Anesthetics are widely applied in cultivation and transportation of aquatic products to reduce the metabolism of fish and avoid some stress reactions. The overdose of using anesthetics or the occurrence of their residues in the aquatic products may negatively affect human health after consumption. A rapid screening procedure and a new analytical method for the simultaneous determination of 14 anesthetics was established in this study. The compounds were extracted from fish and shrimp matrices by sodium acetate buffer solution, cleaned up via solid-phase extraction, and then were analyzed using an ultrahigh-performance liquid chromatography system coupled with a tandem quadrupole-time-of-flight mass spectrometer (UPLC-Q-TOF–MS) in a positive-ion mode. Methodological verification analyses, including linearity, precision, recovery, limit of detection (LOD), and limit of quantification (LOQ), were conducted to validate the method. The correlation coefficient of the calibration curves of each anesthetic exceeded 0.99. The average recoveries of this analysis method varied from 72.36 to 111.27% with relative standard deviations less than 5%. The detection limit of anesthetics reached 0.3–0.7 μg/kg. Agilent Mass Hunter Personal Compound Database Library (PCDL) Manager was used to establish a database of 14 anesthetics and 8 isotope internal standards, which enabled a quick screening and analysis without standard substances. The results of the spiked blank samples demonstrated reliability of the screening method with mass deviations less than 5 ppm and the scores determined by relative molecular mass, retention time, isotope ratio, and distribution all greater than 85. In addition, according to the fragmentation information of mass spectrometry, the fragmentation rules of some anesthetics were summarized. This method was validated with reliable results in quantitative and qualitative analysis and could be used as regulatory measures for anesthetics in aquatic products.

Anaesthetics are widely used in aquaculture as a method of stress relief. However, few studies have reported the influence of anaesthetics on the immune system of fish. Therefore, in the present study, we selected two different anaesthetics, MS-222 (100 mg/L) and eugenol (20 mg/L), to anaesthetize crucian carp and analyse the transcriptome. It was found that over 137 million high-quality reads were generated and de novo assembled into a final set of 97,556 unigenes. A total of eight differentially expressed genes (DEGs) related to the immune response were co-expressed in the two anaesthetic groups. GO and the KEGG revealed that these genes functioned primarily to enrich antigen processing and presentation pathways, including MHCI , MHCIα , p-MHCIIα , P-MHCIIα , CD74 , MHCII antigen and MHCII antigenα . Moreover, LRRFIP2 was found be associated with the immune response. Consistent with the transcriptome findings, qPCR verified the changes in the relative level of expression of these genes. Furthermore, long-term exposure to low concentrations of MS-222 (30 mg/L) and eugenol (8 mg/L) showed an impact on these immune genes. In conclusion, anaesthetics used in high or low concentrations for treatment and transport could affect the immune system in fish species. While eugenol was associated with an earlier activation of immune gene expression, MS-222 exhibited more significant effects on the immune response. These findings improve the understanding of the mechanisms of anaesthetics on immune damage and will be of great value for future studies involving anaesthetic selection and treatment for fish.

Liquid chromatography coupled with quadrupole linear ion trap tandem mass spectrometry (LC-QLIT-MS/MS) technology operated in electrospray ionization (ESI) was developed for tracing anesthetic (AETs) and sedatives (SDTs) in fish. Sampling procedure was achieved by using acetonitrile extraction followed by dispersive solid phase extraction (DSPE) clean-up. Under the optimized laboratory conditions, reliable qualitative confirmation was obtained through the multiple reaction monitoring-information dependent acquisition-enhanced product ion (MRM-IDA- EPI ) mode. Results indicated a favorable linear in the concentration range of 1–100 μg∙kg −1 (0.1–10 μg∙kg −1 for MS-222), with regression coefficient not less than 0.9997. The detection limit ranges from 0.03 to 0.4 μg∙kg −1 (S/N = 3). The validated method was applied to determine AETs and SDTs in fish with satidfied recoveries of 86.3 %–111.7 % and the relative standard deviations (RSD) of 1.9 %–8.9 % (n = 6). Practical samples analysis indicated that the proposed method is simple, rapid, sensitive and accurate for identification of AETs and SDTs.

Herein, uniform magnetic covalent organic framework (MCOF) microflowers were synthesized by facile inlaying aldehyde-functionalized Fe 3 O 4 nanoparticles onto the COF under mechanical stirring at room temperature. The MCOF microflowers kept flower-shaped morphology with high crystallinity and magnetism. In addition, the good water dispersibility and reusability made the MCOF microflowers to be good magnetic solid phase extraction (MSPE) adsorbents for the high-efficiency enrichment of eugenol anesthetics from aquatic products. The MCOF microflowers showed fast adsorption rate (10 min) and high extraction efficiency (above 84.02%) for eugenol anesthetics under optimal conditions. The MCOF microflowers displayed maximum adsorption capacities of 6.67, 12.31, and 16.78 µg g −1 for eugenol, eugenol acetate, and methyl isoeugenol, respectively. The MCOF microflowers based MSPE coupled with high-performance liquid chromatography method was proposed for enrichment and analysis of eugenol anesthetics in tilapia, shrimp and crab samples. The method showed a wide linear of 5.0–500.0 µg kg −1 and low detection limits of 2.4–3.6 μg kg −1 for eugenol anesthetics. The recoveries of spiked eugenol anesthetics ranged from 80.0% to 118.8% with relative standard deviations no>9.2%. Results indicate the high potential of MCOF microflowers to be an adsorbent for efficient extraction and analysis of eugenol anesthetics from complex aquatic product samples.

