Maltose gradient-induced biosensor-based high-throughput screening for directed evolution of maltogenic amylase from Bacillus stearothermophilus

Jiayuan Wang,Laichuang Han,Maofang Teng,Qinghua Li,Jingwen Zhou,Jianghua Li,Guocheng Du,Guoqiang Zhang

Maltogenic amylase is a starch-hydrolyzing enzyme commonly used in bread baking and high-concentration maltose syrup production. However, low catalytic activity limits its industrial application. Improving catalytic activity based on molecular modification and directed evolution requires a High-Throughput Screening (HTS) method. In this study, a maltose gradient-induced (MaGI) biosensor was designed and applied for the directed evolution of maltogenic amylase AmyM, showing a good positive correlation between enzyme activity and fluorescence. The MaGI biosensor detected maltose and maltogenic amylase activity efficiently and specifically. Two mutants, Q440N and S442N/Q661L, were identified through the screening of 3000 mutants using the MaGI biosensor, showing a significant increase in catalytic activity of 35.56 % and 24.51 %, respectively, compared to the wild-type. Meanwhile, the t 1/2 of Q440N and S442N/Q661L at 60 °C increased by 58.53 % and 66.66 %, respectively. In industrial applications, the enhancement of catalytic activity and stability is conducive to improving production efficiency and reducing costs. MD simulation has found that when modifying multidomain enzymes, distal mutations can enhance catalytic activity. In conclusion, the developed MaGI biosensor is a promising tool for high-throughput and specific detection of maltose.

Comparison of three enthalpy relaxation models based on fictive temperature and nonlinear Adam–Gibbs formulation

Wang Xuan,Xiao Kesong,Liu Xiangnong,Wu Hao,Gao Cai

The phenomenological models based on fictive temperature and nonlinear Adam–Gibbs formulation have been widely used to describe the enthalpy relaxation kinetics. Although the parameters in Adam–Gibbs formulation are physically explicable, the reasonabilities of the parameters obtained from curve-fitting procedures have not been systematically discussed yet. In this work, differential scanning calorimetry was employed to measure the heat capacities of maltitol in both the amorphous and the crystalline states, and the data were used to generate the model parameters. Results showed that the configurational ground state plays an important role in extracting the model parameters. Selecting the crystal rather than the glass as the ground state seems to be effective to improve the reasonabilities of the fitted parameters. It suggested that even if the fit quality seems sound, the physical reasonability of the best-fit parameters still needs to be treated with caution.

Dual-functional TFNC polyester membranes utilizing maltitol for dye/salt separation and desalination

Tonghui Zhang,Yumeng Wang,Bo Ge,Shuangshuang Wang,Yanhu Zhan,Xuefen Wang

To achieve a promising application prospect in nanofiltration (NF) membrane separation , a strategic design tailored to particular functions is crucial. Here, tailor-made thin film nanofibrous composite (TFNC) NF membranes for specific applications were fabricated via interfacial polymerization (IP) between maltitol and trimesoyl chloride (TMC) on PAN nanofibrous substrates. Owing to the moderate reactivity of maltitol, a range of sugar alcohol based polyester membranes with different morphologies and cross-linking degrees were obtained by varying the monomer concentration as well as the reaction time. In this case, the finely tuned loose Mal1-TMC0.2-2 membrane exhibited a prominent permeability (110.34 L m −2  h −1  bar −1 ) and an extraordinary selectivity for dye/salt mixed solution, with the rejection of Congo Red up to 99.11 % and NaCl retention of only 7.41 %, indicating its suitability for textile wastewater treatment . Further, the dense Mal2-TMC0.4-10 membrane showed an excellent Na 2 SO 4 rejection of 97.40 % with a good permeability of 18.65 L m −2  h −1  bar −1 , making it a candidate for seawater and brackish water desalination . In addition, both membranes demonstrated reliable chlorine and pressure resistance along with long-term stability. This study provided new insights and inspiration to the design and preparation of NF membranes for specialized applications.

Maltose hydrogenation to maltitol over industrial Raney Ni catalyst: Kinetics and mechanism

Haoan Fan,Bolong Li,Xuezhi Zhao,Zhecheng Fang,Chao Chen,Wenhua Zhou,Wulong Yang,Mian Li,Xiuyang Lu,Jie Fu

The kinetics and mechanisms of selective maltose hydrogenation over Raney Ni remain poorly understood, which hinders the industrial advancement of maltitol production for biorefining. Herein, the impact of hydrogenation conditions and in-depth mechanistic characterizations were carefully investigated in this research. Optimization of reaction conditions illustrated that maltitol formation was favored by moderate reaction temperatures (≤130°C), high hydrogen pressures (>7 MPa), and near-neutral sodium phosphate buffer (pH = 6.47). Kinetic studies demonstrated the E a of 45.5 kJ/mol for the hydrogenation of maltose. The in situ IR revealed that the hydrogenation of maltose over Raney Ni occurs in the order of ring opening and adsorption via CO of the aldose form followed by hydrogenation. The UV–vis spectra demonstrated the near-neutral setting (pH = 6.47) promoted the production of open-chain aldose conformation and selective CO hydrogenation to the corresponding alcohols. The present study on concentrated-maltose hydrogenation paves the way for the industrial development of maltitol manufacturing.

