First determination of extracellular paralytic shellfish poisoning toxins in the culture medium of toxigenic dinoflagellates by HILIC–HRMS

Lijun Liu,Junhui Chen,Xiuping He,Shuang Hao,Ziru Lian,Baodong Wang

Paralytic shellfish poisoning (PSP) toxins have received considerable attention in recent years because of their adverse effects on marine breeding industries and human health. In this study, a reliable method for the analysis of extracellular PSP toxins in the culture medium of marine toxic dinoflagellates was developed for the first time using graphitized carbon black–solid-phase extraction and hydrophilic interaction liquid chromatography–high-resolution mass spectrometry. The limit of quantification of typical PSP toxins in algal culture medium ranged from 0.072 μg/L to 0.151 μg/L under optimal conditions. Satisfactory absolute recoveries (87.5%–102.4%), precision (relative standard deviation ≤ 7.6%), and linearity ( R 2  ≥ 0.9998) were also achieved. In addition, the proposed method was applied to screen and determine the extracellular PSP toxins of two typical toxigenic dinoflagellates, Alexandrium minutum and Alexandrium tamarense . The total concentrations of the extracellular PSP toxins in A. minutum and A. tamarense over the whole growth period were within 2.0–735.5 and 2.0–19.2 μg/L, respectively. The concentrations of extracellular PSP toxins varied remarkably in the different growth stages of A. minutum and A. tamarense , and the contents of some extracellular PSP toxins were substantially higher than those of intracellular PSP toxins. Therefore, the extracellular PSP toxins released by toxigenic red tide algae cannot be ignored, and their environmental fate , bioavailability , and potential harm to aquatic environment need to be investigated in future studies.

Determination of Paralytic Shellfish Toxins in Bivalve Mollusks by Amino-Modified Multiwalled Carbon Nanotube (MWCNT) Solid-Phase Extraction (SPE) and High-Performance Liquid Chromatography–Tandem Mass Spectrometry (HPLC–MS/MS)

Xu-Feng Wang,Qiang Wang,Jin-Lan Yang,Dong-Hao Zhao

An amino-modified multiwalled carbon nanotube (MWCNT) solid-phase extraction-based hydrophilic interaction liquid chromatography–tandem mass spectrometry (HILIC–MS/MS) method was developed for simultaneous determination of 13 paralytic shellfish toxins (PSTs) in bivalve mollusks. Shellfish samples were extracted with 1% acetic acid followed by cleanup using optimized MWCNT-NH2 based solid-phase extraction. Absorbed PSTs were eluted with 2 mL 1% acetic acid–acetonitrile (70/30, v/v). The separation was performed on a Waters BEH Amide column within 7.5 min utilizing 0.1% formic acid–acetonitrile and 2 mM ammonium acetate containing 0.1% formic acid as the mobile phase. The analytes were quantified by matrix-matched external standard curves with correlation coefficients® exceeding 0.997 acquired in multiple reaction-monitoring mode. The limits of detection and quantitation in shellfish samples were from 2.76 to 9.8 μg/kg and 9.11 to 39.3 μg/kg, respectively. Recoveries at three spiked levels ranged from 71.4% to 97.8% with relative standard deviations less than 12.6%. The developed method was satisfactorily applied to the analysis of shellfish.

Magnetic Amino-Modified Multiwalled Carbon Nanotube (MWCNT) Based Magnetic Dispersive Solid-Phase Extraction (m-dSPE) for the Determination of Paralytic Shellfish Toxins in Bivalve Mollusks with Hydrophilic Interaction Liquid Chromatography–Tandem Mass Spectrometry (HILIC-MS/MS)