A highly selective and sensitive method of reversed phase high-performance liquid chromatography (RP-HPLC) coupled with resonance Rayleigh scattering (RRS) was developed for the determination of procaine, bupivacaine and tetracaine. Separation of three local anaesthetics was achieved at 35 °C on a C 18 column. The mobile phase was 30: 70 (v/v) acetonitrile/triethylamine–phosphoric acid buffer (pH 2.9) at flow rate of 0.3 mL/min. The RRS detection was conducted by taking advantage of the strong RRS enhancement of the local anaesthetics with erythrosine reaction in an acidic medium. Under optimum conditions, the limit of detection (S/N = 3) values were in the range of 2.4–11.2 ng/mL. Recoveries from spiked human urine samples were 95.8%–104.5%. The proposed method applied to the determination of local anaesthetics in human urine achieved satisfactory results. In addition, the mechanism of the reaction is fully discussed. The concentration of five rapidly metabolized anesthetics in living tilapias was determined in this study, by the presented method coupling in vivo solid phase microextraction (SPME) to gas chromatography-mass spectrometry (GC-MS), which was the first time that in vivo sampling method was adapted in detecting the anesthetic residue in the living aquatic product. The analytical performance of the developed method was evaluated in homogenized tilapia dorsal muscle, and the results demonstrated that the present method possessed low detection limits 1.7–9.4 ng g −1 ), wide linear ranges (10 or 30–5000 ng g −1 ), and satisfactory reproducibility (relative standard deviations no more than 8.1% and 10.8% for inter-fiber and intra-fiber assays, respectively). Standard curves were established in homogenized tilapia dorsal muscle for calibrating in vivo SPME in living tilapias. And the concentrations determined by in vivo SPME were close to those determined by the liquid extraction. By using the present method, one anesthetic residue was detected above the detection limit in tilapias from the local markets. Comparing to traditional methods, the present one exhibited superior time-efficiency and cost performance, as the extraction time was only ten minutes, which was short to successfully avoid the possible loss of analytes caused by elimination and sample storage. In addition, owing to the time-efficiency of the present method, the elimination of the anesthetics in tilapias was traced successfully in the laboratory.

Local anesthetics have been widely used in clinical analgesia due to their ability to provide effective regional pain management. Accurate measurement of local anesthetics in body fluids is crucial for ensuring patient medication safety and optimizing therapeutic efficacy. Herein, we present a convenient, economical, sensitive, and efficient TLC-SERS method for multiplex determination of six kinds of anesthetics (pro) in human plasma, including procaine hydrochloride (Pro), tetracaine hydrochloride (Tet), dibucaine (Dib), mepivacaine hydrochloride (Mep), lidocaine hydrochloride (Lid), and ropivacaine hydrochloride (Rop). The TLC method was adopted to separate six local anesthetics effectively. In order to improve the sensitivity, TLC spots were concentrated into smaller ones using methanol through solvent-driven enrichment, then Ag NPs staining was applied to enriched spots for a strong and unique SERS response of each anesthetic. As a result, linear calibration curves of SERS intensity ratio versus negative logarithm of spotting amounts sampled on TLC plates were obtained, along with the lowest detectable amounts in this study were 1 ng (Pro), 10 pg (Tet), 10 ng (Dib), 50 ng (Mep), 50 ng (Lid), and 0.1 μg (Rop), which were up to 2 × 104times more sensitive than our previous TLC-Raman method. Moreover, the method was successfully applied to human plasma samples, demonstrating the feasibility and potential for multiplex analysis of local anesthetics in clinical practice, criminal forensics, and aquaculture.

Graphical A ready-to-use collagen triboelectric nanogenerator (TENG) wound dressing system has been developed. The electric field generated by the TENG promotes the release of the analgesic drug from the collagen patch to achieve rapid pain relief. This system provides stable electrical stimulation output and its low cost and portability also offer an effective strategy for emergency on-site applications. Healing of exposed wounds is a key process in the repair of skin injuries and is crucial to prevent infection, maintain fluid balance, and promote tissue regeneration. Conventional dressings have been widely used; however, they are often inconvenient to use and have limited analgesic effects. In this work, we have developed a ready to use, disposable collagen-triboelectric nanogenerator (Col-TENG) wound protection dressing system. A porous collagen patch acts as a drug reservoir and a contact layer for the skin to protect the wound and remove traumatic contaminants. The frictional electrical field generated by TENG effectively promotes the release of analgesic lidocaine drug from the collagen patch. In vitro and in vivo experiments have shown that compared to conventional drug release systems, this novel friction-facilitated drug release device achieves faster and more drug release, as well as superior analgesia effects, which is promising in application of pain management in wild environments.

