Antioxidant and antimicrobial potential of polysaccharides sequentially extracted from Polygonatum cyrtonema Hua

Ling Li,Kiran Thakur,Bu-Yan Liao,Jian-Guo Zhang,Zhao-Jun Wei

We examined the antioxidant and antimicrobial attributes of Polygonatum cyrtonema Hua polysaccharides (HBSS, CHSS, DASS, CASS). The UV spectra and FT-IR results showed the purity and acidic nature of polysaccharides with β -configurations. DASS (78.585% hydroxyl radical scavenging, 92.794% ABTS radical scavenging, 75.648% DPPH radical scavenging activity, 0.582 reducing power and 98.721% Fe 2+ chelation at 5   mg/mL) possessed the best antioxidant potential; while, HBSS (21.113% hydroxyl radical scavenging, 14.412% ABTS radical scavenging, 28.005% DPPH radical scavenging activity, 0.285 reducing power and 48.969% Fe 2+ chelation at 5   mg/mL) was observed with the lowest antioxidant action. The bacterial inhibition effect of four PCHPs on Gram-positive organisms ( B. subtilis and S. aureus with the maximum inhibitory diameters of 18.9 and 15.8   mm, respectively) was better than that of Gram-negative bacteria ( E. coli and S. typhimurium with the maximum inhibitory diameters of 10.9 and 8.4   mm, respectively). The bacterial sensitivity was observed in the following order: B. subtilis   >   S. aureus   >   E. coli   >   S. typhimurium . The trend of antibacterial action of PCHPs was in following order: DASS   >   CHSS   >   CASS   >   HBSS. DASS can be considered as natural anti-oxidant as well as spoilage inhibitors to enhance the nutritive and functional values of various foods.

Microwave-Assisted Degradation of Polysaccharide from Polygonatum sibiricum and Antioxidant Activity

Hui Zhang,Xiu-ting Cai,Qing-hua Tian,Lin-xia Xiao,Zhen Zeng,Xin-tong Cai,Ji-zhong Yan,Qing-yong Li

Four polysaccharide fractions (P-1: 71.40%, P-2: 1.95%, P-3: 1.14%, P-4: 1.64%) were isolated from crude Polygonatum sibiricum polysaccharide (PSP), processed by water extraction, ethanol precipitation, and further separated with diethylaminoethyl cellulose-52 anion-exchange chromatography. Their molecular weights and monosaccharide compositions were characterized by high performance gel chromatography with evaporative light scattering detector and ultraviolet–visible detector. The antioxidant activity of four polysaccharides fractions were assessed by the electron transfer menchanism (DPPH, ferric reducing power, and ABST assays) and chelation of transition metals (Fe 2+ and Cu 2+ chelation ability). The highest content fraction P-1 exhibited the lowest antioxidant activity, and the ranking of antioxidant capacity was P-4 > P-3 > P-2 > PSP > P-1. After processed by microwave-assisted degradation, the molecular weight of P-1 was decreased from 2.99 × 10 5 to 2.33 × 10 3 Da, while the antioxidant activity of degraded P-1 was about eightfold higher than natural P-1. These results indicated that the proposed microwave-assisted degradation approach was an efficacious methodology to improve their bioactivity by lower the molecular weight of polysaccharides. Practical Application This study provided an environmentally friendly, convenient and efficient microwave-assisted degradation technology to process the neutral polysaccharides from Polygonatum sibiricum . The results could be used for the development and utilization of various plant polysaccharides as a kind of food supplement in our daily life.

Yan Lei,Bing Wang,Guanglin Zhu,Guo Feng,Wei Li,Caiyao Han,Qian Qin,Xie-an Yu,Xueli Song,Zhengyan He,Ju Zhang,Hongmei Su,Wenjing Wang

Background: In the research of the pharmacological activity of modern Chinese medicine, a large number of traditional Chinese medicines show anti-inflammatory activity. Asparagi radix is one of the anti-inflammatory drugs. With the deepening of people's research on Asparagi radix, its anti-inflammatory pharmacological effects have been gradually explored, but its specific anti-inflammatory active ingredients are still unclear. Therefore, it is necessary to screen and identify its specific anti-inflammatory active ingredients from Asparagi radix. At present, COX-2 is an important target for the treatment of chronic inflammation, and many drugs targeting this target have been marketed. However, some drugs have serious side effects, so it is very meaningful to study new COX-2 inhibitors. Methods: In this experiment, the fractions of different time periods are separated by high performance liquid chromatography-separation-trapping system (HPLC-CAD-FC). Then, ultra-high performance liquid chromatography-quadrupole time-of-flight/mass spectrometry (UPLC-Q-TOF/MS) and biological activity analysis techniques are used to identify the chemical composition and in vitro anti-inflammatory activity of each fraction. Finally, the data is integrated to screen out the substances with potential anti-inflammatory activity in Asparagi radix. Results: The results showed that the method established in this experiment can successfully isolate the active components of Asparagi radix, and four saponins with potential anti-inflammatory activity are identified. Conclusion: In summary, the analysis method established in this study can quickly and effectively screen out its anti-inflammatory active substances, and its potential anti-inflammatory active ingredients are mainly steroidal saponins.