Xu-Feng Wang,Qiang Wang,Ying-Xia Zhang,Jin-Lan Yang,Dong-Hao Zhao

A sensitive and efficient magnetic amino modified multi-walled carbon nanotubes (m-MWCNT-NH2)-based magnetic dispersive solid-phase extraction (m-dSPE) coupled with hydrophilic interaction liquid chromatography–tandem mass spectrometry (HILIC-MS/MS) strategy is reported for the simultaneous determination of 13 paralytic shellfish toxins (PSTs) in bivalve mollusks. The samples were extracted twice with 1% acetic acid and purified using optimized m-dSPE procedures with Fe3O4 coated MWCNT-NH2 composites as magnetic adsorbents. The adsorbed PSTs on the m-MWCNT-NH2 adsorbents were separated using a Ni-coated neodymium magnet and subsequently eluted with 2 mL water–acetonitrile–acetic acid (80:20:1, v/v/v) and analyzed by HILIC-MS/MS. The correlation coefficients (r) of the targeted toxins obtained in matrix-matched external standard curves were from 0.997 to 0.999. The PSTs were acquired and quantified in the multiple reaction-monitoring mode and the limits of detection and quantitation in bivalve mollusks were 1.10 to 4.51 μg/kg and 3.67 to 13.5 μg/kg, respectively. Spiked recoveries at three concentrations in negative samples were from 73.4% to 92.5% with precision less than 10.6%. The method validation was in accordance with 2002/657/EC guidelines and method application in commercial bivalve mollusk samples. The developed method was rapid, convenient and economical for the determination of the targeted toxins.

Development of magnetic fluorescence aptasensor for sensitive detection of saxitoxin based on Fe3O4@Au-Pt nanozymes

Lin Zhu,Wei Zeng,Yueqing Li,Yu Han,Jing Wei,Long Wu

In this work, on the basis of Fe 3 O 4 @Au-Pt nanozymes (MAP NZs) and aptamer recognition, a magnetic fluorescent aptasensor (MFA) was developed for sensitive and accurate detection of saxitoxin (STX). With the bridge of STX aptamer (Apt STX ) and complementary DNA (cDNA), Apt STX decorated MAP NZs (MAP/Apt) and cDNA modified green quantum dots (cDNA@g-QDs) were connected to form MAP/Apt-cDNA@g-QDs complex. As STX behaves a strong binding ability towards Apt STX , it will compete with cDNA and hybridize with Apt to release cDNA@g-QDs. With the addition of TMB, MAP will catalyze TMB to the oxidized TMB (ox-TMB), thereby quenching the fluorescence of g-QDs due to the inner filter effect. Based on this finding, the quantitative relationship between the change in fluorescence of gQDs and STX concentration was explored with a limit of detection (LOD, S/N = 3) of 0.6 nM. An internal standard signal of oxTMB was adopted and reduced the fluctuation of fluorescence signal output. Besides, the fluorescence probe can selectively recognize and detect STX among five marine toxins. Eventually, the MFA method behaved good performance in detecting seafood samples with recoveries of 82.0 % ∼ 102.6 % as well as coefficient of variations (CV) of 7.2 % ∼ 10.3 %. Therefore, the method with internal signal is hopeful to be a potential candidate for sensitive and accurate detection of STX in seafood.

Ethanol Alleviates Amyloid-β-Induced Toxicity in an Alzheimer’s Disease Model of Caenorhabiditis elegans

Bai Shuju,Wang Wenbo,Zhang Zhiwei,Li Mengyao,Chen Zehan,Wang Jiuqiao,Zhao Yanlin,An Lu,Wang Yuxiang,Xing Shu,Fu Xueqi,Ma Junfeng