In this work, thiocyano-functionalized monodisperse dendritic mesoporous silica microspheres (DMS-SCN) were successfully synthesized and utilized as efficient solid-phase extraction adsorbents for the enrichment of eugenol anesthetics in aquatic products. The thiocyano functionalization significantly improves the adsorption capacity of the adsorbent for the four eugenol anesthetics from 723–1092 μg g −1 to 2180–3885 μg g −1 in virtue of the reinforced permanent dipole interactions, π-π stacking interactions, and electrostatic interactions. By coupling DMS-SCN based solid phase extraction with HPLC (SPE-HPLC) method, an analytical method for determination of eugenol anesthetics in tilapia, carp and shrimp was successfully developed. This method achieves low detection limits ranging from 0.32 ng mL −1 to 0.82 ng mL −1 and exhibits a wide linear range from 1 ng mL −1 to 200ng mL −1 . The recoveries for the four eugenol anesthetics range from 85.16 % to 109.1 %, with relative standard deviations between 1.18 % and 8.37 %. The method demonstrates minimal matrix effects and high accuracy. The DMS-SCN based SPE-HPLC method is anticipated to be a valuable tool for the analysis of eugenol anesthetics in complex sample matrices.

Local anesthetics, drugs that only affect a restricted area of the body, are widely used in daily clinical practice. Less studied but equally important is the distribution of local anesthetics inside organisms. Here, we present a rapid in situ testing method of drug distribution in various organs. The temporal and spatial distribution of anesthetics in mice was measured by solid-phase microextraction (SPME), thermal desorption (TD), and dielectric barrier discharge ionization (DBDI) atmospheric pressure mass spectrometry. A coated SPME probe using a tungsten wire as the support covered with a carbonaceous material was prepared by a simple, low-cost flame method. An in-line structure of the inlet allows TD and DBDI to share the same capillary tube, which greatly improves the transmission efficiency. Nine kinds of anesthetics, such as lidocaine and dyclonine, were detected, and the limit of detection was determined to be as low as 13 pg/mL. In addition, the time-dependent distribution of drugs in mice organs was studied. We also found that macromolecules in organisms do not noticeably interfere with the detection. This method is convenient and efficient because it does not require tissue homogenates and allows direct in situ detection. Compared with the conventional analytical methods, this method is simple and rapid, works in situ, and allows microscale analysis of trace analytes in biological organisms with high sensitivity.

Examining the author’s own experiences of narcotics addiction reveals certain aspects of the addicted mentality that have strong ethical valence. In general, this shows that addiction is not a state fundamentally characterized by lack. The rudiments of this position are found in some contemporary philosophy of addiction; also, it is contrasted with a common widely held mistaken view. Addiction should instead be understood in continuity with and as illuminating the nature of human personhood and subjectivity. Under a phenomenology specific to the author’s experience, addiction appears as a mode of experience that has an unmanageable overflow of narratives created as discourses concerning people, events, thoughts, and feelings; narratives embodied in assemblages of objects; and narratives appearing as mental images. These considerations suggest that pre-reflective connection to the world can be profoundly illuminative but also can isolate is from the world and, further, that our ethical values form from within our lives and not as an artificial addition. Our historical, narrative self-understanding has existential and moral import. Thus, addiction by its extremity exemplifies the ceaseless ethical activity of personhood.

Hydroxy-α-sanshool (HAS), extracted from Zanthoxylum piperitum, is commonly used in oral surgery to relief pain. However, the application of HAS is limited in clinical practice due to its poor stability. This study focuses on the design of a novel nano-formulation delivery system for HAS to improve its stability and local anesthetic effect. Hydroxy-α-sanshool loaded nanostructured lipid carriers (HAS-NLCs) were prepared by melting emulsification and ultra-sonication using monostearate (GMS) and oleic acid (OA) as lipid carriers, and poloxamer-188 (F68) as a stabilizer. Besides, the formulation was optimized by response surface methodology (RSM). Then, the best formulation was characterized for particle size, polydispersity index (PDI), zeta potential, entrapment efficiency (EE%), drug loading (DL%), differential scanning calorimetry (DSC), and morphology (transmission electron microscopy, TEM). The obtained HAS-NLCs were homogeneous, near spherical particles with high DL% capacity. The stability of HAS-NLCs against oxygen, light, and heat was greatly improved over 10.79 times, 3.25 times, and 2.09 times, respectively, compared to free HAS. In addition, HAS-NLCs could exhibit sustained release in 24 h following a double-phase kinetics model in vitro release study. Finally, HAS-NLCs had excellent anesthetic effect at low dose in formalin test compared with free HAS and lidocaine, which indicated HAS-NLCs were a potential local anesthesia formulation in practice.
