Tomato yellow leaf curl virus (TYLCV), a major plant virus, infects multiple plant species, severely threatening global food security. Arbutin, a natural product used in cosmetics to reduce pigmentation, also exhibits antibacterial and anti-inflammatory properties. However, its potential in plant protection remains undocumented. This study reveals arbutin’s ability to inhibit TYLCV infection. In Nicotiana benthamiana, 100 μg/mL arbutin inhibited viral gene accumulation by up to 76.8%, surpassing ningnanmycin (65.8%) and ribavirin (39.5%). Besides, microscale thermophoresis indicated that arbutin bound strongly to the TYLCV coat protein (CP). Molecular docking indicated that arbutin interacted with ARG58, VAL65, and CYS69. RT-qPCR and Western blot experiments confirmed the crucial roles of these amino acids, especially VAL65, in viral infection. Transcriptome analysis revealed that mutating VAL65 affected plant–pathogen interaction pathways and MAPK signaling in host defense mechanisms. This study unveils arbutin’s novel antiviral function, providing crucial insights for developing new biopesticides against plant viruses.

Tomato yellow leaf curl disease (TYLCD) significantly affects tomato yield. The jasmonic acid (JA) pathway is crucial in the defence response of plants; however, its role in plant resistance to TYLCD remains undefined. In production, CCC (chlorocholine chloride) is often used to cultivate strong seedlings to enhance seedling vitality and improve stress resistance. However, the mechanism through which CCC enhances disease resistance in tomatoes remains unclear. In this study, tomato seedlings were exogenously sprayed with 300 mg/L CCC before and after inoculation with tomato yellow leaf curl virus (TYLCV). The results indicated that no significant tomato yellow virus disease phenotype was observed in tomato seedlings after spraying with CCC and subsequent inoculation with the virus. Spraying CCC on seedlings inoculated with the virus and exhibiting typical phenotypes can significantly alleviate the yellowing and curling symptoms of new leaves and improve photosynthesis-related indicators in tomato plants. The detection of virus copy numbers within the plants revealed that the virus copy numbers in plants treated with CCC were significantly lower than those in the control group. Transcriptomic analysis revealed that, after spraying CCC, the key enzyme genesAOS2andAOCin the JA synthesis pathway in tomatoes were significantly upregulated, whereas the expressions ofJAZ2andMYC2genes, which negatively regulate JA synthesis, were significantly downregulated. In the stable state, JAZ proteins interact withMYC2and inhibit its transcriptional activity ofMYC2. Tomatoes overexpressingMYC2andJAZ2exhibit a significant decrease in TYLCD resistance. These results indicated that exogenous spraying CCC affected the expression of genes such asMYC2andJAZ2, and then regulated JA pathway, increased the endogenous JA content in plants, and enhanced the disease resistance of tomato plants to TYLCD. This study provides a scientific reference for effectively preventing and controlling TYLCD in tomato production and reducing the influence of TYLCD on tomato yield and quality.

Tea polysaccharides have attracted widespread attention in regulating gut microbiota. A highly efficient enzymolysis and ultrasonic-assisted extraction method was developed and optimized for the extraction of polysaccharides from Zhonghuang No. 1 etiolated-green tea (GTP) with a-single factor and response surface methodology, i.e, with a pH of 5.5, an enzyme concentration of 7%, an ultrasonic time of 2.5 h, a solid-liquid ratio of 1:20 g/mL, and an ultrasonic temperature of 80°C. GTP exhibited a triple helix structure and demonstrated the ability to bind with Congo red. Its surface was observed to be spherical with slender chain fragments. GTP remained intact throughout the simulated in vitro digestive process, which encompassed the oral, gastric, and small intestinal phases. The relative abundance of Bacteroidota was significantly increased, while the Firmicutes/Bacteroidota ratio was reduced following the administration of GTP. The contents of Faecalibacterium and Bacteroides were increased, while the contents of Escherichia_Shigella and Dorea were reduced after GTP treatment. Furthermore, GTP were degraded and utilized by gut microbiota to produce SCFAs and reducing pH value. GTP regulated the composition of the gut microbiota and has the potential to act as a prebiotic, with the possibility of its use as a functional food.

Celery is an important leafy vegetable crop and is widely used in fresh food consumption and processing. Therefore, the preservation of fresh cut celery is warranted. As a food preservative, sodium hydrosulfide has been used to maintain postharvest quality of crops. However, the role of sodium hydrosulfide in the preservation of postharvest celery remains unclear. Here, by regulating endogenous hydrogen sulfide content with sodium hydrosulfide and hypotaurine, we evaluated the effects of sodium hydrosulfide on postharvest celery quality. Sodium hydrosulfide treatment promoted endogenous hydrogen sulfide accumulation by triggering AgLCD and AgSAT expression, as well as increased activity of L-cysteine desulfhydrase, D-cysteine desulfhydrase, O-acetylserine (thiol)lyase, and serine acetyltransferase. Simultaneously, sodium hydrosulfide treatment enhanced the expression of AgSOD1, AgPOD, and AgCAT , improved the activities of superoxide dismutase, peroxidase, and catalase, and inhibited the accumulation of malondialdehyde, hydrogen peroxide, and superoxide anion, thereby reducing electrolyte leakage of the cell membrane. AgPAL and AgCAD expressions were up-regulated, and phenylalanine ammonia-lyase and cinnamyl alcohol dehydrogenase activities were increased to promote lignin synthesis and alleviate the softening process. AgPAO and AgPPH expression was down-regulated, polyamine oxidases and pheophytinase activities were decreased to inhibit chlorophyll degradation and delay yellowing. Meanwhile, sodium hydrosulfide treatment effectively inhibited nutrient loss, including soluble sugar, soluble protein, vitamin C, carotenoids, and total phenols. Our findings suggest that hydrogen sulfide plays an essential role in delaying quality deterioration of postharvest celery, while sodium hydrosulfide treatment is a promising method for celery preservation.