Amyloid-β, a hallmark of Alzheimer’s disease, forms toxic intracellular oligomers and extracellular senile plaques resulting in neuronal toxicity. Ethanol is widely consumed worldwide. Moderate ethanol consumption has numerous benefits in humans. We found that ethanol could significantly extend the lifespan of Caenorhabiditis elegans in a previous study. Based on that study, we tested the effect of ethanol on Alzheimer’s disease transgenic Caenorhabiditis elegans strain CL4176, which expresses amyloid-β1-42 peptide in body wall muscle cells. Ethanol delayed paralysis and reduced amyloid-β oligomers in Caenorhabiditis elegans worms of the CL4176 strain. Moreover, ethanol could induce the nuclear translocation of DAF-16 in the nematodes. However, in worms that were fed daf-16 RNAi bacteria, ethanol no longer delayed the paralysis. The qPCR assays showed that ethanol increases the expression of daf-16, hsf-1 and their common target genes- small heat shock protein genes. In addition, we also found that ethanol could increase lysosome mass in the CL4176 worms. In summary, our study indicated that ethanol attenuated amyloid-β toxicity in the Alzheimer’s disease model of Caenorhabiditis elegans via increasing the level of lysosomes to promote amyloid-β degradation and upregulating the levels of small heat shock protein genes to reduce amyloid-β aggregation.

Rising CO2 will increase toxicity of marine dinoflagellate Alexandrium minutum

Ziru Lian,Fang Li,Xiuping He,Junhui Chen,Ren-Cheng Yu

Ocean acidification caused by increasing emission of carbon dioxide (CO 2 ) is expected to have profound impacts on marine ecological processes, including the formation and evolution of harmful algal blooms (HABs). We designed a set of experiments in the laboratory to examine the effects of increasing CO 2 on the growth and toxicity of a toxic dinoflagellate Alexandrium minutum producing paralytic shellfish toxins (PSTs). It was found that high levels of CO 2 (800 and 1200 ppm) significantly promoted the growth of A. minutum compared to the group (400 ppm) representing the current CO 2 level. The total yields of PSTs by A. minutum , including both intracellular and extracellular toxins, were significantly enhanced, probably due to the induction of core enzyme activity and key amino acids synthesis for PST production. More interestingly, high level of CO 2 promoted the transformation from gonyautoxin2&3 to gonyautoxin1&4 and depressed the release of PSTs from inside to outside of the cells. All these processes collectively led to an apparent increase of A. minutum toxicity. Our study demonstrated that rising CO 2 would increase the risk of toxic A. minutum based on the comprehensive analyses of different processes including algal growth and toxin synthesis, transformation and release.

Detection, occurrence, influencing factors and environmental risks of paralytic shellfish toxins in seawater in a typical mariculture bay

Lei Pan,Xiuping He,Junhui Chen,Jinhui Jeanne Huang,Yuning Wang,Shengkang Liang,Baodong Wang

Paralytic shellfish toxins (PSTs) producing algae are widely distributed in the global coastal aquatic environment, posing a threat to coastal ecosystem health and mariculture safety. However, the levels and potential environmental risks of PSTs frequently detected in shellfish remain largely unexplored in seawater of mariculture zones. In this study, a new method for trace detection of 13 common PSTs (<1.0 ng/L) in seawater was established based on off-line solid phase extraction (SPE) and on-line SPE-liquid chromatography-tandem mass spectrometry (on-line SPE-LC-MS/MS), and a systematic investigation of PSTs in seawater of the Laizhou Bay, a typical aquaculture bay in China, was conducted to understand their pollution status, environmental impact factors and ecological risks for the first time. Eleven PSTs were detected in the seawater of Laizhou Bay with total concentrations ranging from 0.75 to 349.47 ng/L (mean, 176.27 ng/L), which indicates the rich diversity of PSTs in the mariculture bay and demonstrates the reliability of the proposed analytical method. C1, C2, GTX2, GTX3, dc GTX2, and dc GTX3 were found to be the predominant PSTs, which refreshed the knowledge of PST contamination in the coastal aquatic environment. PST levels in seawater exhibited the highest levels in the southeastern mouth of Laizhou Bay and decreased toward the inner bay. Correlation analyses showed that climatic factors , nutrient status and hydrological conditions had significant effects on the distribution of PST in mariculture bay. Preliminary environmental risk assessments revealed that aquatic organisms throughout the waters of Laizhou Bay are at risk of chronic PST toxicity. These findings imply that the risk of PST in seawater of mariculture bay has previously been grossly underestimated, and that the coastal aquatic environment in North China and even the world may be at more serious risk of PST pollution, which should be taken seriously.

