An eco-friendly depressant derived from grape seed extract (GSE) as depressant for flotation separation of pyrite from chalcopyrite under low-alkalinity conditions

Shiqin Dong,Haiyang He,Zhilian Qiu,Liguo Peng,Shuang Qin,Jianjun Fang,Dianwen Liu,Peilun Shen

This study explores the selective depression mechanism of pyrite by proanthocyanidins (PC) during chalcopyrite-pyrite flotation separation. The selective inhibition effect of inhibitor PC was examined through flotation experiments. The depression mechanism of PC on pyrite was analyzed employing X-ray photoelectron spectroscopy (XPS) analysis, selective adsorption tests, adsorption experiments, contact angle measurement, and zeta potential measurement. The artificial mixed mineral flotation test of chalcopyrite-pyrite at a ratio of 1:12 was conducted under pH 7, PC concentration of 75 mg/L, and PAX concentration of 18.75 mg/L. The copper concentrate exhibited a recovery rate of 88.97%, with a copper grade of 29.43%. The sulfur grade in copper concentrate was 40.35%, and the pyrite recovery rate was 1.97%. PC can be used as a potential depressant of pyrite. adsorption experiments and Zeta potential measurement indicated that PC treatment significantly impeded PAX adsorption on the pyrite surface, as confirmed by XPS test results and Ion adsorption analysis.

In vitro inhibitory effect of tea extracts on starch digestibility

Ishmael Ayim,Haile Ma,Evans Adingba Alenyorege,Yuqing Duan

The effect of brown, oolong, and green teas’ extract on the in vitro inhibition of starch digestibility of cassava, corn, plantain, and yam was explored. The starches exhibited moderately low hydrolysis rate and had moderate resistant starch ratios. Tea metabolites after extraction at different temperatures were quantified and correlated to starch inhibition at 1% introduction using principal component analysis (PCA). Total polyphenols (TP), (−)-epigallocatechin (EGC), (−)-gallocatechin gallate (GCG), (−)-catechin gallate (CG), and (−)-epigallocatechin gallate (EGCG) were positively correlated with starch digestion inhibition especially in the first and second hours of digestion. The (+)-catechin (C) and Total flavonoids (TF) in the teas, however, had a negative correlation with starch inhibition. Green tea extracts were more effective in starch digestion inhibition followed by brown tea and then oolong. TP, EGC, GCG, CG, and EGCG from green tea had the strongest inhibitory effect on starch digestion and that purified extracts of these compounds could be used at a statistically optimized combination to formulate diets with improved hypoglycemic potential. Practical applications In order to identify key components in tea responsible for hypoglycemic effect, tea extracts from three different source and their starch inhibitory properties were analyzed using starches from four plant sources. To this end, key components in the tea varieties were identified with potential hypoglycemic effect. Therefore, diets with potential hypoglycemic effect could be formulated using purified extracts from tea such as TP, (−)-epigallocatechin, (−)-gallocatechin gallate, (−)-catechin gallate, and (−)-epigallocatechin gallate under optimized conditions.

Preparation and Antioxidant Activity of Ethyl-Linked Anthocyanin-Flavanol Pigments from Model Wine Solutions

Lingxi Li,Minna Zhang,Shuting Zhang,Yan Cui,Baoshan Sun

Anthocyanin-flavanol pigments, formed during red wine fermentation and storage by condensation reactions between anthocyanins and flavanols (monomers, oligomers, and polymers), are one of the major groups of polyphenols in aged red wine. However, knowledge of their biological activities is lacking. This is probably due to the structural diversity and complexity of these molecules, which makes the large-scale separation and isolation of the individual compounds very difficult, thus restricting their further study. In this study, anthocyanins (i.e., malvidin-3-glucoside, cyanidin-3-glucoside, and peonidin-3-glucoside) and (–)-epicatechin were first isolated at a preparative scale by high-speed counter-current chromatography. The condensation reaction between each of the isolated anthocyanins and (–)-epicatechin, mediated by acetaldehyde, was conducted in model wine solutions to obtain ethyl-linked anthocyanin-flavanol pigments. The effects of pH, molar ratio, and temperature on the reaction rate were investigated, and the reaction conditions of pH 1.7, molar ratio 1:6:10 (anthocyanin/(–)-epicatechin/acetaldehyde), and reaction temperature of 35 °C were identified as optimal for conversion of anthocyanins to ethyl-linked anthocyanin-flavanol pigments. Six ethyl-linked anthocyanin-flavanol pigments were isolated in larger quantities and collected under optimal reaction conditions, and their chemical structures were identified by HPLC-QTOF-MS and ECD analyses. Furthermore, DPPH, ABTS, and FRAP assays indicate that ethyl-linked anthocyanin-flavanol pigments show stronger antioxidant activities than their precursor anthocyanins. Keywords:anthocyanin;antioxidant activity;ethyl-linked anthocyanin-flavanol pigment