Synergistic effect of Ca/Zn main stabilizer and maltitol auxiliary stabilizer on the thermal stability, mechanical properties, and transparency of poly(vinyl chloride)

Xuhui Wang,Guojie Zhang,Huanhuan Dong,Yong Zhang

Maltitol, a natural compound, offers a potentially eco-friendly alternative to conventional auxiliary thermal stabilizers. Its auxiliary stabilization effect was studied by discoloration tests, Congo red tests, thermogravimetric analysis, and UV–visible spectroscopy. Additionally, x-ray photoelectron spectroscopy and discoloration tests were used to study the stabilizing mechanisms of maltitol. Compared to dipentaerythritol, commonly used in the PVC industry, maltitol as an auxiliary stabilizer exhibits better improvement in thermal stability. This improvement is related to its melting point, ability to complex with zinc chloride, and content of hydroxyl groups. Furthermore, the addition of maltitol in PVC not only maintains good transparency of PVC basically unchanged but also improves the mechanical properties. Maltitol, as a natural, eco-friendly, and efficient auxiliary thermal stabilizer, exhibits a promising application prospect in PVC materials.

Microneedles fabricated from alginate and maltose for transdermal delivery of insulin on diabetic rats

Yang Zhang,Guohua Jiang,Weijiang Yu,Depeng Liu,Bin Xu

To reduce the inconvenient and painful of subcutaneous needle injection, the calcium ion cross-linked alginate/maltose (Ca 2   + /Alg-Mal) composite microneedles have been fabricated by a template method. The as-prepared microneedles exhibited strong mechanical properties with the highest failure force around 0.41   N/needle. The biological activity and stability of loaded insulin in microneedles were investigated. Due to the good mechanical properties and excellent biocompatibility, the as-prepared microneedles have been applied for transdermal delivery of insulin on diabetic Sprague-Dawley (SD) rat models in vivo . After transdermal administration to the diabetic rats, the released insulin from biodegradable composite microneedles exhibit an obvious and effective hypoglycemic effect with relative pharmacological availability (RPA) and relative bioavailability (RBA) at 94.1   ±   5.6% and 93.7   ±   4.7% compared with that of subcutaneous injection route. This work suggests that as-prepared Ca 2   + /Alg-Mal microneedles can be used to encapsulate insulin and have a potential application in diabetes treatment via transdermal ingestion.

Research on the Preparation and Performance of Binary Modified Maltodextrin Water-Reducing Agent

Jingzhi Wu,Min Qiao,Zhifeng Lyu,Qianping Ran

Using maltodextrin (MD) of different dextrose equivalent (DE) values, 1,3-propanesultone, and maleic anhydride as raw materials, a novel binary modified maltodextrin (BMMD) was synthesized and further applied as a water-reducing agent. Its structure was characterized by Fourier transform infrared (FT-IR) and UV. The rheological behavior of the sample solution and strengths for concrete were also determined and the adsorption was tested by TOC. The influence of the process parameters to degree of substitution (DS) and the dosage on the fluidity of cement paste were investigated. The results show that the optimal conditions of sulfonation were the MD of DE 15, (sulfonated agent)/ (MD) of 1.4, the catalyst amount of 1% by mass MD, and the reaction time of 12 h; the optimal conditions of esterification were (esterified agent)/ (SMD) of 0.6, the reaction temperature of 90°C, and the reaction time of 4 h. The optimal dosages of sulfonated maltodextrin (SMD) and BMMD were 0.475% and 0.45%, respectively. In this dosage, the main dispersion capacity of BMMD attributes to two kinds of anchoring groups () and the appropriate molecular weight of MD.

Effects of maltose on stability and rheological properties of orange oil-in-water emulsion formed by OSA modified starch

Chao Li,Xiong Fu,Faxing Luo,Qiang Huang

Effects of maltose on the stability and rheological properties of orange oil-in-water (O/W) emulsion formed by octenyl succinic anhydride modified waxy corn starch (OS-starch) were investigated. Emulsion samples were prepared using a jet homogenizer. Creaming behavior, droplet size distribution and rheological properties of emulsions during storage were studied. Results showed that the emulsion with no maltose was unstable over a quiescent storage period of one month, while the emulsions containing maltose appeared relatively stable. The OS-starch played a critical role in stabilizing emulsion by adsorbing at oil–water interface. Maltose assisted the emulsion against creaming, flocculation , and coalescence. Rheological results showed that the viscoelastic properties of the emulsions increased with increasing maltose content. Maltose increased the viscosity of emulsion, and enhanced the elastic structure of emulsion through interaction with OS-starch and water. Increase of viscoelasticity of the emulsion system retarded oil droplets free motion and strengthened the mutual acting force between droplets, thereby delaying creaming, flocculation, and coalescence of oil droplets.