Elevated pCO2 may increase the edible safety risk of clams exposed to toxic Alexandrium spp.

Min Pang,Yuchen Gong,Hongju Chen,Ying Shi,Zhao Li,Xiuping He,Junhui Chen,Xuexi Tang,Zongling Wang,Xuelei Zhang,Pei Qu

Toxic harmful algal blooms (HABs) have received increasing attention owing to their threat to the health of aquatic life and seafood consumers. This study evaluated the impacts of elevated atmospheric partial pressure of CO 2 ( p CO 2 ) on the production of paralytic shellfish toxins (PSTs) in different Alexandrium spp. strains, together with its further effects on the bioaccumulation/elimination dynamics of PSTs in bivalves contaminated with PSTs from toxic dinoflagellates. Our results showed that elevated p CO 2 stimulated the growth of the two Alexandrium spp. ( A. catenella and A. pacificum ) isolated from the northern and southern coastal areas of China, respectively, and affected PST production including content and toxicity of the two strains differently. Further PSTs bioaccumulation/elimination in PSTs-contaminated Manila clam, Ruditapes philippinarum under high p CO 2 also occurred. It is worth noting the biotransformation of neosaxitoxin (NEO) with high toxicity through trophic transfer with effect of elevated p CO 2 . When in microalgae cultured under the control (410 ppm) and elevated p CO 2 conditions (495 and 850 ppm), the proportion of NEO in the PST content produced by A. catenella was reduced from 11.1 to 6.4 and 2.6 %, while the proportion of NEO in A. pacificum was increased from 3.1 to 3.6 and 4.7 %, respectively. NEO accounted for >50 % of total PST contents in clams, which were biotransformed via transfer from dinoflagellates and higher p CO 2 enhanced this biotransformation leading to increased NEO accumulation. The negatively affected elimination of PSTs, especially NEO, in clams fed with A. catenella or A. pacificum , indicates that the detoxification of PSTs-contaminated clams may be more difficult under elevated p CO 2 . This study provides reference for developing models to assess the safety of bivalves under the co-stress of environmental change and toxic HABs, suggesting that ocean acidification may lead to the higher safety risk of Manila clams exposed to toxic HAB dinoflagellates.

Portable Smartphone-based Colorimetric Analyzer with Enhanced Gold Nanoparticles for On-site Tests of Seafood Safety

Longjie ZHONG,Jiadi SUN,Ying GAN,Shuqi ZHOU,Zijian WAN,Quchao ZOU,Kaiqi SU,Ping WANG

Saxitoxin (STX) is one of the paralytic shellfish poisons (PSP) that endanger people’s health. It is necessary to develop methods for the on-site rapid detection for STX in order to prevent safety accidents. An enzyme-linked immunosorbent assay (ELISA) is timesaving and effective, but it is not suitable for large-scale in-field tests due to the expensiveness of commercial ELISA kits and the bulkiness of a microtiter plate reader (MTPR). In this study, a portable smartphone-based colorimetric analyzer (SBCA) with a cost-effictive enhanced gold nanoparticle-based ELISA (EGNB-ELISA) was proposed for STX detection. In a bicinchoninic acid (BCA) protein assay (R2 = 0.9939) and a glucose assay (R2 = 0.9937), SBCA was shown to be in good agreement with MTPR. EGNB-ELISA had a 12.5-fold lower detection limit (0.4 ng/mL) and a lower detection range (1 – 50 ng/mL, Y = 0.4037X + 0.3564, R2 = 0.9797) than the classical ELISA. The recovery rate ranged over 89.1 – 112.2%. The whole detection system, combining both homemade SBCA and ENGB-ELISA, is expected to satisfy the needs of on-site STX sample tests to guarantee seafood safety.