Isolicoflavonol alleviates UVB-induced photodamage via protecting mitochondria and blocking the activation of NLRP3 inflammasome

Xing-Jie Zhang,Peng-Yun Yang,Ling Ding,Jun Wang,Xiao-Li Li,Wei-Lie Xiao

Photodamage, a type of skin inflammation caused by excessive exposure to solar radiation, leads to skin redness, inflammation, and even the development of skin cancer, posing a severe threat to individuals living at high altitudes. UVB radiation is considered the primary factor contributing to photodamage. It stimulates macrophages within the epidermis, triggers inflammasome activation, and increases the inflammatory cytokine interleukin-1β (IL-1β) production. This study examined the protective effects of the compound isolicoflavonol (ILF) and its mechanism against UVB-induced photodamage. We irradiated UVB to create a photodamage model in mice and macrophages. Next, we assessed ILF's ability to protect the skin and cells from UVB photodamage and its inhibitory effects on UVB-mediated NLRP3 inflammasome. Our findings indicated that ILF reduced UVB-induced skin injury and inflammation in mouse skin, decreased cell death, NLRP3 inflammasome activation, ROS production, and mitochondrial dysfunction. These results suggest that ILF may be a potent agent for protecting the skin against UVB-induced photodamage.

S-equol, a metabolite of dietary soy isoflavones, alleviates lipopolysaccharide-induced depressive-like behavior in mice by inhibiting neuroinflammation and enhancing synaptic plasticity

Cong Lu,Rongjing Gao,Yingyu Zhang,Ning Jiang,Ying Chen,Jing Sun,Qiong Wang,Bei Fan,Xinmin Liu,Fengzhong Wang

Systemic injection with lipopolysaccharide can lead to depressive-like behavior in experimental animals by inducing neuroinflammation and is considered to be a classic model of depression. S-equol is a major metabolite of dietary soy isoflavones with antioxidant and anti-inflammatory effects, and it has many beneficial effects on human health, including alleviation of menopausal symptoms, osteoporosis, cancer, obesity, chronic kidney disease, and cognitive dysfunction. A recent study reported that S-equol inhibited lipopolysaccharide-stimulated neuroinflammation in astrocytes. However, there is no research on the antidepressant-like effects of S-equol. Therefore, the present study was conducted to evaluate the antidepressant-like effects of S-equol in a lipopolysaccharide-induced depression model in mice and explore its underlying mechanisms. Our results demonstrated that treatment with S-equol (10, 20 and 40 mg kg−1) for 19 days markedly reversed the behavior of acute LPS (1.0 mg kg−1) treated mice in sucrose preference, tail suspension and forced swimming tests, exerting antidepressant-like effects. In addition, S-equol administration significantly decreased the levels of pro-inflammatory cytokines (tumor necrosis factor, interleukin-6, interleukin-10, interleukin-1β), increased the levels of 5-hydroxytryptamine and norepinephrine, and normalized the release of tryptophan and kynurenine in the hippocampi of lipopolysaccharide-treated mice. Moreover, treatment with S-equol significantly up-regulated the expression of synaptic plasticity-related proteins (phospho synapsin, synapsin, postsynaptic density-95) and down-regulated the toll-like receptor 4/nuclear factor kappa B signaling pathway in the hippocampi of lipopolysaccharide-treated mice. These findings demonstrated that S-equol significantly alleviated the depressive-like behavior induced by acute systemic injection of LPS, and its antidepressant action was related to mediation of neuroinflammation via the TLR4/NF-κB signaling pathway, normalization of the monoamine neurotransmitter levels, reversal of tryptophan metabolism dysfunction, and enhancement of synaptic plasticity. The current study provides insight into the potential of S-equol in the prevention of depression.

Nonvolatile metabolism in postharvest tea (Camellia sinensis L.) leaves: Effects of different withering treatments on nonvolatile metabolites, gene expression levels, and enzyme activity

Xinlei Yu,Yuchuan Li,Chang He,Jingtao Zhou,Yuqiong Chen,Zhi Yu,Pu Wang,Dejiang Ni

The influence mechanism of different withering methods (CK, indoor natural spreading; LTD, low-temperature plus dark; LTY, low-temperature plus yellow-light; LTCD, low-temperature plus CO 2 ) on non-volatile compounds in postharvest tea leaves was investigated by UHPLC-Q‐TOF/MS-based non-targeted metabolomic and transcriptomic analyses. Compared with CK, low-temperature withering could slow down polyphenol oxidation by inhibiting polyphenol oxidase activity and keeping the expression of genes for flavanol synthesis. After withering, the proteinaceous amino acid content increased significantly, especially for LTCD and LTY, mainly due to increased peptidase activity and up-regulation of genes involved in the biosynthesis of valine, leucine, aspartic acid, glutamic acid, phenylalanine, and proline. Moreover, LTCD and LTY enhanced the synthesis of γ-aminobutyric acid and metabolism of phenylalanine-methyl salicylate and tryptophan-indole, respectively. Meanwhile, the transformation of theobromine to caffeine was accelerated under low-temperature withering. This research provides a   genetic   metabolic   basis for the application of low-temperature withering to actual green tea processing.

