabetic wounds. In this study, quaternized oxidized sodium alginate (QOSA) and carboxymethyl chitosan (CMCS) formed hydrogel through Schiff base reaction, and the composite hydrogel was prepared by adding the antioxidant activity of deer antler blood polypeptide (D). The hydrogel exhibits favorable attributes, including a high swelling ratio, biocompatibility, and noteworthy antioxidant, antibacterial, and hemostatic properties. Finally, it was used to evaluate its effectiveness in repairing diabetic wounds. Upon evaluation, this hydrogel can effectively promote diabetic wound healing. It facilitates cell proliferation at the wound site, mitigates inflammatory responses, and enhances the expression of growth factors at the wound site. This suggests that this hydrogel holds significant promise as an ideal candidate for advanced wound dressings.

Effect of velvet antler on the immune activity of cyclophosphamide-induced immunosuppressed mice

Liu Song-xin,Li Zhi-man,Shao Zi-jun,Xia Yun-shi,Zhao Li-juan,Ren Duo-duo,Sun Yin-shi

To study the antioxidant and immune protection activity of velvet antler (VA) in cyclophosphamide-induced immunocompromised mice, BALB/c mice were intragastrically administered 100, 300, or 500 mg/(kg·d) VA for 30 days, and were intraabdominally injected with cyclophosphamide to create a hypoimmune murine model. Cellular immunity, macrophage phagocytic activity, natural killer (NK) cell activity, white blood cell count, splenic lymphocyte subsets and the mRNA expression levels of serum cytokines were analysed. We found that the IC50 for removing 2,2'-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS), OH, and 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radicals of VA were 0.476, 4.33, and 37.92 mg/mL, respectively. VA could enhance innate and adaptive immunity of mice by improving cellular immunity, macrophage phagocytic ability, and NK cell activity; promoting the production of IL-4, IFN-γ, TNF-α and other cytokines; increasing the content and ratio of lymphocyte subsets. These results indicate that VA has good antioxidant activity and can improve the immune function of cyclophosphamide-induced immunodeficient mice.

Mechanism of Velvet Antler Peptide in Preventing Mild Cognitive Impairment Based on Non-Targeted Metabolomics

Xu Huibo,Yang Runze,Liu Yuexin,Wang Jinji,Lan Tianye,Lin Zhe,Zhu Enzhi,Huang Xiaowei

Velvet antler peptide (VAP) is one of the main active ingredients in Velvet antler (VA), and its mechanism in preventing mild cognitive impairment (MCI) needs to be further studied. In this research, rats model with MCI was established, non-targeted metabonomics method was performed. Total 50 wistar rats were divided into: control, model, piracetam (PIR), high- and low-dose VAP groups, and were administered for 18 days. Scopolamine (2 mg/kg) was injected intraperitoneally to establish the MCI rat model. The contents of superoxide dismutase (SOD) and malondialdehyde (MDA) in rat serum were detected by the enzyme-linked immunosorbent assay (ELISA). The expression of B-cell lymphoma/leukemia-2 (Bcl-2), Bcl-2-associated death promoter (Bad) and Bcl-2-associated X protein (Bax) in hippocampus of rats were detected by Western blot analysis. The brain and kidney of MCI rats were detected by ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC-Q/TOF-MS) metabolomics technology, and the differences of metabolites and the dynamic changes of metabolic pathway were analyzed based on the results of Principal Component Analysis (PCA) and Orthogonal partial least squares discriminant analysis (OPLS-DA) The correlation analysis of significantly different metabolites was then analyzed by Cluster analysis (CA), and then the differential metabolite pathway enrichment analysis was performed. According to the statistical analysis of univariate and multivariates, the models constructed between the groups are stable and reliable, the variability and separation trend are good, and there are differences in the metabolomes. In results, VAP increased the content of serum SOD, reduced the content of MDA, increased the expression level of Bcl-2 and reduced the expression level of Bad and Bax protein in MCI model rats. The relevant potential markers in the brain and kidney of MCI rats mainly included folic acid, acetyl L-carnitine (ALC), Prostaglandin E1 (PGE1), 3′,5′-cyclic-adenosine-monophosphate (cAMP) and various amino acids. Therefore, VAP could inhibit oxidative stress response, reduce hippocampal neuron apoptosis and protect neurons in MCI rats. Its mechanism may be related to regulating folate and lipid metabolism, cyclic adenosine phosphate energy cycle, amino acid biosynthesis and renal secretion of sex hormones.