Effects of Extraction Methods on the Physicochemical Properties and Biological Activities of Polysaccharides from Polygonatum sibiricum

Yongshuai Jing,Meng Yan,Hao Zhang,Dongbo Liu,Xiaoyue Qiu,Beibei Hu,Danshen Zhang,Yuguang Zheng,Lanfang Wu

Polygonatum sibiricumpolysaccharides (PSPs) have important biological functions, such as antioxidation, immunomodulatory, and hypolipidemic functions. Different extraction methods have effects on their structures and activities. In this study, six extraction methods, including hot water extraction (HWE), alkali extraction (AAE), ultrasound-assisted extraction (UAE), enzyme-assisted extraction (EAE), microwave-assisted extraction (MAE), and freeze–thaw-assisted extraction (FAE) were used to extract PSPs, and their structure–activity relationships were analyzed. The results showed that all six PSPs had similar functional group compositions, thermal stability, and glycosidic bond compositions. PSP-As (PSPs extracted by AAE) exhibited better rheological properties due to their higher molecular weight (Mw). PSP-Es (PSPs extracted by EAE) and PSP-Fs (PSPs extracted by FAE) had better lipid-lowering activity due to their lower Mw. PSP-Es and PSP-Ms (PSPs extracted by MAE), which do not contain uronic acid and have a moderate Mw, had better 1,1-diphenyl-2-picrylhydrazyl (DPPH)-radical-scavenging activity. On the contrary, PSP-Hs (PSPs extracted by HWE) and PSP-Fs, with the Mw of uronic acid, had the best OH-radical-scavenging activity. The high-Mw PSP-As had the best Fe2+-chelating ability. In addition, mannose (Man) may play an important role in the immunomodulatory activity. These results indicate that different extraction methods affect the structure and biological activity of polysaccharides to varying degrees, and these results are helpful for understanding the structure–activity relationship of PSPs.Keywords:Polygonatum sibiricum;polysaccharides;extraction methods;physicochemical properties;biological activity

Blood-Enriching Effects and Immune-Regulation Mechanism of Steam-Processed Polygonatum Sibiricum Polysaccharide in Blood Deficiency Syndrome Mice

Wang Juan,Wang Furong,Yuan Lixia,Ruan Hongsheng,Zhu Zhibiao,Fan Xiaoling,Zhu Lingyan,Peng Xin

Polygonatum sibiricum Red. has been used as a medicinal herb and nutritional food in traditional Chinese medicine for a long time. It must be processed prior to clinical use for safe and effective applications. However, the present studies mainly focused on crude Polygonatum sibiricum (PS). This study aimed to investigate the chemical properties, blood-enriching effects and mechanism of polysaccharide from the steam-processed Polygonatum sibiricum (SPS), which is a common form of PS in clinical applications. Instrumentation analyses and chemistry analyses revealed the structure of SPS polysaccharide (SPSP). A mice model of blood deficiency syndrome (BDS) was induced by acetylphenylhydrazine (APH) and cyclophosphamide (CTX). Blood routine test, spleen histopathological changes, serum cytokines, etc. were measured. The spleen transcriptome changes of BDS mice were detected by RNA sequencing (RNA-seq). The results showed that SPSP consists predominantly of Gal and GalA together with fewer amounts of Man, Glc, Ara, Rha and GlcN. It could significantly increase peripheral blood cells, restore the splenic trabecular structure, and reverse hematopoietic cytokines to normal levels. RNA-seq analysis showed that 122 differentially expressed genes (DEGs) were obtained after SPSP treatment. GO and KEGG analysis revealed that SPSP-regulated DEGs were mainly involved in hematopoiesis, immune regulation signaling pathways. The reliability of transcriptome profiling was validated by quantitative real-time PCR and Western blot, and the results indicated that the potential molecular mechanisms of the blood-enriching effects of SPSP might be associated with the regulating of JAK1-STAT1 pathway, and elevated the hematopoietic cytokines (EPO, G-CSF, TNF-α and IL-6). This work provides important information on the potential mechanisms of SPSP against BDS.