Luteolin and apigenin derived glycosides from Alphonsea elliptica abrogate LPS-induced inflammatory responses in human plasma

Ali Attiq,Juriyati Jalil,Khairana Husain,Hazni Falina Mohamad,Abrar Ahmad

Ethnopharmacological relevance Numerous Alphonsea species including Alphonsea elliptica (mempisang) leaves and fruits are indigenously used in inflammatory conditions such as postpartum swelling and rheumatism in southeast Asian countries. In our previous in-vitro findings, A. elliptica methanol extract exhibited platelet-activating factor inhibition, suggesting the presence of phyto-constituents with anti-inflammatory potential. Aim of the study However, so far there is no literature available on the anti-inflammatory activity of this species. Henceforth, based on the above background and our previous laboratory findings, we hypothesize that phytoconstituents of A. elliptica could possess anti-inflammatory potential against inflammatory mediators including prostaglandin-E 2 (PGE 2 ), cyclooxegenase-2 (COX-2) and cytokines (IL-1β and IL-6). Materials and methods Vacuum and column chromatography techniques were employed for the isolation of phytoconstituents. The structure elucidation was carried out using HRESI-MS, 1 H and 13 C-NMR analysis and compared with the published literature. For cytotoxicity analysis, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay was performed on peripheral blood mononuclear cells. In-vitro anti-inflammatory activities were evaluated against the levels of PGE 2 , COX-2, IL-1β and IL-6 in lipopolysaccharide (LPS)-induced human plasma using enzyme-linked immunosorbent assay and radioimmunoassay . Results Unprecedentedly, chromatographic purification of methanolic leaves extract afforded five flavones namely vitexin , isovitexin , orientin, isoorientin , schaftoside with three flavanols ; kaempferol , myricetin and rutin from A elliptica . In cell viability analysis, isolates did not present cytotoxicity up to 50 μM. In anti-inflammatory evaluation, orientin and isoorientin exhibited strong (≥70%), while isovitexin and vitexin produced strong to moderate (50–69%) PGE 2 , COX-2, IL-1β and IL-6 inhibition at 25 and 50 μM. Isoorientin, orientin, isovitexin, and vitexin showed significant (p < 0.05) and concentration-dependent PGE 2 inhibition with IC 50 values (μM) of 11.40, 14.71, 17.70 and 20.58 against indomethacin (8.80). Furthermore, isoorientin, orientin, isovitexin, and vitexin produced significant concentration-dependent inhibition with IC 50 values (μM) of COX-2: 7.13, 9.51, 12.81, 16.61; IL-1β 4.80, 6.20, 10.85, 14.51; IL-6: 4.01, 5.90, 11.51 and 14.88 as compared to dexamethasone : 5.29, 2.93, 3.72, respectively (p < 0.05). Conclusion Conclusively, isolated phytoconstituents are reported for the first time from the A. elliptica. Moreover, isovitexin, vitexin orientin and isoorientin abrogated LPS-induced inflammatory responses in human plasma at non-cytotoxic concentrations.

Leaf extract from Vitis vinifera L. reduces high fat diet-induced obesity in mice

Li Meng,Yuehua Jiao,Xiaohong Zhou,Cong Liang,Kexin Yan,Ye Zhao,Xiujie Deng,Xue Han,Yanyan Yang,Hanmin Liu,Pimin Gong,Lanwei Zhang

Despite the health benefits of Vitis vinifera L. leaves, its anti-obesity potential has not been fully explored. In this work, we showed that Vitis vinifera L. leaf extract (VLE) inhibits the pancreatic lipase activity. Intragastric administration of VLE to mice led to a significant decrease in the body weight, tissue fat accumulation, levels of cholesterol, low-density lipoprotein and triglyceride compared to mice fed with high fat diet. We also found a lower level of neuropeptide-Y (NPY) in the serum and hypothalamus and a higher level of fibroblast growth factor 15 in mice supplemented with VLE. These results suggested that VLE regulates both the NPY-mediated pathway and the bile acid-FGF15 pathway to control energy metabolism and body weight gain. The composition of VLE was further investigated by a targeted metabolomics approach, which identified 21 compounds including phenolic acids, flavones, flavanols, flavanones, coumarins, and stilbenes. Taken together, we demonstrated the capacity of grape leaves in reducing obesity, which could be mediated by NPY and bile acids. Identification of putative active compounds in VLE also open the path for further studies to determine their effectiveness individually to treat obesity.