Effects of maltose and lysine treatment on coffee aroma by flash gas chromatography electronic nose and gas chromatography–mass spectrometry

Yuqin He,Haide Zhang,Nana Wen,Rongsuo Hu,Guiping Wu,Ying Zeng,Xiong Li,Xiaodan Miao

BACKGROUND Arabica coffee is a sub-tropical agricultural product in China. Coffee undergoes a series of thermal reactions to form abundant volatile profiles after roasting, so it loses a lot of reducing sugars and amino acids. Adding carbonyl compounds with amino acids before roasting could ensure the nutrition and flavour of coffee. The technology is versatile for the development of coffee roasting process. This investigation evaluates the effects of combining maltose and lysine (Lys) to modify coffee aroma and the possibly related mechanisms. Arabica coffee was pretreated with a series of solvent ratios of maltose and Lys with an identical concentration (0.25 mol L −1 ) before microwave heating. RESULTS It was found that the combination of maltose and Lys significantly ( P ≤ 0.05) influenced quality indices of coffee (pH and browning degree). Ninety-six aromatic volatiles have been isolated and identified. Twelve volatile profiles revealed the relationship between fragrance difference and compound content in coffee. Moreover, coffee aroma was modified by a large number of volatiles with different chemical classes and character. CONCLUSION Thus, our results suggest that the combination of reagents changed overall aroma quality through a series of complex thermal reactions, especially the ratio of Lys/maltose over 2:1. © 2017 Society of Chemical Industry

Experimental and Theoretical Investigations on the Supermolecular Structure of Isoliquiritigenin and 6-O-α-D-Maltosyl-β-cyclodextrin Inclusion Complex

Bin Li,Benguo Liu,Jiaqi Li,Huizhi Xiao,Junyi Wang,Guizhao Liang

Isoliquiritigenin (ILTG) possesses many pharmacological properties. However, its poor solubility and stability in water hinders its wide applications. The solubility of bioactive compounds can often be enhanced through preparation and delivery of various cyclodextrin (CD) inclusion complexes. The 6-O-α-d-maltosyl-β-CD (G2-β-CD), as one of the newest developments of CDs, has high aqueous solubility and low toxicity, especially stable inclusion characteristics with bioactive compounds. In this work, we for the first time construct and characterize the supermolecular structure of ILTG/G2-β-CD by scanning electron microscopy (SEM), ultraviolet-visible spectroscopy (UV), Fourier transform infrared spectroscopy (FT-IR), and X-ray diffractometry (XRD). The solubility of ILTG in water at 25 °C rises from 0.003 to 0.717 mg/mL by the encapsulation with G2-β-CD. Our experimental observations on the presence of the ILTG/G2-β-CD inclusion complex are further supported by the ONIOM(our Own N-layer Integrated Orbital molecular Mechanics)-based QM/MM (Quantum Mechanics/Molecular Mechanics) calculations, typically substantiating these supermolecular characteristics, such as detailed structural assignments, preferred binding orientations, selectivity, solvent effects, interaction energies and forces of the ILTG/G2-β-CD inclusion complex. Our results have elucidated how ILTG interacts with G2-β-CD, demonstrating the primary host-guest interactions between ILTG and G2-β-CD, characterized by hydrogen bonds, hydrophobic interactions, electrostatic forces, and conformational effects, are favored for the formation of the ILTG/G2-β-CD inclusion.Keywords:isoliquiritigenin;6-O-α-d-Maltosyl-β-CD;inclusion complex;ONIOM calculation

Directional Mining and Thermostability Modification of Novel Acid-Resistant Maltogenic Amylase from Lactobacillus paralimentarius

Qian Hou,Zongxiao Huang,Huicong Chen,Weining Huang,Ning Li,Xiaoqing Mu

Due to the low pH and high temperature in dough rising and baking processes of starchy foods, high thermostability and acidic resistance have become the most important characteristics for commercial maltogenic amylase as a baking antiaging additive. Therefore, a novel maltogenic amylase from Lactobacillus paralimentarius, exhibiting an activity of 274.79 U·mg–1 under optimal conditions at 50 °C and pH 5.0, was screened through EnzymeMiner-assisted preliminary screening, microbial sources-assisted reselection, and structure analysis-assisted final selection, which was based on the bioinformatics. Data-driven rational design was carried out to construct a mutation library for the improvement of thermostability; the optimal reaction temperature and activity of mutant A375P/S257P were increased to 65 °C and 430.15 U·mg–1, respectively, while the half-life at 60 °C increased by 7.09-fold. The structural analysis indicated that the formation of new hydrogen bonds and the increase in rigidity reduce the free energy of mutant A375P/S257P, thereby further enhancing its thermostability.