Antiaging effect of Curcuma longa L. essential oil on ultraviolet-irradiated skin

Yating Zheng,Chunxing Pan,Zejun Zhang,Wenqian Luo,Xiaoxin Liang,Yaohui Shi,Linjie Liang,Xi Zheng,Lanyue Zhang,Zhiyun Du

Objective Essential oil derived from the rhizome of Curcumin longa (CL-EO) has marked anti-inflammatory, antioxidant, antimicrobial, anticancer, and antiviral activities. However, its effect on photodamaged skin has not yet been evaluated. In this study, the anti-skin photoaging activity of CL-EO was determined by performing an ultraviolet B (UVB)-induced skin aging assay. Methods CL-EO was extracted by hydrodistillation and characterized using gas chromatography–mass spectrometry. The anti-skin aging effect was determined by topically applying 150 μl of CL-EO diluted to 1, 5, and 10% with ethanol to the dorsal area of UVB-irradiated nude mice every day except on Sunday for 8 weeks. Histological and immunohistochemical analyses were performed. Results In total, 56 compounds, which accounted for 94.36% of the contents of CL-EO, were detected. The major compounds in CL-EO were ar-turmerone (36.04%), curlone (8.78%), β-turmerone (7.05%), 8,9-dehydro-9-formyl-cycloisolongifolene (5.69%), β-sesquiphellandrene (5.39%), germacrone (4.51%), ar-curcumene (2.19%), α-himachalene (2.14%), and ledane (2.13%). Hematoxylin and eosin staining of tissue sections revealed that CL-EO decreased epidermal skin thickness. Immunohistochemistry showed that CL-EO inhibited interleukin-1β and tumor necrosis factor-α expression. Conclusions Thus, CL-EO can reduce cutaneous photoaging in a UVB-irradiated nude mouse model. Therefore, CL-EO could be used in the formulation of skin care and functional cosmetic products.

A Novel Method for the Pre-Column Derivatization of Saccharides from Polygonatum cyrtonema Hua. by Integrating Lambert–Beer Law and Response Surface Methodology

Hui Liu,Yuanyuan Zhao,Leijing Chen,Jiao Du,Hongyan Guo,Bin Wang

Traditional Chinese medicine (TCM) safety and effectiveness can be ensured by establishing a suitable quality assessment system. This work aims to develop a pre-column derivatization HPLC method forPolygonatum cyrtonemaHua. quality control. In this study, 1-(4′-cyanophenyl)-3-methyl-5-pyrazolone (CPMP) was synthesized and reacted with monosaccharides derived from P. cyrtonema polysaccharides (PCPs), followed by HPLC separation. According to the Lambert–Beer law, CPMP has the highest molar extinction coefficient of all synthetic chemosensors. A satisfactory separation effect was obtained under a detection wavelength of 278 nm using a carbon-8 column and gradient elution over 14 min, with a flow rate of 1 mL per minute. Glucose (Glc), galactose (Gal), and mannose (Man) make up the majority of the monosaccharide components in PCPs, and their molar ratios are 1.73:0.58:1. The confirmed HPLC method has outstanding precision and accuracy, establishing a quality control method for PCPs. Additionally, the CPMP showed a visual improvement from colorless to orange after the detection of reducing sugars, allowing for further visual analysis. Keywords:quality control;chemosensor;pre-column derivatization;P. cyrtonemaHua. polysaccharides

Zhao Yuanyuan,Wang Zhen,Fu Ranze,Xie Ruonan,Wang Bin,Li Qinglin

Introduction: Polygonatum cyrtonema Hua. (PC) is a traditional Chinese herb with a history of use in both food and medicine. For clinical use, processed PC pieces are most commonly used, while present research has focused on crude PC polysaccharides (PCPs).Methods: In this study, a new polysaccharide, PCP-F1, with a molecular weight of 37.46 kDa, was separated from four-time processed PCPs by column chromatography and evaluated by antioxidant activity. It was composed of glucose, mannose, galactose, rhamnose, and galacturonic acid with a molar ratio of 3.5: 2.5: 1.3: 1.8: 0.8.Results and Discussion: The methylation analysis and two-dimensional NMR measurement revealed that the configuration of PCP-F1 contained nine residues in the primary structural unit by the chain of →3)-α-D-Glcp, →2)-α-D-Glcp (6→, →1)-ꞵ-D-Glcp (2→, →2)-α-D-GalAp (3,4→, →1) -ꞵ-D-Manp (3→, →2)-α-D-Glcp (3→, branched for →3)-α-D-Glcp, →2)-ꞵ-D-Galp (4→, →1)-ꞵ-D-Glcp (2→, →2,4)-α-D-Manp (6→, →3)-α-L-Rhap (4→. Radical scavenging assays indicated that PCP-F1 could scavenge radicals with a high scavenging rate, suggesting PCP-F1 possesses good antioxidant activity. The study confirms the importance of processed PC and offers the potential for exploiting it as a functional food.

Novel Insights into Water-Soluble Carbohydrates Exploration in Different Species and Parts (Rhizome, Root, Leaf, Stem, Fruit) of Polygonatum Mill.