Understanding the fundamental interaction of nanoparticles at plant interfaces is critical for reaching field-scale applications of nanotechnology-enabled plant agriculture, as the processes between nanoparticles and root interfaces such as root compartments and root exudates remain largely unclear. Here, using iron deficiency–induced plant chlorosis as an indicator phenotype, we evaluated the iron transport capacity of Fe3O4 nanoparticles coated with citrate (CA) or polyacrylic acid (PAA) in the plant rhizosphere. Both nanoparticles can be used as a regulator of plant hormones to promote root elongation, but they regulate iron deficiency in plant in distinctive ways. In acidic root exudates secreted by iron-deficient Arabidopsis thaliana, CA-coated particles released fivefold more soluble iron by binding to acidic exudates mainly through hydrogen bonds and van der Waals forces and thus, prevented iron chlorosis more effectively than PAA-coated particles. We demonstrate through roots of mutants and visualization of pH changes that acidification of root exudates primarily originates from root tips and the synergistic mode of nanoparticle uptake and transformation in different root compartments. The nanoparticles entered the roots mainly through the epidermis but were not affected by lateral roots or root hairs. Our results show that magnetic nanoparticles can be a sustainable source of iron for preventing leaf chlorosis and that nanoparticle surface coating regulates this process in distinctive ways. This information also serves as an urgently needed theoretical basis for guiding the application of nanomaterials in agriculture.

The distinct qualities of etiolated leaves from chlorotic tea cultivars are significantly related to their developmental stages during harvesting. In this study, Hjy ( Camellia sinensis L. cv . Huangjinye) as a light sensitive etiolated tea variety had a higher concentration of theanine (99.36 mg/g) and lower total catechins (119.15 mg/g) than normal green tea plant. The shading treatment could turn the yellow leaves to green and alter the levels of catechins, caffeine, and theanine of tea shoots in Hjy. Additionally, the buds and 2nd leaves of Hjy tea shoots had the highest content of theanine, whereas the 3rd leaves contained the highest caffeine and total catechins. Moreover, the three different developmental stages of Hjy tea leaves had different levels of pigments, total free amino acids, total polyphenols, and soluble protein. Transcriptome and metabolome analyses identified a total of 58,175 genes and 414 metabolites in the three developmental stages, which were most enriched in photosynthesis, primary metabolism, lipid metabolism, and flavonoid metabolism. The multi-omics analysis revealed that the accumulations of L- glutamic acid and L-glutamine were up-regulated by GOGATs and GDHs and down-regulated by GSs, TS , and ALaDC. WRKY24, WRKY27, MYB105, and some other genes related to plant development and ion balance were highly correlated with L- glutamic acid biosynthesis. The decreased expressions of LARs and ANRs contributed to the reduction of catechin, epigallocatechin, and epigallocatechin gallate. WRKY35 , W RKY53, MYB62 , and some other genes related to photosynthesis, plant development, monooxygenases, and glycosylation exhibit roles in regulating catechins biosynthesis. This study provided the novel information about the Hjy tea plant, and the integrated strategy of multi-omics allowed a mechanistic understanding of the differential accumulation of L- glutamic acid and catechins during the development of etiolated tea leaves.

The whitefly Bemisia tabaci ( B. tabaci ) stands as the exclusive identified vector for tomato yellow leaf curl virus (TYLCV), the causative agent of tomato yellow leaf curl disease (TYLCD). Current disease management strategies face significant challenges due to the absence of effective antiviral chemicals, necessitating heavy reliance on whitefly population control. However, the occurrence of insecticide resistance and the invasiveness of B. tabaci have severely compromised conventional control methods, highlighting an urgent demand for innovative disease management approaches. This study presents a kind of LDH superpolymer as a nano-insecticide to control B. tabaci . It was generated through self-assembly of LDH SLNSs gel with supplement of matrine (MT) to form nanohybrids. We found that the 150 nm LDH nanosheets displayed a high efficiency to permeate tomato leaf stomata, enabling accumulation in intercellular cavities and mesophyll cell cytoplasm. The loading efficiency of MT-LDH nanohybrids achieved to 16 %. Compared to conventional MT formulations, foliar application of 500 mg/L MT-LDH increased 50.39–75.17 % phyllospheric MT retention in tomato leaves. The nanomaterial exhibited efficient showing 2.83-fold bioactivity against B. tabaci adults than that of aqueous MT solutions. Moreover, we revealed that the biocontrol activity of the MT-LDH nanohybrids conferred dual protective benefits: (1) activation of plant defenses to inhibit mitigation of TYLCV; and (2) enhanced systemic acquired resistance against viral infection. These findings establish a novel antiviral paradigm that both killed vector insect and activate host defence. The MT-LDH nanohybrids offered a potential solution to control TYLCV epidemics with reduced pesticide reliance.