Pilose antler peptide protects osteoblasts from inflammatory and oxidative injury through EGF/EGFR signaling

Yang Chunhui,Cai Wenjun,Wen Hui,Sha Liquan,Zhao Changwei,Zhang Tianzhu,Zhao Wenhai

Epidermal growth factor (EGF)/EFG receptor (EGFR) signaling plays an important role in the osteoblastogenesis. The potential effects of pilose antler peptide (PAP) on osteoblast cell damages was investigated in our present study through EGF/EGFR signaling. In MC3T3-E1 osteoblastic cells, PAP treatment significantly inhibited the production of inflammatory cytokines by decreasing the levels of serum proinflammatory cytokines interleukin-1β (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α). PAP treatment also alleviated the oxidative responses as indicated by increased activities of catalase (SOD) and decreased levels of malondialdehyde (MDA). EGF inhibition, by siRNA knockdown, almost abolished PAP-induced osteoblast cytoprotection against inflammation and oxidant stress. Further, our results showed that PAP stimulated the nuclear erythroid factor 2-related factor 2 (Nrf2)2/heme oxygenase-1(HO-1) signaling, and inhibited the activation of uclear factor kappa B (NF-κB) pathway in MC3T3-E1 cells. On the other hand, EGF siRNA knockdown inhibited PAP-induced cytoprotection, which decreased the expression of Nrf-2, HO-1 and increased the level of p -NF-κBp65, p -IκBα in MC3T3-E1 cells. Thus, our research demonstrated that PAP protects osteoblasts from inflammatory and oxidative injury through EGF/EGFR signaling.

Quantitative Proteomics Based on Data-Independent Acquisition Reveals Differential Protein Expression in Sika Deer Antler-Derived Traditional Chinese Medicine Across Different Growth Periods

Yihao Jiang,Lei Liang,Zheng Hu,Zhangfeng Ding,Zhibiao Yu

Deer antler-derived medicinal materials, including antler velvet, antlers, and deer antler base, exhibit differential therapeutic efficacy across developmental stages, though their molecular mechanisms at the proteomic level remain uncharacterized. This study employed Data-Independent Acquisition (DIA) quantitative proteomics to systematically analyze protein profiles in sika deer antler velvet, antlers, and deer antler base. Comparative analysis revealed 3154 differentially expressed proteins (DEPs, 95% upregulated) between antler velvet and antlers, which were significantly enriched in Ribosome Biogenesis (e.g., Polyadenylate-binding protein), oxidative phosphorylation, and neurodegenerative disease pathways. In the comparison of deer antler base versus antlers, 1024 DEPs (92% upregulated) were identified, primarily involved in proteolysis (e.g., ACTC protein), glycolysis, and complement and coagulation cascades. Between deer antler base and antler velvet, 2749 DEPs (87% downregulated) were enriched in Thioredoxin domains, cytoskeleton regulation, and RNA-binding functions. Subcellular localization demonstrated antler velvet proteins predominantly distributed in the cytoplasm (37.6%) and nucleus (19.6%), while deer antler base proteins showed marked enrichment in extracellular regions (19.7%) and cytoskeletal components. As the first comprehensive proteomic characterization of these materials, this study identifies ribosomal proteins and complement pathway-related proteins as key biomarkers, thus establishing a scientific foundation for precise authentication, quality control, and efficacy–mechanism interpretation of deer antler-derived medicines. It further highlights antler velvet’s neuroprotective potential and deer antler base’s immunomodulatory applications.