Chentao Huang,Jiabei Xia,Jiayun Xu,Xingyu Mei,Huan Cheng,Shiguo Chen,Jinping Si,Xingqian Ye,Jianle Chen

Polygonatum a widely recognized medicinal and edible plant, exhibits several confirmed biological activities and holds significant market potential. Numerous studies have identified polysaccharides are one of the primary bioactive components in the rhizomes of Polygonatum . Our previous research identified a notable presence of fructo-oligosaccharides in the rhizomes of Polygonatum sibiricum , highlighting its largely overlooked potential as a natural source of bioactive compounds. Therefore, a more systematic and comprehensive analysis of water-soluble carbohydrates across different species and plant parts of Polygonatum is essential. The aim of this study was to investigate the carbohydrate composition in four different Polygonatum species, focusing on the rhizomes, roots, leaves, stems, and fruits. In particular, we utilized a self-established method for the determination of fructo-oligosaccharides. The results indicated the presence of small amounts of mono/disaccharides and polysaccharides, as well as a considerable quantity of fructo-oligosaccharides in rhizome, root, fruit of Polygonatum . The low-molecular-weight glycans was the predominant carbohydrate in the rhizomes of Polygonatum Mill. spp., comprising over 80% of the total soluble carbohydrates, with polysaccharides contributing approximately 11%, and mono/disaccharides less than 10%. Our study offers valuable insights and a meaningful basis for the excavation of the functional carbohydrate resources of the Polygonatum Mill.

Polysaccharides from Dicliptera chinensis (L.) Juss. Attenuates drug-induced liver injury by phosphorylating AMPK and thus facilitating the entry of FOXO3 into the cell nucleus

Chaoyue Yang,Mingli Zhong,Xuefei Jiao,Houkang Cao,Yandan Qin,Hengjie Xu,Fengyue Guo,Zixuan Tang,Tiansong Lv,Lilin Guan,Yongwang Wang,Ya Gao,Kefeng Zhang

Background Isoniazid and rifampicin, frontline tuberculosis drugs, frequently induce drug-induced liver injury (DILI), marked by hepatitis and hepatocyte necrosis. Polysaccharides from Dicliptera chinensis (L.) Juss. (DCP) exhibit anti-inflammatory, antioxidant, and hepatoprotective properties, but their effects on DILI remain unexplored Objective This study investigated DCP therapeutic potential against DILI and elucidated its molecular mechanisms Methods In vivo (using C57BL/6 mice) and in vitro (using HepG2 cells) DILI models were established and treated with DCP. Transcriptomics, qRT-PCR, and Western blotting were employed to analyze pathway regulation Results DCP significantly attenuated hepatocyte apoptosis, inflammation, and oxidative stress in DILI mice. Transcriptomic analysis linked DCP'S effects to the modulation of AMPK-FOXO3, p53, and NF-κB pathways, alongside regulation of antioxidant and cell cycle genes. In HepG2 cells, DCP similarly protected against DILI by enhancing AMPK phosphorylation, which facilitated the FOXO3 nuclear translocation. Both models demonstrated DCP'S suppression of p53 and NF-κB activation, restoration of antioxidant defenses, and correction of cell cycle dysregulation Conclusion DCP mitigates DILI by reducing apoptosis, oxidative stress, and inflammation through activation of the AMPK-FOXO3 pathway, inhibition of p53/NF-κB signaling, and stabilization of the cell cycle. These findings highlight DCP'S potential as a therapeutic agent for DILI prevention and treatment

Efficient extraction of flavonoids from Polygonatum sibiricum using a deep eutectic solvent as a green extraction solvent

Hongli Zhang,Feilong Hao,Zhifang Yao,Junling Zhu,Xu Jing,Xiaowen Wang

Deep eutectic solvents (DESs) are becoming eco-friendly alternatives for extracting biologically-active substances from natural products. In this study, a DES is used as an eco-friendly solvent to extract flavonoids from Polygonatum sibiricum with the assistance of ultrasound. The antioxidant activity of the extracts was then investigated and the recyclability and reusability of the DES evaluated. Optimizing the DES extraction process produced the following optimum conditions: a betaine–acetic acid molar ratio of 1:4, water content of 30%, solid–liquid ratio of 1: 100 g mL −1 , extraction temperature of 50 °C, and extraction time of 30 min. The yield of extracted flavonoids reached 25.4 mg g −1 , which is about eight times that achieved using ethanol extraction. Furthermore, the antioxidant ability of the DES extract was more potent than that of the extract obtained using ethanol. Used DES can be efficiently recovered using ultrahigh capacitance porous activated carbon (the recovery yield of DES was around 92% of that of the original DES after ten reuses). In conclusion, DESs can be used to establish green ultrasound-assisted extraction methods for efficient extraction of plant flavonoids.
